C57BL/6 mice have attenuated allergic airway hyperresponsiveness (AHR) when compared with Balb/c mice but the underlying mechanisms remain unclear. SP-D, an innate immune molecule with potent ...immunosuppressive activities may have an important modulatory role in the allergic airway response and the consequent physiological changes. We hypothesized that an elevated SP-D production is associated with the impaired ability of C57BL/6 mice to develop allergic AHR.
SP-D mRNA and protein expression was investigated during development of allergic airway changes in a model of Aspergillus fumigatus (Af)-induced allergic inflammation. To study whether strain dependency of allergic AHR is associated with different levels of SP-D in the lung, Balb/c and C57BL/6 mice were compared.
Sensitization and exposure to Af induced significant airway inflammation in both mouse strains in comparison with naïve controls. AHR to acetylcholine however was significantly attenuated in C57BL/6 mice in spite of increased eosinophilia and serum IgE when compared with Balb/c mice (p < 0.05). Af challenge of sensitized C57BL/6 mice induced a markedly increased SP-D protein expression in the SA surfactant fraction (1,894 +/- 170% of naïve controls) that was 1.5 fold greater than the increase in Balb/c mice (1,234 +/- 121% p < 0.01). These changes were selective since levels of the hydrophobic SP-B and SP-C and the hydrophilic SP-A were significantly decreased following sensitization and challenge with Af in both strains. Further, sensitized and exposed C57BL/6 mice had significantly lower IL-4 and IL-5 in the BAL fluid than that of Balb/c mice (p < 0.05).
These results suggest that enhanced SP-D production in the lung of C57BL/6 mice may contribute to an attenuated AHR in response to allergic airway sensitization. SP-D may act by inhibiting synthesis of Th2 cytokines.

Humans with primary biliary cirrhosis (PBC), a disease characterized by the destruction of small bile ducts, exhibit signature autoantibodies against mitochondrial Pyruvate Dehydrogenase Complex E2 ...(PDC-E2) that crossreact onto the homologous enzyme of Novosphingobium aromaticivorans, an ubiquitous alphaproteobacterium. Here, we show that infection of mice with N. aromaticivorans induced signature antibodies against microbial PDC-E2 and its mitochondrial counterpart but also triggered chronic T cell-mediated autoimmunity against small bile ducts. Disease induction required NKT cells, which specifically respond to N. aromaticivorans cell wall alpha-glycuronosylceramides presented by CD1d molecules. Combined with the natural liver tropism of NKT cells, the accumulation of N. aromaticivorans in the liver likely explains the liver specificity of destructive responses. Once established, liver disease could be adoptively transferred by T cells independently of NKT cells and microbes, illustrating the importance of early microbial activation of NKT cells in the initiation of autonomous, organ-specific autoimmunity.
Although extremely rare, rupture of an unscarred gravid uterus poses significant morbidity and mortality to both fetus and mother. In the past, loss of uterine contraction was thought to be ...characteristic of uterine rupture, while recent evidence shows that uterine contraction pattern is not associated with uterine rupture. We report two cases of rupture in the unscarred term uterus. Both patients were multiparous and denied any past medical complications, previous cesarean delivery or myomectomy. Uterine rupture occurred in the latent phase of labor at 1 cm dilation (Case 1) and during an intravenous oxytocin infusion after three doses of intravaginal misoprostol (Case 2). Case 2 required cesarean hysterectomy and blood transfusion. Case 1 was monitored with an external tocodynamometer, while Case 2 was monitored with an internal pressure transducer. External monitoring demonstrated the classic sign of complete loss of uterine tone. In contrast, internal monitoring demonstrated an increase in uterine resting tone. Both techniques revealed a stepwise gradual decrease in contraction amplitude followed by sudden onset of profound and prolonged fetal bradycardia (staircase sign). In cases of uterine rupture, differing baseline characteristics between contraction patterns were dependant on uterine monitoring technique. In both techniques a stepwise gradual decrease in contraction amplitude was followed by prolonged fetal bradycardia.
An eddy covariance (EC) system with a tunable diode laser trace gas analyzer was used in a field setting in Ireland to measure N2O emissions on a continuous basis over an eight‐month period, spanning ...a range of seasonal conditions. Intensely‐grazed grassland fields within the footprint area of the EC sensors were subject to chemical fertilizer and slurry applications in order to boost grassland yield, and the amounts of these applications were documented by the farmers on a monthly basis. Three major emission events, covering a timeframe of 16 days (6.6% of the measurement period) contributed to over half (51.1%) of the observed cumulative flux. Two of these events occurred during the summer, while the third occurred during the winter, with vastly different soil moisture and soil temperature conditions associated with these times of the year. The type of N applications (fertilizer vs. slurry), soil moisture and temperature status had implications for controlling the short‐term rates of N2O emissions. Cumulative N2O emissions, however, were driven by fertilizer and slurry N applications, as the emission factor of approximately 3.0% displayed consistency throughout the eight‐month period.
Pasture decline is considered to be a serious challenge to agricultural productivity of subterranean clover across southern Australia. Root disease is a significant contributing factor to pasture ...decline. However, root disease assessments are generally carried out in the early part of the growing season and in areas predominantly sown to permanent pastures. For this reason, in spring 2004, a survey was undertaken to determine the severity of root disease in mature subterranean clover plants in pastures located in the wheatbelt of Western Australia. DNA-based soil assays were used to estimate population density in the soil of a variety of soil-borne pathogens known to commonly occur in the Mediterranean-type environments of southern Australia. The relationships between severity of disease on tap and lateral roots and root diameter, root length, nodulation, and total rainfall were determined. The survey showed, for the first time, that severe root disease is widespread in spring across the wheatbelt of Western Australia. There was a positive correlation between rainfall and tap root disease, and between tap root disease and average root diameter of the entire root system. Despite the high levels of root disease present across the sites, the DNA of most root disease pathogens assayed was detected in trace concentrations. Only Pythium Clade F showed high DNA concentrations in the soil. DNA concentrations in the soil, in particular for Phytophthora clandestina and Rhizoctonia solani AG 2.1 and AG 2.2, were higher in the smaller autumn sampling in 2006. This study suggests that the productivity of subterranean clover-based pastures is severely compromised by root rot diseases throughout the growing season in the wheatbelt of Western Australia.

