This issue of the ARCHIVES includes previously unpublished protocols for the examination of specimens removed from patients with cancer of diverse sites. We provide a historical context for the ...development of these protocols and describe the process of development and approval. General information about the structure and content of the protocols is also provided. Cancer protocol development is an important step in the process of standardized cancer reporting. The value of such standardized reporting is discussed.
The objective of this study is to transport and deliver resveratrol to intracellular sites using apolipoprotein E3 (apoE3). Reconstituted high-density lipoprotein (rHDL) bearing resveratrol ...(rHDL/res) was prepared using phospholipids and the low-density lipoprotein receptor (LDLr)-binding domain of apoE3. Biophysical characterization revealed that resveratrol was partitioned into the phospholipid bilayer of discoidal rHDL/res particles (~19 nm diameter). Co-immunoprecipitation studies indicated that the LDLr-binding ability of apoE3 was retained. Cellular uptake of resveratrol to intracellular sites was evaluated in glioblastoma A-172 cells by direct fluorescence using chemically synthesized NBD-labeled resveratrol (res/NBD) embedded in rHDL/res. Competition and inhibition studies indicate that the uptake is by receptor mediated endocytosis via the LDLr, with co-localization of apoE3 and res/NBD in late endosomes/lysosomes. We propose that rHDL provides an ideal hydrophobic milieu to sequester resveratrol and that rHDL containing apoE3 serves as an effective "nanovehicle" to transport and deliver resveratrol to targeted intracellular sites.

Abstract EX Lupi, a low-mass young stellar object, went into an accretion-driven outburst in 2022 March. The outburst caused a sudden phase change of ∼112° ± 5° in periodically oscillating multiband ...lightcurves. Our high-resolution spectra obtained with the High Resolution Spectrograph (HRS) on board the Southern African Large Telescope also revealed a consistent phase change in the periodically varying radial velocities (RVs), along with an increase in the RV amplitude of various emission lines. The phase change and increase in RV amplitude morphologically translates to a change in the azimuthal and latitudinal location of the accretion hotspot over the stellar surface, which indicates a reconfiguration of the accretion funnel geometry. Our three-dimensional magnetohydrodynamic simulations reproduce the phase change for EX Lupi. To explain the observations, we explored the possibility of forward shifting of the dipolar accretion funnel as well as the possibility of the emergence of a new accretion funnel. During the outburst, we also found evidence of the hotspot’s morphology extending azimuthally asymmetrically with a leading hot edge and cold tail along the stellar rotation. Further, our high-cadence photometry showed that the accretion flow has clumps. We also detected possible clumpy accretion events in the HRS spectra that showed episodically highly blueshifted wings in the Ca ii IR triplet and Balmer H lines.
