Abstract
We analyse Chandra X-ray Observatory observations of a set of galaxy clusters selected by the South Pole Telescope using a new publicly available forward-modelling projection code, mbproj2, ...assuming hydrostatic equilibrium. By fitting a power law plus constant entropy model we find no evidence for a central entropy floor in the lowest entropy systems. A model of the underlying central entropy distribution shows a narrow peak close to zero entropy which accounts for 60 per cent of the systems, and a second broader peak around $130 \rm \, keV\rm \, cm^{2}$. We look for evolution over the 0.28–1.2 redshift range of the sample in density, pressure, entropy and cooling time at 0.015R500 and at 10 kpc radius. By modelling the evolution of the central quantities with a simple model, we find no evidence for a non-zero slope with redshift. In addition, a non-parametric sliding median shows no significant change. The fraction of cool-core clusters with central cooling times below 2 Gyr is consistent above and below z = 0.6 (∼30–40 per cent). Both by comparing the median thermodynamic profiles, centrally biased towards cool cores, in two redshift bins, and by modelling the evolution of the unbiased average profile as a function of redshift, we find no significant evolution beyond self-similar scaling in any of our examined quantities. Our average modelled radial density, entropy and cooling-time profiles appear as power laws with breaks around 0.2R500. The dispersion in these quantities rises inwards of this radius to around 0.4 dex, although some of this scatter can be fitted by a bimodal model.
We consider the problem of self-regulated heating and cooling in galaxy clusters and the implications for cluster magnetic fields and turbulence. Viscous heating of a weakly collisional magnetized ...plasma is regulated by the pressure anisotropy with respect to the local direction of the magnetic field. The intracluster medium is a high-beta plasma, where pressure anisotropies caused by the turbulent stresses and the consequent local changes in the magnetic field will trigger very fast microscale instabilities. We argue that the net effect of these instabilities will be to pin the pressure anisotropies at a marginal level, controlled by the plasma beta parameter. This gives rise to local heating rates that turn out to be comparable to the radiative cooling rates. Furthermore, we show that a balance between this heating and bremsstrahlung cooling is thermally stable, unlike the often conjectured balance between cooling and thermal conduction. Given a sufficient (and probably self-regulating) supply of turbulent power, this provides a physical mechanism for mitigating cooling flows and preventing cluster core collapse. For observed density and temperature profiles, the assumed balance of viscous heating and radiative cooling allows us to predict magnetic field strengths, turbulent velocities and turbulence scales as functions of distance from the centre. Specific predictions and comparisons with observations are given for several different clusters. Our predictions can be further tested by future observations of cluster magnetic fields and turbulent velocities.

Additionally, many small molecules, e.g. 991, have been identified that bind between the kinase domain and the carbohydrate-binding module of the β subunit, stabilising their interaction and leading to activation. It was reported recently that non-phosphorylated Thr
AMPK is activated by AMP and A769662. We present here the crystal structure of non-phosphorylated Thr
AMPK in complex with AMP and 991. This structure reveals that the activation loop, as well as the complex overall, is similar to the Thr
phosphorylated complex. We find that in the presence of AMP and 991 non-phosphorylated Thr
, AMPK is much less active than the Thr
phosphorylated enzyme. In human cells, the basal level of Thr
phosphorylation is very low (∼1%), but is increased 10-fold by treatment with 2-deoxyglucose. In cells lacking the major Thr
kinases, LKB1 and CaMKKβ, Thr
phosphorylation is almost completely abolished, and AMPK activity is virtually undetectable. Our data show that AMP and 991 binding to non-phosphorylated Thr
AMPK can induce an ordered, active-like, conformation of the activation loop explaining how AMPK activity can be measured
without Thr
phosphorylation. However, in a cellular context, phosphorylation of Thr
is critical for significant activation of AMPK.
ABSTRACT
We study the late-time evolution of the central regions of two Milky Way (MW)-like simulations of galaxies formed in a cosmological context, one hosting a fast bar and the other a slow one. ...We find that bar length, Rb, measurements fluctuate on a dynamical time-scale by up to 100 per cent, depending on the spiral structure strength and measurement threshold. The bar amplitude oscillates by about 15 per cent, correlating with Rb. The Tremaine–Weinberg method estimates of the bars’ instantaneous pattern speeds show variations around the mean of up to $\sim \!20{{\ \rm per\ cent}}$, typically anticorrelating with the bar length and strength. Through power spectrum analyses, we establish that these bar pulsations, with a period in the range ∼60–200 Myr, result from its interaction with multiple spiral modes, which are coupled with the bar. Because of the presence of odd spiral modes, the two bar halves typically do not connect at exactly the same time to a spiral arm, and their individual lengths can be significantly offset. We estimated that in about 50 per cent of bar measurements in MW-mass external galaxies, the bar lengths of SBab-type galaxies are overestimated by $\sim \!15{{\ \rm per\ cent}}$ and those of SBbc types by $\sim \!55{{\ \rm per\ cent}}$. Consequently, bars longer than their corotation radius reported in the literature, dubbed ‘ultrafast bars’, may simply correspond to the largest biases. Given that the Scutum–Centaurus arm is likely connected to the near half of the MW bar, recent direct measurements may be overestimating its length by 1–1.5 kpc, while its present pattern speed may be 5–10 $\rm km\ s^{-1}\ kpc^{-1}$ smaller than its time-averaged value.
