Directly detecting thermal emission from young extrasolar planets allows measurement of their atmospheric compositions and luminosities, which are influenced by their formation mechanisms. Using the ...Gemini Planet Imager, we discovered a planet orbiting the ∼20-million-year-old star 51 Eridani at a projected separation of 13 astronomical units. Near-infrared observations show a spectrum with strong methane and water-vapor absorption. Modeling of the spectra and photometry yields a luminosity (normalized by the luminosity of the Sun) of 1.6 to 4.0 × 10–6 and an effective temperature of 600 to 750 kelvin. For this age and luminosity, "hot-start" formation models indicate a mass twice that of Jupiter. This planet also has a sufficiently low luminosity to be consistent with the "cold-start" core-accretion process that may have formed Jupiter.
There has been increasing interest in finding non-invasive biomarkers for neurodegenerative diseases such as Alzheimer's disease (AD). This observational study investigated AD-specific biomarkers in ...tear fluid. Tear fluid was collected from a total of 65 subjects, including 23 patients with subjective cognitive decline (SCD), 22 patients with mild cognitive impairment (MCI), 11 dementia patients and 9 healthy controls (HC). Levels of amyloid-beta peptides (AB38, AB40, AB42), total-tau (t-tau) and phosphorylated-tau (p-tau) were determined using multiplex immunoassays. Levels of AB40 and t-tau were detectable in the vast majority (> 94%) of tear fluid samples. Cerebrospinal fluid (CSF) was available from a subset of patients. In this group, tear t-tau levels were significantly higher in people with dementia compared to SCD patients. Tear t-tau levels were elevated in patients with neurodegeneration (classified according to the A/T/N system) compared to patients without neurodegeneration. Negative correlations were found between CSF AB42 and CSF t-tau, and between CSF AB42 and tear t-tau. In summary, this study shows the potential of tau proteins in tear fluid to be associated with disease severity and neurodegeneration.
Alzheimer's disease (AD) is the most common cause of dementia, and due to increasing life expectancy the number of patients is expected to grow. The diagnosis of AD involves the use of biomarkers ...determined by an amyloid PET scan or cerebrospinal fluid analyses that are either invasive or expensive, and not available in each hospital, thus limiting their usage as a front-line screener. The TearAD study aims to use tear fluid as a potential source for AD biomarkers. In previous reports, we demonstrated that AD biomarkers amyloid-beta and tau, are measurable in tear fluid and are associated with disease severity and neurodegeration. This study aims to validate previous results in a larger cohort and evaluate the diagnostic accuracy of tear biomarkers to discriminate between individuals with and without neurodegeneration as determined by hippocampal atrophy.
The TearAD study is an observational longitudinal multi-center study that will enroll 50 cognitively healthy controls, 50 patients with subjective cognitive decline, 50 patients with mild cognitive impairment and 50 patients with AD dementia from the memory clinic. Participants will be examined at baseline, after one year, and after two years follow-up. Study assessments include neuropsychological tests and ophthalmic examination. All participants will receive a MRI scan, and a subset of the study population will undergo cerebral spinal fluid collection and an amyloid PET scan. Tear fluid will be collected with Schirmer strips and levels of Aβ38, Aβ40, Aβ42, t-tau and p-tau in tear fluid will be determined using multiplex immunoassays. Blood samples will be collected from all participants. Images of the retina will be obtained with a standard, hyperspectral and ultra-wide field fundus camera. Additionally, macular pigment optical density will be measured with the macular pigment reflectometer, and cross-sectional images of the retina will be obtained through optical coherence tomography imaging.
The TearAD study will provide insight into the potential diagnostic use of tear biomarkers as a minimally invasive and low cost tool for the screening and diagnosis of AD.

Hydrogen-poor superluminous supernovae (SLSNe-I) have been predominantly found in low-metallicity, star-forming dwarf galaxies. Here we identify Gaia17biu/SN 2017egm as an SLSN-I occurring in a ..."normal" spiral galaxy (NGC 3191) in terms of stellar mass (several times 1010 M ) and metallicity (roughly solar). At redshift z = 0.031, Gaia17biu is also the lowest-redshift SLSN-I to date, and the absence of a larger population of SLSNe-I in dwarf galaxies of similar redshift suggests that metallicity is likely less important to the production of SLSNe-I than previously believed. With the smallest distance and highest apparent brightness for an SLSN-I, we are able to study Gaia17biu in unprecedented detail. Its pre-peak near-ultraviolet to optical color is similar to that of Gaia16apd and among the bluest observed for an SLSN-I, while its peak luminosity (Mg = −21 mag) is substantially lower than that of Gaia16apd. Thanks to the high signal-to-noise ratios of our spectra, we identify several new spectroscopic features that may help to probe the properties of these enigmatic explosions. We detect polarization at the ∼0.5% level that is not strongly dependent on wavelength, suggesting a modest, global departure from spherical symmetry. In addition, we put the tightest upper limit yet on the radio luminosity of an SLSN-I with <5.4 × 1026 erg s−1 Hz−1 at 10 GHz, which is almost a factor of 40 better than previous upper limits and one of the few measured at an early stage in the evolution of an SLSN-I. This limit largely rules out an association of this SLSN-I with known populations of gamma-ray-burst-like central engines.
