The use of cocoa pod husk hemicellulose hydrolysate (CPHHH) was evaluated for the production of xylitol by Candida boidinii XM02G yeast isolated from soil of cocoa-growing areas and decaying bark, as ...an alternative means of reusing this type of waste. Xylitol was obtained in concentrations of 11.34 g.L-1, corresponding to a yield (Yp/s) of 0.52 g.g-1 with a fermentation efficiency (ε) of 56.6%. The yeast was tolerant to inhibitor compounds present in CPHHH without detoxification in different concentration factors, and was able to tolerate phenolic compounds at approximately 6 g.L-1. The yeast was also able to metabolize more than 99% (p/v) of furfural and hydroxymethylfurfural present in the non-detoxified CPHHH without extension of the cell-growth lag phase, showing the potential of this microorganism for the production of xylitol. The fermentation of cocoa pod husk hydrolysates appears to provide an alternative use which may reduce the impact generated by incorrect disposal of this waste.

The objective of this study was to describe the profile of bicycle users, their perceived difficulty to cycle, and the most frequent trip origins and destinations in Aracaju, Northeast Brazil. Our ...cross-sectional study sampled 1001 participants and we collected information through structured interviews. Aged ≥15 years, participants were residents of all Aracaju's neighborhoods and used a bicycle for commuting to work or for leisure. We observed that bicycle users in Aracaju are predominantly employed male subjects, aged between 18 and 40 years, and were the heads of their households. Most of the them reported "work" as the main reason for their bicycle trips and, "health" and "practicality" aspects as their main motivations for using bicycles. In general, the neighborhoods in the north and center of the city were identified as the most difficult for cycling, and the easiest trips occurred in places with cycle paths. As a conclusion of this study, we reaffirm the need for intersectoral actions that create favorable environments for active commuting and more sustainable cities.
Coagulation abnormalities in COVID-19 patients have not been addressed in depth.
To perform a longitudinal evaluation of coagulation profile of patients admitted to the ICU with COVID-19.
...Conventional coagulation tests, rotational thromboelastometry (ROTEM), platelet function, fibrinolysis, antithrombin, protein C and S were measured at days 0, 1, 3, 7 and 14. Based on median total maximum SOFA score, patients were divided in two groups: SOFA ≤ 10 and SOFA > 10.
Thirty patients were studied. Some conventional coagulation tests, as aPTT, PT and INR remained unchanged during the study period, while alterations on others coagulation laboratory tests were detected. Fibrinogen levels were increased in both groups. ROTEM maximum clot firmness increased in both groups from Day 0 to Day 14. Moreover, ROTEM-FIBTEM maximum clot firmness was high in both groups, with a slight decrease from day 0 to day 14 in group SOFA ≤ 10 and a slight increase during the same period in group SOFA > 10. Fibrinolysis was low and decreased over time in all groups, with the most pronounced decrease observed in INTEM maximum lysis in group SOFA > 10. Also, D-dimer plasma levels were higher than normal reference range in both groups and free protein S plasma levels were low in both groups at baseline and increased over time, Finally, patients in group SOFA > 10 had lower plasminogen levels and Protein C than patients with SOFA <10, which may represent less fibrinolysis activity during a state of hypercoagulability.
COVID-19 patients have a pronounced hypercoagulability state, characterized by impaired endogenous anticoagulation and decreased fibrinolysis. The magnitude of coagulation abnormalities seems to correlate with the severity of organ dysfunction. The hypercoagulability state of COVID-19 patients was not only detected by ROTEM but it much more complex, where changes were observed on the fibrinolytic and endogenous anticoagulation system.

