On 24 August 2016 a magnitude ML 6.0 occurred in the Central Apennines (Italy) between Amatrice and Norcia causing nearly 300 fatalities. The main shock ruptured a NNW‐SSE striking, WSW dipping ...normal fault. We invert waveforms from 26 three‐component strong motion accelerometers, filtered between 0.02 and 0.5 Hz, within 45 km from the fault. The inferred slip distribution is heterogeneous and characterized by two shallow slip patches updip and NW from the hypocenter, respectively. The rupture history shows bilateral propagation and a relatively high rupture velocity (3.1 km/s). The imaged rupture history produced evident directivity effects both N‐NW and SE of the hypocenter, explaining near‐source peak ground motions. Fault dimensions and peak slip values are large for a moderate‐magnitude earthquake. The retrieved rupture model fits the recorded ground velocities up to 1 Hz, corroborating the effects of rupture directivity and slip heterogeneity on ground shaking and damage pattern.

Muography is an imaging technique based on the measurement of absorption profiles for muons as they pass through rocks and earth. Muons are produced in the interactions of high-energy cosmic rays in ...the Earth's atmosphere. The technique is conceptually similar to usual X-ray radiography, but with extended capabilities of investigating over much larger thicknesses of matter thanks to the penetrating power of high-energy muons. Over the centuries a complex system of cavities has been excavated in the yellow tuff of Mt. Echia, the site of the earliest settlement of the city of Naples in the 8th century BC. A new generation muon detector designed by us, was installed under a total rock overburden of about 40 metres. A 26 days pilot run provided about 14 millions of muon events. A comparison of the measured and expected muon fluxes improved the knowledge of the average rock density. The observation of known cavities proved the validity of the muographic technique. Hints on the existence of a so far unknown cavity was obtained. The success of the investigation reported here demonstrates the substantial progress of muography in underground imaging and is likely to open new avenues for its widespread utilisation.
We image the rupture history of the 2009 L'Aquila (central Italy) earthquake using a nonlinear joint inversion of strong motion and GPS data. This earthquake ruptured a normal fault striking along ...the Apennines axis and dipping to the SW. The inferred slip distribution is heterogeneous and characterized by a small, shallow slip patch located up‐dip from the hypocenter (9.5 km depth) and a large, deeper patch located southeastward. The rupture velocity is larger in the up‐dip than in the along‐strike direction. This difference can be partially accounted by the crustal structure, which is characterized by a high velocity layer above the hypocenter and a lower velocity below. The latter velocity seems to have affected the along strike propagation since the largest slip patch is located at depths between 9 and 14 km. The imaged slip distribution correlates well with the on‐fault aftershock pattern as well as with mapped surface breakages.

