We studied the epidemiologic triad-related factors influencing human papilloma virus (HPV) persistence in Mexican women with systemic lupus erythematosus (SLE). Patients aged ≥18 years with SLE ...(American College of Rheumatology criteria), with and without HPV persistence, were selected. Groups were analyzed by (1) host: clinical disease characteristics; (2) agent: (I) infectious (prevalence, incidence, HPV genotype and co-infections (≥2 HPV genotypes or mycoplasmas)), (II) chemical (contraceptives and immunosuppressive drugs) and (III) physical (vitamin D deficiency) and (3) environment. A total of 121 SLE patients were selected over a two-year period. (1) Host: mean age 45.8 years and disease duration 12.7 years. (2) Agent: (I) infectious. HPV infection prevalence in the second sample was 26.4%, high-risk HPV genotypes 21.5% and co-infections 7.4%. HPV infection incidence was 13.2%, persistence 13.2% and clearance 15.7%. (II) Chemical: use of oral hormonal contraceptives 5% and immunosuppressive treatment 97.5%. (III) Physical: Vitamin D levels were similar in both groups. (3) Environment: (I) natural. A total of 60.6% of patients were residents of Puebla City. (II) Social: The mean education level was 10.9. Poverty levels were: III degree 52.4%, IV degree 28% and II degree 17%. (III) Cultural behavioral: Onset of sexual life was 20.5 years, 10% had ≥3 sexual partners and 51.2% were postmenopausal. In conclusion, no factor of the epidemiologic triad was associated with HPV infection prevalence.

Aldo-keto reductases (AKRs) metabolize a wide range of substrates, including polycyclic aromatic hydrocarbons (PAHs), generating metabolites (o-quinones) and reactive oxygen species (ROS), which are ...capable of initiating and promoting carcinogenesis. Exposure to PAHs, their metabolites, and ROS further increase AKRs isoform expression that may amplify oxidative damage. Human AKR enzymes are highly polymorphic, and allelic variants may contribute to different AKRs expression in individuals. Despite the importance of AKRs in PAHs metabolism, there are no studies that evaluate, in general human populations, the effect of PAHs on AKRs expression in peripheral blood lymphocytes (PBLs). The aim of this study was to determine the effect of tobacco smoke exposure, and AKR1A1*2 and AKR1C3*2 polymorphisms, on AKR1A1 and AKR1C1–AKR1C3 messenger RNA (mRNA) levels in PBLs from smokers. In the smoker group, there is a statistically significant positive association between AKR1A1, AKR1C1, and AKR1C3 mRNA induction and urine cotinine levels in individuals with a body mass index (BMI) less than 25. However, AKR1A1*2 and AKR1C3*2 alleles did not influence AKR1A1 and AKR1C1–AKR1C3 mRNA levels. These results suggest that AKRs induction by PAHs in smokers’ PBLs is associated with BMI; therefore, the role of adipose tissue accumulation in PAHs’ effects needs further investigation.

We report on the experimental observation of magnetic hysteresis up to room temperature in thiol-capped Au nanoparticles with 1.4 nm size. The coercive field ranges from 860 Oe at 5 K to 250 Oe at ...300 K. It is estimated that the Au atoms exhibit a magnetic moment of mu=0.036mu(B). However, Au nanoparticles with similar size but stabilized by means of a surfactant, i.e., weak interaction between protective molecules and Au surface atoms, are diamagnetic, as bulk Au samples are. The apparent ferromagnetism is consequently associated with 5d localized holes generated through Au-S bonds. These holes give rise to localized magnetic moments that are frozen in due to the combination of the high spin-orbit coupling (1.5 eV) of gold and the symmetry reduction associated with two types of bonding: Au-Au and Au-S.