Usher Syndrome classification takes into account the absence of vestibular function but its correlation with genotype is not well characterized. We intend to investigate whether video Head Impulse ...Test (vHIT) is useful in screening and to differentiate Usher Syndrome types. 29 Usher patients (USH) with a genetically confirmed diagnosis and 30 healthy controls were studied with vHIT and dizziness handicap inventory questionnaire (DHI). Statistical significant differences between USH1, USH2 and controls were found in the vestibulo-ocular-reflex (VOR) gain of all SCCs, with USH1 patients consistently presenting smaller gains. VOR gain of the right lateral SCC could discriminate controls from USH1, and USH2 from USH1 with an overall diagnostic accuracy of 90%. USH1 DHI correlated with VOR (ρ = - 0,971, p = 0.001). Occurrence rate of covert and overt lateral semicircular canals refixation saccades (RS) was significantly different between groups, being higher in USH1 patients (p < 0.001). USH1 peak velocity of covert and overt saccades was higher for lateral semicircular canals (p < 0.05 and p = 0.001) compared with USH2 and controls. Covert saccades occurrence rate for horizontal SCCs could discriminate USH1 from USH2 patients and controls with a diagnostic accuracy of 85%. vHIT is a fast and non-invasive instrument which allowed us to screen and distinguish Usher patients from controls with a high precision. Importantly, its use allowed further discrimination between USH1 from USH2 groups. Moreover, VOR gain seems to correlate with vertigo-related quality of life in more severe phenotypes.
Microalgae cultivation, when compared to the growth of higher plants, presents many advantages such as faster growth, higher biomass productivity, and smaller land area requirement for cultivation. ...For this reason, microalgae are an alternative platform for carotenoid production when compared to the traditional sources. Currently, commercial microalgae production is not well developed but, fortunately, there are several studies aiming to make the large-scale production feasible by, for example, employing different cultivation systems. This review focuses on the main carotenoids from microalgae, comparing them to the traditional sources, as well as a critical analysis about different microalgae cultivation regimes that are currently available and applicable for carotenoid accumulation. Throughout this review paper, we present relevant information about the main commercial microalgae carotenoid producers; the comparison between carotenoid content from food, vegetables, fruits, and microalgae; and the great importance and impact of these molecule applications, such as in food (nutraceuticals and functional foods), cosmetics and pharmaceutical industries, feed (colorants and additives), and healthcare area. Lastly, the different operating systems applied to these photosynthetic cultivations are critically discussed, and conclusions and perspectives are made concerning the best operating system for acquiring high cell densities and, consequently, high carotenoid accumulation.
Antibiotic-induced dysbiosis is a key factor predisposing intestinal infection by Clostridium difficile. Here, we show that interventions that restore butyrate intestinal levels mitigate clinical and ...pathological features of C. difficile-induced colitis. Butyrate has no effect on C. difficile colonization or toxin production. However, it attenuates intestinal inflammation and improves intestinal barrier function in infected mice, as shown by reduced intestinal epithelial permeability and bacterial translocation, effects associated with the increased expression of components of intestinal epithelial cell tight junctions. Activation of the transcription factor HIF-1 in intestinal epithelial cells exerts a protective effect in C. difficile-induced colitis, and it is required for butyrate effects. We conclude that butyrate protects intestinal epithelial cells from damage caused by C. difficile toxins via the stabilization of HIF-1, mitigating local inflammatory response and systemic consequences of the infection.

Fachi et al. demonstrate that butyrate is able to protect the intestinal epithelium from the damage caused by Clostridium difficile toxins by stabilizing HIF-1 and increasing tight junctions, which reduces intestinal epithelial permeability, thus inhibiting intestinal inflammation and bacterial translocation.
The cross-cultural adaptation and validation of the Sniffin`Sticks test for the Portuguese population is described. Over 270 people participated in four experiments. In Experiment 1, 67 participants ...rated the familiarity of presented odors and seven descriptors of the original test were adapted to a Portuguese context. In Experiment 2, the Portuguese version of Sniffin`Sticks test was administered to 203 healthy participants. Older age, male gender and active smoking status were confirmed as confounding factors. The third experiment showed the validity of the Portuguese version of Sniffin`Sticks test in discriminating healthy controls from patients with olfactory dysfunction. In Experiment 4, the test-retest reliability for both the composite score (r71 = 0.86) and the identification test (r71 = 0.62) was established (p<0.001). Normative data for the Portuguese version of Sniffin`Sticks test is provided, showing good validity and reliability and effectively distinguishing patients from healthy controls with high sensitivity and specificity. The Portuguese version of Sniffin`Sticks test identification test is a clinically suitable screening tool in routine outpatient Portuguese settings.

This work cross-correlated rheological, thermodynamic, and conformational features of several natural polysaccharides to their cryoprotective performance. The basis of cryoprotection of FucoPol, ...pectin, and agar revealed a causal combination of (i) an emerging sol–gel transition (p = 0.014) at near-hypothermia (4 °C), (ii) noncolligative attenuated supercooling of the kinetic freezing point of water (p = 0.026) supporting ice growth anticipation, and (iii) increased conformational order (p < 0.0001), where helix-/sheet-like features boost cryoprotection. FucoPol, of highest cryoprotective performance, revealed a predominantly helical structure (α/β = 1.5) capable of forming a gel state at 4 °C and the highest degree of supercooling attenuation (TH = 6.2 °C). Ice growth anticipation with gel-like polysaccharides suggests that the gel matrix neutralizes elastic deformations and lethal cell volumetric fluctuations during freezing, thus preventing the loss of homeostasis and increasing post-thaw viability. Ultimately, structured gels capable of attenuated supercooling enable cryoprotective action at the polymer–cell interface, in addition to polymer–ice interactions. This rationale potentiates implementing alternative, biobased, noncytotoxic polymers in cryobiology.
To determine the association between the number of flares systemic lupus erythematosus (SLE) patients experience and damage accrual, independently of other known risk factors.
SLE patients (34 ...centres, nine Latin American countries) with a recent diagnosis (≤2 years) and ≥3 evaluations were studied. Disease activity was ascertained with the Systemic Lupus Erythematosus Disease Activity Index (SLEDAI) and damage with the SLICC/ACR Damage Index (SDI). Flare was defined as an increase ≥4 points in the SLEDAI between two study visits. An ambidirectional case- crossover design was used to determine the association between the number of flares and damage accrual.
901 patients were eligible for the study; 500 of them (55.5%) experienced at least one flare, being the mean number of flares 0.9 (SD: 1.0). 574 intervals from 251 patients were included in the case-crossover design since they have case and control intervals, whereas, the remaining patients did not. Their mean age at diagnosis was 27.9 years (SD: 11.1), 213 (84.9%) were women. The mean baseline SDI and SLEDAI were 1.3 (1.3) and 13.6 (8.1), respectively. Other features were comparable to those of the entire sample. After adjusting for possible confounding variables, the number of flares, regardless of their severity, was associated with damage accrual (SDI) OR 2.05, 95% CI 1.43 to 2.94, p<0.001 (OR 2.62, 95% CI 1.31 to 5.24, p=0.006 for severe and OR 1.91, 95% CI 1.28 to 2.83, p=0.001 for mild-moderate).

