Photonic components with adjustable parameters, such as variable-focal-length lenses or spectral filters, which can change functionality upon optical stimulation, could offer numerous useful ...applications. Tuning of such components is conventionally achieved by either micro- or nanomechanical actuation of their constituent parts, by stretching or by heating. Here, we report a novel approach for making reconfigurable optical components that are created with light in a non-volatile and reversible fashion. Such components are written, erased and rewritten as two-dimensional binary or greyscale patterns into a nanoscale film of phase-change material by inducing a refractive-index-changing phase transition with tailored trains of femtosecond pulses. We combine germanium-antimony-tellurium-based films with a diffraction-limited resolution optical writing process to demonstrate a variety of devices: visible-range reconfigurable bichromatic and multi-focus Fresnel zone plates, a super-oscillatory lens with subwavelength focus, a greyscale hologram, and a dielectric metamaterial with on-demand reflection and transmission resonances.
The past decade has seen an intensive effort to achieve optical imaging resolution beyond the diffraction limit. Apart from the Pendry-Veselago negative index superlens, implementation of which in ...optics faces challenges of losses and as yet unattainable fabrication finesse, other super-resolution approaches necessitate the lens either to be in the near proximity of the object or manufactured on it, or work only for a narrow class of samples, such as intensely luminescent or sparse objects. Here we report a new super-resolution microscope for optical imaging that beats the diffraction limit of conventional instruments and the recently demonstrated near-field optical superlens and hyperlens. This non-invasive subwavelength imaging paradigm uses a binary amplitude mask for direct focusing of laser light into a subwavelength spot in the post-evanescent field by precisely tailoring the interference of a large number of beams diffracted from a nanostructured mask. The new technology, which--in principle--has no physical limits on resolution, could be universally used for imaging at any wavelength and does not depend on the luminescence of the object, which can be tens of micrometres away from the mask. It has been implemented as a straightforward modification of a conventional microscope showing resolution better than λ/6.
Molecular understanding of neutralizing antibody responses to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) could accelerate vaccine design and drug discovery. We analyzed 294 ...anti-SARS-CoV-2 antibodies and found that immunoglobulin G heavy-chain variable region 3-53 (IGHV3-53) is the most frequently used IGHV gene for targeting the receptor-binding domain (RBD) of the spike protein. Co-crystal structures of two IGHV3-53-neutralizing antibodies with RBD, with or without Fab CR3022, at 2.33- to 3.20-angstrom resolution revealed that the germline-encoded residues dominate recognition of the angiotensin I converting enzyme 2 (ACE2)-binding site. This binding mode limits the IGHV3-53 antibodies to short complementarity-determining region H3 loops but accommodates light-chain diversity. These IGHV3-53 antibodies show minimal affinity maturation and high potency, which is promising for vaccine design. Knowledge of these structural motifs and binding mode should facilitate the design of antigens that elicit this type of neutralizing response.
Abstract
This study investigates the role of the parameterized boundary layer structure in hurricane intensity change using two retrospective HWRF forecasts of Hurricane Earl (2010) in which the ...vertical eddy diffusivity Km was modified during physics upgrades. Earl undergoes rapid intensification (RI) in the low-Km forecast as observed in nature, while it weakens briefly before resuming a slow intensification at the RI onset in the high-Km forecast. Angular momentum budget analysis suggests that Km modulates the convergence of angular momentum in the boundary layer, which is a key component of the hurricane spinup dynamics. Reducing Km in the boundary layer causes enhancement of both the inflow and convergence, which in turn leads to stronger and more symmetric deep convection in the low-Km forecast than in the high-Km forecast. The deeper and stronger hurricane vortex with lower static stability in the low-Km forecast is more resilient to shear than that in the high-Km forecast. With a smaller vortex tilt in the low-Km forecast, downdrafts associated with the vortex tilt are reduced, bringing less low-entropy air from the midlevels to the boundary layer, resulting in a less stable boundary layer. Future physics upgrades in operational hurricane models should consider this chain of multiscale interactions to assess their impact on model RI forecasts.

