How can we comprehend people who pay for an experience marketed as painful? On one hand, consumers spend billions of dollars every year to alleviate different kinds of pain. On the other hand, ...millions of individuals participate in extremely painful leisure pursuits. In trying to understand this conundrum, we ethnographically study a popular adventure challenge where participants subject themselves to electric shocks, fire, and freezing water. Through sensory intensification, pain brings the body into sharp focus, allowing individuals to rediscover their corporeality. In addition, painful extraordinary experiences operate as regenerative escapes from the self. By flooding the consciousness with gnawing unpleasantness, pain provides a temporary relief from the burdens of self-awareness. Finally, when leaving marks and wounds, pain helps consumers create the story of a fulfilled life. In a context of decreased physicality, market operators play a major role in selling pain to the saturated selves of knowledge workers, who use pain as a way to simultaneously escape reflexivity and craft their life narrative.
Biomaterials have been extensively used to leverage beneficial outcomes in various therapeutic applications, such as providing spatial and temporal control over the release of therapeutic agents in ...drug delivery as well as engineering functional tissues and promoting the healing process in tissue engineering and regenerative medicine. This Perspective presents important milestones in the development of polymeric biomaterials with defined structures and properties. Contemporary studies of biomaterial design have been reviewed with focus on constructing materials with controlled structure, dynamic functionality, and biological complexity. Examples of these polymeric biomaterials enabled by advanced synthetic methodologies, dynamic chemistry/assembly strategies, and modulated cell–material interactions have been highlighted. As the field of polymeric biomaterials continues to evolve with increased sophistication, current challenges and future directions for the design and translation of these materials are also summarized.

The movements of some long-distance migrants are driven by innate compass headings that they follow on their first migrations (e.g., some birds and insects), while the movements of other first-time ...migrants are learned by following more experienced conspecifics (e.g., baleen whales). However, the overall roles of innate, learned, and social behaviors in driving migration goals in many taxa are poorly understood. To look for evidence of whether migration routes are innate or learned for sea turtles, here for 42 sites around the world we compare the migration routes of >400 satellite-tracked adults of multiple species of sea turtle with ∼45 000 Lagrangian hatchling turtle drift scenarios. In so doing, we show that the migration routes of adult turtles are strongly related to hatchling drift patterns, implying that adult migration goals are learned through their past experiences dispersing with ocean currents. The diverse migration destinations of adults consistently reflected the diversity in sites they would have encountered as drifting hatchlings. Our findings reveal how a simple mechanism, juvenile passive drift, can explain the ontogeny of some of the longest migrations in the animal kingdom and ensure that adults find suitable foraging sites.
The mountain of modern interpretation to which the language of the Fourteenth Amendment of the United States Constitution has been subjected tends to overshadow the multiple concepts of ...antidiscrimination that were actually circulating at the time of its drafting. Moreover, as authors on race and law have pointed out, Congress itself lacked any African American representatives during the 1866–68 moment of transitional justice. The subsequent development of a “state action doctrine” limiting the reach of federal civil rights enforcement, in turn, eclipsed important contemporary understandings of the harms that Reconstruction-era initiatives sought to combat. In contrast to the oblique language of the Fourteenth Amendment, a dignity-based legal theory of affirmative equal rights had by 1867 taken center stage in the cosmopolitan city of New Orleans. Activists formulated the concept of “public rights” as a claim to participation without discrimination in the entire sphere of “common life.” Elections for delegates to Louisiana's Constitutional Convention of 1867–68, held under the broad suffrage mandated by the Military Reconstruction Acts, yielded a convention in which half of the members were men of African descent. Seeking the “impartial treatment of all men” in “churches, hotels, cars, steamboats, theaters, stores, even schools,” the convention crafted a Bill of Rights that affirmatively guaranteed to all of the state's citizens “the same civil, political, and public rights,” independent of race or color. These innovations in the defense of human rights under law drew from a deep well of anti-caste thinking developed in domestic and transnational discussions conducted in both French and English, with participants from both sides of the Atlantic and the Caribbean. Cosmopolitan progressives such as Edouard Tinchant and Jean-Charles Houzeau worked with Louisiana-born activists including Louis Charles Roudanez, Simeon Belden, and Paul Trévigne to develop and advance the idea of public rights. Legislators crafted and passed state statutes that provided for civil penalties for violation of these rights, along with a private cause of action that could yield both actual and exemplary damages. Throughout the 1870s, however, advocates met a fierce white-supremacist counterattack, one that fused obstructionist litigation, vote suppression, and vigilante violence. A claim to equal treatment under the 1868 constitution was won in the state courts by Josephine Decuir, but was overturned in 1877 at the United States Supreme Court. With the ascent of the Democratic Party, white supremacists–including the lawyer/vigilante Robert Hardin Marr-took their seats on the state Supreme Court. By 1879, the public rights guarantees had been expunged from the state's constitution. Nonetheless, for a crucial decade, the cross-racial politics of Louisiana had overcome many of the deficits of legitimacy that often undercut moments of transitional lawmaking. Delegates to the 1867–68 Constitutional Convention took the opportunity to spell out specific positive rights that they saw as essential to full civil freedom. And at the center, they placed their insistence that the state had an obligation to assure that men and women of color would not be subjected to forced indignity in the public sphere.