Cosmic-ray muon radiography (muography), an imaging technique that can provide measurements of rock densities within the top few 100 m of a volcanic cone, has now achieved a spatial resolution of the ...order of 10 m in optimal detection conditions. Muography provides images of the top region of a volcano edifice with a resolution that is considerably better than that typically achieved with other conventional methods (i.e. gravimetric). We expect such precise measurements, to provide us with information on anomalies in the rock density distribution, which can be affected by dense lava conduits, low-density magma supply paths or the compression with the depth of the overlying soil. The MUon RAdiography of VESuvius (MURAVES) project is now in its final phase of construction and deployment. Up to four muon hodoscopes, each with a surface of roughly 1 m
, will be installed on the slope of Vesuvius and take data for at least 12 months. We will use the muographic profiles, combined with data from gravimetric and seismic measurement campaigns, to determine the stratigraphy of the lava plug at the bottom of the Vesuvius crater, in order to infer potential eruption pathways. While the MURAVES project unfolds, others are using emulsion detectors on Stromboli to study the lava conduits at the top of the volcano. These measurements are ongoing: they have completed two measurement campaigns and are now performing the first data analysis.This article is part of the Theo Murphy meeting issue 'Cosmic-ray muography'.
SOCS (suppressor of cytokine signalling) proteins are negative-feedback regulators of the JAK (Janus kinase)/STAT (signal transducer and activator of transcription) pathway. Their expression levels ...are low under physiological conditions, but they are up-regulated in response to cytokine stimulation in many immune and inflammatory processes. Overexpression of SOCS1 in keratinocyte clones abrogates the IFNγ (interferon γ)-induced expression of many pro-inflammatory genes and the release of related chemokines by blocking the JAK/STAT pathway. SOCS1 inhibits JAK2 kinase activity by binding the catalytic site of JAK2, with its KIR (kinase-inhibitory region) acting as a pseudo-substrate of the enzyme. In the present study, we screened a focused combinatorial peptide library of KIR to identify new peptides able to mimic its function with an improved affinity towards the JAK2 catalytic site. Using an alanine-scanning method, KIR residues that are crucial for the interaction with JAK2 were unveiled. In this way, the KIR sequence was restricted to a shorter segment and ‘non-essential’ residues were replaced by different amino acids following a simplified combinatorial approach. We selected a new unnatural sequence able to bind to JAK2 with Kd values in the nanomolar range. This peptide was tested in human keratinocyte cultures and reduced the phosphorylation of STAT1 and the expression levels of IRF-1 (interferon regulatory factor-1).
A variety of peptides active in biological pathways have been identified e.g. receptor antagonists or inhibitors of protein-protein interactions and several peptide or peptide-derived compounds are ...on the drug market or in clinical trials. Through the rational design or the combinatorial preparation and High-throughput screening of arrays of compounds, peptides play a pivotal role for the rapid identification of ligands, but, despite these favorable properties, they often present poorer bioavailability and lower metabolic stability respect to traditional drugs. The process of conversion of a peptide in a small molecule provides the reduction of the peptide to the minimum active sequence (MAS) testing truncated peptides from the C- and N- termini alternatively. Then the influence of individual amino acid on the biological activity is determined by systematically replacing each residue in the peptide with specific amino acids. After structure-activity relationship (SAR) of each amino acid in the sequence has been assessed, the bioactive conformational flexibility is reduced by introducing constraints at various positions. These features are used for the design of a pharmacophore model in which functional groups crucial for activity are pre-positioned. Here we propose a panoramic review of the common principles for the conversion of peptides into small organic molecules and the most interesting findings in peptide-based leads of the last decades.
We present the first high-quality catalog of early aftershocks of the three mainshocks of the 2016 central Italy Amatrice-Visso-Norcia normal faulting sequence. We located 10,574 manually picked ...aftershocks with a robust probabilistic, non-linear method achieving a significant improvement in the solution accuracy and magnitude completeness with respect to previous studies. Aftershock distribution and relocated mainshocks give insight into the complex architecture of major causative and subsidiary faults, thus providing crucial constraints on multi-segment rupture models. We document reactivation and kinematic inversion of a WNW-dipping listric structure, referable to the inherited Mts Sibillini Thrust (MST) that controlled segmentation of the causative normal faults. Spatial partitioning of aftershocks evidences that the MST lateral ramp had a dual control on rupture propagation, behaving as a barrier for the Amatrice and Visso mainshocks, and later as an asperity for the Norcia mainshock. We hypothesize that the Visso mainshock re-activated also the deep part of an optimally oriented preexisting thrust. Aftershock patterns reveal that the Amatrice Mw5.4 aftershock and the Norcia mainshock ruptured two distinct antithetic faults 3-4 km apart. Therefore, our results suggest to consider both the MST cross structure and the subsidiary antithetic fault in the finite-fault source modelling of the Norcia earthquake.
The hAPE1 (human apurinic/apyrimidinic endonuclease 1) is an essential enzyme, being the main abasic endonuclease in higher eukaryotes. However, there is strong evidence to show that hAPE1 can ...directly bind specific gene promoters, thus modulating their transcriptional activity, even in the absence of specific DNA damage. Recent findings, moreover, suggest a role for hAPE1 in RNA processing, which is modulated by the interaction with NPM1 (nucleophosmin). Independent domains account for many activities of hAPE1; however, whereas the endonuclease and the redox-active portions of the protein are well characterized, a better understanding of the role of the unstructured N-terminal region is needed. In the present study, we characterized the requirements for the interaction of hAPE1 with NPM1 and undamaged nucleic acids. We show that DNA/RNA secondary structure has an impact on hAPE1 binding in the absence of damage. Biochemical studies, using the isolated N-terminal region of the protein, reveal that the hAPE1 N-terminal domain represents an evolutionary gain of function, since its composition affects the protein's stability and ability to interact with both nucleic acids and NPM1. Although required, however, this region is not sufficient itself to stably interact with DNA or NPM1.
We study the 30 October 2016 Norcia earthquake (MW 6.5) to retrieve the rupture history by jointly inverting seismograms and coseismic Global Positioning System displacements obtained by dense local ...networks. The adopted fault geometry consists of a main normal fault striking N155° and dipping 47° belonging to the Mt. Vettore‐Mt. Bove fault system (VBFS) and a secondary fault plane striking N210° and dipping 36° to the NW. The coseismic rupture initiated on the VBFS and propagated with similar rupture velocity on both fault planes. Updip from the nucleation point, two main slip patches have been imaged on these fault segments, both characterized by similar peak‐slip values (~3 m) and rupture times (~3 s). After the breakage of the two main slip patches, coseismic rupture further propagated southeastward along the VBFS, rupturing again the same fault portion that slipped during the 24 August earthquake. The retrieved coseismic slip distribution is consistent with the observed surface breakages and the deformation pattern inferred from interferometric synthetic aperture radar measurements. Our results show that three different fault systems were activated during the 30 October earthquake. The composite rupture model inferred in this study provides evidences that also a deep portion of the NNE trending section of the Olevano‐Antrodoco‐Sibillini thrust broke coseismically, implying the kinematic inversion of a thrust ramp. The obtained rupture history indicates that in this sector of the Apennines, compressional structures inherited from past tectonics can alternatively segment boundaries of NW trending active normal faults or break coseismically during moderate‐to‐large magnitude earthquakes.

Muon radiography, also known as muography, is an imaging technique that provides information on the mass density distribution inside large objects. Muons are naturally produced in the interactions of ...cosmic rays in the Earth's atmosphere. The physical process exploited by muography is the attenuation of the muon flux, that depends on the thickness and density of matter that muons cross in the course of their trajectory. A particle detector with tracking capability allows the measurement of the muons flux as a function of the muon direction. The comparison of the measured muon flux with the expected one gives information on the distribution of the density of matter, in particular, on the presence of cavities. In this article, the measurement performed at Mt. Echia in Naples (Saracino 2017
