End-point RT-PCR is a suitable alternative diagnostic technique since it is cheaper than RT-qPCR tests and can be implemented on a massive scale in low- and middle-income countries. In this work, a ...bioinformatic approach to guide the design of PCR primers was developed, and an alternative diagnostic test based on end-point PCR was designed. End-point PCR primers were designed through conservation analysis based on kmer frequency in SARS-CoV-2 and human respiratory pathogen genomes. Highly conserved regions were identified for primer design, and the resulting PCR primers were used to amplify 871 nasopharyngeal human samples with a previous RT-qPCR based SARS-CoV-2 diagnosis. The diagnostic test showed high accuracy in identifying SARS-CoV-2-positive samples including B.1.1.7, P.1, B.1.427/B.1.429 and B.1.617.2/ AY samples with a detection limit of 7.2 viral copies/µL. In addition, this test could discern SARS-CoV-2 infection from other viral infections with COVID-19-like symptomatology. The designed end-point PCR diagnostic test to detect SARS-CoV-2 is a suitable alternative to RT-qPCR. Since the proposed bioinformatic approach can be easily applied in thousands of viral genomes and over highly divergent strains, it can be used as a PCR design tool as new SARS-CoV-2 variants emerge. Therefore, this end-point PCR test could be employed in epidemiological surveillance to detect new SARS-CoV-2 variants as they emerge and propagate.
Upon agonist stimulation,
-adrenergic receptors couple to G
proteins, calcium signaling and protein kinase C activation; subsequently, the receptors are phosphorylated, desensitized, and ...internalized. Internalization seems to involve scaffolding proteins, such as
-arrestin and clathrin. However, the fine mechanisms that participate remain unsolved. The roles of protein kinase C and the small GTPase, Rab9, in
-AR vesicular traffic were investigated by studying
-adrenergic receptor-Rab protein interactions, using Förster resonance energy transfer (FRET), confocal microscopy, and intracellular calcium quantitation. In human embryonic kidney 293 cells overexpressing
spp. red fluorescent protein (DsRed)-tagged
-ARs and enhanced green fluorescent protein--tagged Rab proteins, pharmacological protein kinase C activation mimicked
-AR traffic elicited by nonrelated agents, such as sphingosine 1-phosphate (i.e., transient
-AR-Rab5 FRET signal followed by a sustained
-AR-Rab9 interaction), suggesting brief receptor localization in early endosomes and transfer to late endosomes. This latter interaction was abrogated by blocking protein kinase C activity, resulting in receptor retention at the plasma membrane. Similar effects were observed when a dominant-negative Rab9 mutant (Rab9-GDP) was employed. When
-adrenergic receptors that had been mutated at protein kinase C phosphorylation sites (S396A, S402A) were used, phorbol ester-induced desensitization of the calcium response was markedly decreased; however, interaction with Rab9 was only partially decreased and internalization was observed in response to phorbol esters and sphingosine 1-phosphate. Finally, Rab9-GDP expression did not affect adrenergic-mediated calcium response but abolished receptor traffic and altered desensitization. Data suggest that protein kinase C modulates
-adrenergic receptor transfer to late endosomes and that Rab9 regulates this process and participates in G protein-mediated signaling turn-off.
•A new experimental device for the study of drop deformation dynamics is presented.•The drop is deformed under flow fields not accessible until now.•A nonlinear control scheme is implemented.•The ...experimental device allows the study of drops for long experimental times.
In this paper, we present for the deformation of a drop under flow, using an experimental device based on the Two-Roll Mill geometry. To study the drop deformation for extended periods of time, the use of this device requires implementation of a non-linear control scheme that has not previously been used. The device and control scheme generate well characterized flow fields, which present a richness of behavior of the drop dynamics that smoothly varies from that observed in simple shear flows, where the vorticity dominates, to those observed in purely Hyperbolic flows. The Two-Roll Mill flow cell allows detailed studies such as the critical capillary number versus the critical viscosities ratio for elongational flows with significant vorticity. As well, the experimental device is able to track the time evolution of the drop shape permitting to study its complex time evolutions. Experiments were performed for sub-millimeter size drops with the determination of the surface tension with high accuracy.
Mexico has the highest 30-day mortality due to acute myocardial infarction (AMI), which constitutes one of the main causes of mortality in the country: 28 % versus 7.5 % on average for the ...Organization for Economic Co-operation and Development member countries.
To establish critical pathways and essential interinstitutional pharmacological strategies for the care of patients with AMI in Mexico, regardless of their socioeconomic status.
A group of experts in AMI diagnosis and treatment, representatives of the main public health institutions in Mexico, as well as the Mexican cardiology societies, the Mexican Red Cross and representatives of the Spanish Society of Cardiology, were brought together in order to optimize strategies based on the best existing evidence.
An interinstitutional clinical practice guideline was designed for early diagnosis and timely treatment of AMI with ST-segment elevation, following the clinical horizon of the disease, with the proposal of algorithms that improve the prognosis of patients who attend the emergency services due to an AMI.
With these clinical practice guidelines, the group of experts proposes to universalize AMI diagnosis and treatment, regardless of patient socioeconomic status.
During 2006, laboratory studies were conducted to evaluate the pathogenicity of seven isolates of Pandora neoaphidis (Remaudière & Hennebert) against the cabbage aphid, Brevicoryne brassicae (L.). ...Two isolates were obtained from the aphids Rhopalosiphum padi (L.) (TEXRP1), and Myzus persicae (Sulzer) (TEXMP1), infesting broccoli (Brassica napus (L.)) in Texcoco, Mexico; the rest of the isolates were obtained from Rhopalosiphum maidis (Fitch) infesting sorghum (Sorghum bicolor (L.) Moench), in five localities in Tamaulipas, Mexico. The greatest percentages of mortality were caused by isolates RBF, TEXRP1, y RBB, at a rate of 87, 76, y 72%, respectively, 240 hours after the initiation of the treatments. These results show the potential of P. neoaphidis as an agent of biological control against B. brassicae, depending of the place and the origin of the isolate.
