Plastic pollution in aquatic ecosystems is an emerging environmental risk, as it may negatively impacts ecology, endangers aquatic species, and causes economic damage. Rivers are known to play a ...crucial role in transporting land‐based plastic waste to the world's oceans, but riverine ecosystems are also directly affected by plastic pollution. To better quantify global plastic pollution transport and to effectively reduce sources and risks, a thorough understanding of origin, transport, fate, and effects of riverine plastic debris is crucial. In this overview paper, we discuss the current scientific state on plastic debris in rivers and evaluate existing knowledge gaps. We present a brief background of plastics, polymer types typically found in rivers, and the risk posed to aquatic ecosystems. Additionally, we elaborate on the origin and fate of riverine plastics, including processes and factors influencing plastic debris transport and its spatiotemporal variation. We present an overview of monitoring and modeling efforts to characterize riverine plastic transport, and give examples of typical values from around the world. Finally, we present an outlook to riverine plastic research. With this paper, we aim to present an inclusive and comprehensive overview of riverine plastic debris research to date and suggest multiple ways forward for future research.

Gemcitabine has limited clinical benefits in pancreatic ductal adenocarcinoma. The solvent-based traditional taxanes docetaxel and paclitaxel have not shown clinical results superior to gemcitabine. ...Nab-paclitaxel, a water-soluble albumin-bound paclitaxel, may carry superior distribution properties into the tumor microenvironment and has shown efficacy in multiple tumor types. We evaluated nab-paclitaxel effects compared with gemcitabine or docetaxel. For pancreatic ductal adenocarcinoma cells AsPC-1, BxPC-3, MIA PaCa-2 and Panc-1, gemcitabine IC50 ranged from 494nM to 23.9 μM; docetaxel IC50 range was from 5 to 34nM; nab-paclitaxel IC50 range was from 243nM to 4.9 μM. Addition of IC25 dose of docetaxel or nab-paclitaxel decreased gemcitabine IC50. Net tumor growth inhibition after gemcitabine, docetaxel or nab-paclitaxel was 67, 31 and 72%, which corresponded with intratumoral proliferative and apoptotic indices. Tumor stromal density was decreased by nab-paclitaxel and to a lesser extent by docetaxel as measured through reduction in α-smooth muscle actin, S100A4 and collagen 1 expression. Animal survival was prolonged after nab-paclitaxel treatment (41 days, P < 0.002) compared with gemcitabine (32 days, P = 0.005), docetaxel (32 days, P = 0.005) and controls (20 days). Survival in nab-paclitaxel/gemcitabine and docetaxel/gemcitabine sequential treatment groups was not superior to nab-paclitaxel alone. Low-dose combination of gemcitabine with nab-paclitaxel or docetaxel was more effective compared with controls or gemcitabine alone but not superior to regular dose nab-paclitaxel alone. Combination treatment of gemcitabine+nab-paclitaxel or gemcitabine+docetaxel increased gemcitabine concentration in plasma and tumor. The superior antitumor activity of nab-paclitaxel provides a strong rationale for considering nab-paclitaxel as first-line monotherapy in pancreatic ductal adenocarcinoma.
Here we report concentrations of pollutants in floating plastics from the North Pacific accumulation zone (NPAC). We compared chemical concentrations in plastics of different types and sizes, ...assessed ocean plastic potential risks using sediment quality criteria, and discussed the implications of our findings for bioaccumulation. Our results suggest that at least a fraction of the NPAC plastics is not in equilibrium with the surrounding seawater. For instance, “hard plastic” samples had significantly higher PBDE concentrations than “nets and ropes” samples, and 29% of them had PBDE composition similar to a widely used flame-retardant mixture. Our findings indicate that NPAC plastics may pose a chemical risk to organisms as 84% of the samples had at least one chemical exceeding sediment threshold effect levels. Furthermore, our surface trawls collected more plastic than biomass (180 times on average), indicating that some NPAC organisms feeding upon floating particles may have plastic as a major component of their diets. If gradients for pollutant transfer from NPAC plastic to predators exist (as indicated by our fugacity ratio calculations), plastics may play a role in transferring chemicals to certain marine organisms.
