Mycobacterium bovis (M. bovis) is a causative agent of bovine tuberculosis, a significant source of morbidity and mortality in the global cattle industry. The Randomised Badger Culling Trial was a ...field experiment carried out between 1998 and 2005 in the South West of England. As part of this trial, M. bovis isolates were collected from contemporaneous and overlapping populations of badgers and cattle within ten defined trial areas. We combined whole genome sequences from 1,442 isolates with location and cattle movement data, identifying transmission clusters and inferred rates and routes of transmission of M. bovis. Most trial areas contained a single transmission cluster that had been established shortly before sampling, often contemporaneous with the expansion of bovine tuberculosis in the 1980s. The estimated rate of transmission from badger to cattle was approximately two times higher than from cattle to badger, and the rate of within-species transmission considerably exceeded these for both species. We identified long distance transmission events linked to cattle movement, recurrence of herd breakdown by infection within the same transmission clusters and superspreader events driven by cattle but not badgers. Overall, our data suggests that the transmission clusters in different parts of South West England that are still evident today were established by long-distance seeding events involving cattle movement, not by recrudescence from a long-established wildlife reservoir. Clusters are maintained primarily by within-species transmission, with less frequent spill-over both from badger to cattle and cattle to badger.

A new 1.4 GHz, 19-element, dual-polarization, cryogenic phased-array feed (PAF) radio astronomy receiver has been developed for the Robert C. Byrd Green Bank Telescope (GBT) as part of the Focal ...L-band Array for the GBT (FLAG) project. Commissioning observations of calibrator radio sources show that this receiver has the lowest reported beam-formed system temperature (Tsys) normalized by aperture efficiency ( ) of any phased-array receiver to date. The measured Tsys/ is 25.4 2.5 K near 1350 MHz for the boresight beam, which is comparable to the performance of the current 1.4 GHz cryogenic single-feed receiver on the GBT. The degradation in Tsys/ at ∼4′ (required for Nyquist sampling) and ∼8′ offsets from the boresight is, respectively, ∼1% and ∼20% of the boresight value. The survey speed of the PAF with seven formed beams is larger by a factor between 2.1 and 7 compared to a single-beam system, depending on the observing application. The measured performance, both in frequency and offset from the boresight, qualitatively agrees with predictions from a rigorous electromagnetic model of the PAF. The astronomical utility of the receiver is demonstrated by observations of the pulsar B0329+54 and an extended H ii region, the Rosette Nebula. The enhanced survey speed with the new PAF receiver will enable the GBT to carry out exciting new science, such as more efficient observations of diffuse, extended neutral hydrogen emission from galactic inflows and searches for fast radio bursts.
Reactive oxygen species (ROS)-mediated oxidative stress and DNA damage have recently been recognized as contributing to the efficacy of most bactericidal antibiotics, irrespective of their primary ...macromolecular targets. Inhibitors of targets involved in both combating oxidative stress as well as being required for in vivo survival may exhibit powerful synergistic action. This study demonstrates that the de novo arginine biosynthetic pathway in Mycobacterium tuberculosis (Mtb) is up-regulated in the early response to the oxidative stress-elevating agent isoniazid or vitamin C. Arginine deprivation rapidly sterilizes the Mtb de novo arginine biosynthesis pathway mutants ΔargB and ΔargF without the emergence of suppressor mutants in vitro as well as in vivo. Transcriptomic and flow cytometry studies of arginine-deprived Mtb have indicated accumulation of ROS and extensive DNA damage. Metabolomics studies following arginine deprivation have revealed that these cells experienced depletion of antioxidant thiols and accumulation of the upstream metabolite substrate of ArgB or ArgF enzymes. ΔargB and ΔargF were unable to scavenge host arginine and were quickly cleared from both immunocompetent and immunocompromised mice. In summary, our investigation revealed in vivo essentiality of the de novo arginine biosynthesis pathway for Mtb and a promising drug target space for combating tuberculosis.
Bovine tuberculosis (bTB) caused by Mycobacterium bovis is a re-emerging problem in both livestock and humans. The association of some M. bovis strains with hyper-virulence, MDR-TB and disseminated ...disease makes it imperative to understand the biology of the pathogen.
Mycobacterium bovis (15) among 1755 M. tuberculosis complex (MTBC) isolated between 2012 and 2014 were characterized and analyzed for associated patient demography and other risk factors. Five of the M. bovis isolates were whole-genome sequenced and comparatively analyzed against a global collection of published M. bovis genomes.
Mycobacterium bovis was isolated from 3/560(0.5%) females and 12/1195(1.0%) males with pulmonary TB. The average age of M. bovis infected cases was 46.8 years (7-72years). TB patients from the Northern region of Ghana (1.9%;4/212) had a higher rate of infection with M. bovis (OR = 2.7,p = 0.0968) compared to those from the Greater Accra region (0.7%;11/1543). Among TB patients with available HIV status, the odds of isolating M. bovis from HIV patients (2/119) was 3.3 higher relative to non-HIV patients (4/774). Direct contact with livestock or their unpasteurized products was significantly associated with bTB (p<0.0001, OR = 124.4,95% CI = 30.1-508.3). Two (13.3%) of the M. bovis isolates were INH resistant due to the S315T mutation in katG whereas one (6.7%) was RIF resistant with Q432P and I1491S mutations in rpoB. M. bovis from Ghana resolved as mono-phyletic branch among mostly M. bovis from Africa irrespective of the host and were closest to the root of the global M. bovis phylogeny. M. bovis-specific amino acid mutations were detected among MTBC core genes such as mce1A, mmpL1, pks6, phoT, pstB, glgP and Rv2955c. Additional mutations P6T in chaA, G187E in mgtC, T35A in Rv1979c, S387A in narK1, L400F in fas and A563T in eccA1 were restricted to the 5 clinical M. bovis from Ghana.
