Coffee silverskin (a coffee roasting by-product) contains high amounts of dietary fibre (49% insoluble and 7% soluble) and protein (19%). Potassium (∼5g/100g), magnesium (2g/100g) and calcium (0.6g/100g) are the major macrominerals. The vitamin E profile of silverskin comprises α-tocopherol, β-tocopherol, ɣ-tocopherol, δ-tocopherol, β-tocotrienol, ɣ-tocotrienol, and δ-tocotrienol. The fatty acid profile is mainly saturated (C16:0 and C22:0), but the total amount of fat is low (2.4%). Caffeine (1.25g/100g), chlorogenic acid (246mg/100g), and 5-hydroxymethylfurfural (5.68mg/100g) are also present in silverskin. Total phenolics and flavonoids are partially responsible for the in vitro antioxidant activity. Silverskin extracts protected erythrocytes from oxidative AAPH- and H2O2-induced hemolysis, but at high concentrations a pro-oxidant effect on erythrocyte morphology was observed.

The disruption of a transportation network can have a high social and economic impact on the welfare of a society, as it can significantly affect the daily routines of a community. Although ...many studies have focused on the estimation of physical risk in the components that compose these networks, only a limited number have analyzed their interconnections and impact in the traffic flow. The present study analyzes how earthquake damage can disrupt the road network in an urban environment, and how this will influence the ability of the population to travel. Traffic due to daily commutes is modeled for different layouts of the network, corresponding to possible disruptions caused by earthquake damage. The duration and length of each trip were calculated both for the undamaged network conditions and for the disrupted network. The increase in the median duration and length of each trip allows estimating the economic loss for each event due to drivers' delay. By combining the probability of a specific road being blocked with its number of users, the average number of affected vehicles was estimated, and the most critical segments identified. The methodology was applied to a case study concerning the road network of the area around the Italian city of Messina in Sicily. The results were calculated for both a repetition of the well‐known historical event of 1908 and a set of simulated earthquakes consistent with the national probabilistic seismic hazard model of Italy.
Pulmonary regurgitation is the most common complication in repaired tetralogy of Fallot patients. Severe chronic pulmonary regurgitation can be tolerated for decades, but if not treated, it can ...progress to symptomatic, irreversible right ventricular dilatation and dysfunction. We investigated clinical associations with pulmonary valve replacement among patients with significative pulmonary regurgitation and how interventional developments can change their management.
All adult patients with repaired tetralogy of Fallot who were followed at an adult CHD Clinic at a single centre from 1980 to 2022 were included on their first outpatient visit. Follow-up was estimated from the time of correction surgery until one of the following events occurred first: pulmonary valve replacement, death, loss to follow-up or conclusion of the study.
We included 221 patients (116 males) with a median age of 19 (18-25). At a median age of 33 (10) years old, 114 (51%) patients presented significant pulmonary regurgitation. Among patients with significant pulmonary regurgitation, pulmonary valve replacement was associated with male gender, older age at surgical repair, and longer QRS duration in adulthood. Pulmonary valve replacement was performed in 50 patients, including four transcatheter pulmonary valve implantations, at a median age of 34 (14) years.
Pulmonary regurgitation affects a large percentage of tetralogy of Fallot adult patients, requiring a long-term clinical and imaging follow-up. Sex, age at surgical repair and longer QRS are associated with the need of PVR among patients with significative pulmonary regurgitation. Clinical practice and current literature support TPVI as the future gold standard intervention.
The residue generated from the processing of
(cumbeba) fruit pulp represents a large amount of material that is discarded without proper application. Despite that, it is a raw material that is source ...of ascorbic acid, carotenoids and phenolic compounds, which are valued in nutraceutical diets for allegedly combating free radicals generated in metabolism. This research paper presents a study focused on the mathematical modeling of drying kinetics and the effect of the process on the level of bioactive of cumbeba residue. The experiments of cumbeba residue drying (untreated or whole residue (WR), crushed residue (CR) and residue in the form of foam (FR)) were carried out in a fixed-bed dryer at four air temperatures (50, 60, 70 and 80 °C). Effective water diffusivity (D
) was determined by the inverse method and its dependence on temperature was described by an Arrhenius-type equation. It was observed that, regardless of the type of pretreatment, the increase in air temperature resulted in higher rate of water removal. In relation to bioactive compounds, it was observed that for a fixed temperature the whole residue had higher retention of bioactive compounds, especially phenolic compounds, whereas the crushed residue and the residue in the form of foam had intermediate and lower levels, respectively. This study provides evidence that cumbeba residue in its whole form can be used for the recovery of natural antioxidant bioactive compounds, mainly phenolic compounds, with the possibility of application in the food and pharmaceutical industries.
Despite use of 7-valent pneumococcal conjugate vaccine, incidence of pleural effusion and empyema (pediatric complicated pneumococcal pneumonia PCPP) is reportedly increasing globally. We cultured ...and performed PCR on 152 pleural fluid samples recovered from pediatric patients in Portugal during 2010-2015 to identify and serotype Streptococcus pneumoniae. We identified only 17 cases by culture, but molecular methods identified S. pneumoniae in 68% (92/135) of culture-negative samples. The most frequent serotypes were 3, 1, and 19A, together accounting for 62% (68/109) of cases. Nineteen cases attributable to 13-valent pneumococcal conjugate vaccine (PCV13) serotypes (mostly serotype 3) were detected among 22 children age-appropriately vaccinated with PCV13. The dominance of the additional serotypes included in PCV13 among PCPP cases in Portugal continues, even with PCV13 available on the private market (without reimbursement) since 2010 and with average annual coverage of 61% among age-eligible children. Our data suggest reduced effectiveness of PCV13 against serotype 3 PCPP.

