Objectives
The purpose of this study was to investigate the effect of photo‐induced hydrophilic titanium dioxide (TiO2) on serum fibronectin (sFN) attachment, and further to evaluate initial ...osseointegration responses in the dog mandibles.
Materials and methods
To apply the anatase TiO2 film, plasma source ion implantation (PSII) method followed by annealing was employed for the titanium disks and implants, which were then illuminated with UV‐A for 24 h for the experimental groups. Non‐deposited titanium disks and implants were prepared for the control group. Surface characterization was performed using the interferometer and contact angle analyzer. The attachments of sFN were evaluated using fluorescence emission analysis. Thereafter both groups of implants were placed in the mandible of six beagle dogs. Bone response was investigated with histological and histomorphometrical analyses after periods of 2 and 4 weeks.
Results
The experimental groups exhibited strong hydrophilicity under UV‐A illumination and showed significant improvement in sFN attachment. And further, the experimental implants enhanced the bone formation with the bone‐to‐implant contact of 42.7% after 2 weeks of healing (control: 28.4%).
Conclusions
The combined applications of plasma fibronectin and PSII to produce hydrophilic titanium surfaces could accelerate early osseointegration.
This study aimed to evaluate the effects of chemical conditioning and self-adhesive resins (SARs) on the bonding of mechanically conditioned high-strength composite resin block (HSCRB).
Eighteen ...sections of HSCRB (KZR-CAD HR 3 Gammatheta, Yamakin) were treated with alumina air abrasion and randomly divided into 3 groups according to the SARs for bonding: RelyX Unicem 2 (RXU), SA Luting Plus (SAL), and G-Cem ONE (GCO). The sections were further divided into 3 subgroups according to the chemical conditioning of the adherend surfaces: no conditioning (C), universal adhesive (UA), and a mixture of γ-MPTS and 10-MDP (MM). After the surface conditioning, the sections were cemented with the SARs. Each cemented section was cut into 40 beams. Half of the beams were thermocycled (4°C/60°C, 10,000 cycles). The micro-tensile bond strength (μTBS) values were measured using a universal testing machine. Scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDS), contact angles, and surface roughness measurements were performed on the adherend surfaces of each subgroup.
RXU showed the highest μTBS values among the 3 SARs tested, while MM application exhibited the highest μTBS values among the 3 chemical conditioning methods tested. After thermocycling, the samples in the RXU/MM, RXU/UA, and GCO/MM groups showed no significant changes in the μTBS values, whereas the others showed a significant reduction.
The bond strength of HSCRB was influenced by the chemical conditioning, SARs, and aging. γ-MPTS and 10-MDP application yielded higher μTBS values of mechanically treated HSCRB than the UA.
Definitive treatment strategies for bisphosphonate-related osteonecrosis of the jaw (BRONJ) have not been developed. Cell-based therapy is an attractive treatment method for intractable diseases in ...the medical and dental fields; however, approval has been challenging in dentistry. Recently, we developed quality- and quantity (QQ)-controlled peripheral blood mononuclear cells (PBMNCs) that have anti-inflammatory and pro-angiogenesis effects. The aim of this study was to investigate the effects of QQ-controlled PBMNC transplantation on BRONJ-like lesions in mice.
To create high-prevalence BRONJ-like lesions, cyclophosphamide (CY) and zoledronate (ZA) were used with tooth extraction. Drug treatment was performed for 5 weeks. QQ-controlled PBMNC transplantation was performed immediately following tooth extraction of both maxillary first molars at 3 weeks after drug administration. Mice were euthanized at 2 weeks post-extraction. Histomorphometric and immunohistochemical analyses, microcomputed tomography assessment, and quantitative polymerase chain reaction evaluation were conducted using maxillae and long bones.
ZA effects on long bones were noted, regardless of CY. Severely inhibited osseous and soft tissue wound healing of tooth extraction sockets was induced by CY/ZA combination therapy, which was diagnosed as BRONJ-like lesions. QQ-controlled PBMNC transplantation reduced BRONJ-like lesions by improving soft tissue healing with increased M1 and M2 macrophages and enhanced neovascularization in the connective tissue of tooth extraction sockets. QQ-controlled PBMNC transplantation also reduced inflammation by decreasing polymorphonuclear cells and TNF-α expression in the tooth extraction sockets. Additionally, QQ-controlled PBMNC transplantation partially improved osseous healing of tooth extraction sockets. Interestingly, only 20,000 QQ-controlled PBMNCs per mouse induced these transplantation effects. QQ-controlled PBMNC transplantation did not affect the systemic microenvironment.
Our findings suggest that transplantation of a small amount of QQ-controlled PBMNCs may become novel therapeutic or prevention strategies for BRONJ without any adverse side effects.
