The potato rot nematode (Ditylenchus destructor) is a very economically important nematode in agronomic and horticultural plants worldwide. In this study, 43 populations of D. destructor were ...collected from different hosts across China, including 37 populations from Chinese herbal medicine plants. Obtained sequences of ITS-rDNA and D2–D3 of 28S-rDNA genes of D. destructor were compared and analyzed. Nine types of significant length variations in ITS sequences were observed among all populations. The differences in ITS1 length were mainly caused by the presence of repetitive elements with substantial base substitutions. Reconstructions of ITS1 secondary structures showed that the minisatellites formed a stem structure. Ten haplotypes were observed in all populations based on mutations and variations of helix H9. Among them, 3 known haplotypes (A–C) were found in 7 populations isolated from potato, sweet potato, and Codonopsis pilosula, and 7 unique haplotypes were found in other 36 populations collected from C. pilosula and Angelica sinensis compared with 7 haplotypes (A–G) according to Subbotin’ system. These unique haplotypes were different from haplotypes A–G, and we named them as haplotypes H–N. The present results showed that a total of 14 haplotypes (A–N) of ITS-rDNA have been found in D. destructor. Phylogenetic analyses of ITS-rDNA and D2–D3 showed that all populations of D. destructor were clustered into two major clades: one clade only containing haplotype A from sweet potato and the other containing haplotypes B–N from other plants. For further verification, PCR-ITS-RFLP profiles were conducted on 7 new haplotypes. Collectively, our study suggests that D. destructor populations on Chinese medicinal materials are very different from those on other hosts and this work provides a paradigm for relevant researches.
Gaseous disks have been proposed as a mechanism for facilitating mergers of binary black holes. We explore circumbinary disk systems to determine the evolution of the central binary. To do so, we ...perform 3D, hydrodynamic, locally isothermal simulations of circumbinary disks on a Cartesian grid. We focus on binaries of equal mass ratios on fixed circular orbits. To investigate the orbital evolution of the binary, we examine the various torques exerted on the system. For the case where the disk plane and binary orbital plane are aligned, we find that the total torque is positive so that the semimajor axis of the binary increases. For the misaligned case, we run simulations with the binary orbital plane and disk midplane misaligned by 45° and find the same results-the binary grows. The timescale for the circumbinary disk to realign to the plane of the binary is consistent with the global viscous timescale of the disk.
Polyoxometalate is explored as a redox shuttle for non‐electrochromic charge‐balancing material of the conjugated polymer (CP)‐based electrochromic window (ECW). H3PW12O40 is electrochemically ...deposited on a TiO2 nanoparticle film that is coated on a fluorine doped tin oxide glass. The content of PW12 is increased as the TiO2 film is thicker. The PW12‐coated TiO2 film (PWTNF) is applied as a counter electrode (CE) of a CP‐based ECW in an ionic liquid electrolyte, to increase the potential distribution at the CP‐coated working electrode, thereby decreasing the operation voltage of the ECW. This could be ascribed to the function of PW12 as an electron acceptor and redox shuttle that allows effective charge transport from/to the CE and nearby TiO2 nanoparticles. For the optimized ECW with PWTNF as a CE, the maximum optical contrast is obtained at the applied voltage of 1.5 to −2.0 V (68%), along with long cyclability (2500 cycles), bistability (> 2.35 h) and UV stability (> 60 h). The ECW with PWTNF exhibits reproducible capacitive properties, along with high figure of merit for color change with a high power output, in acid‐free electrolyte for the first time.

Enantiomerically enriched indole-containing heterocycles play a vital role in bioscience, medicine, and chemistry. As one of the most attractive subtypes of indole alkaloids, highly substituted ...tetrahydro-γ-carbolines are the basic structural unit in many natural products and pharmaceuticals. However, the syntheses of tetrahydro-γ-carbolines with high functionalities from readily available reagents are significant challenging. In particular, the stereodivergent syntheses of tetrahydro-γ-carbolines containing multi-stereogenic centers remain quite difficult. Herein, we report an expedient and stereodivergent assembly of tetrahydro-γ-carbolines with remarkably high levels of stereoselective control in an efficient cascade process from aldimine esters and indolyl allylic carbonates via a synergistic Cu/Ir catalyst system. Control experiments-guided optimization of synergistic catalysts and mechanistic investigations reveal that a stereodivergent allylation reaction and a subsequent highly stereoselective iso-Pictet-Spengler cyclization are the key elements to success.
FOXM1 (forkhead box protein M1) is a critical proliferation-associated transcription factor that is widely spatiotemporally expressed during the cell cycle. It is closely involved with the processes ...of cell proliferation, self-renewal, and tumorigenesis. In most human cancers, FOXM1 is overexpressed, and this indicates a poor prognosis for cancer patients. FOXM1 maintains cancer hallmarks by regulating the expression of target genes at the transcriptional level. Due to its potential role as molecular target in cancer therapy, FOXM1 was named the Molecule of the Year in 2010. However, the mechanism of FOXM1 dysregulation remains indistinct. A comprehensive understanding of FOXM1 regulation will provide novel insight for cancer and other diseases in which FOXM1 plays a major role. Here, we summarize the transcriptional regulation, post-transcriptional regulation and post-translational modifications of FOXM1, which will provide extremely important implications for novel strategies targeting FOXM1.
Natural killer (NK) cells play a critical role in the innate antitumor immune response. Recently, NK cell dysfunction has been verified in various malignant tumors, including hepatocellular carcinoma ...(HCC). However, the molecular biological mechanisms of NK cell dysfunction in human HCC are still obscure.
The expression of circular ubiquitin-like with PHD and ring finger domain 1 RNA (circUHRF1) in HCC tissues, exosomes, and cell lines was detected by qRT-PCR. Exosomes were isolated from the culture medium of HCC cells and plasma of HCC patients using an ultracentrifugation method and the ExoQuick Exosome Precipitation Solution kit and then characterized by transmission electronic microscopy, NanoSight and western blotting. The role of circUHRF1 in NK cell dysfunction was assessed by ELISA. In vivo circRNA precipitation, RNA immunoprecipitation, and luciferase reporter assays were performed to explore the molecular mechanisms of circUHRF1 in NK cells. In a retrospective study, the clinical characteristics and prognostic significance of circUHRF1 were determined in HCC tissues.
Here, we report that the expression of circUHRF1 is higher in human HCC tissues than in matched adjacent nontumor tissues. Increased levels of circUHRF1 indicate poor clinical prognosis and NK cell dysfunction in patients with HCC. In HCC patient plasma, circUHRF1 is predominantly secreted by HCC cells in an exosomal manner, and circUHRF1 inhibits NK cell-derived IFN-γ and TNF-α secretion. A high level of plasma exosomal circUHRF1 is associated with a decreased NK cell proportion and decreased NK cell tumor infiltration. Moreover, circUHRF1 inhibits NK cell function by upregulating the expression of TIM-3 via degradation of miR-449c-5p. Finally, we show that circUHRF1 may drive resistance to anti-PD1 immunotherapy in HCC patients.
Exosomal circUHRF1 is predominantly secreted by HCC cells and contributes to immunosuppression by inducing NK cell dysfunction in HCC. CircUHRF1 may drive resistance to anti-PD1 immunotherapy, providing a potential therapeutic strategy for patients with HCC.

