The natural history of prostate cancer is highly variable and it is difficult to predict. We showed previously that a cell cycle progression (CCP) score was a robust predictor of outcome in a ...conservatively managed cohort diagnosed by transurethral resection of the prostate. A greater need is to predict outcome in patients diagnosed by needle biopsy.
Total RNA was extracted from paraffin specimens. A CCP score was calculated from expression levels of 31 genes. Clinical variables consisted of centrally re-reviewed Gleason score, baseline prostate-specific antigen level, age, clinical stage, and extent of disease. The primary endpoint was death from prostate cancer.
In univariate analysis (n=349), the hazard ratio (HR) for death from prostate cancer was 2.02 (95% CI (1.62, 2.53), P<10(-9)) for a one-unit increase in CCP score. The CCP score was only weakly correlated with standard prognostic factors and in a multivariate analysis, CCP score dominated (HR for one-unit increase=1.65, 95% CI (1.31, 2.09), P=3 × 10(-5)), with Gleason score (P=5 × 10(-4)) and prostate-specific antigen (PSA) (P=0.017) providing significant additional contributions.
For conservatively managed patients, the CCP score is the strongest independent predictor of cancer death outcome yet described and may prove valuable in managing clinically localised prostate cancer.
In a joint NRL/Manila Observatory mission, as part of the Seven SouthEast Asian Studies program (7-SEAS), a 2-week, late September 2011 research cruise in the northern Palawan archipelago was ...undertaken to observe the nature of southwest monsoonal aerosol particles in the South China Sea/East Sea (SCS/ES) and Sulu Sea region. Previous analyses suggested this region as a receptor for biomass burning from Borneo and Sumatra for boundary layer air entering the monsoonal trough. Anthropogenic pollution and biofuel emissions are also ubiquitous, as is heavy shipping traffic. Here, we provide an overview of the regional environment during the cruise, a time series of key aerosol and meteorological parameters, and their interrelationships. Overall, this cruise provides a narrative of the processes that control regional aerosol loadings and their possible feedbacks with clouds and precipitation. While 2011 was a moderate El Nino-Southern Oscillation (ENSO) La Nina year, higher burning activity and lower precipitation was more typical of neutral conditions. The large-scale aerosol environment was modulated by the Madden-Julian Oscillation (MJO) and its associated tropical cyclone (TC) activity in a manner consistent with the conceptual analysis performed by Reid et al. (2012). Advancement of the MJO from phase 3 to 6 with accompanying cyclogenesis during the cruise period strengthened flow patterns in the SCS/ES that modulated aerosol life cycle. TC inflow arms of significant convection sometimes span from Sumatra to Luzon, resulting in very low particle concentrations (minimum condensation nuclei CN < 150 cm-3, non-sea-salt PM2.5 < 1 mu g m-3). However, elevated carbon monoxide levels were occasionally observed suggesting passage of polluted air masses whose aerosol particles had been rained out. Conversely, two drier periods occurred with higher aerosol particle concentrations originating from Borneo and Southern Sumatra (CN > 3000 cm-3 and non-sea-salt PM2.5 10-25 mu g m-3). These cases corresponded with two different mechanisms of convection suppression: lower free-tropospheric dry-air intrusion from the Indian Ocean, and large-scale TC-induced subsidence. Veering vertical wind shear also resulted in aerosol transport into this region being mainly in the marine boundary layer (MBL), although lower free troposphere transport was possible on the western sides of Sumatra and Borneo. At the hourly time scale, particle concentrations were observed to be modulated by integer factors through convection and associated cold pools. Geostationary satellite observations suggest that convection often takes the form of squall lines, which are bowed up to 500 km across the monsoonal flow and 50 km wide. These squall lines, initiated by cold pools from large thunderstorms and likely sustained by a veering vertical wind shear and aforementioned mid-troposphere dry layers, propagated over 1500 km across the entirety of the SCS/ES, effectively cutting large swaths of MBL aerosol particles out of the region. Our conclusion is that while large-scale flow patterns are very important in modulating convection, and hence in allowing long-range transport of smoke and pollution, more short-lived phenomena can modulate cloud condensation nuclei (CCN) concentrations in the region, resulting in pockets of clean and polluted MBL air. This will no doubt complicate large scale comparisons of aerosol-cloud interaction.
