Recent work in cognitive neuroscience has focused on analyzing the brain as a network, rather than as a collection of independent regions. 016009 suggests that these inconsistencies may be explained on the grounds that high-modularity networks favor performance on simple tasks whereas low-modularity networks favor performance on more complex tasks. The current study tests these predictions by relating modularity from resting-state fMRI to performance on a set of simple and complex behavioral tasks. Complex and simple tasks were defined on the basis of whether they did or did not draw on executive attention. Consistent with predictions, we found a negative correlation between individuals' modularity and their performance on a composite measure combining scores from the complex tasks but a positive correlation with performance on a composite measure combining scores from the simple tasks. These results and theory presented here provide a framework for linking measures of whole-brain organization from network neuroscience to cognitive processing.

In cognitive network neuroscience, the connectivity and community structure of the brain network is related to measures of cognitive performance, like attention and memory. Research in this emerging ...discipline has largely focused on two measures of connectivity-modularity and flexibility-which, for the most part, have been examined in isolation. The current project investigates the relationship between these two measures of connectivity and how they make separable contribution to predicting individual differences in performance on cognitive tasks. Using resting state fMRI data from 52 young adults, we show that flexibility and modularity are highly negatively correlated. We use a Brodmann parcellation of the fMRI data and a sliding window approach for calculation of the flexibility. We also demonstrate that flexibility and modularity make unique contributions to explain task performance, with a clear result showing that modularity, not flexibility, predicts performance for simple tasks and that flexibility plays a greater role in predicting performance on complex tasks that require cognitive control and executive functioning. The theory and results presented here allow for stronger links between measures of brain network connectivity and cognitive processes.
Bilingual language control may involve cognitive control, including inhibition and switching. These types of control have been previously associated with neural activity in the inferior frontal gyrus ...(IFG) and the anterior cingulate cortex (ACC). In previous studies, the DRD2 gene, related to dopamine availability in the striatum, has been found to play a role in neural activity during cognitive control tasks, with carriers of the gene’s A1 allele showing different patterns of activity in inferior frontal regions during cognitive control tasks than non-carriers. The current study sought to extend these findings to the domain of bilingual language control. Forty-nine Spanish-English bilinguals participated in this study by providing DNA samples through saliva, completing background questionnaires, and performing a language production task (picture-naming), a non-verbal inhibition task (Simon task), and a non-verbal switching task (shape-color task) in the fMRI scanner. The fMRI data were analyzed to determine whether variation in the genetic background or bilingual language background predicts neural activity in the IFG and ACC during these three tasks. Results indicate that genetic and language background variables predicted neural activity in the IFG during English picture naming. Variation in only the genetic background predicted neural activity in the ACC during the shape-color switching task; variation in only the language background predicted neural activity in the ACC and IFG during the Simon task. These results suggest that variation in the DRD2 gene should not be ignored when drawing conclusions about bilingual verbal and non-verbal cognitive control.