There is also increasing evidence that microorganisms are used by higher eukaryotes not only for the supply of essential factors like vitamins but also as biological weapons to protect themselves or to kill other organisms. Excellent examples for such systems are entomopathogenic nematodes of the genera

that live in mutualistic symbiosis with bacteria of the genera
Photorhabdus

Xenorhabdus
, respectively. Although these systems have been used successfully in organic farming on an industrial scale, it was only shown during the last 15 years that several different natural products (NPs) produced by the bacteria play key roles in the complex life cycle of the bacterial symbionts, the nematode host and the insect prey that is killed by and provides nutrients for the nematode-bacteria pair. Since the bacteria can switch from mutualistic to pathogenic lifestyle, interacting with two different types of higher eukaryotes, and since the full system with all players can be established in the lab, they are promising model systems to elucidate the natural function of microbial NPs. This review summarizes the current knowledge as well as open questions for NPs from

Xenorhabdus
and tries to assign their roles in the tritrophic relationship.

The Hengduan Mountains (HDM) biodiversity hotspot exhibits exceptional alpine plant diversity. Here, we investigate factors driving intraspecific divergence within a HDM alpine species Salix ...brachista (Cushion willow), a common component of subnival assemblages. We produce a high-quality genome assembly for this species and characterize its genetic diversity, population structure and pattern of evolution by resequencing individuals collected across its distribution. We detect population divergence that has been shaped by a landscape of isolated sky island-like habitats displaying strong environmental heterogeneity across elevational gradients, combined with population size fluctuations that have occurred since approximately the late Miocene. These factors are likely important drivers of intraspecific divergence within Cushion willow and possibly other alpine plants with a similar distribution. Since intraspecific divergence is often the first step toward speciation, the same factors can be important contributors to the high alpine species diversity in the HDM.
Polyoxometalates (POMs) can act as an electron reservoir, giving rise to mixed-valence colored state species without their structural changes. This unique feature of POMs has led to the development ...of POM-based electrochromic devices (ECDs), which show several advantages over organic and inorganic material-based ECDs. For example, the EC properties of POMs can be tuned through molecular design. Furthermore, POMs show high chemical and UV stabilities. In this review, POM structures and the preparation methods for the EC films of POMs were comprehensively introduced. To avoid the dissolution of POM films in electrolyte media or to prepare a homogeneous transparent POM film, a stabilizer was introduced. Several types of stabilizers, including ionic types and carbon materials, were also reviewed for POM films. The EC properties of POMs depend primarily on their structural diversity. Thus, we reviewed the EC properties of different POM films prepared from diverse structures. In addition, we described the EC properties of the POM films that were obtained
via
different preparation methods and exhibited different morphologies. Furthermore, several types of POM-based ECDs were introduced so that not only electrochromic functions but also capacitive functions could be integrated in the ECD. This review may be of interest to researchers in the ECD field devoted to improving the material stability, optical contrast, and design of new POM structures as well as those engaged in the extension of POMs to functional materials.

Myopia is a health issue that has attracted global attention due to its high prevalence and vision-threatening complications. It is well known that the onset and progression of myopia are related to ...both genetic and environmental factors: more than 450 common genetic loci have been found to be associated with myopia, while near work and outdoor time are the main environmental risk factors. As for many complex traits, gene-environment interactions are implicated in myopia development. To date, several genetic loci have been found to interact with near work or educational level. Gene-environment interaction research on myopia could yield models that provide more accurate risk predictions, thus improving targeted treatments and preventive strategies. Additionally, such investigations might have the potential to reveal novel genetic information. In this review, we summarised the findings in this field and proposed some topics for future investigations.
