In 2009, 12.8% of the Dutch boys and 14.8% of the Dutch girls aged 2-21 years were overweight and 1.8% of the boys and 2.2% of the girls were classified as obese. This is a two to three fold higher prevalence in overweight and four to six fold increase in obesity since 1980. Since 1997, a substantial rise took place, especially in obesity, which increased 1.4 times in girls and doubled in boys. There was no increase in mean BMI SDS in the major cities since 1997.
Overweight and obesity prevalences in 2009 were substantially higher than in 1980 and 1997. However, the overweight prevalence stabilized in the major cities. This might be an indication that the rising trend in overweight in The Netherlands is starting to turn.

There is an urgent need for accurate and rapid diagnostic tests to identify carbapenemase producing enterobacteria (CPE). Here, we have evaluated the Carbapenem Inactivation Method (CIM) test to ...detect CPEs from cultured colonies.
A total of 256 enterobacterial isolates were used to evaluate the performance of the CIM in comparison to Carba NP test and molecular detection used a reference method. Ninety three well-characterized isolates (including 29 non-CPE and 63 CPEs of worldwide origin) with decreased susceptibility to at least one carbapenem were used to (i) evaluate the efficacy of CIM test and (ii) to compare it to the Carba NP test. We also tested different carbapenems to determine the best substrate for this test. Finally, the CIM test was then evaluated prospectively against 164 isolates referred to the French National Reference Center (NRC) for Antimicrobial Resistance from may 2016 to july 2016.
Based on the results of this retrospective study, sensitivity and specificity of the CIM and the Carba NP test were 92.1% and 100%, respectively. We demonstrated that the meropenem was the best substrate to perform the CIM test since sensitivity and specificity were 81.1% and 100% using ertapenem disk, and 100% and 65,6% using imipenem disk, and respectively. Taking in account the results of retrospective and prospective studies, CIM and Carba NP tests have similar sensitivity, specificity, positive predictive value and negative predictive values being 96.3%, 98.9%, 99.0% and 98.4% for the CIM test versus 96.9%, 100%, 100% and 100% for the Carba NP test.
Our results confirm that the CIM test may be a useful tool for the reliable confirmation of carbapenemase-activity in enterobacterial isolates, especially in clinical microbiological laboratories with limited resources, no trained personnel, and no specialized equipment.

Genes encoding extended-spectrum β-lactamases (ESBLs) have been reported in a variety of Gram-negative species, mostly in Enterobacteriaceae, Pseudomonas aeruginosa and Acinetobacter baumannii. They are either TEM- or SHV-derivatives, CTX-M-like enzymes and less frequently of the GES, PER, or VEB types. The mechanisms at the origin of their acquisition are diverse, and mostly are related to insertion sequences, transposons and class 1 integrons. This diversity of genetic vehicles at the origin of these mobilization/acquisition processes may explain spread of ESBLs worldwide.
To assess and compare the secular trend in age at menarche in Dutch girls (1955-2009) and girls from Turkish and Moroccan descent living in the Netherlands (1997-2009).
Data on growth and maturation ...were collected in 20,867 children of Dutch, Turkish and Moroccan descent in 2009 by trained health care professionals. Girls, 9 years and older, of Dutch (n = 2138), Turkish (n = 282), and Moroccan (n = 295) descent were asked whether they had experienced their first period. We compared median menarcheal age in 2009 with data from the previous Dutch Nationwide Growth Studies in 1955, 1965, 1980 and 1997. Age specific body mass index (BMI) z-scores were calculated to assess differences in BMI between pre- and postmenarcheal girls in different age groups.
Median age at menarche in Dutch girls, decreased significantly from 13.66 years in 1955 to 13.15 years in 1997 and 13.05 years in 2009. Compared to Dutch girls there is a larger decrease in median age of menarche in girls of Turkish and Moroccan descent between 1997 and 2009. In Turkish girls age at menarche decreased from 12.80 to 12.50 years and in Moroccan girls from 12.90 to 12.60 years. Thirty-three percent of Turkish girls younger than 12 years start menstruating in primary school. BMI-SDS is significantly higher in postmenarcheal girls than in premenarcheal girls irrespective of age.
There is a continuing secular trend in earlier age at menarche in Dutch girls. An even faster decrease in age at menarche is observed in girls of Turkish and Moroccan descent in the Netherlands.

