The BCL-2 inhibitor venetoclax combined with hypomethylating agents or low-dose cytarabine represents an important new therapy for older or unfit patients with acute myeloid leukemia (AML). We ...analyzed 81 patients receiving these venetoclax-based combinations to identify molecular correlates of durable remission, response followed by relapse (adaptive resistance), or refractory disease (primary resistance). High response rates and durable remissions were typically associated with NPM1 or IDH2 mutations, with prolonged molecular remissions prevalent for NPM1 mutations. Primary and adaptive resistance to venetoclax-based combinations was most commonly characterized by acquisition or enrichment of clones activating signaling pathways such as FLT3 or RAS or biallelically perturbing TP53. Single-cell studies highlighted the polyclonal nature of intratumoral resistance mechanisms in some cases. Among cases that were primary refractory, we identified heterogeneous and sometimes divergent interval changes in leukemic clones within a single cycle of therapy, highlighting the dynamic and rapid occurrence of therapeutic selection in AML. In functional studies, FLT3 internal tandem duplication gain or TP53 loss conferred cross-resistance to both venetoclax and cytotoxic-based therapies. Collectively, we highlight molecular determinants of outcome with clinical relevance to patients with AML receiving venetoclax-based combination therapies.
•Adaptive resistance is often associated with TP53 abnormalities or kinase activation, particularly FLT3 internal tandem duplication.•NPM1 mutation is associated with excellent survival prospects and durable molecular remission after venetoclax-based combination therapy.

The nucleus accumbens is a major input structure of the basal ganglia and integrates information from cortical and limbic structures to mediate goal-directed behaviors. Chronic exposure to several ...classes of drugs of abuse disrupts plasticity in this region, allowing drug-associated cues to engender a pathologic motivation for drug seeking. A number of alterations in glutamatergic transmission occur within the nucleus accumbens after withdrawal from chronic drug exposure. These drug-induced neuroadaptations serve as the molecular basis for relapse vulnerability. In this review, we focus on the role that glutamate signal transduction in the nucleus accumbens plays in addiction-related behaviors. First, we explore the nucleus accumbens, including the cell types and neuronal populations present as well as afferent and efferent connections. Next we discuss rodent models of addiction and assess the viability of these models for testing candidate pharmacotherapies for the prevention of relapse. Then we provide a review of the literature describing how synaptic plasticity in the accumbens is altered after exposure to drugs of abuse and withdrawal and also how pharmacological manipulation of glutamate systems in the accumbens can inhibit drug seeking in the laboratory setting. Finally, we examine results from clinical trials in which pharmacotherapies designed to manipulate glutamate systems have been effective in treating relapse in human patients. Further elucidation of how drugs of abuse alter glutamatergic plasticity within the accumbens will be necessary for the development of new therapeutics for the treatment of addiction across all classes of addictive substances.
We examine the weakening of the Atlantic Meridional Overturning Circulation (AMOC) in response to increasing CO
2
at different horizontal resolutions in a state-of-the-art climate model and in a ...small ensemble of models with differing resolutions. There is a strong influence of the ocean mean state on the AMOC weakening: models with a more saline western subpolar gyre have a greater formation of deep water there. This makes the AMOC more susceptible to weakening from an increase in CO
2
since weakening ocean heat transports weaken the contrast between ocean and atmospheric temperatures and hence weaken the buoyancy loss. In models with a greater proportion of deep water formation further north (in the Greenland-Iceland-Norwegian basin), deep-water formation can be maintained by shifting further north to where there is a greater ocean-atmosphere temperature contrast. We show that ocean horizontal resolution can have an impact on the mean state, and hence AMOC weakening. In the models examined, those with higher resolutions tend to have a more westerly location of the North Atlantic Current and stronger subpolar gyre. This likely leads to a greater impact of the warm, saline subtropical Atlantic waters on the western subpolar gyre resulting in greater dense water formation there. Although there is some improvement of the higher resolution models over the lower resolution models in terms of the mean state, both still have biases and it is not clear which biases are the most important for influencing the AMOC strength and response to increasing CO
2
.

