Spinal muscular atrophy is a rare, autosomal recessive, neuromuscular disease caused by biallelic loss of the survival motor neuron 1 (SMN1) gene, resulting in motor neuron dysfunction. In this ...STR1VE-EU study, we aimed to evaluate the safety and efficacy of onasemnogene abeparvovec gene replacement therapy in infants with spinal muscular atrophy type 1, using broader eligibility criteria than those used in STR1VE-US.
STR1VE-EU was a multicentre, single-arm, single-dose, open-label phase 3 trial done at nine sites (hospitals and universities) in Italy (n=4), the UK (n=2), Belgium (n=2), and France (n=1). We enrolled patients younger than 6 months (180 days) with spinal muscular atrophy type 1 and the common biallelic pathogenic SMN1 exon 7–8 deletion or point mutations, and one or two copies of SMN2. Patients received a one-time intravenous infusion of onasemnogene abeparvovec (1·1 × 1014 vector genomes vg/kg). The outpatient follow-up consisted of assessments once per week starting at day 7 post-infusion for 4 weeks and then once per month until the end of the study (at age 18 months or early termination). The primary outcome was independent sitting for at least 10 s, as defined by the WHO Multicentre Growth Reference Study, at any visit up to the 18 months of age study visit, measured in the intention-to-treat population. Efficacy was compared with the Pediatric Neuromuscular Clinical Research (PNCR) natural history cohort. This trial is registered with ClinicalTrials.gov, NCT03461289 (completed).
From Aug 16, 2018, to Sept 11, 2020, 41 patients with spinal muscular atrophy were assessed for eligibility. The median age at onasemnogene abeparvovec dosing was 4·1 months (IQR 3·0–5·2). 32 (97%) of 33 patients completed the study and were included in the ITT population (one patient was excluded despite completing the study because of dosing at 181 days). 14 (44%, 97·5% CI 26–100) of 32 patients achieved the primary endpoint of functional independent sitting for at least 10 s at any visit up to the 18 months of age study visit (vs 0 of 23 untreated patients in the PNCR cohort; p<0·0001). 31 (97%, 95% CI 91–100) of 32 patients in the ITT population survived free from permanent ventilatory support at 14 months compared with six (26%, 8–44) of 23 patients in the PNCR natural history cohort (p<0·0001). 32 (97%) of 33 patients had at least one adverse event and six (18%) had adverse events that were considered serious and related to onasemnogene abeparvovec. The most common adverse events were pyrexia (22 67% of 33), upper respiratory infection (11 33%), and increased alanine aminotransferase (nine 27%). One death, unrelated to the study drug, occurred from hypoxic-ischaemic brain damage because of a respiratory tract infection during the study.
STR1VE-EU showed efficacy of onasemnogene abeparvovec in infants with symptomatic spinal muscular atrophy type 1. No new safety signals were identified, but further studies are needed to show long-term safety. The benefit–risk profile of onasemnogene abeparvovec seems favourable for this patient population, including those with severe disease at baseline.

The reversible intercalation of solvated Na-ions into graphite and the concomitant formation of ternary Na–graphite intercalation compounds (GICs) are studied using several in operando techniques, ...such as X-ray-diffraction (XRD), electrochemical scanning tunnelling microscopy (EC-STM) and electrochemical quartz crystal microbalance techniques (EQCM). Linear ethylene glycol dimethyl ether homologues (“glymes”) G x with x + 1 O-atoms were used as solvents, where x is 1–4. The intercalation mechanism of Na + (G x ) y -complexes was investigated with a focus on the phase transitions and diffusion rates of the Na + (G x ) y -complexes inside the graphite lattice. For the four shortest glymes (G 1 to G 4 ), it is found using XRD that an intermediate stage 2 Na–GIC (NaC 48 ) is formed upon the partial sodiation of the graphite electrode. At full sodiation stage 1 Na–GIC (NaC 18 , 112 mA h g −1 ) is obtained for G 1 , G 2 and G 4 , while the G 3 system also forms a stage 1 Na–GIC but with less Na incorporated (NaC 30 , 70 mA h g −1 ). The phase transitions of a battery electrode upon ion-intercalation are visualised using STM on the atomic scale for the first time. In addition, the local diffusion rates of the intercalated species inside the electrode were determined, a unique approach for determining kinetic effects in batteries on the atomic scale. The formation of a solid electrolyte interphase (SEI) is observed with STM as well as with an EQCM, while the latter technique is used for novel in situ hydrodynamic spectroscopy, giving further insight into the intercalation mechanism.

