The parabrachial nucleus (PBN), which is located in the pons and is dissected by one of the major cerebellar output tracks, is known to relay sensory information (visceral malaise, taste, ...temperature, pain, itch) to forebrain structures including the thalamus, hypothalamus, and extended amygdala. The availability of mouse lines expressing Cre recombinase selectively in subsets of PBN neurons and viruses for Cre-dependent gene expression is beginning to reveal the connectivity and functions of PBN component neurons. This review focuses on PBN neurons expressing calcitonin gene-related peptide (CGRPPBN) that play a major role in regulating appetite and transmitting real or potential threat signals to the extended amygdala. The functions of other specific PBN neuronal populations are also discussed. This review aims to encourage investigation of the numerous unanswered questions that are becoming accessible.

CGRPPBN neurons relay a wide variety of aversive signals (e.g., food poisoning, pain, itch, and hypercapnia) to the central nucleus of the amygdala, the bed nucleus of the stria terminalis, and other brain regions.
Activation of CGRPPBN neurons is necessary and sufficient to establish robust foot-shock and taste memories.
Chronic activation of CGRPPBN neurons can lead to severe anorexia and starvation.
Other PBN neurons transmit taste, temperature, respiratory, satiety, thirst, salt-appetite, or glucose counter-regulatory signals.
Plants have evolved sophisticated mechanisms to recycle intracellular constituents, which are essential for developmental and metabolic transitions; for efficient nutrient reuse; and for the proper ...disposal of proteins, protein complexes, and even entire organelles that become obsolete or dysfunctional. One major route is autophagy, which employs specialized vesicles to encapsulate and deliver cytoplasmic material to the vacuole for breakdown. In the past decade, the mechanics of autophagy and the scores of components involved in autophagic vesicle assembly have been documented. Now emerging is the importance of dedicated receptors that help recruit appropriate cargo, which in many cases exploit ubiquitylation as a signal. Although operating at a low constitutive level in all plant cells, autophagy is upregulated during senescence and various environmental challenges and is essential for proper nutrient allocation. Its importance to plant metabolism and energy balance in particular places autophagy at the nexus of robust crop performance, especially under suboptimal conditions.
Why did the wave of democracy that swept the former Soviet Union and Eastern Europe starting more than a decade ago develop in ways unexpected by observers who relied on existing theories of ...democracy? In Postcommunism and the Theory of Democracy, four distinguished scholars conduct the first major assessment of democratization theory in light of the experience of postcommunist states. Richard Anderson, Steven Fish, Stephen Hanson, and Philip Roeder not only apply theory to practice, but using a wealth of empirical evidence, draw together the elements of existing theory into new syntheses. The authors each highlight a development in postcommunist societies that reveals an anomaly or lacuna in existing theory. They explain why authoritarian leaders abandon authoritarianism, why democratization sometimes reverses course, how subjects become citizens by beginning to take sides in politics, how rulers become politicians by beginning to seek popular support, and not least, how democracy becomes consolidated. Rather than converging on a single approach, each author shows how either a rationalist, institutionalist, discursive, or Weberian approach sheds light on this transformation. They conclude that the experience of postcommunist democracy demands a rethinking of existing theory. To that end, they offer rich new insights to scholars, advanced students, policymakers, and anyone interested in postcommunist states or in comparative democratization.

Data accumulated over the past few years now show that it targets numerous intracellular regulators that have central roles in hormone signalling, the regulation of chromatin structure and transcription, tailoring morphogenesis, responses to environmental challenges, self recognition and battling pathogens.