Mutual aid groups have been an indispensable part of the public response to the COVID-19 pandemic. They have provided many forms of support, in particular grocery shopping which has enabled people to ...self-isolate if required. While community solidarity during emergencies and disasters is common, previous studies have shown that such solidarity behaviors tend to decline over time, even when needs remain high. In this study, we address how mutual aid groups can be sustained over time in the context of the COVID-19 pandemic. We conducted 32 interviews with organizers of COVID-19 mutual aid and community support groups in the United Kingdom between September 2020 and January 2021. Based on a reflexive thematic analysis, we identified several community and group level experiences and strategies that were related to sustained participation in COVID-19 mutual aid groups. Meeting community needs over time with localized action and resources and building trust and community-based alliances were foundational elements in the COVID-19 mutual aid groups. Group processes strategies, such as a culture of care and support and regular group meetings, were used to help to sustain involvement. Some experiences resulting from participation in COVID-19 mutual aid groups were also related to sustained participation, including positive emotions (e.g., joy, pride), well-being and sense of efficacy, and an increasing sense of local community belonging and cohesion. Based on these findings, we propose four practical recommendations for sustaining mutual aid groups to assist public engagement with protective behaviors in the COVID-19 pandemic and beyond. We recommend providing practical and financial support to COVID-19 mutual aid groups; to mobilize the knowledge and the experiences acquired by COVID-19 mutual aid groups for developing programs and interventions for addressing the medium and long-term impacts of COVID-19; to prioritize community-level interventions; and to recognize the role of group processes as these have the potential to lead to long-term community responses. These approaches will be key for ensuring that communities effectively recover from the COVID-19 pandemic.
This research analyses the pronominal system of Soikkola Ingrian. The main goal of the article is descriptive, because existing publications on Soikkola Ingrian give only a superficial analysis of ...pronouns and there are no detailed descriptions based on data from the 21st century. The article focuses on both interspeaker and intraspeaker variation in four groups of Ingrian pronouns: personal, demonstrative, reflexive and reciprocal. The field data collected by the authors in the 21st century are compared with the data presented in grammars written in the 19th and 20th centuries. The research concludes that although the pronominal system has not changed significantly, there are a number of innovations in Ingrian pronominal forms and their variation. The most frequent type of variation is the variation between individual speakers, but not between sub-dialectal groups. Free variation is also common, while other types of variation are less relevant.

Artikkel on olemuselt kirjeldav. Olemasolevad Soikkola murret puudutavad uurimused esitavad üsna pealiskaudse pronoomenite analüüsi ning puuduvad 21. sajandi andmetel põhinevad üksikasjalikud kirjeldused. Artikkel keskendub nii kõnelejasisesele kui ka kõnelejatevahelisele varieerumisele neljas isuri pronoomenite rühmas: personaal-, demonstratiiv-, refleksiiv- ja retsiprookpronoomenid. Autorite poolt 21. sajandil kogutud välitööde andmeid võrreldakse 19. ja 20. sajandil kirjutatud grammatikate andmetega. Uurimuses jõutakse järeldusele, et kuigi pronoomenite süsteem pole oluliselt muutunud, on isuri pronoomenite vormides ja nende varieerumises mitmeid uuendusi. Kõige sagedamini esineb varieerumist üksikute kõnelejate vahel, kuid mitte murde allrühmade vahel. Levinud on ka vaba varieerumine, samas kui muud tüüpi varieerumised on vähem olulised.
Land-based macroplastic is considered one of the major sources of marine plastic debris. However, estimations of plastic emission from rivers into the oceans remain scarce and uncertain, mainly due ...to a severe lack of standardized observations. To properly assess global plastic fluxes, detailed information on spatiotemporal variation in river plastic quantities and composition are urgently needed. In this paper, we present a new methodology to characterize riverine macroplastic dynamics. The proposed methodology was applied to estimate the plastic emission from the Saigon River, Vietnam. During a two-week period, hourly cross-sectional profiles of plastic transport were made across the river width. Simultaneously, sub-hourly samples were taken to determine the weight, size and composition of riverine macroplastics (>5cm). Finally, extrapolation of the observations based on available hydrological data yielded new estimates of daily, monthly and annual macroplastic emission into the ocean. Our results suggest that plastic emissions from the Saigon River are up to 4 times higher than previously estimated. Importantly, our flexible methodology can be adapted to local hydrological circumstances and data availability, thus enabling a consistent characterization of macroplastic dynamics in rivers worldwide. Such data will provide crucial knowledge for the optimization of future mediation and recycling efforts.
Hormone-sensitive lipase (HSL) plays a crucial role in intracellular lipolysis, and loss of HSL leads to diacylglycerol (DAG) accumulation, reduced FA mobilization, and impaired PPARγ signaling. Hsl ...knockout mice exhibit adipose tissue inflammation, but the underlying mechanisms are still not clear. Here, we investigated if and to what extent HSL loss contributes to endoplasmic reticulum (ER) stress and adipose tissue inflammation in Hsl knockout mice. Furthermore, we were interested in how impaired PPARγ signaling affects the development of inflammation in epididymal white adipose tissue (eWAT) and inguinal white adipose tissue (iWAT) of Hsl knockout mice and if DAG and ceramide accumulation contribute to adipose tissue inflammation and ER stress. Ultrastructural analysis showed a markedly dilated ER in both eWAT and iWAT upon loss of HSL. In addition, Hsl knockout mice exhibited macrophage infiltration and increased F4/80 mRNA expression, a marker of macrophage activation, in eWAT, but not in iWAT. We show that treatment with rosiglitazone, a PPARγ agonist, attenuated macrophage infiltration and ameliorated inflammation of eWAT, but expression of ER stress markers remained unchanged, as did DAG and ceramide levels in eWAT. Taken together, we show that HSL loss promoted ER stress in both eWAT and iWAT of Hsl knockout mice, but inflammation and macrophage infiltration occurred mainly in eWAT. Also, PPARγ activation reversed inflammation but not ER stress and DAG accumulation. These data indicate that neither reduction of DAG levels nor ER stress contribute to the reversal of eWAT inflammation in Hsl knockout mice.
