Successful plant establishment is critical to the development of high-yielding crops. Detailed molecular maps for a range of wheat doubled-haploid populations (Cranbrook/Halberd, Sunco/Tasman, CD87/Katepwa and Kukri/Janz) were used to identify genomic regions affecting coleoptile characteristics length, cross-sectional area and degree of spiralling across contrasting soil temperatures. Genotypic variation was large and distributions of genotype means were approximately normal with evidence for transgressive segregation. Narrow-sense heritabilities were high for coleoptile length and cross-sectional area indicating a strong genetic basis for differences among progeny. In contrast, heritabilities for coleoptile spiralling were small. Molecular marker analyses identified a number of significant quantitative trait loci (QTL) for coleoptile growth. Many of the coleoptile growth QTL mapped directly to the Rht-B1 or Rht-D1 dwarfing gene loci conferring reduced cell size through insensitivity to endogenous gibberellins. Other QTL for coleoptile growth were identified throughout the genome. Epistatic interactions were small or non-existent, and there was little evidence for any QTL x temperature interaction. Gene effects at significant QTL were approximately one-half to one-quarter the size of effects at the Rht-B1 and Rht-D1 regions. However, selection at these QTL could together alter coleoptile length by up to 50 mm. In addition to Rht-B1b and Rht-D1b, genomic regions on chromosomes 2B, 2D, 4A, 5D and 6B were repeatable across two or more populations suggesting their potential value for use in breeding and marker-aided selection for greater coleoptile length and improved establishment.
The psychological impact of COVID-19 on Health Care Workers (HCWs) has been widely reported. Few studies have sought to examine HCWs personal models of COVID-19 utilising an established theoretical ...framework. We undertook a mixed methods study of beliefs about COVID-19 held by HCWs in the Mid-West and South of Ireland during the first and third waves of COVID-19. Template analysis was undertaken on the free text responses of 408 HCWs about their perceptions of the Cause of COVID-19 as assessed by the Brief Illness Perception Questionnaire (B-IPQ). Responses were re-examined in the same cohort for stability at 3 months follow-up (n = 100). This analytic template was subsequently examined in a new cohort (n = 253) of HCWs in the third wave. Female HCWs perceived greater emotional impact of COVID-19 than men (t = −4.31, df405, p < 0.01). Differences between occupational groups were evident in relation to Timeline (F4,401 = 3.47, p < 0.01), Treatment Control (F4,401 = 5.64, p < 0.001) and Concerns about COVID-19 (F4,401 = 3.68, p < 0.01). Administration staff believed that treatment would be significantly more helpful and that COVID-19 would last a shorter amount of time than medical/nursing staff and HSCP. However, administration staff were significantly more concerned than HSCP about COVID-19. Template analysis on 1059 responses to the Cause items of the B-IPQ identified ten higher order categories of perceived Cause of COVID-19. The top two Causes identified at both Waves were 'individual behavioural factors' and 'overseas travel'. This study has progressed our understanding of the models HCWs hold about COVID-19 over time, and has highlighted the utility of the template analysis approach in analysing free-text questionnaire data. We suggest that group and individual occupational identities of HCWs may be of importance in shaping HCWs responses to working through COVID-19.
Saposin A (SapA) lipoprotein discs, also known as picodiscs (PDs), represent an attractive method to solubilize glycolipids for protein interaction studies in aqueous solution. Recent electrospray ...ionization mass spectrometry (ESI-MS) data suggest that the size and composition of 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC)-containing PDs at neutral pH differs from those of N,N-dimethyldodecylamine N-oxide determined by X-ray crystallography. Using high-resolution ESI-MS, multiangle laser light scattering (MALLS), and molecular dynamics (MD) simulations, the composition, heterogeneity, and structure of POPC–PDs in aqueous ammonium acetate solutions at pH 4.8 and 6.8 were investigated. The ESI-MS and MALLS data revealed that POPC–PDs consist predominantly of (SapA dimer + iPOPC) complexes, with i = 23–29, and have an average molecular weight (MW) of 38.2 ± 3.3 kDa at pH 4.8. In contrast, in freshly prepared solutions at pH 6.8, POPC–PDs are composed predominantly of (SapA tetramer + iPOPC) complexes, with i = 37–60, with an average MW of 68.0 ± 2.7 kDa. However, the (SapA tetramer + iPOPC) complexes are unstable at neutral pH and convert, over a period of hours, to (SapA trimer + iPOPC) complexes, with i = 29–36, with an average MW of 51.1 ± 2.9 kDa. The results of molecular modeling suggest spheroidal structures for the (SapA dimer + iPOPC), (SapA trimer + iPOPC), and (SapA tetramer + iPOPC) complexes in solution. Comparison of measured collision cross sections (Ω) with values calculated for gaseous (SapA dimer + 26POPC)8+, (SapA trimer + 33POPC)12+, and (SapA tetramer + 42POPC)16+ ions produced from modeling suggests that the solution structures are largely preserved in the gas phase, although the lipids do not maintain regular bilayer orientations.
The nucleon axial charge is calculated as a function of the pion mass in full QCD. Using domain wall valence quarks and improved staggered sea quarks, we present the first calculation with pion ...masses as light as 354 MeV and volumes as large as (3.5 fm)3. We show that finite volume effects are small for our volumes and that a constrained fit based on finite volume chiral perturbation theory agrees with experiment within 7% statistical errors.
Summary
Background Psoriasis has been associated with excessive alcohol consumption and psychological distress.
Objectives To investigate the level of alcohol use in a psoriasis population and to ...determine whether alcohol consumption is linked to psychological distress in patients with psoriasis.
Methods Ninety‐five patients with chronic plaque psoriasis completed validated questionnaires designed to assess psychological distress, current and past alcohol consumption and self‐reported physical severity of psoriasis.
Results Using different measures of alcohol consumption, between 17% and 30% of patients were classified as having difficulties with alcohol. Thirteen per cent and 18% of patients with psoriasis believed that they had a current or past drinking problem, respectively. There was a modest but significant association between levels of anxiety and depression and weekly alcohol consumption (r = 0·29, P < 0·01 and r = 0·24, P = 0·03, respectively). Those patients who believed that they had an alcohol problem had higher levels of anxiety (P = 0·03), depression (P < 0·01) and psoriasis‐associated disability (P = 0·04). There was a modest but significant association between physical severity of psoriasis and weekly alcohol consumption (r = 0·27, P = 0·02).
Conclusions A significant minority of patients with psoriasis consumes excessive alcohol. Patients with psoriasis should be assessed for excessive alcohol use and appropriate interventions initiated. Further studies are required to investigate whether such interventions are effective in combating alcohol abuse and in improving the psychological and physical aspects of psoriasis.