Our data indicate potential zoonotic transmission of bTB in Ghana and hence calls for intensified public education on bTB, especially among risk groups.

-band receiver. These commissioning runs establish FLAG as the preeminent PAF receiver currently available for spectral-line observations on the world’s major radio telescopes.
Insight into the trophic ecology of marine predators is vital for understanding their ecosystem role and predicting their responses to environmental change. Juvenile southern elephant seals (SES) ...Mirounga leonina are considered generalist predators within the Southern Ocean. Although mesopelagic fish and squid dominate their stomach lavage samples, the stable isotope profile captured along the length of sampled vibrissae of young SES at Macquarie Island, southwest Pacific Ocean (54.5° S, 158.9° E) recently emphasized the contribution of crustaceans to their diet (likely Euphausia superba). Herein, we used the stable isotope values of sampled vibrissal regrowths with known growth histories to assess the diet of juvenile SES at Marion Island, southern Indian Ocean (46.8° S, 37.8° E) on a temporally integrated basis. We specifically aimed to quantify the possible contribution of crustaceans to the diet of juvenile SES. Sequentially (chronologically) sampled vibrissal regrowths of 14 juvenile SES produced fine-scale dietary information spanning up to 9 mo. The depleted stable isotope signatures of nitrogen (δ15N) (8.5 ± 0.6‰) and carbon (δ13C) (−20.3 ± 0.1‰) measured during the period of independent foraging suggested the use of a lower trophic level diet within the Polar Frontal Zone. A mixing model predicted that up to 76% of juvenile SES diet comprised crustaceans, consisting of 2 crustacean groups, each contributing 26% (credible interval, CI: 13–39%) and 50% (CI: 35–64%) to their diets, presumably representing subantarctic krill species. This first utilisation of the isotopic signature captured along the length of vibrissal regrowths confirms the inclusion and importance of crustaceans in the diet of juvenile SES.
Objectives: This study aimed to expand on the limited South African malnutrition prevalence data and investigate the feasibility of mid-upper-arm circumference (MUAC) as a malnutrition screening ...tool.
Design: A cross-sectional, multi-centre, descriptive design was adopted.
Setting: The study was undertaken in three tertiary public hospitals in the same urban area within the Eastern Cape of South Africa.
Subjects: Adult hospitalised patients volunteered to participate (n = 266).
Methods: Data were collected using interviewer-administered questionnaires; obtaining anthropometric measurements; and consulting medical files. For maximum accuracy of various MUAC cut-off points, receiver operating characteristic curves were generated and area under the curve determined.
Results: Both body mass index (BMI) and MUAC identified 21% of participants as underweight or malnourished, and 39% as overweight or obese. The Malnutrition Universal Screening Tool (MUST) found 23% at increased malnutrition risk. Nurses or doctors detected and referred only 19% of underweight patients (BMI < 18.5 kg/m
2
), to dietetics services. Direct measurements of BMI and MUST were unobtainable in 38% and 43% of patients respectively, whilst MUAC was obtainable in 100%.
A statistically significant relationship (p < 0.001) exists between MUAC, BMI and MUST to detect malnutrition or malnutrition risk. MUAC cut-offs for undernutrition were determined at < 23 cm (BMI < 16 kg/m
2
) and < 24 cm (BMI < 18.5 kg/m
2
), respectively, for the study's population groups.
Conclusion: Malnutrition prevalence was high in this study, but often unidentified, with only a fifth referred to dietetic services. MUAC is a feasible method to identify adult malnutrition and should be considered as a malnutrition screening tool and key nutritional status indicator in South African public hospitals.
Non-tuberculous mycobacteria (NTM) are environmental microorganisms and opportunistic pathogens in individuals with pre-existing lung conditions such as cystic fibrosis (CF) and non-CF ...bronchiectasis. While recent studies of
have identified transmission within single CF centres as well as nationally and globally, transmission of other NTM species is less well studied.
To investigate the potential for transmission of the
complex (MAC) we sequenced 996 isolates from 354 CF and non-CF patients at the Royal Brompton Hospital (London, UK; collected 2013-2016) and analysed them in a global context. Epidemiological links were identified from patient records. Previously published genomes were used to characterise global population structures.
We identified putative transmission clusters in three MAC species, although few epidemiological links could be identified. For
, lineages were largely limited to single countries, while for
, global transmission clusters previously associated with heater-cooler units (HCUs) were found. However, the immediate ancestor of the lineage causing the major HCU-associated outbreak was a lineage already circulating in patients.
CF and non-CF patients shared transmission chains, although the lack of epidemiological links suggested that most transmission is indirect and may involve environmental intermediates or asymptomatic carriage in the wider population.