Aims. The goal of this paper is to analyse the behaviour of the gas-to-dust mass ratio (G/D) of local Universe galaxies over a wide metallicity range. We especially focus on the low-metallicity part ...of the G/D vs metallicity relation and investigate several explanations for the observed relation and scatter. Methods. We assembled a total of 126 galaxies, covering a 2 dex metallicity range and with 30% of the sample with 12 + log(O/H)≤ 8.0. We homogeneously determined the dust masses with a semi-empirical dust model including submm constraints. The atomic and molecular gas masses have been compiled from the literature. We used two XCO scenarios to estimate the molecular gas mass: the Galactic conversion factor, XCO,MW, and a XCO that depends on the metallicity XCO,Z (∝Z-2). We modelled the observed trend of the G/D with metallicity using two simple power laws (slope of –1 and free) and a broken power law. Correlations with morphological type, stellar masses, star formation rates, and specific star formation rates are also discussed. We then compared the observed evolution of the G/D with predictions from several chemical evolution models and explored different physical explanations for the observed scatter in the G/D values. Results. We find that out of the five tested galactic parameters, metallicity is the main physical property of the galaxy driving the observed G/D. The G/D versus metallicity relation cannot be represented by a single power law with a slope of –1 over the whole metallicity range. The observed trend is steeper for metallicities lower than ~8.0. A large scatter is observed in the G/D values for a given metallicity: in metallicity bins of ~0.1 dex, the dispersion around the mean value is ~0.37 dex. On average, the broken power law reproduces the observed G/D best compared to the two power laws (slope of –1 or free) and provides estimates of the G/D that are accurate to a factor of 1.6. The good agreement of observed values of the G/D and its scatter with respect to metallicity with the predicted values of the three tested chemical evolution models allows us to infer that the scatter in the relation is intrinsic to galactic properties, reflecting the different star formation histories, dust destruction efficiencies, dust grain size distributions, and chemical compositions across the sample. Conclusions. Our results show that the chemical evolution of low-metallicity galaxies, traced by their G/D, strongly depends on their local internal conditions and individual histories. The large scatter in the observed G/D at a given metallicity reflects the impact of various processes occurring during the evolution of a galaxy. Despite the numerous degeneracies affecting them, disentangling these various processes is now the next step.
Abstract Psoriasis is an inflammatory disease of the skin that is sometimes accompanied by an auto-inflammatory arthritis called psoriatic arthritis (PsA). Psoriasis and PsA are multifactorial ...diseases that result from complex interactions of environmental and genetic risk factors. Epigenetic marks, which are labile chemical marks with diverse functions, form a layer of biological information that sits at the interface of genetics and the environment. Aberrant epigenetic regulation has been previously implicated in other rheumatological disorders. The purpose of this review is to summarize and critically evaluate the nascent literature on epigenetics in psoriasis and PsA. A systematic review yielded 52 primary articles after applying inclusion and exclusion criteria. Data were extracted using a standardized template and study quality assessed using a methodological quality checklist. Studies reflect a broad range of epigenetic sub-disciplines, the most common being DNA methylation, followed by the parent of origin effect or genomic imprinting, expression or activity of epigenetic modifying enzymes, and histone modifications. Epidemiological studies demonstrating excessive paternal transmission provided the earliest evidence of epigenetic deregulation in psoriatic disease, however few studies have examined its molecular mechanisms. Methylation studies evolved rapidly from low resolution global to targeted analyses of known psoriatic disease susceptibility loci such as HLA-C*0602 . The recent explosion of epigenome-wide association studies has provided us with novel insights into psoriasis pathogenesis, and the mechanism of action of UVB, methotrexate, and anti-TNF therapies, as well as molecular signatures of psoriasis that may have clinical relevance. Finally, recent studies of pharmacological inhibitors of epigenetic modifier enzymes demonstrate their potential applicability as novel treatment modalities for psoriasis. Challenges of epigenetics research in psoriasis and PsA were identified and future perspectives are discussed herein.

We compared the height of young Dutch people aged 0-21 y as determined on the basis of the growth study of 2009, with the height data from growth studies conducted in 1955, 1965, 1980, and 1997.
