"Unter dem Titel "Erprobung neu strukturierter Ausbildungsformen im Rahmen des Ausbildungskonsenses 2007-2010" führte das Hamburger Institut für berufliche Bildung (HIBB) in Zusammenarbeit mit der ...Beruflichen Schule für Büro- und Personalmanagement Bergedorf (H17) und der Staatlichen Gewerbeschule für Informations- und Elektrotechnik, Chemie- und Automatisierungstechnik (G18) seit August 2008 einen Schulversuch durch, in dem eine alternative Form der beruflichen Ausbildung erprobt wurde. Die wissenschaftliche Begleitung des Schulversuchs wurde durch ein Konsortium des Instituts für Berufs- und Wirtschaftspädagogik der Universität Hamburg gewährleistete. Ziel des Schulversuchs war es, das Konzept der bisherigen vollqualifizierenden Berufsfachschule mit der Erlangung der Fachhochschulreife (FHR) und einem regulären dualen Ausbildungsabschluss zu verknüpfen und damit die bisher übliche Verweildauer vieler Jugendlicher im Schulsystem wesentlich zu verkürzen. Dazu absolvierten die Schüler und Schülerinnen zunächst einen 2-jährigen schulischen Ausbildungsabschnitt mit den Abschlüssen Technische Assistenz Informatik (an der G18) bzw. Kaufmännische Assistenz (an der H17) und der Fachhochschulreife. Integrierte Praktika ermöglichten den Erwerb der erforderlichen Praxisanteile zur FHR. Bei erfolgreichem Bestehen folgte ein 1,5-jähriger, rein betrieblicher Ausbildungsabschnitt mit dem Kammerabschluss zum Kaufmann für Bürokommunikation bzw. Fachinformatiker Systemintegration." Wie chancengerecht und leistungsfähig ist die berufliche Bildung in Deutschland? Anhand von Indikatoren vergleicht die Studie das gesamte Berufsausbildungssystem in Deutschland, von der dualen und ...vollzeitschulischen Ausbildung bis zum Übergangssystem. Die Ergebnisse zeigen, dass in allen Bundesländern die Zahl der Ausbildungsplätze und die Zahl der Ausbildungsbewerber:innen sinken. Jugendliche mit Hauptschulabschluss oder ohne deutsche Staatsangehörigkeit haben Probleme bei der Suche nach einer Lehrstelle oder einem Platz in einer Fachschule. Im Vergleich der Bundesländer spielt der Wohnort eine wichtige Rolle, da die Bildungssysteme der Länder auch in der beruflichen Ausbildung unterschiedlich aufgestellt sind. Der Ländermonitor macht Handlungsbedarfe der Bildungspolitik auf Bundes- und Landesebene sichtbar und stellt Lösungsmöglichkeiten vor. Grundlage der Studie sind statistische Daten des Bundes und der Länder, der Bundesagentur für Arbeit und des Bundesinstituts für Berufsbildung, die von einer Autorengruppe unter Leitung des Soziologischen Forschungsinstituts Göttingen ausgewertet wurden.

Double-strand break repair by recombination requires a homology search. In yeast, induced breaks move significantly more than undamaged loci. To examine whether DNA damage provokes an increase in ...chromatin mobility generally, we tracked undamaged loci under DNA-damaging conditions. We found that the yeast checkpoint factors Mec1, Rad9, and Rad53 are required for genome-wide increases in chromatin mobility, but not the repair protein Rad51. Mec1 activation by targeted Ddc1/Ddc2 enhances chromatin mobility even in the absence of damage. Finally, the INO80 chromatin remodeler is shown to act downstream from Mec1 to increase chromatin mobility, highlighting an additional damage-related role of this nucleosome remodeling complex.
Chromatin is organized and segmented into a landscape of domains that serve multiple purposes. In contrast to transcription, which is controlled by defined sequences at distinct sites, DNA damage can ...occur anywhere. Repair accordingly must occur everywhere, yet it is inevitably affected by its chromatin environment. In this review, we summarize recent work investigating how changes in chromatin organization facilitate and/or guide DNA double-strand break repair. In addition, we examine new live cell studies on the dynamics of chromatin and the mechanisms that regulate its movement.
Recent advances in both the technologies used to measure chromatin movement and the biophysical analysis used to model them have yielded a fuller understanding of chromatin dynamics and the polymer ...structure that underlies it. Changes in nucleosome packing, checkpoint kinase activation, the cell cycle, chromosomal tethers, and external forces acting on nuclei in response to external and internal stimuli can alter the basal mobility of DNA in interphase nuclei of yeast or mammalian cells. Although chromatin movement is assumed to be necessary for many DNA-based processes, including gene activation by distal enhancer-promoter interaction or sequence-based homology searches during double-strand break repair, experimental evidence supporting an essential role in these activities is sparse. Nonetheless, high-resolution tracking of chromatin dynamics has led to instructive models of the higher-order folding and flexibility of the chromatin polymer. Key regulators of chromatin motion in physiological conditions or after damage induction are reviewed here.
Chromatin moves with subdiffusive and spatially constrained dynamics within the cell nucleus. Here, we use single-locus tracking by time-lapse fluorescence microscopy to uncover information regarding ...the forces that influence chromatin movement following the induction of a persistent DNA double-strand break (DSB). Using improved time-lapse imaging regimens, we monitor trajectories of tagged DNA loci at a high temporal resolution, which allows us to extract biophysical parameters through robust statistical analysis. Polymer modeling based on these parameters predicts chromatin domain expansion near a DSB and damage extrusion from the domain. Both phenomena are confirmed by live imaging in budding yeast. Calculation of the anomalous exponent of locus movement allows us to differentiate forces imposed on the nucleus through the actin cytoskeleton from those that arise from INO80 remodeler-dependent changes in nucleosome organization. Our analytical approach can be applied to high-density single-locus trajectories obtained in any cell type.

Amitai et al. present a robust analytical workflow for the analysis of single-particle trajectories. The extracted biophysical parameters allow accurate modeling of chromatin dynamics. The authors predict and show that, at double-strand breaks, chromatin decompacts and induces break relocation, while nuclear oscillation arises from cytoskeleton forces.