The structure and evolution of Hurricane Earl (2010) during its rapid intensification as sampled by aircraft is studied here. Rapid intensification occurs in two stages. During the early stage, ...covering similar to 24 h, Earl was a tropical storm experiencing moderate northeasterly shear with an asymmetric distribution of convection, and the symmetric structure was shallow, broad, and diffuse. The upper-level circulation center was significantly displaced from the lower-level circulation at the beginning of this stage. Deep, vigorous convection-termed convective bursts-was located on the east side of the storm and appeared to play a role in positioning the upper-level cyclonic circulation center above the low-level center. By the end of this stage the vortex was aligned and extended over a deep layer, and rapid intensification began. During the late stage, rapid intensification continued as Earl intensified similar to 20 m s super(-1) during the next 24 h. The vortex remained aligned in the presence of weaker vertical shear, although azimuthal asymmetries persisted that were characteristic of vortices in shear. Convective bursts concentrated near the radius of maximum winds, with the majority located inside the radius of maximum winds. Each of the two stages described here raises questions about the role of convective- and vortex-scale processes in rapid intensification. During the early stage, the focus is on the role of convective bursts and their associated mesoscale convective system on vortex alignment and the onset of rapid intensification. During the late stage, the focus is on the processes that explain the observed radial distribution of convective bursts that peak inside the radius of maximum winds.

Technological advances have enabled the identification of an increasingly large spectrum of single nucleotide variants within the human genome, many of which may be associated with monogenic disease ...or complex traits. Here, we propose an integrative approach, named FATHMM-MKL, to predict the functional consequences of both coding and non-coding sequence variants. Our method utilizes various genomic annotations, which have recently become available, and learns to weight the significance of each component annotation source.
We show that our method outperforms current state-of-the-art algorithms, CADD and GWAVA, when predicting the functional consequences of non-coding variants. In addition, FATHMM-MKL is comparable to the best of these algorithms when predicting the impact of coding variants. The method includes a confidence measure to rank order predictions.
Countermeasures to prevent and treat coronavirus disease 2019 (COVID-19) are a global health priority. We enrolled a cohort of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)-recovered ...participants, developed neutralization assays to investigate antibody responses, adapted our high-throughput antibody generation pipeline to rapidly screen more than 1800 antibodies, and established an animal model to test protection. We isolated potent neutralizing antibodies (nAbs) to two epitopes on the receptor binding domain (RBD) and to distinct non-RBD epitopes on the spike (S) protein. As indicated by maintained weight and low lung viral titers in treated animals, the passive transfer of a nAb provides protection against disease in high-dose SARS-CoV-2 challenge in Syrian hamsters. The study suggests a role for nAbs in prophylaxis, and potentially therapy, of COVID-19. The nAbs also define protective epitopes to guide vaccine design.
Airborne Doppler radar observations of the wind field in the tropical cyclone boundary layer (TCBL) during the landfall of Hurricane Ida (2021) are examined here. Asymmetries in tangential and radial ...flow are governed by tropical cyclone (TC) motion and vertical wind shear prior to landfall, while frictional effects dominate the asymmetry location during landfall. Strong TCBL inflow on the offshore‐flow side of Ida occurs during landfall, while the location of the peak tangential wind at the top of the TCBL during this period is located on the onshore‐flow side. A comparison of these observations with a numerical simulation of TC landfall shows many consistencies with the modeling study, though there are some notable differences that may be related to differences in the characteristics of the land surface between the simulation and the observations here.
Plain Language Summary
The structure of the wind field in the lowest levels of a hurricane when it makes landfall plays a significant role in determining damage. This study uses aircraft observations to document the changes that occur in the winds in the lowest levels of a hurricane prior to and as it makes landfall. The location of the strongest inflow into the storm center and winds around the center are determined by characteristics like storm motion and environmental winds when the storm is over open water, but at landfall these wind peaks are dominated by the differences in friction between the land surface and water. At landfall, the inflow near the surface is strongest on the side of the hurricane experiencing offshore flow, while winds moving around the storm center a few thousand feet above the surface are strongest on the side experiencing onshore flow. These results are mostly consistent with computer modeling studies, allowing for studies on physical processes to be conducted using these models.