In the present study, sol–gel derived nanoparticle calcium silicate bioactive glass was added to the resin-modified light cure glass-ionomer cement to assess the influence of additional bioactive ...glass nanoparticles on the mechanical and biological properties of resin-modified glass-ionomer cement. The fabricated bioactive glass nanoparticles added resin-modified glass-ionomer cements (GICs) were immersed in the phosphate buffer solution for 28 days to mimic real condition for the mechanical properties. Resin-modified GICs containing 3, 5 and 10 % bioactive glass nanoparticles improved the flexural strength compared to the resin-modified glass-ionomer cement and the samples containing 15 and 20 % bioactive glass nanoparticles before and after immersing in the phosphate buffer solution. Characterization of the samples successfully expressed the cause of the critical condition for mechanical properties. Cell study clarified that resin-modified glass-ionomer cement with high concentrations of bioactive glass nanoparticles has higher cell viability and better cell morphology compare to control groups. The results for mechanical properties and toxicity approved that the considering in selection of an optimum condition would have been a more satisfying conclusion for this study.
Background
Bone mechanical function is regulated by bone quality and bone mineral density (BMD) that reflect bone strength. The preferential alignment of biological apatite (BAp) c‐axis/collagen ...fibers and osteocytes is a determinant factor of bone quality. However, the effect of mechanical loading on bone quality around dental implants is unclear.
Purpose
The aim of this study was to clarify the effects of mechanical loading on osseointegration, bone volume BMD, and bone quality around dental implants.
Materials and Methods
Twenty anodized Ti‐6Al‐4V alloy implants (KYOCERA Co., Kyoto, Japan) were placed in the proximal tibial metaphysis of 10 rabbits. Twelve weeks after surgery, mechanical loading was applied along the long axis of the implant (50 N, 3 Hz, 1,800 cycles, 2 days/week) for 8 weeks. Osseointegration, bone volume, BMD, and bone quality were evaluated using light microscopy, microcomputed tomography, polarized light microscopy, and microbeam X‐ray diffractometer.
Results
Mechanical loading increased osseointegration, bone volume, and BMD. Bone quality around dental implant was altered with increased osteocyte numbers and the preferential alignment direction and degree of BAp c‐axis/collagen fibers.
Conclusions
These findings suggest that mechanical loading effectively induces bone anabolic responses around dental implants. Altered bone quality may upregulate bone strength, contributing to long‐term implant stability.
Objective The objective of this study was to evaluate the effects of combinations of silane primers and adhesive agents on the bond strength of a composite block for a computer-aided design and ...manufacturing system. Material and Methods Three silane primers Clearfil Ceramic Primer (CP), Super-Bond PZ Primer (PZ), and GC Ceramic Primer II (GP) were used in conjunction with five adhesive agents G-Premio Bond (P-Bond), Repair Adhe Adhesive (R-Adhesive), Super-Bond D-Liner Dual (SB-Dual), Super-Bond C&B (SB-Self), and SB-Dual without tributylborane derivative (SB-Light). The surface of a composite block (Gradia Block) was ground with silicon carbide paper. After treatment with a silane primer, a adhesive agent was applied to each testing specimen. The specimens were then bonded with a light-curing resin composite. After 24 h, the shear bond strength values were determined and compared using a post hoc test (α=0.05, n=8/group). We also prepared control specimens without primer (No primer) and/or without adhesive agent (No adhesive). Results PZ/SB-Dual and GP/SB-Dual presented the highest bond strength, followed by GP/P-Bond, CP/SB-Dual, CP/R-Adhesive, No primer/SB-Dual, GP/R-Adhesive, CP/P-Bond, No primer/R-Adhesive, PZ/R-Adhesive, CP/SB-Self, PZ/P-Bond, PZ/SB-Self, and GP/SB-Self in descending order of bond strength. No primer/P-Bond, No primer/SB-Self, and all specimens in the SB-Light and No adhesive groups presented the lowest bond strengths. Conclusion A dual-curing adhesive agent (SB-Dual) containing a tributylborane derivative in combination with a silane primer (GP or PZ) presents a greater bond strength between the composite block and the repairing resin composite than the comparators used in the study.