Chlorpyrifos and methyl parathion are among the most widely used insecticides in the world. Human populations are constantly exposed to low doses of both due to their extensive use and presence in food and drinking water. Glutathione S-transferase (GST) catalyzes the conjugation of glutathione on electrophilic substrates and is an important line of defense in the protection of cellular components from reactive species. GST alpha1 (GSTA1) is the predominant isoform of GST expressed in the human liver; thus, determining the effect of insecticides on GSTA1 transcription is very important. In the present study, we analyzed the effects of methyl parathion and chlorpyrifos on GSTA1 gene expression in HepG2 cells using real time PCR, and activity and immunoreactive protein assays. The results demonstrated that exposure to methyl parathion and chlorpyrifos increased the level of GSTA1 mRNA, GSTA1 immunoreactive protein and GST activity relative to a control. These results demonstrated that these insecticides can increase the expression of GSTA1. In conclusion, HepG2 cell cultures treated with methyl parathion and chlorpyrifos could be a useful model for studying the function of GSTA1 and its role in the metabolism of xenobiotics in the liver.

In this cross-sectional study of the cohort of T2D patients, a total of 224 individuals were selected for interaction studies. Clinical and anthropometric data were obtained from pre-designed medical records. Dietary intake was assessed by validated three-day food consumption records. Biochemical measurements were determined by automated methods.
genotyping was performed by a real-time allelic discrimination assay. Gene-diet interactions were tested by corrected multiple linear regression analyses, which were adjusted by potential confounding factors such as age, sex, energy intake, BMI and anti-hyperglycemic therapy (Metformin, Glibenclamide or Insulin), and years with T2D.
Seventy-six percent of patients with T2D were on Metformin therapy. The frequencies of the
alleles were ε2 (5.8%), ε3 (74.1%) and ε4 (20.1%). After statistical settings, significant
alleles-by-diet interactions in relation to the metabolic profile were found. Interestingly, higher blood levels of total cholesterol (p int. = 0.016), non-HDL-c (p int. = 0.024), and LDL-c (p int. = 0.030) were found only in carriers of the
ε2 allele with a low consumption of MUFA. In contrast, carriers of the
ε4 allele with a high ω-6:ω-3 PUFA ratio in the diet had higher %HbA1c blood concentrations (p int. = 0.035).
This study suggests a differential metabolic impact of
alleles on lipid/glycemic phenotypes depending on the dietary intake, with important potential implications in the personalized medicine and nutritional management of patients with type 2 diabetes mellitus.
EMMPRIN, also known as Basigin or CD147, is a transmembrane glycoprotein member of the immunoglobulin superfamily. It is expressed basally in cells that regulate physiological processes of the ...cardiovascular, nervous, and immune systems. However, EMMPRIN is also capable of interacting with different proteins, like VEGFR, SMAD4, Integrin, MCT, CyPA, GLUT1, CAIV, Annexin II, Cav-1, CAML, etc., and regulating signaling pathways that stimulate the cell processes of proliferation, apoptosis, metabolism, adhesion, invasion, migration, metastasis, tumor immune response, and angiogenesis processes, which favors the development of different types of cancer. EMMPRIN is the first protein reported that favors cancer development due to its ability to interact with extracellular, intracellular, and membrane proteins. In conclusion, EMMPRIN regulates several proteins associated with the development of tumor processes. Therefore, blocking the expression of EMMPRIN can be a therapeutic target, and the analysis of its expression can be used as an important biomarker in cancer.
The Satellite for Orbital Aerodynamics Research (SOAR) is a CubeSat mission, due to be launched in 2021, to investigate the interaction between different materials and the atmospheric flow regime in ...very low Earth orbits (VLEO). Improving knowledge of the gas–surface interactions at these altitudes and identification of novel materials that can minimise drag or improve aerodynamic control are important for the design of future spacecraft that can operate in lower altitude orbits. Such satellites may be smaller and cheaper to develop or can provide improved Earth observation data or communications link-budgets and latency. In order to achieve these objectives, SOAR features two payloads: (i) a set of steerable fins which provide the ability to expose different materials or surface finishes to the oncoming flow with varying angle of incidence whilst also providing variable geometry to investigate aerostability and aerodynamic control; and (ii) an ion and neutral mass spectrometer with time-of-flight capability which enables accurate measurement of the in-situ flow composition, density, velocity. Using precise orbit and attitude determination information and the measured atmospheric flow characteristics the forces and torques experienced by the satellite in orbit can be studied and estimates of the aerodynamic coefficients calculated. This paper presents the scientific concept and design of the SOAR mission. The methodology for recovery of the aerodynamic coefficients from the measured orbit, attitude, and in-situ atmospheric data using a least-squares orbit determination and free-parameter fitting process is described and the experimental uncertainty of the resolved aerodynamic coefficients is estimated. The presented results indicate that the combination of the satellite design and experimental methodology are capable of clearly illustrating the variation of drag and lift coefficient for differing surface incidence angle. The lowest uncertainties for the drag coefficient measurement are found at approximately 300 km, whilst the measurement of lift coefficient improves for reducing orbital altitude to 200 km.