Cord blood (CB) mononuclear cell populations have demonstrated significant promise in biomaterials-based regenerative therapies; however, the contributions of monocyte and macrophage subpopulations ...towards proper tissue healing and regeneration are not well understood, and the phenotypic responses of macrophage to microenvironmental cues have not been well-studied. In this work, we evaluated the effects of cytokine stimulation and altered substrate stiffness. Macrophage derived from CB CD14+ monocytes adopted distinct inflammatory (M1) and anti-inflammatory (M2a and M2c) phenotypes in response to cytokine stimulation (M1: lipopolysaccharide (LPS) and interferon (IFN-γ); M2a: interleukin (IL)-4 and IL-13; M2c: IL-10) as determined through expression of relevant cell surface markers and growth factors. Cytokine-induced macrophage readily altered their phenotypes upon sequential administration of different cytokine cocktails. The impact of substrate stiffness on macrophage phenotype was evaluated by seeding CB-derived macrophage on 3wt%, 6wt%, and 14wt% poly(ethylene glycol)-based hydrogels, which exhibited swollen shear moduli of 0.1, 3.4, and 10.3 kPa, respectively. Surface marker expression and cytokine production varied depending on modulus, with anti-inflammatory phenotypes increasing with elevated substrate stiffness. Integration of specific hydrogel moduli and cytokine cocktail treatments resulted in the differential regulation of macrophage phenotypic biomarkers. These data suggest that CB-derived macrophages exhibit predictable behaviors that can be directed and finely tuned by combinatorial modulation of substrate physical properties and cytokine profiles.

Adventitial fibroblasts (AFs) are critical mediators of vascular remodeling. However, the contributions of AFs towards development of vasculature and the specific mechanisms by which these cells ...regulate physiological expansion of the vasa vasorum, the specialized microvasculature that supplies nutrients to the vascular wall, are not well understood. To determine the regulatory role of AFs in microvascular endothelial cell (MVEC) neovasculogenesis and to investigate the regulatory pathways utilized for communication between the two cell types, AFs and MVECs were cultured together in poly(ethylene glycol)-based hydrogels. Following preliminary evaluation of a set of cell adhesion peptides (AG10, AG73, A2G78, YIGSR, RGD), 7.5wt% hydrogels containing 3 mM RGD were selected as these substrates did not initiate primitive tubule structures in 3D MVEC monocultures, thus providing a passive platform to study AF-MVEC interaction. The addition of AFs to hydrogels promoted MVEC viability; however, increasing AF density within hydrogels stimulated MVEC proliferation, increased microvessel density and size, and enhanced deposition of basement membrane proteins, collagen IV and laminin. Importantly, AF-MVEC communication through the transforming growth factor beta (TGF-β)/activin receptor-like kinase 5 (ALK5) signaling pathway was observed to mediate microvessel formation, as inhibition of ALK5 significantly decreased MVEC proliferation, microvessel formation, mural cell recruitment, and basement membrane production. These data indicate that AFs regulate MVEC neovasculogenesis and suggest that therapeutics targeting the TGF-β/ALK5 pathway may be useful for regulation of vasculogenic and anti-vasculogenic responses.

Whilst several explanations have been proposed for the absence of fire-related behaviours at well preserved Lower Palaeolithic sites, much of the emphasis of previous research has concentrated on our ability to find fire in the archaeological record. Furthermore, evolutionary models of early hominin fire engagement have often been developed and discussed in the context of early African hominins. Here we explore the role of fire in the behaviours, choices and lives of hominins during the earliest occupations of temperate regions, with a focus on Europe. We consider fire use in the context of Europe’s specific palaeoenvironmental conditions and discuss whether a long or short fire chronology model best fits the current evidence for the use of controlled fire in these regions during the Lower Palaeolithic.
