In this study, we assessed the immunosuppressive potential of curcumin, a pharmacologically safe and cost-effective naturally occurring polyphenolic phytochemical, on the induction of Th1 cytokines ...that are frequently overexpressed in patients experiencing rejection after renal transplant.
Peripheral blood lymphocytes obtained from 68 renal transplant recipients and 17 healthy controls were treated with curcumin before stimulation with phorbol myristate acetate and were analyzed with flow cytometry for interferon-alpha and interleukin 4 positive cells.
Patients experiencing acute rejection exhibited a high level of interferon-alpha (38.3% +/- 11.2%) and a low level of interleukin 4 (4.2% +/- 2.0%) in their activated peripheral blood lymphocytes. The use of curcumin dose-dependently decreased interferon-alpha induction in cultures from healthy controls (28.1% +/- 4.8%-10.7% +/- 5.3%, P < .001), patients experiencing acute rejection (38.3%-18.3%, P < .001), and those experiencing chronic rejection (40.6%-12.9%, P = .01) when compared with corresponding untreated cultures. In contrast, curcumin exerted only a marginal effect on interleukin 4 expression. Interestingly, curcumin was found to inhibit nuclear factor kappa beta activation by blocking the degradation of the inhibitory unit I kappa B alpha. We also noted the synergistic inhibitory effect of in vitro treatment with curcumin in combination with cyclosporine on the peripheral blood lymphocytes of patients experiencing acute rejection.
These data provide a rationale for the use of curcumin as an affordable, pharmacologically safe, adjuvant immunosuppressant when used with cyclosporine and suggest that curcumin can effectively suppress Th1 cytokine induction after renal transplant.
Background: Breast carcinoma is the most common malignancy in female worldwide, leading cause of death in women. Immunohistochemistry plays a very important role in the prognostication and treatment ...determination of breast carcinoma patients. Objective: The objective of the study was to analyze the immunohistochemical markers in invasive carcinoma of breast and to correlate the expression of hormonal receptors with age of the patient, tumor size, histological grade, and lymph node metastasis. Materials and Methods: The study was conducted on 88 infiltrating ductal breast carcinoma sample in a tertiary care hospital of Southern Assam for a period of 2 year (January2018-December 2019). Data including age, tumor size, and histologic grade and lymph node status retrieved from pathology department. Chi-square was used to determine the statistical significance between estrogen receptors and progesterone receptors (ER/PR) status human epidermal receptor growth factor 2 (HER2/neu) status along with their correlation with various clinicopathological parameters with respect to infiltrating ductal breast carcinoma. Result: The mean age of the patients was 56.6 years. We observed correlation between ER and PR expression with age, tumor size, and tumor grade. There was correlation between HER2/neu expression and age only. None of the markers showed correlation with lymph node involvement (P > 0.05). Conclusion: Our findings showed the importance of biomarkers (ER, PR, and HER2/neu) expression as prognostic factors for therapeutic decision.

Vitiligo is an acquired cutaneous hypomelanosis with a 0.5-2% incidence world wide, without predilection of sex and ethinicity. 16- 35% patients with vitiligo experience significance psychiatric ...morbidity, depression, sleep disturbance, suicidal thoughts, sucidial attempts and anxiety. It also leads to difficulties in forming relationships, avoidance of certain social situations and difficulty in sexual relationships (Porter J et al., 1990). Vitiligo patients present with progressive depigmentation involving progressive loss of melanocytes from skin. The present review will present the accounts of the mechanism and the work done up to till date against vitiligo. This review includes the complete pathomechanism of vitiligo, the treatment available for this disorder. And the drugs recommended for the disorder till date and all the natural products studied till date.

Evidences have suggested that
Tectona grandis (TG) attenuates gastric mucosal injury; however its mechanism has not yet been established. The aim of present study was to evaluate the gastroprotective ...mechanism of ethanolic extract of TG (E-EtOH), butanolic fraction (Fr-Bu) and to identify its active constituents. Anti-ulcer activities were evaluated against cold restraint (CRU) and pyloric ligation (PL) induced gastric ulcer models and further confirmed through H
+ K
+-ATPase inhibitory activity. Cytoprotective activity was evaluated in alcohol (AL) induced gastric ulcer model and further through PGE
2 level. E-EtOH and Fr-Bu attenuated ulcer formation in CRU. Moreover E-EtOH and Fr-Bu displayed potent anti-secretory activity as evident through reduced free acidity and pepsin activity in PL, confirmed further by
in vitro inhibition of H
+ K
+-ATPase activity. In addition cytoprotective potential of E-EtOH and Fr-Bu were apparent with protection in AL model, increased PGE
2 content and enhanced mucin level in PL. Phytochemical investigations of Fr-Bu yielded terpenoides and a phenolic glycoside, verbascoside. The anti-secretory mechanism of verbascoside mediated apparently through inhibition of H
+ K
+-ATPase with corresponding decrease in plasma gastrin level, is novel to our finding. Gastroprotection elicited by TG might be through proton pump inhibition and consequent augmentation of the defensive mechanism.
Compound verbascoside isolated from the butanolic fraction of
Tectona grandis showed significant anti-ulcer activity and this study represents the first report of its anti-secretory potential.

A HP20 resin-based unique method was adopted to get an active fraction of the hydroalcoholic extract of G. glabra roots. The fraction showed potent cytotoxicity against cancer cell line and was ...further subjected to detailed phytochemical investigation to obtain ten biomarkers. The isolated compounds were also tested for the cytotoxicity against the C6 glioma cell line in vitro using MTT assay. Among the isolated compounds, glycyrrhetic acid (1), glabrol (6), and glabridin (9) exhibited significant cytotoxicity. The compounds showed a dose-dependent decrease in cell viability. The active compounds were subjected to molecular docking study against topoisomerase I and topoisomerase II to support the mechanism of antitumor activity.