Nanoparticle albumin-bound paclitaxel (nab-paclitaxel, NPT) has recently shown efficacy in pancreatic ductal adenocarcinoma (PDAC). Targeting tumor angiogenesis is a sensible combination therapeutic ...strategy for cancer, including PDAC. We tested the hypothesis that NPT response in PDAC can be enhanced by the mechanistically different antiangiogenic agents bevacizumab (Bev) or sunitinib (Su), despite its inherently increased tumor penetration and drug delivery. Compared with controls (19 days), median animal survival was increased after NPT therapy (32 days, a 68% increase, P = 0.0008); other regimens with enhanced survival were NPT+Bev (38 days, a 100% increase, P = 0.0004), NPT+Su (37 days, a 95% increase, P = 0.0004), and NPT+Bev+Su (49 days, a 158% increase, P = 0.0001) but not bevacizumab, sunitinib, or Bev+Su therapy. Relative to controls (100 ± 22.8), percentage net local tumor growth was 28.2 ± 23.4 with NPT, 55.6 ± 18 (Bev), 38.8 ± 30.2 (Su), 11 ± 7.2 (Bev+Su), 32.8 ± 29.2 (NPT+Bev), 6.6 ± 10.4 (NPT+Su), and 13.8 ± 12.5 (NPT+Bev+Su). Therapeutic effects on intratumoral proliferation, apoptosis, microvessel density, and stromal density corresponded with tumor growth inhibition data. In AsPC-1 PDAC cells, NPT IC(50) was reduced >6-fold by the addition of sunitinib (IC(25)) but not by bevacizumab. In human umbilical vein endothelial cells (HUVEC), NPT IC(50) (82 nmol/L) was decreased to 41 nmol/L by bevacizumab and to 63 nmol/L by sunitinib. In fibroblast WI-38 cells, NPT IC(50) (7.2 μmol/L) was decreased to 7.8 nmol/L by sunitinib, but not by bevacizumab. These findings suggest that the effects of one of the most active cytotoxic agents against PDAC, NPT, can be enhanced with antiangiogenic agents, which clinically could relate to greater responses and improved antitumor results.
Myeloid cells are key factors in the progression of bronchopulmonary dysplasia (BPD) pathogenesis. Endothelial monocyte-activating polypeptide II (EMAP II) mediates myeloid cell trafficking. The ...origin and physiological mechanism by which EMAP II affects pathogenesis in BPD is unknown. The objective was to determine the functional consequences of elevated EMAP II levels in the pathogenesis of murine BPD and to investigate EMAP II neutralization as a therapeutic strategy. Three neonatal mouse models were used: (1) BPD (hyperoxia), (2) EMAP II delivery, and (3) BPD with neutralizing EMAP II antibody treatments. Chemokinic function of EMAP II and its neutralization were assessed by migration in vitro and in vivo. We determined the location of EMAP II by immunohistochemistry, pulmonary proinflammatory and chemotactic gene expression by quantitative polymerase chain reaction and immunoblotting, lung outcome by pulmonary function testing and histological analysis, and right ventricular hypertrophy by Fulton's Index. In BPD, EMAP II initially is a bronchial club-cell-specific protein-derived factor that later is expressed in galectin-3
macrophages as BPD progresses. Continuous elevated expression corroborates with baboon and human BPD. Prolonged elevation of EMAP II levels recruits galectin-3
macrophages, which is followed by an inflammatory state that resembles a severe BPD phenotype characterized by decreased pulmonary compliance, arrested alveolar development, and signs of pulmonary hypertension. In vivo pharmacological EMAP II inhibition suppressed proinflammatory genes Tnfa, Il6, and Il1b and chemotactic genes Ccl2 and Ccl9 and reversed the severe BPD phenotype. EMAP II is sufficient to induce macrophage recruitment, worsens BPD progression, and represents a targetable mechanism of BPD development.
The skin serves as an important barrier protecting the body from physical, chemical and pathogenic hazards as well as regulating the bi-directional transport of water, ions and nutrients. In order to ...improve the knowledge on skin structure and function as well as on skin diseases, animal experiments are often employed, but anatomical as well as physiological interspecies differences may result in poor translatability of animal-based data to the clinical situation. In vitro models, such as human reconstructed epidermis or full skin equivalents, are valuable alternatives to animal experiments. Enormous advances have been achieved in establishing skin models of increasing complexity in the past. In this review, human skin structures are described as well as the fast evolving technologies developed to reconstruct the complexity of human skin structures in vitro.