Sequestration of carbon dioxide by steelmaking slag was studied in an atmospheric three-phase system containing industrial slag particles, water, and CO

gas. Batch-type reactors were used to ...measure the rate of aqueous alkaline leaching and slag particle carbonization independently. Four sizes of slag particles were tested for the Ca leaching rate in deionized water at a constant 7.5 pH in an argon atmosphere and for carbonate conversion with CO
2
bubbled through an aqueous suspension. Conversion data (fraction of Ca leached or converted to carbonate) were evaluated to determine the rate-limiting step based on the shrinking core model. For Ca leaching, the chemical reaction is the controlling mechanism during the initial period of time, which then switches to diffusion through the developed porous layer as the rate-limiting step. Carbonate conversion proceeded much slower than leaching conversion and was found to be limited by diffusion through the product calcium carbonate layer. The calculated value of diffusivity was found to be 5 × 10
−9
cm

/s, which decreased by an order of magnitude with increasing carbonization conversion as a result of changing density of the product layer. The experimental data fit the shrinking core model well after correction for the particle specific surface area.

The development of safe diagnostic protocols for working with SARS-CoV-2 clinical samples at Biosafety Level 2 (BSL2) requires understanding of the effect of heat-treatment on SARS-CoV-2 viability and downstream RT-PCR sensitivity. In this study heating SARS-CoV-2/England/2/2020 to 56 °C and 60 °C for 15, 30 and 60 min reduced the virus titre by between 2.1 and 4.9 log10 pfu/mL (as determined by plaque assay). Complete inactivation did not occur and there was significant variability between replicates. Viable virus was detected by plaque assay after heat-treatment at 80 °C for 15 or 30 min but not 60 or 90 min. After heat-treatment at 80 °C for 60 min infectious virus was only detected by more sensitive virus culture. No viable virus was detected after heating to 80 °C for 90 min or 95 °C for 1 or 5 min. RT-PCR sensitivity was not compromised by heating to 56 °C and 60 °C. However, RT-PCR sensitivity was reduced (≥3 Ct value increase) after heating the virus to 80 °C for 30 min or longer, or 95 °C for 1 or 5 min. In summary we found that the efficacy of heat-inactivation varies greatly depending on temperature and duration. Local validation of heat-inactivation and its effects downstream is therefore essential for molecular testing.
Asymmetry in vertical stiffness has been associated with increased injury incidence and impaired performance. The determinants of vertical stiffness asymmetry have not been previously investigated. ...Eighteen healthy men performed three unilateral drop jumps during which vertical stiffness and joint stiffness of the ankle and knee were calculated. Reactive strength index was also determined during the jumps using the ratio of flight time to ground contact time. “Moderate” differences in vertical stiffness (t17 = 5.49; P < 0.001), “small” differences in center of mass displacement (t17 = −2.19; P = 0.043), and “trivial” differences in ankle stiffness (t17 = 2.68; P = 0.016) were observed between stiff and compliant limbs. A model including ankle stiffness and reactive strength index symmetry angles explained 79% of the variance in vertical stiffness asymmetry (R2 = 0.79; P < 0.001). None of the symmetry angles were correlated to jump height or reactive strength index. Results suggest that asymmetries in ankle stiffness may play an important role in modulating vertical stiffness asymmetry in recreationally trained men.
Coconut rhinoceros beetle (
Oryctes rhinoceros
; CRB) was discovered in 2015 in a small outbreak in Honiara, Guadalcanal, Solomon Islands. This was the first record of CRB from Solomon Islands and a ...response plan was prepared. An awareness programme was launched and where CRB sightings were confirmed, delimitation surveys were carried out. Following the launch of the awareness programme, later in 2015, CRB was also reported from the Shortland Islands in the Western Province. Other islands were surveyed from 2015 to 2020. If CRB presence was confirmed, beetles were collected and analysed for haplotype and presence of the classical biological control agent, Oryctes rhinoceros nudivirus (OrNV). A distribution map and timeline of invasion were developed. The initial populations belonged to two distinct haplotypes: CRB-G (clade I
A
) in Honiara and CRB-S (clade II) in Shortland Islands. Despite control measures, by 2020 CRB-G had spread to islands in eight provinces and CRB-S had spread to islands in seven provinces. CRB-S and CRB-G co-occur in Guadalcanal and Malaita provinces. In 2019, OrNV was detected from field collected CRB from Guadalcanal and has since spread to Malaita Island. In both cases the virus was detected where CRB-G and CRB-S co-occur. Other outbreak areas in Solomon Islands remain OrNV-free. The two haplotypes appear to have spread following patterns of air and sea movement between the point of origin (CRB-G from Honiara and CRB-S from the Shortland Islands) to other islands/provinces.