ABSTRACT We conducted Very Large Array C-configuration observations to measure positions and luminosities of Galactic Class II 6.7 GHz methanol masers and their associated ultra-compact H ii regions. ...The spectral resolution was 3.90625 kHz and the continuum sensitivity reached 45 Jy beam−1. We mapped 372 methanol masers with peak flux densities of more than 2 Jy selected from the literature. Absolute positions have nominal uncertainties of 0 3. In this first paper on the data analysis, we present three catalogs; the first gives information on the strongest feature of 367 methanol maser sources, and the second provides information on all detected maser spots. The third catalog presents derived data of the 127 radio continuum counterparts associated with maser sources. Our detection rate of radio continuum counterparts toward methanol masers is approximately one-third. Our catalogs list properties including distance, flux density, luminosity, and the distribution in the Galactic plane. We found no significant relationship between luminosities of masers and their associated radio continuum counterparts, however, the detection rate of radio continuum emission toward maser sources increases statistically with the maser luminosities.
It has become apparent that beer (both alcoholic and nonalcoholic) contains appreciable amounts of non-starch polysaccharides, a broad subgroup of dietary fiber. It is worth noting that the ...occurrence of non-starch polysaccharides in alcoholic beer does not imply this should be consumed as a source of nutrition. But the popularity of nonalcoholic beer is growing, and the lessons learnt from non-starch polysaccharides in brewing can be largely translated to nonalcoholic beer. For context, we briefly review the origins of dietary fiber, its importance within the human diet and the significance of water-soluble dietary fiber in beverages. We review the relationship between non-starch polysaccharides and brewing, giving focus to the techniques used to quantify non-starch polysaccharides in beer, how they affect the physicochemical properties of beer and their influence on the brewing process. The content of non-starch polysaccharides in both regular and low/nonalcoholic beer ranges between 0.5 − 4.0 g/L and are predominantly composed of arabinoxylans and β-glucans. The process of malting, wort production and filtration significantly affect the soluble non-starch polysaccharide content in the final beer. Beer viscosity and turbidity are strongly associated with the content of non-starch polysaccharides.
A range of carbohydrates has been rapidly and selectively converted to 5-chloromethyl furfural using microwave heating in a biphasic reaction system with a range of organic solvents. Fructose and ...inulin were especially effective for production of this valuable bio-platform molecule, with yields of >70% obtained in 15 minutes under microwave heating. Yields from cellulose were dramatically increased with ball mill pre-treatment, this being associated with a reduction in polysaccharide crystallinity.
Background: Histone deacetylase blockade can promote heat shock protein 90 (HSP90) acetylation, abrogating androgen receptor signaling. A phase II trial of the histone deacetylase inhibitor (HDACi) ...romidepsin was conducted in patients with progressing, metastatic, castration-resistant prostate cancer (CRPC). Patients and methods: A dose of 13 mg/m2 was administered i.v. over 4 h on days 1, 8 and 15 every 28 days. The primary end point was rate of disease control defined as no evidence of radiological progression at 6 months. A sample size of 16 assessable patients in stage 1 and nine assessable patients in stage 2 was selected; progression to stage 2 required one or more patients with disease control in stage 1 (Ho = 0.10, Ha = 0.30; α and β = 0.10). Results: Thirty-five patients were enrolled. Two patients achieved a confirmed radiological partial response (RECIST) lasting ≥6 months, along with a confirmed prostate-specific antigen decline of ≥50%. Eleven patients experienced toxicity necessitating early discontinuation. The commonest adverse events were nausea (30 patients; 85.7%), fatigue (28 patients; 80.0%), vomiting (23 patients; 65.7%) and anorexia (20 patients; 57.1%). There was no significant cardiac toxicity. Conclusions: At the dose and schedule selected, romidepsin demonstrated minimal antitumor activity in chemonaive patients with CRPC. Further studies of improved HDACi, alone and in combination with other therapies, should nevertheless be investigated.
Here we present the first steps in developing a global multi-model aerosol forecasting ensemble intended for eventual operational and basic research use. Drawing from members of the International ...Cooperative for Aerosol Prediction (ICAP) latest generation of quasi-operational aerosol models, 5-day aerosol optical thickness (AOT) forecasts are analyzed for December 2011 through November 2012 from four institutions: European Centre for Medium-Range Weather Forecasts (ECMWF), Japan Meteorological Agency (JMA), NASA Goddard Space Flight Center (GSFC), and Naval Research Lab/Fleet Numerical Meteorology and Oceanography Center (NRL/FNMOC). For dust, we also include the National Oceanic and Atmospheric Administration-National Geospatial Advisory Committee (NOAA NGAC) product in our analysis. The Barcelona Supercomputing Centre and UK Met Office dust products have also recently become members of ICAP, but have insufficient data to be included in this analysis period. A simple consensus ensemble of member and mean AOT fields for modal species (e.g., fine and coarse mode, and a separate dust ensemble) is used to create the ICAP Multi-Model Ensemble (ICAP-MME). The ICAP-MME is run daily at 00:00 UTC for 6-hourly forecasts out to 120 h. Basing metrics on comparisons to 21 regionally representative Aerosol Robotic Network (AERONET) sites, all models generally captured the basic aerosol features of the globe. However, there is an overall AOT low bias among models, particularly for high AOT events. Biomass burning regions have the most diversity in seasonal average AOT. The Southern Ocean, though low in AOT, nevertheless also has high diversity. With regard to root mean square error (RMSE), as expected the ICAP-MME placed first over all models worldwide, and was typically first or second in ranking against all models at individual sites. These results are encouraging; furthermore, as more global operational aerosol models come online, we expect their inclusion in a robust operational multi-model ensemble will provide valuable aerosol forecasting guidance.