Percutaneous mitral valve commissurotomy (PMC) is a viable alternative to mitral valve (MV) surgery in the treatment of patients with rheumatic mitral stenosis (RMS). In this single-center ...retrospective study of consecutive patients with RMS submitted to PMC from 1991 to 2008, we analyzed clinical, echocardiographic, and hemodynamic data and events during follow-up (FUP) until December 2021. Major adverse cardiovascular events (MACE) were a combined endpoint of all-cause death, cardiovascular hospitalization, and MV re-intervention. A total of 124 patients were enrolled: 108 (87.1%) were female, with a mean age at PMC of 46 standard deviation (SD) 11 years. PMC was successful in 91.1%, with a mean reduction in invasive transmitral pressure gradient (TMPG) of 8 (SD 7) mmHg at PMC time. During the mean FUP of 20 (SD 6) years, 51 (41.1%) patients had MV re-intervention (86.3% surgery and 13.7% redo-PMC), 37 (29.8%) were hospitalized, and 30 (24.2%) died. Approximately 75% of patients remained MACE-free after 10 years, and this percentage decreased to around 40% after 20 years; at this time mark, about 8 in 10 patients were alive. A reduction of <5 mmHg in TMPG at PMC time was associated with a 2.7-fold greater rate of MACE compared to a reduction of ≥5 mmHg, independent of MV regurgitation after PMC and moderate disease of other valves (adjusted hazard ratio 2,7; 95% confidence interval 1.395-5.298, p=0.003). In this cohort with favorable long-term results after PMC, a reduction of <5 mmHg in TMPG at PMC time was associated with MACE during FUP. More studies are needed to validate this independent predictor.
O estágio curricular supervisionado é um momento essencial e deliberativo dos cursos de licenciatura, no qual o docente em formação inicial desenvolve sua capacidade de análise, reflexão e ...investigação em locais e situações reais de exercício da profissão. Esse momento possibilita a construção de uma visão crítica acerca da dinâmica das relações do campo institucional, preparando o futuro docente para o enfrentamento dos problemas que emergem dos contextos concretos do ensino. Assim, neste relato buscamos compreender algumas dimensões envolvidas no processo do estágio curricular supervisionado do Curso de Licenciatura em Ciências da Natureza, da Universidade Federal da Integração Latino- -Americana (organização, atuação dos docentes orientadores e supervisores do estágio, obstáculos, saberes e sentimentos desenvolvidos pelos discentes). Para isso, realizamos uma análise interpretativa das perspectivas manifestadas por 9 licenciandos estagiários em resposta a um questionário. As reflexões proporcionadas a partir da análise fortalecem o entendimento de que o estágio é um momento de construção da identidade profissional e de prática reflexiva, em que o discente visualiza problemas reais de sua profissão e se conscientiza da necessidade de se buscar e desenvolver constantemente novos saberes profissionais, além de aprender mecanismos para lidar com sentimentos, como de medo e ansiedade, associados ao início da profissão.

In the last years, sustainable practices have been developed to minimize the negative effects of production and excessive consumption on the environment. The textile and clothing industry is one of ...the most polluting industries globally and needs to rethink its strategies. The fast-fashion caused an increase in production, and the environmental weight associated with the textile industry also increased. The problems range from the enormous expenditure of water resources to the carbon and greenhouse gas emissions to reaching the consumer. This review focuses on the eco-friendly approaches taken by the industry towards supportable apparel manufacturing, from the choice of raw materials to the last step in the textile industry.
Mesenchymal stromal cells (MSC) hold great promise for tissue engineering and cell-based therapies due to their multilineage differentiation potential and intrinsic immunomodulatory and trophic ...activities. Over the past years, increasing evidence has proposed extracellular vesicles (EVs) as mediators of many of the MSC-associated therapeutic features. EVs have emerged as mediators of intercellular communication, being associated with multiple physiological processes, but also in the pathogenesis of several diseases. EVs are derived from cell membranes, allowing high biocompatibility to target cells, while their small size makes them ideal candidates to cross biological barriers. Despite the promising potential of EVs for therapeutic applications, robust manufacturing processes that would increase the consistency and scalability of EV production are still lacking. In this work, EVs were produced by MSC isolated from different human tissue sources bone marrow (BM), adipose tissue (AT), and umbilical cord matrix (UCM). A serum-/xeno-free microcarrier-based culture system was implemented in a Vertical-Wheel
TM
bioreactor (VWBR), employing a human platelet lysate culture supplement (UltraGRO
TM
-PURE), toward the scalable production of MSC-derived EVs (MSC-EVs). The morphology and structure of the manufactured EVs were assessed by atomic force microscopy, while EV protein markers were successfully identified in EVs by Western blot, and EV surface charge was maintained relatively constant (between −15.5 ± 1.6 mV and −19.4 ± 1.4 mV), as determined by zeta potential measurements. When compared to traditional culture systems under static conditions (T-flasks), the VWBR system allowed the production of EVs at higher concentration (i.e., EV concentration in the conditioned medium) (5.7-fold increase overall) and productivity (i.e., amount of EVs generated per cell) (3-fold increase overall). Overall, we demonstrate that this scalable culture system can robustly manufacture EVs from MSC derived from different tissue sources, toward the development of novel therapeutic products.