Summary Background Clinical trials have shown that trastuzumab, a recombinant monoclonal antibody against HER2 receptor, significantly improves overall survival and disease-free survival in women ...with HER2-positive early breast cancer, but long-term follow-up data are needed. We report the results of comparing observation with two durations of trastuzumab treatment at a median follow-up of 11 years, for patients enrolled in the HERA (HERceptin Adjuvant) trial. Methods HERA (BIG 1-01) is an international, multicentre, open-label, phase 3 randomised trial of 5102 women with HER2-positive early breast cancer, who were enrolled from hospitals in 39 countries between Dec 7, 2001, and June 20, 2005. After completion of all primary therapy (including, surgery, chemotherapy, and radiotherapy as indicated), patients were randomly assigned (1:1:1) to receive trastuzumab for 1 year (once at 8 mg/kg of bodyweight intravenously, then 6 mg/kg once every 3 weeks) or for 2 years (with the same dose schedule), or to the observation group. Primary endpoint is disease-free survival, and analyses are in the intention-to-treat population. Hazard ratios (HRs) were estimated from Cox models, and survival curves were estimated by the Kaplan-Meier method. Comparison of 2 years versus 1 year of trastuzumab is based on 366-day landmark analyses. This study is registered with ClinicalTrials.gov ( NCT00045032 ). Findings Of the 5102 women randomly assigned in the HERA trial, three patients had no evidence of having provided written informed consent to participate. We followed up the intention-to-treat population of 5099 patients (1697 in observation, 1702 in 1-year trastuzumab, and 1700 in 2-years trastuzumab groups). After a median follow-up of 11 years (IQR 10·09–11·53), random assignment to 1 year of trastuzumab significantly reduced the risk of a disease-free survival event (HR 0·76, 95% CI 0·68–0·86) and death (0·74, 0·64–0·86) compared with observation. 2 years of adjuvant trastuzumab did not improve disease free-survival outcomes compared with 1 year of this drug (HR 1·02, 95% CI 0·89–1·17). Estimates of 10-year disease-free survival were 63% for observation, 69% for 1 year of trastuzumab, and 69% for 2 years of trastuzumab. 884 (52%) patients assigned to the observation group selectively crossed over to receive trastuzumab. Cardiac toxicity remained low in all groups and occurred mostly during the treatment phase. The incidence of secondary cardiac endpoints was 122 (7·3%) in the 2-years trastuzumab group, 74 (4·4%) in the 1-year trastuzumab group, and 15 (0·9%) in the observation group. Interpretation 1 year of adjuvant trastuzumab after chemotherapy for patients with HER2-positive early breast cancer significantly improves long-term disease-free survival, compared with observation. 2 years of trastuzumab had no additional benefit. They explore why everywhere, growing economic inequality and the proliferation of precarious, low-wage jobs pose dilemmas for the second generation. They also evaluate perspectives often proposed to explain the success of immigrant integration in certain countries, including nationally specific models, the political economy, and the histories of Canada and the United States as settler societies.

We summarize recent contributions in the broad area of age of information (AoI). In particular, we describe the current state of the art in the design and optimization of low-latency cyberphysical ...systems and applications in which sources send time-stamped status updates to interested recipients. These applications desire status updates at the recipients to be as timely as possible; however, this is typically constrained by limited system resources. We describe AoI timeliness metrics and present general methods of AoI evaluation analysis that are applicable to a wide variety of sources and systems. Starting from elementary single-server queues, we apply these AoI methods to a range of increasingly complex systems, including energy harvesting sensors transmitting over noisy channels, parallel server systems, queueing networks, and various single-hop and multi-hop wireless networks. We also explore how update age is related to MMSE methods of sampling, estimation and control of stochastic processes. The paper concludes with a review of efforts to employ age optimization in cyberphysical applications.
For quartz-rich tectonites two types of deformation thermometer are currently commonly employed: 1) The quartz c-axis fabric opening-angle thermometer that provides an estimate of deformation ...temperatures when fabrics were ‘locked in’ during dislocation creep and dynamic recrystallization. 2) The quartz recrystallization thermometer that indicates a range of likely deformation temperatures based on observed microstructures and inferred mechanisms of dynamic recrystallization. A critically important caveat in applying both thermometers is the assumption that deformation temperature is the primary controlling factor in recrystallization mechanisms and fabric development. However, fabric opening-angles and recrystallization mechanisms are also sensitive to other variables such as strain rate and water weakening. In this paper the development of these thermometers is reviewed, and their potential sensitivities to competing factors such as temperature, strain rate, water weakening and (in the case of opening-angles) 3D strain type are discussed. Examples of the application of these potential thermometers to naturally deformed quartz-rich rocks are given, and case studies of correlations between deformation temperatures estimated by these thermometers and temperatures of synkinematic metamorphism determined by petrology-based thermobarometers are highlighted. In the review, attention is focused on problems associated with applying these thermometers to natural deformation, and examples of such problems are discussed.