The phenomenon of street children in Latin America is examined, and some answers to commonly asked questions are provided. The term "street children" refers to the homeless, abandoned or runaways.
Since the late 1970s, space-borne microwave radiometers have been providing measurements of radiation emitted by the Earth’s surface. From these measurements it is possible to derive vegetation ...optical depth (VOD), a model-based indicator related to the density, biomass, and water content of vegetation. Because of its high temporal resolution and long availability, VOD can be used to monitor short- to long-term changes in vegetation. However, studying long-term VOD dynamics is generally hampered by the relatively short time span covered by the individual microwave sensors. This can potentially be overcome by merging multiple VOD products into a single climate data record. However, combining multiple sensors into a single product is challenging as systematic differences between input products like biases, different temporal and spatial resolutions, and coverage need to be overcome. Here, we present a new series of long-term VOD products, the VOD Climate Archive (VODCA). VODCA combines VOD retrievals that have been derived from multiple sensors (SSM/I, TMI, AMSR-E, WindSat, and AMSR2) using the Land Parameter Retrieval Model. We produce separate VOD products for microwave observations in different spectral bands, namely the Ku-band (period 1987–2017), X-band (1997–2018), and C-band (2002–2018). In this way, our multi-band VOD products preserve the unique characteristics of each frequency with respect to the structural elements of the canopy. Our merging approach builds on an existing approach that is used to merge satellite products of surface soil moisture: first, the data sets are co-calibrated via cumulative distribution function matching using AMSR-E as the scaling reference. To do so, we apply a new matching technique that scales outliers more robustly than ordinary piecewise linear interpolation. Second, we aggregate the data sets by taking the arithmetic mean between temporally overlapping observations of the scaled data. The characteristics of VODCA are assessed for self-consistency and against other products. Using an autocorrelation analysis, we show that the merging of the multiple data sets successfully reduces the random error compared to the input data sets. Spatio-temporal patterns and anomalies of the merged products show consistency between frequencies and with leaf area index observations from the MODIS instrument as well as with Vegetation Continuous Fields from the AVHRR instruments. Long-term trends in Ku-band VODCA show that since 1987 there has been a decline in VOD in the tropics and in large parts of east-central and north Asia, while a substantial increase is observed in India, large parts of Australia, southern Africa, southeastern China, and central North America. In summary, VODCA shows vast potential for monitoring spatial–temporal ecosystem changes as it is sensitive to vegetation water content and unaffected by cloud cover or high sun zenith angles. As such, it complements existing long-term optical indices of greenness and leaf area. The effect of flow distortion on the measurements produced by a LiDAR or SoDAR in close proximity to either complex terrain or a structure creating localised flow distortion is difficult to determine ...by analytical means. Also, as LiDARs and SoDARs are not point measurement devices, the techniques they employ for velocity measurements leads to complexities in the estimation of the effect of flow distortion on the accuracy of the measurements they make. This paper presents a method by which the effect of flow distortion on measurements made by a LiDAR in a distorted flow field may be determined using computational fluid dynamics. The results show that the error created by the flow distortion will cause the vector measured by a LiDAR to differ significantly from an equivalent point measurement. However, the results of the simulation show that, if the LiDAR is being used to measure the undisturbed flow field above a structure which creates highly localised flow distortion, the LiDAR results are less affected by the distortion of the local flow field than data acquired by a point measurement technique such as a cup anemometer.

The eddy covariance method is regularly used for measuring gas fluxes over agricultural fields and natural ecosystems. For many applications, it is desirable to partition the measured fluxes into constitutive components: the water vapor flux into transpiration and direct evaporation components, and the carbon dioxide flux into photosynthesis and respiration components. The flux variance similarity (FVS) partitioning method is based on flux variance similarity relationships and correlation analyses of high-frequency eddy covariance data (Scanlon and Sahu, 2008; Scanlon and Kustas, 2010, 2012). The FVS method is relatively complex computationally, and that complexity has likely been an impediment to greater use and testing of the procedure. In this work, we present a new algebraic solution to the key computational task in the partitioning algorithm, which significantly simplifies the FVS method. We also introduce Fluxpart, a free and open source Python 3 module that implements the FVS partitioning procedure. Example flux partitioning calculations are presented.