Purpose: This systematic review aimed to evaluate the effects of implant placement sites on the biomechanical behavior of implant-assisted removable partial dentures (IARPDs) using finite element ...analysis (FEA).Study selection: Two reviewers independently conducted manual searches of the PubMed, Scopus, and ProQuest databases for articles investigating implant location in IARPDs using FEA, according to the 2020 Systematic Reviews and Meta-analyses statement. Studies published in English up to August 1, 2022, were included in the analysis based on the critical question.Results: Seven articles meeting the inclusion criteria were systematically reviewed. Six studies investigated mandibular Kennedy Class I and one study investigated mandibular Kennedy Class II. Implant placement reduced the displacement and stress distribution of the IARPD components, including dental implants and abutment teeth, regardless of the Kennedy Class type and dental implant placement site. Most of the included studies showed that, based on the biomechanical behavior, the molar region, rather than the premolar region, is the preferred implant placement site. None of the selected studies investigated the maxillary Kennedy Class I and II.Conclusions: Based on the FEA regarding mandibular IARPDs, we concluded that implant placement in both the premolar and molar regions improves the biomechanical behaviors of IARPD components, regardless of the Kennedy Class. Implant placement in the molar region results in more suitable biomechanical behaviors compared with implant placement in the premolar region in Kennedy Class I. No conclusion was reached for Kennedy Class II due to the lack of relevant studies.
Purpose: This scoping review aimed to systematically map research regarding implant-assisted removable partial dentures (IARPDs), and identify existing gaps in knowledge.Study selection: Two ...reviewers independently conducted a search of the MEDLINE-PubMed and Scopus databases according to the Preferred Reporting Items for Systematic Review and Meta-Analyses (PRISMA) extension for Scoping Review and included articles published in English up to August 31, 2022, including human studies, reviews, and in vitro studies. Expert opinions, animal studies, and clinical studies involving complete overdentures were excluded, and ten aspects for establishing the treatment strategy for IARPDs were examined.Results: One hundred and twelve articles were chosen. There were two treatment modalities: IARPDs retained by implant- and tooth-supported surveyed single crowns (SCs) or fixed partial dentures (FPDs). In IARPDs retained by tooth-supported surveyed SCs or FPDs, the survival rate of dental implants for IARPDs was relatively higher with a wide range of marginal bone loss and many complications, but with improved functional performance, oral health-related quality of life, and patient satisfaction. There were limited data on survival or success rates and designs of IARPDs, attachment selections, length and diameter, inclination, placement sites, and loading protocols of implants, regardless of prosthetic types. There was limited information on maxillary IARPDs except for survival rates of implants.Conclusions: Although IARPDs could become a useful treatment strategy, there is limited scientific consensus with gaps in knowledge about their use. Additional well-designed clinical and in vitro studies are necessary to scientifically establish IARPDs as definitive prostheses in implant dentistry.
The aim of this study was to develop an effective method for cleaning implant abutments made by computer-aided design and computer-aided manufacturing techniques and to investigate the effect of ...decontamination in vitro. Briefly, a newly developed reagent (PK) and/or vacuum plasma (Plasma) were used to clean the surfaces of zirconia disks, and the effects of this decontamination were evaluated by X-ray photoelectron spectroscopy. Human gingival fibroblasts (HGFs) were cultured on sample disks for 6, 24, and 48 h. We evaluated cell attachment and gene expression of the acute inflammatory cytokines interleukin-6 and vascular endothelial growth factor A, and type 1 collagen. In the PK and PK+Plasma groups, surface contaminants were reduced by washing. In addition, HGF attachments was increased in the PK and PK+Plasma groups. Gene expressions of interleukin-6 and vascular endothelial growth factor A were lower at 6 h. Gene expression of type 1 collagen was increased at all time points after seeding. These results suggest that decontamination of implant abutment surfaces is important in initial HGF attachment and may improve the biological seal of peri-implant soft tissue.
Purpose The aim of the present study was to investigate the effects of chemotherapeutic/bisphosphonate combination therapy with tooth extraction on gene expression patterns of fresh extraction wounds ...during initial stages prior to their diagnosis as bisphosphonate-related osteonecrosis of the jaw (BRONJ)-like lesions in mice.Methods Female C57BL/6J mice were used. To create a high-prevalence BRONJ mouse model, combination therapy with the chemotherapy drug cyclophosphamide (CY) and zoledronic acid (ZA) was performed (CY/ZA). Both maxillary first molars were extracted 3 weeks after drug therapy. Saline was used as the control (VC). Soft tissues near the fresh extraction wounds were dissected at 72 h postextraction to investigate the gene expression patterns. Maxillae and long bones at 2 and 4 weeks postextraction were also analyzed.Results CY/ZA significantly increased the relative expression levels of IL-6 and decreased those of IL-10 and IGF-1 when compared with those in VC. Moreover, CY/ZA significantly reduced the relative expression levels of CCR-7, cxcl12, cxcr4, and CD105 when compared with those in VC, whereas the level of F4/80 was significantly increased by CY/ZA. Furthermore, CY/ZA significantly decreased the relative expression levels of VEGFA, VEGFB, and VEGFC at 72 h postextraction compared with those in VC.Conclusions Considering that the present study lacked adequate in vitro models, CY/ZA markedly changed the gene expression patterns associated with wound healing from the initial stages prior to onset of BRONJ-like lesions, which may help us to understand the pathophysiology of BRONJ in humans.