Phoca vitulina
), a mammal with broadly sensitive amphibious hearing. Scanning electron micrographs of the apical surface of the outer hair cells (OHCs) and inner hair cells (IHCs) within the organ of Corti were obtained from five individuals and analyzed by linear and geometric morphometrics. Measurements were taken at regular locations along the cochlea. The spiral shape of the seal cochlea contained two and a half turns. The organ of Corti had an average length of 27.7 mm with 12,628 OHCs (12,400-12,900). Six linear morphometric parameters showed significant patterns of change associated with their location within the cochlear spiral. Likewise, these trends were similarly expressed in cell configuration (cell blocks with 57 landmarks in 12 representative cells) revealed by geometric morphometry. Cell configuration varied predictably with position in the cochlea according to clustering analyses and Procrustes ANOVA (F= 25.936, p<0001). Changes associated with OHCs were primarily responsible for observed changes in cell configuration. An integration trend in cell shape change was also observed in which IHCs and OHCs share features in their morphological variation by the two-block partial least squares analysis (CR=0.987, p<0.001) and the modularity hypothesis (CV=0.99, p=0.05). These descriptive and quantitative findings provide a baseline for the morphology and morphometry of the sensory cells of the organ of Corti in harbor seals, allowing for comparisons between normal and pathological features. This initial morphological description should enable the correlation between position, morphometric features, and frequency coding along the spiral of the inner ear in this species, whose hearing ability is well studied.
Written language is a human invention that our brains did not evolve for. Yet, most research has focused on finding a single theory of reading, identifying the common set of cognitive and neural ...processes shared across individuals, neglecting individual differences. In contrast, we investigated variation in single word reading. Using a novel statistical method for analyzing heterogeneity in multi-subject task-based functional magnetic resonance imaging (fMRI), we clustered readers based on their brain's response to written stimuli. Separate behavioral testing and neuroimaging analysis shows that these clusters differed in the role of the sublexical pathway in processing written language, but not in reading skill. Taken together, these results suggest that individuals vary in the cognitive and neural mechanisms involved in word reading. In general, neurocognitive theories need to account not only for what tends to be true of the population, but also the types of variation that exist, even within a neurotypical population.
The goal of the present study is to reveal how the neural mechanisms underlying non-native speech perception change throughout childhood. In a pre-attentive listening fMRI task, English monolingual ...and Spanish–English bilingual children – divided into groups of younger (6–8yrs) and older children (9–10yrs) – were asked to watch a silent movie while several English syllable combinations played through a pair of headphones. Two additional groups of monolingual and bilingual adults were included in the analyses. Our results show that the neural mechanisms supporting speech perception throughout development differ in monolinguals and bilinguals. While monolinguals recruit perceptual areas (i.e., superior temporal gyrus) in early and late childhood to process native speech, bilinguals recruit perceptual areas (i.e., superior temporal gyrus) in early childhood and higher-order executive areas in late childhood (i.e., bilateral middle frontal gyrus and bilateral inferior parietal lobule, among others) to process non-native speech. The findings support the Perceptual Assimilation Model and the Speech Learning Model and suggest that the neural system processes phonological information differently depending on the stage of L2 speech learning.

There has been considerable progress in the management of acute lymphoblastic leukemia (ALL) but further improvement is needed to increase long-term survival. The thiopurine agent 6-mercaptopurine ...(6-MP) used for ALL maintenance therapy has a key influence on clinical outcomes and relapse prevention. Genetic inheritance in thiopurine metabolism plays a major role in interindividual clinical response variability to thiopurines; however, most cases of thiopurine resistance remain unexplained.
We used lymphoblastoid cell lines (LCLs) from healthy donors, selected for their extreme thiopurine susceptibility. Thiopurine metabolism was characterized by the determination of TPMT and HPRT activity. We performed genome-wide expression profiling in resistant and sensitive cell lines with the goal of elucidating the mechanisms of thiopurine resistance.
We determined a higher TPMT activity (+44%; P = 0.024) in resistant compared to sensitive cell lines, although there was no difference in HPRT activity. We identified a 32-gene transcriptomic signature that predicts thiopurine resistance. This signature includes the GTPBP4 gene coding for a GTP-binding protein that interacts with p53. A comprehensive pathway analysis of the genes differentially expressed between resistant and sensitive cell lines indicated a role for cell cycle and DNA mismatch repair system in thiopurine resistance. It also revealed overexpression of the ATM/p53/p21 pathway, which is activated in response to DNA damage and induces cell cycle arrest in thiopurine resistant LCLs. Furthermore, overexpression of the p53 target gene TNFRSF10D or the negative cell cycle regulator CCNG2 induces cell cycle arrest and may also contribute to thiopurine resistance. ARHGDIA under-expression in resistant cell lines may constitute a novel molecular mechanism contributing to thiopurine resistance based on Rac1 inhibition induced apoptosis and in relation with thiopurine pharmacodynamics.
Our study provides new insights into the molecular mechanisms underlying thiopurine resistance and suggests a potential research focus for developing tailored medicine.