We propose two models for hominin fire behaviours in the temperate latitudes, using a heuristic ‘macroscale to microscale’ approach to understanding the needs for — and the use of — fire during this period. We argue that such holistic approaches must combine experimental work, experiential observations and cost-benefit approaches and should consider site context and function, fire function, social behaviour, and mobility, to evaluate the limited evidence for fire use in the Lower Palaeolithic. We highlight that, varying with seasonality, fire function (and the associated costs and benefits) was of particular importance and may explain the overall paucity of evidence for fire use in temperate regions prior to the Middle Palaeolithic. This has implications for other potential survival strategies that are invisible in the early archaeological record, such as shelter, clothing, and the putrefaction of meat for later consumption.
This article explores a core question in the law of slavery: how was an individual's status as slave or free socially discerned and formally adjudicated? Under the doctrine of “freedom by ...prescription,” a person who had in good faith “lived as free” could argue that the absence of exercise of ownership for a specified term of years extinguished a prior owner's title. In the medieval Siete Partidas of Alfonso the Wise, which continued as a legal point of reference in Louisiana well after the end of Spanish rule, both the law of status and the law of property confirmed this path to freedom. From 1808 onward, Louisiana jurists and legislators sought to eliminate the remnants of the doctrine, but it lingered in popular and even judicial consciousness. The 1853 kidnapping of a woman named Eulalie Oliveau, six of her children, and eleven of her grandchildren for sale in the New Orleans slave market brought the question of “freedom by prescription” back into the courts. The awkward resolution of that case, and the uncertain fate of Eulalie Oliveau and her children, foreshadowed Reconstruction-era struggles over the content of legal freedom and the rights that freedom might bring to those who had once been held as property.
The LIM kinase family consists of just two members: LIM kinase 1 (LIMK1) and LIM kinase 2 (LIMK2). With uniquely organised signalling domains, LIM kinases are regulated by several upstream signalling ...pathways, principally acting downstream of Rho GTPases to influence the architecture of the actin cytoskeleton by regulating the activity of the cofilin family proteins cofilin1, cofilin2 and destrin. Although the LIM kinases are very homologous, particularly when comparing kinase domains, there is emerging evidence that each may be subject to different regulatory pathways and may contribute to both distinct and overlapping cellular and developmental functions. Normal central nervous system development is reliant upon the presence of LIMK1, and its deletion has been implicated in the development of the human genetic disorder Williams syndrome. Normal testis development, on the other hand, is disrupted by the deletion of LIMK2. In addition, the possible involvement of each kinase in cardiovascular disorders as well as cancer has recently emerged. The LIM kinases have been proposed to play an important role in tumour-cell invasion and metastasis; fine-tuning the balance between phosphorylated and non-phosphorylated cofilin may be a significant determinant of tumour-cell metastatic potential. In this review, we outline the structure, regulation and function of LIM kinases and their functions at cellular and organismal levels, as well as their possible contributions to human disease.
It is fundamentally important for many animal ecologists to quantify the costs of animal activities, although it is not straightforward to do so. The recording of triaxial acceleration by ...animal‐attached devices has been proposed as a way forward for this, with the specific suggestion that dynamic body acceleration (DBA) be used as a proxy for movement‐based power.
Dynamic body acceleration has now been validated frequently, both in the laboratory and in the field, although the literature still shows that some aspects of DBA theory and practice are misunderstood. Here, we examine the theory behind DBA and employ modelling approaches to assess factors that affect the link between DBA and energy expenditure, from the deployment of the tag, through to the calibration of DBA with energy use in laboratory and field settings.
Using data from a range of species and movement modes, we illustrate that vectorial and additive DBA metrics are proportional to each other. Either can be used as a proxy for energy and summed to estimate total energy expended over a given period, or divided by time to give a proxy for movement‐related metabolic power. Nonetheless, we highlight how the ability of DBA to predict metabolic rate declines as the contribution of non‐movement‐related factors, such as heat production, increases.
Overall, DBA seems to be a substantive proxy for movement‐based power but consideration of other movement‐related metrics, such as the static body acceleration and the rate of change of body pitch and roll, may enable researchers to refine movement‐based metabolic costs, particularly in animals where movement is not characterized by marked changes in body acceleration.
Quantifying the costs of animal activities is important yet difficult to achieve. Dynamic body acceleration, derived from triaxial acceleration sensors, is a powerful method with which to examine the energetic implications of animal decision‐making in a wide range of systems, including with respect to the effects of individual experience, state and the abiotic/ biotic environment.