The analysis sample included 5,811 boys and 6,194 girls. Height by age was the same as in 1997. Mean final height was 183.8 cm (SD = 7.1 cm) in boys and 170.7 cm (SD = 6.3 cm) in girls. The educational levels of both children and their parents are positively correlated with mean height. Since 1997, differences between geographical regions have decreased but not vanished, with the northern population being the tallest.
The world's tallest population has stopped growing taller after a period of 150 y, the cause of which is unclear. The Dutch may have reached the optimal height distribution. Alternatively, growth-promoting environmental factors may have stabilized in the past decade, preventing the population from attaining its full growth potential.
Objective
Biomarkers that can predict the development of psoriatic arthritis (PsA) in patients with psoriasis would be useful in clinical practice. The aim of this study was to assess whether CXCL10 ...could be a predictive biomarker of PsA prior to its onset.
Methods
Psoriasis patients without arthritis were followed up prospectively and assessed annually for development of PsA by a rheumatologist. Patients in whom PsA developed were designated as converters, while those in whom PsA did not develop were termed nonconverters. Baseline serum concentrations of CXCL10 were measured by Luminex assay in 46 converters and 45 nonconverters.
Results
The level of CXCL10 was significantly higher in converters (median 493 pg/ml interquartile range (IQR) 356–984) than in nonconverters (median 371 pg/ml IQR 263–578; P = 0.005). In contrast, C‐reactive protein (CRP) levels were not significantly different between converters and nonconverters at baseline. CXCL10 was associated with conversion status after adjustment for age, sex, duration of psoriasis, and duration of follow‐up (odds ratio 1.3, 95% confidence interval 1.1–1.5, P = 0.004). In a subset of converters, the CXCL10 level was significantly higher at baseline (median 927.4 pg/ml IQR 547.6–1,243) than after PsA diagnosis (491.5 pg/ml IQR 323.2–607; P < 0.0001), while CRP levels were lower at baseline (26.6 μg/ml IQR 16.37–62.75) than after PsA diagnosis (36.1 μg/ml IQR 14.74–101.7; P = 0.003). CXCL10 gene expression was increased 17.3‐fold in synovial fluid (SF) compared with blood from PsA patients (P = 0.01) and 44.3‐fold in the SF of PsA patients compared with the SF of patients with gout (P = 0.001).
Conclusion
CXCL10 may be involved in PsA pathogenesis and is a candidate predictive biomarker for PsA in patients with psoriasis.
The blaNDM-1 gene has been reported to be often located on broad-host-range plasmids of the IncA/C type in clinical but also environmental bacteria recovered from the New Delhi, India, area. ...IncA/C-type plasmids are the main vehicles for the spread of the cephalosporinase gene blaCMY-2, frequently identified in the United States, Canada, and Europe. In this study, we completed the sequence of IncA/C plasmid pNDM-KN carrying the blaNDM-1 gene, recovered from a Klebsiella pneumoniae isolate from Kenya. This sequence was compared with those of three IncA/C-type reference plasmids from Escherichia coli, Yersinia ruckeri, and Photobacterium damselae. Comparative analysis showed that the blaNDM-1 gene was located on a widely diffused plasmid scaffold known to be responsible for the spread of blaCMY-2-like genes and consequently for resistance to broad-spectrum cephalosporins. Considering that IncA/C plasmids possess a broad host range, this scaffold might support a large-scale diffusion of the blaNDM-1 gene among Gram-negative rods.
N-methyl-D-aspartate receptors (NMDARs) are critically involved in basic brain functions and neurodegeneration as well as tumor invasiveness. Targeting specific subtypes of NMDARs with distinct ...activities has been considered an effective therapeutic strategy for neurological disorders and diseases. However, complete elimination of off-target effects of small chemical compounds has been challenging and thus, there is a need to explore alternative strategies for targeting NMDAR subtypes. Here we report identification of a functional antibody that specifically targets the GluN1-GluN2B NMDAR subtype and allosterically down-regulates ion channel activity as assessed by electrophysiology. Through biochemical analysis, x-ray crystallography, single-particle electron cryomicroscopy, and molecular dynamics simulations, we show that this inhibitory antibody recognizes the amino terminal domain of the GluN2B subunit and increases the population of the non-active conformational state. The current study demonstrates that antibodies may serve as specific reagents to regulate NMDAR functions for basic research and therapeutic objectives.