The Aerospace Corporation's Nightglow Imager (ANI) observes nighttime OH emission (near 1.6 μm) every 2 s over an approximate 73° field of view. ANI had previously been used to study instability ...features seen over Maui. Here we describe observations of instabilities seen from 5 to 8 UT on 24 March 2012 over Cerro Pachon, Chile, and compare them with previous results from Maui, with theory, and with Direct Numerical Simulations (DNS). The atmosphere had reduced stability because of the large negative temperature gradients measured by a Na lidar. Thus, regions of dynamical and convective instabilities are expected to form, depending on the value of the Richardson number. Bright primary instabilities are formed with a horizontal wavelength near 9 km and showed the subsequent formation of secondary instabilities, rarely seen over Maui, consistent with the primaries being dynamical instabilities. The ratio of the primary to secondary horizontal wavelength was greater over Chile than over Maui. After dissipation of the instabilities, smaller‐scale features appeared with sizes in the buoyancy subrange between 1.5 and 6 km. Their size spectra were consistent with the model of Weinstock (1978) if the turbulence is considered to be increasing. The DNS results produce secondary instabilities with sizes comparable to what is seen in the images although their spectra are somewhat steeper than is observed. However, the DNS results also show that after the complete decay of the primary features, scale sizes considerably smaller than 1 km are produced and these cannot be seen by the ANI instrument.

The SERCA-LVAD trial was a phase 2a trial assessing the safety and feasibility of delivering an adeno-associated vector 1 carrying the cardiac isoform of the sarcoplasmic reticulum calcium ATPase ...(AAV1/SERCA2a) to adult chronic heart failure patients implanted with a left ventricular assist device. The SERCA-LVAD trial was one of a program of AAV1/SERCA2a cardiac gene therapy trials including CUPID1, CUPID 2 and AGENT trials. Enroled subjects were randomised to receive a single intracoronary infusion of 1 × 10
DNase-resistant AAV1/SERCA2a particles or a placebo solution in a double-blinded design, stratified by presence of neutralising antibodies to AAV. Elective endomyocardial biopsy was performed at 6 months unless the subject had undergone cardiac transplantation, with myocardial samples assessed for the presence of exogenous viral DNA from the treatment vector. Safety assessments including ELISPOT were serially performed. Although designed as a 24 subject trial, recruitment was stopped after five subjects had been randomised and received infusion due to the neutral result from the CUPID 2 trial. Here we describe the results from the 5 patients at 3 years follow up, which confirmed that viral DNA was delivered to the failing human heart in 2 patients receiving gene therapy with vector detectable at follow up endomyocardial biopsy or cardiac transplantation. Absolute levels of detectable transgene DNA were low, and no functional benefit was observed. There were no safety concerns in this small cohort. This trial identified some of the challenges of performing gene therapy trials in this LVAD patient cohort which may help guide future trial design.
Supernova (SN) 2008ax in NGC 4490 was discovered within hours after shock breakout, presenting the rare opportunity to study a core-collapse SN beginning with the initial envelope-cooling phase ...immediately following shock breakout. We present an extensive sequence of optical and near-infrared spectra, as well as three epochs of optical spectropolarimetry. Our initial spectra, taken two days after shock breakout, are dominated by hydrogen Balmer lines at high velocity. However, by maximum light, He I lines dominated the optical and near-infrared spectra, which closely resembled those of normal Type Ib supernovae (SNe Ib) such as SN 1999ex. This spectroscopic transition defines Type IIb SNe, but the strong similarity of SN 2008ax to normal SNe Ib beginning near maximum light, including an absorption feature near 6270 A due to H Delta *a at high velocities, suggests that many objects classified as SNe Ib in the literature may have ejected similar amounts of hydrogen as SN 2008ax, roughly a few X 0.01 M . Only the unusually early discovery of SN 2008ax allowed us to observe the spectroscopic signatures of the hydrogen-rich outer ejecta. Early-time spectropolarimetry (six and nine days after shock breakout) revealed strong line polarization modulations of 3.4% across H Delta *a, indicating the presence of large asphericities in the outer ejecta and possibly that the spectrum of SN 2008ax could be dependent on the viewing angle. After removal of interstellar polarization, the continuum shares a common polarization angle with the hydrogen, helium, and oxygen lines, while the calcium and iron absorptions are oriented at different angles. This is clear evidence of deviations from axisymmetry even in the outer ejecta. Intrinsic continuum polarization of 0.64% only nine days after shock breakout shows that the outer layers of the ejecta were quite aspherical. A single epoch of late-time spectropolarimetry as well as the shapes of the nebular line profiles demonstrate that asphericities extended from the outermost layers all the way down to the center of this core-collapse SN. SN 2008ax may in some ways be an extragalactic analog of the explosion giving rise to Cassiopeia A, which has recently been determined to be a remnant of an SN IIb.