The reintroduction of a species that is extinct in the wild demands caution because reintroduction locations may be associated with threats, such as hunting, poor‐quality habitat, and climate change. ...This is the case for Cyanopsitta spixii (Spix's Macaw), which has been extinct in the wild since 2000. The few living individuals were created in captivity and will be used in a reintroduction project within the species’ original distribution area, the Caatinga domain (Brazil). Because the occurrence records for this bird are old and inaccurate, we investigated the current and future environmental suitability of the 14 plant species used by C. spixii as resource. These plants are key elements for the long‐term reestablishment of the species in the wild, so the use of models helps in the assessment of the effects of climate change on the availability of these resources for the species and informs selection of the best places for reintroduction. We based our models of environmental suitability on 19 bioclimatic variables and nine physical soil and topography variables. Climate projections were created for the present and for the year 2070 with an optimistic (SSP2‐4.5) and a pessimistic (SSP5‐8.5) climate scenario. Both future climate scenarios lead to a reduction in area of environmental suitability that overlapped for all the plant species: 33% reduction for SSP2‐4.5 and 63% reduction for SSP5‐8.5. If our projections materialize, climate change could thus affect the distribution of key resources, and the maintenance of C. spixii would depend on restoration of degraded areas, especially riparian forests, and the preservation of already existing natural areas. The Caatinga domain is very threatened by habitat loss and, for the success of this reintroduction project, the parties involved must act to protect the species and the resources it uses.
Evaluación de los Sitios de Reintroducción de Especies con base en la Futura Idoneidad Climática para los Recursos Alimenticios

La reintroducción de una especie que se encuentra extinta en vida libre exige precaución pues las localidades de reintroducción pueden estar asociadas con amenazas como la cacería, hábitats de mala calidad y el cambio climático. Éste es el caso para Cyanopsitta spixii (Guacamaya de Spix), que ha estado extinta en vida libre desde el 2000. Los pocos individuos vivos nacieron en cautiverio y se usarán para un proyecto de reintroducción dentro del área original de distribución de la especie: el dominio Caatinga (Brasil). Ya que los registros de la presencia de esta ave son viejos e imprecisos, investigamos la idoneidad ambiental actual y a futuro de 14 especies de plantas que C. spixii usa como recurso. Estas plantas son elementos importantes para el restablecimiento a largo plazo de las especies en vida silvestre, así que el uso de modelos asiste en la evaluación de los efectos del cambio climático sobre la disponibilidad de recursos para la especie e informa la selección de los mejores lugares para la reintroducción. Basamos nuestros modelos de la idoneidad ambiental en 19 variables bioclimáticas y 9 variables físicas del suelo y la topografía. Las proyecciones climáticas fueron creadas para el presente y para el año 2070 con un escenario climático optimista (SSP2‐4.5) y uno pesimista (SSP5‐8.5) Ambos escenarios climáticos futuros llevan a una reducción en el área de idoneidad ambiental sobrepuesta para todas las especies de plantas: 33% de reducción para SSP2‐4.5 y 63% de reducción para SSP5‐8.5. Si nuestras proyecciones se materializan, el cambio climático podría entonces afectar la distribución de los recursos importantes, por lo que la conservación de C. spixii dependería de la restauración de las áreas degradadas, especialmente los bosques riparios y la preservación de las áreas naturales existentes. El dominio Caatinga se encuentra muy amenazado por la pérdida del hábitat y, para el éxito de este proyecto de reintroducción, las partes involucradas deben actuar para proteger a la especie y los recursos que utiliza.