•The design of the experimental CubeSat SOAR (Satellite for Orbital Aerodynamics) is presented.•SOAR is due to be launched in early 2021 to the ISS and subsequently deployed into orbit.•The mission will perform the aerodynamic characterisation of materials in very low Earth orbit (VLEO).•The aim is to improve understanding of the fundamental gas–surface interactions in the VLEO environment.•Materials able to reduce aerodynamic drag and improve aerodynamics-based control will be tested.
Aims.
We study 147 star clusters in the Large Magellanic Cloud (LMC) in order to determine their mean metallicities and ages, as well as the mean metallicities of 80 surrounding fields. We construct ...an age–metallicity relation (AMR) for the clusters in the LMC.
Methods.
For this purpose, we used Strömgren photometry obtained with the SOI camera on the 4.1 m SOAR telescope. We derived the metallicities of individual stars utilizing a metallicity calibration of the Strömgren (
b
−
y
) and
m
1 colors from the literature. Cluster ages were determined from the isochrone fitting.
Results.
We found the mean metallicity and age for 110 star clusters. For the remaining 37, we provide an age estimation only. To the best of our knowledge, for 29 clusters from our sample, we provide both the metallicity and age for the first time, whereas for 66 clusters, we provide a first determination of the metallicity, and for 43 clusters, the first estimation of the age. We also calculated the mean metallicities for stars from 80 fields around the clusters. The results were then analyzed for spatial metallicity and age distributions of clusters in the LMC, as well as their AMR. The old, metal-poor star clusters occur both in and out of the LMC bar region, while intermediate-age clusters are located mostly outside of the bar. The majority of star clusters younger than 1 Gyr are located in the bar region. We find a good agreement between our AMR and theoretical models of the LMC chemical enrichment, as well as with AMRs for clusters from the literature. Next, we took advantage of 26 stellar clusters from our sample which host Cepheid variables and used them as an independent check of the correctness of our age determination procedure. We used period-age relations for Cepheids to calculate the mean age of a given cluster and compared it with the age obtained from isochrone fitting. We find good agreement between these ages, especially for models taking into account additional physical processes (e.g., rotation). We also compared the AMR of the LMC and Small Magellanic Cloud (SMC) derived in a uniform way and we note that they indicate possible former interaction between these two galaxies. The Strömgren photometry obtained for this study has been made publicly available.
Rituximab has emerged as an efficacious option for drug-resistant myasthenia gravis (MG). However, reports published only describe the short-term follow-up of patients treated and little is known ...about their long-term clinical and immunologic evolution. Our objective was to report the clinical and immunologic long-term follow-up of 17 patients (6 MuSK+MG and 11 AChR+MG) and compare the response between AChR+MG and MuSK+MG patients.
Myasthenia Gravis Foundation America postintervention status and changes in treatment and antibody titers were periodically determined. Lymphocyte subpopulations, total immunoglobulin, immunoglobulin G (IgG) anti-MuSK subclasses, and anti-tetanus toxoid IgG before and after treatment were also studied.
After a mean post-treatment period of 31 months, 10 of the AChR+MG patients improved but 6 of them needed reinfusions. In contrast, all MuSK+MG patients achieved a remission (4/6) or minimal manifestations (2/6) status and no reinfusions were needed. Consequently, in the MuSK+MG group, prednisone doses were significantly reduced and concomitant immunosuppressants could be withdrawn. Clinical improvement was associated with a significant decrease in the antibody titers only in the 6 MuSK+MG patients. At last follow-up MuSK antibodies were negative in 3 of these patients and showed a decrease of over 80% in the other 3.
In view of the long-lasting benefit observed in MuSK+MG patients, we recommend to use rituximab as an early therapeutic option in this group of patients with MG if they do not respond to prednisone.
This study provides Class IV evidence that IV rituximab improves the clinical and immunologic status of patients with MuSK+MG.