We report the results of spectroscopic and photometric observations of the emission-line object AS 386. For the first time we found that it exhibits the Be phenomenon and fits the definition of an FS ...CMa type object. The optical spectrum shows the presence of a B-type star with the following properties: Teff = 11,000 500 K, log L/L = 3.7 0.3, a mass of 7 1 M , and a distance D = 2.4 0.3 kpc from the Sun. We detected regular radial velocity variations of both absorption and emission lines with the following orbital parameters: Porb =131.27 0.09 days, semiamplitude K1 = 51.7 3.0 km s−1, systemic radial velocity γ = −31.8 2.6 km s−1, and a mass function of f(m) = 1.9 0.3 M . AS 386 exhibits irregular variations of the optical brightness (V = 10.92 0.05 mag), while the near-IR brightness varies up to ∼0.3 mag following the spectroscopic period. We explain this behavior by a variable illumination of the dusty disk inner rim by the B-type component. Doppler tomography based on the orbital variations of emission-line profiles shows that the material is distributed near the B-type component and in a circumbinary disk. We conclude that the system has undergone a strong mass transfer that created the circumstellar material and increased the B-type component mass. The absence of any traces of a secondary component, whose mass should be ≥7 M , suggests that it is most likely a black hole.
Accretion is a fundamental process which establishes the dynamics of the protoplanetary disk and the final properties of the forming star. In solar-type stars, the star-disk coupling is determined by ...the magnetic field structure, which is responsible for funneling material from the disk midplane to higher latitudes on the star. Here, we use pan-chromatic data for the Herbig Ae star MWC 480 to address whether similar processes occur in intermediate-mass stars. MWC 480 has X-ray emission typical of actively accreting Herbig Ae stars, but with {approx}10x more photoelectric absorption than expected from optical and FUV data. We consider three sources for the absorption: the disk, absorption in a wind or jet, and accretion. While we detect the disk in scattered light in a re-analysis of archival Hubble Space Telescope data, the data are consistent with grazing illumination of the dust disk. We find that MWC 480's disk is stratified, geometrically thin, and is not responsible for the observed photoelectric absorption. MWC 480 drives a bipolar jet, but with a mass-loss rate that is low compared to other Herbig Ae stars, where the outflow is more favorably oriented and enhanced photoelectric absorption is not seen. This excludes a jet or wind origin for the enhanced photoelectric absorption. We compare MWC 480's O VI emission with other Herbig Ae stars. The distribution of the emission in inclination, and lack of a correlation of profile shape and system inclination excludes equatorially confined accretion for the FUSE Herbig Ae stars. The photoelectric absorption data further suggest that the accretion footprint on MWC 480 and other Herbig Ae stars is located at high-temperate, rather than polar, latitudes. These findings support the presence of funneled accretion in MWC 480 and Herbig Ae stars, strengthening the parallel to T Tauri stars.
Background Linezolid is an oxazolidinone antibiotic that is increasingly used to treat drug-resistant, gram-positive pathogens. The mechanism of action is inhibition of bacterial protein synthesis. ...Optic and/or peripheral neuropathy and lactic acidosis are reported side effects, but the underlying pathophysiological mechanism has not been unravelled. Methods. We studied mitochondrial ultrastructure, mitochondrial respiratory chain enzyme activity, and mitochondrial DNA (mtDNA) in muscle, liver, and kidney samples obtained from a patient who developed optic neuropathy, encephalopathy, skeletal myopathy, lactic acidosis, and renal failure after prolonged use of linezolid. In addition, we evaluated mtDNA, respiratory chain enzyme activity, and protein amount in muscle and liver samples obtained from experimental animals that received linezolid or placebo. Results. In the patient, mitochondrial respiratory chain enzyme activity was decreased in affected tissues, without ultrastructural mitochondrial abnormalities and without mutations or depletion of mtDNA. In the experimental animals, linezolid induced a dose- and time-dependent decrease of the activity of respiratory chain complexes containing mtDNA-encoded subunits and a decreased amount of protein of these complexes, whereas the amount of mtDNA was normal. Conclusion. These results provide direct evidence that linezolid inhibits mitochondrial protein synthesis with potentially severe clinical consequences. Prolonged courses of linezolid should be avoided if alternative treatment options are available.