Metronidazole was until recently used as a first-line treatment for potentially life-threatening Clostridioides difficile (CD) infection. Although cases of metronidazole resistance have been ...documented, no clear mechanism for metronidazole resistance or a role for plasmids in antimicrobial resistance has been described for CD. Here, we report genome sequences of seven susceptible and sixteen resistant CD isolates from human and animal sources, including isolates from a patient with recurrent CD infection by a PCR ribotype (RT) 020 strain, which developed resistance to metronidazole over the course of treatment (minimal inhibitory concentration MIC = 8 mg L
). Metronidazole resistance correlates with the presence of a 7-kb plasmid, pCD-METRO. pCD-METRO is present in toxigenic and non-toxigenic resistant (n = 23), but not susceptible (n = 563), isolates from multiple countries. Introduction of a pCD-METRO-derived vector into a susceptible strain increases the MIC 25-fold. Our finding of plasmid-mediated resistance can impact diagnostics and treatment of CD infections.
AMP-activated protein kinase (AMPK) is the downstream component of a protein kinase cascade that plays a major role in maintaining energy homoeostasis. Within individual cells, AMPK is activated by a ...rise in the AMP/ATP ratio that occurs following a fall in ATP levels. AMPK is also regulated by the adipokines, adiponectin and leptin, hormones that are secreted from adipocytes. AMPK regulates a wide range of metabolic pathways, including fatty acid oxidation, fatty acid synthesis, glycolysis and gluconeogenesis. In peripheral tissues, activation of AMPK leads to responses that are beneficial in counteracting the deleterious effects that arise in the metabolic syndrome. Recent studies have demonstrated that modulation of AMPK activity in the hypothalamus plays a role in feeding. A decrease in hypothalamic AMPK activity is associated with decreased feeding, whereas activation of AMPK leads to increased food intake. Furthermore, signalling pathways occurring in the hypothalamus lead to changes in AMPK activity in peripheral tissues, such as skeletal muscle, via the sympathetic nervous system. AMPK, therefore, provides a mechanism for monitoring changes in energy metabolism within individual cells and at the level of the whole body. Activation of AMPK requires phosphorylation of threonine 172 (Thr-172) within the catalytic subunit. Recent studies have shown that both LKB1 and Ca2+/calmodulin-dependent protein kinase kinase- (CaMKK) play important roles in phosphorylating and activating AMPK. In addition, there is evidence that AMPK can be activated by other upstream kinases, although the physiological significance of this is not clear at present. This review focuses on the role of LKB1 and CaMKK in the regulation of AMPK.
The effects of different thermo-chemical pre-treatment methods were determined on the biodegradability and hydrolysis rate of lignocellulosic biomass. Three plant species, hay, straw and bracken were ...thermo-chemically pre-treated with calcium hydroxide, ammonium carbonate and maleic acid. After pre-treatment, the plant material was anaerobically digested in batch bottles under mesophilic conditions for 40 days. From the pre-treatment and subsequent anaerobic digestion experiments, it was concluded that when the lignin content of the plant material is high, thermo-chemical pre-treatments have a positive effect on the biodegradability of the substrate. Calcium hydroxide pre-treatment improves the biodegradability of lignocellulosic biomass, especially for high lignin content substrates, like bracken. Maleic acid generates the highest percentage of dissolved COD during pre-treatment. Ammonium pre-treatment only showed a clear effect on biodegradability for straw.

In 1967, Dan Janzen published “Why Mountain Passes Are Higher in the Tropics” in The American Naturalist. Janzen’s seminal article has captured the attention of generations of biologists and ...continues to inspire theoretical and empirical work. The underlying assumptions and derived predictions are broadly synthetic and widely applicable. Consequently, Janzen’s “seasonality hypothesis” has proven relevant to physiology, climate change, ecology, and evolution. To celebrate the fiftieth anniversary of this highly influential article, we highlight the past, present, and future of this work and include a unique historical perspective from Janzen himself.
AMP-activated protein kinase (AMPK) plays a major role in regulating cellular energy balance by sensing and responding to increases in AMP/ADP concentration relative to ATP. Binding of AMP causes ...allosteric activation of the enzyme and binding of either AMP or ADP promotes and maintains the phosphorylation of threonine 172 within the activation loop of the kinase. AMPK has attracted widespread interest as a potential therapeutic target for metabolic diseases including type 2 diabetes and, more recently, cancer. A number of direct AMPK activators have been reported as having beneficial effects in treating metabolic diseases, but there has been no structural basis for activator binding to AMPK. Here we present the crystal structure of human AMPK in complex with a small molecule activator that binds at a site between the kinase domain and the carbohydrate-binding module, stabilising the interaction between these two components. The nature of the activator-binding pocket suggests the involvement of an additional, as yet unidentified, metabolite in the physiological regulation of AMPK. Importantly, the structure offers new opportunities for the design of small molecule activators of AMPK for treatment of metabolic disorders.
The blue carbon paradigm has evolved in recognition of the high carbon storage and sequestration potential of mangrove, saltmarsh and seagrass ecosystems. However, fluxes of the potent greenhouse ...gases CH4 and N2O, and lateral export of carbon are often overlooked within the blue carbon framework. Here, we show that the export of dissolved inorganic carbon (DIC) and alkalinity is approximately 1.7 times higher than burial as a long-term carbon sink in a subtropical mangrove system. Fluxes of methane offset burial by approximately 6%, while the nitrous oxide sink was approximately 0.5% of burial. Export of dissolved organic carbon and particulate organic carbon to the coastal zone is also significant and combined may account for an atmospheric carbon sink similar to burial. Our results indicate that the export of DIC and alkalinity results in a long-term atmospheric carbon sink and should be incorporated into the blue carbon paradigm when assessing the role of these habitats in sequestering carbon and mitigating climate change.