Objective To determine the outcomes of Canadian children with biliary atresia. Study design Health records of infants born in Canada between January 1, 1985 and December 31, 1995 (ERA I) and between ...January 1, 1996 and December 31, 2002 (ERA II) who were diagnosed with biliary atresia at a university center were reviewed. Results 349 patients were identified. Median patient age at time of the Kasai operation was 55 days. Median age at last follow-up was 70 months. The 4-year patient survival rate was 81% (ERA I = 74%; ERA II = 82%; P = not significant NS). Kaplan-Meier survival curves for patients undergoing the Kasai operation at age ≤30, 31 to 90, and >90 days showed 49%, 36%, and 23%, respectively, were alive with their native liver at 4 years ( P < .0001). This difference continued through 10 years. The 2- and 4-year post-Kasai operation native liver survival rates were 47% and 35% for ERA I and 46% and 39% for ERA II ( P = NS). A total of 210 patients (60%) underwent liver transplantation; the 4-year transplantation survival rate was 82% (ERA I = 83%, ERA II = 82%; P = NS). Conclusions This is the largest outcome series of North American children with biliary atresia at a time when liver transplantation was available. Results in each era were similar. Late referral remains problematic; policies to ensure timely diagnosis are required. Nevertheless, outcomes in Canada are comparable to those reported elsewhere.
Despite widespread concern about declines in pollination services, little is known about the patterns of change in most pollinator assemblages. By studying bee and hoverfly assemblages in Britain and ...the Netherlands, we found evidence of declines (preversus post-1980) in local bee diversity in both countries; however, divergent trends were observed in hoverflies. Depending on the assemblage and location, pollinator declines were most frequent in habitat and flower specialists, in univoltine species, and/or in nonmigrants. In conjunction with this evidence, outcrossing plant species that are reliant on the declining pollinators have themselves declined relative to other plant species. Taken together, these findings strongly suggest a causal connection between local extinctions of functionally linked plant and pollinator species.
Fourteen research flights were conducted in the Pacific Dust Experiment (PACDEX) during April and May 2007 to sample pollution and dust outbreaks from east Asia as they traveled across the northern ...Pacific Ocean into North America and interacted with maritime storms. Significant concentrations of black carbon (BC, consisting of soot and other light‐absorbing particles measured with a soot photometer 2 instrument) and dust were observed both in the west and east Pacific Ocean from Asian plumes of dust and pollution. BC particles were observed through much of the troposphere, but the major finding is that the percentage of these particles compared with the total number of accumulation mode particles increased significantly (by a factor of 2–4) with increasing altitude, with peak values occurring between 5 and 10 km. Dust plumes had only a small impact on total cloud condensation nuclei at the sampling supersaturations but did exhibit high concentrations of ice nuclei (IN). IN concentrations in dust plumes exceeded typical tropospheric values by 4–20 times and were similar to previous studies in the Saharan aerosol layer when differences in the number concentrations of dust are accounted for. Enhanced IN concentrations were found in the upper troposphere off the coast of North America, providing a first direct validation of the transport of high‐IN‐containing dust layers near the tropopause entering the North American continent from distant sources. A source‐specific chemical transport model was used to predict dust and other aerosols during PACDEX. The model was able to predict several features of the in situ observations, including the general altitudes where BC was found and a peak in the ratio of BC to sulfate between 5 and 10 km.
ABSTRACT
The BRITICE‐CHRONO Project has generated a suite of recently published radiocarbon ages from deglacial sequences offshore in the Celtic and Irish seas and terrestrial cosmogenic nuclide and ...optically stimulated luminescence ages from adjacent onshore sites. All published data are integrated here with new geochronological data from Wales in a revised Bayesian analysis that enables reconstruction of ice retreat dynamics across the basin. Patterns and changes in the pace of deglaciation are conditioned more by topographic constraints and internal ice dynamics than by external controls. The data indicate a major but rapid and very short‐lived extensive thin ice advance of the Irish Sea Ice Stream (ISIS) more than 300 km south of St George's Channel to a marine calving margin at the shelf break at 25.5 ka; this may have been preceded by extensive ice accumulation plugging the constriction of St George's Channel. The release event between 25 and 26 ka is interpreted to have stimulated fast ice streaming and diverted ice to the west in the northern Irish Sea into the main axis of the marine ISIS away from terrestrial ice terminating in the English Midlands, a process initiating ice stagnation and the formation of an extensive dead ice landscape in the Midlands.
We project onto the light front the pion's Poincaré-covariant Bethe-Salpeter wave function obtained using two different approximations to the kernels of quantum chromodynamics' Dyson-Schwinger ...equations. At an hadronic scale, both computed results are concave and significantly broader than the asymptotic distribution amplitude, φ(π)(asy)(x)=6x(1-x); e.g., the integral of φ(π)(x)/φ(π)(asy)(x) is 1.8 using the simplest kernel and 1.5 with the more sophisticated kernel. Independent of the kernels, the emergent phenomenon of dynamical chiral-symmetry breaking is responsible for hardening the amplitude.