A refined technique for observing the complete evaporation behaviour of free-falling droplets, from droplet generation to complete solvent evaporation, with ultra-high time resolution is introduced ...and benchmarked. High-resolution phase-delay stroboscopic imaging is employed to simultaneously resolve the evolving droplet morphology, geometric and aerodynamic diameters, throughout the evaporative lifetime with a user-controlled < μs timescale. This allows rapid, complex morphological changes, such as crystallisation events, to be clearly observed and the corresponding mechanisms to be inferred. The dried particles are sampled for offline SEM analysis and the observed morphologies compared to the inflight imaging. Density changes can be calculated directly from the deviation between the geometric and aerodynamic diameters. The full capabilities of the new technique are demonstrated by examination of the different evaporation behaviours and crystallisation mechanisms for aqueous sodium chloride droplets evaporating under different ambient relative humidity (RH) conditions. The crystallisation window, defined as the time taken from initial to complete crystallisation, is shown to be RH dependent, extending from 0.03 s at 20% RH and 0.13 s at 40% RH. The different crystallisation mechanisms observed during the experiments are also clearly reflected in the final structure of the dry particles, with multi-crystal structures produced at low RH compared to single-crystal structures at higher RH. It is anticipated that this technique will unlock measurements which explore the evaporation behaviour and crystallisation mechanisms for rapid, complex droplet drying events, and with increasingly non-ideal solutions, relevant to industrial applications.
High-resolution inflight imaging and measurements of drying droplets enable the evaporation kinetics to be resolved and complex particle crystallisation mechanisms to be observed.
Aerosol mixtures composed of coarse mode desert dust combined with fine mode combustion generated aerosols (from fossil fuel and biomass burning sources) were investigated at three locations that are ...in and/or downwind of major global aerosol emission source regions. Multiyear monitoring data at Aerosol Robotic Network sites in Beijing (central eastern China), Kanpur (Indo-Gangetic Plain, northern India), and Ilorin (Nigeria, Sudanian zone of West Africa) were utilized to study the climatological characteristics of aerosol optical properties. Multiyear climatological averages of spectral single scattering albedo (SSA) versus fine mode fraction (FMF) of aerosol optical depth at 675 nm at all three sites exhibited relatively linear trends up to 50% FMF. This suggests the possibility that external linear mixing of both fine and coarse mode components (weighted by FMF) dominates the SSA variation, where the SSA of each component remains relatively constant for this range of FMF only. However, it is likely that a combination of other factors is also involved in determining the dynamics of SSA as a function of FMF, such as fine mode particles adhering to coarse mode dust. The spectral variation of the climatological averaged aerosol absorption optical depth (AAOD) was nearly linear in logarithmic coordinates over the wavelength range of 440-870 nm for both the Kanpur and Ilorin sites. However, at two sites in China (Beijing and Xianghe), a distinct nonlinearity in spectral AAOD in logarithmic space was observed, suggesting the possibility of anomalously strong absorption in coarse mode aerosols increasing the 870 nm AAOD.
The solubility of cellulose has been studied as a function of composition in the binary mixture of 1,1,3,3-tetramethylguanidine and propionic acid. In amine-rich compositions, greater quantities of ...cellulose can be dissolved than in the equimolar composition, a.k.a. the protic ionic liquid TMGHOPr. By applying a methodology of a short period of heating followed by cooling, similar concentrations of cellulose can be achieved in a much shorter time period. Finally, regeneration of cellulose from solution can be achieved by altering the acid:amine molar ratio. In comparison to cellulose regenerated from these solutions using water as an antisolvent, cellulose regenerated with propionic acid exhibit a lower crystallinity as inferred from x-ray diffractometry, but a greater average molecular weight as inferred from gel permeation chromatography.