A large-scale Scintillating Fibre Tracker has been developed and constructed for the current LHCb Upgrade 1. The high radiation by fast neutrons reduces the light yield of the detector in the course ...of LHC Run 3, and is challenging the tracker’s hit detection efficiency. To mitigate radiation-induced ageing effects, the central fibre mats and photodetectors will be replaced during the next long shutdown (2026–2028). For this purpose, microlens-enhanced silicon photomultipliers with an improved photon detection efficiency are being developed. The first prototypes have been produced based on the outcome of dedicated simulation studies, and the results of the first light yield measurements are presented.
Allosteric regulation of the essential anaplerotic enzyme, pyruvate carboxylase (PC), is vital for metabolic homeostasis. PC catalyzes the bicarbonate- and ATP-dependent carboxylation of pyruvate to ...form oxaloacetate. Dysregulation of PC activity can impact glucose and redox metabolism, which contributes to the pathogenicity of many diseases. To maintain homeostasis, PC is allosterically activated by acetyl-CoA and allosterically inhibited by l-aspartate. In this study, we further characterize the molecular basis of allosteric regulation in Staphylococcus aureus PC (SaPC) using slowly/nonhydrolyzable dethia analogues of acetyl-CoA and site-directed mutagenesis of residues at the biotin carboxylase homodimer interface. The dethia analogues fully activate SaPC but demonstrate significantly reduced binding affinities relative to acetyl-CoA. Residues Arg21, Lys46, and Glu418 of SaPC are located at the biotin carboxylase dimer interface and play a critical role in both allosteric activation and inhibition. A structure of R21A SaPC in complex with acetyl-CoA reveals an intact molecule of acetyl-CoA bound at the allosteric site, offering new molecular insights into the acetyl-CoA binding site. This study demonstrates that the biotin carboxylase domain dimer interface is a critical allosteric site in PC, serving as a convergence point for allosteric activation by acetyl-CoA and inhibition by l-aspartate.
We report the direct observation of muon neutrino interactions with the SND@LHC detector at the Large Hadron Collider. A dataset of proton-proton collisions at sqrts=13.6 TeV collected by SND@LHC in ...2022 is used, corresponding to an integrated luminosity of 36.8 fb^{-1}. The search is based on information from the active electronic components of the SND@LHC detector, which covers the pseudorapidity region of 7.2<η<8.4, inaccessible to the other experiments at the collider. Muon neutrino candidates are identified through their charged-current interaction topology, with a track propagating through the entire length of the muon detector. After selection cuts, 8 ν_{μ} interaction candidate events remain with an estimated background of 0.086 events, yielding a significance of about 7 standard deviations for the observed ν_{μ} signal.
The Vertex Locator (VELO) is a silicon microstrip detector that surrounds the proton-proton interaction region in the LHCb experiment. The performance of the detector during the first years of its ...physics operation is reviewed. The system is operated in vacuum, uses a bi-phase CO sub(2) cooling system, and the sensors are moved to 7 mm from the LHC beam for physics data taking. The performance and stability of these characteristic features of the detector are described, and details of the material budget are given. The calibration of the timing and the data processing algorithms that are implemented in FPGAs are described. The system performance is fully characterised. The sensors have a signal to noise ratio of approximately 20 and a best hit resolution of 4 mu m is achieved at the optimal track angle. The typical detector occupancy for minimum bias events in standard operating conditions in 2011 is around 0.5%, and the detector has less than 1% of faulty strips. The proximity of the detector to the beam means that the inner regions of the n super(+)-on-n sensors have undergone space-charge sign inversion due to radiation damage. The VELO performance parameters that drive the experiment's physics sensitivity are also given. The track finding efficiency of the VELO is typically above 98% and the modules have been aligned to a precision of 1 mu m for translations in the plane transverse to the beam. A primary vertex resolution of 13 mu m in the transverse plane and 71 mu m along the beam axis is achieved for vertices with 25 tracks. An impact parameter resolution of less than 35 mu m is achieved for particles with transverse momentum greater than 1 GeV/c.
