Metabolic complications in an obese state can be aggravated by an abnormal inflammatory response and enhanced production of reactive oxygen species. Pro-inflammatory response is known to be ...associated with the formation of toxic reactive oxygen species and subsequent generation of oxidative stress. Indeed, adipocytes from obese individuals display an altered adipokine profile, with upregulated expression and secretion of pro-inflammatory cytokines such as tumor necrosis factor alpha (TNF-α) and interleukin (IL-6). Interestingly, natural compounds, including phenolic enriched foods are increasingly explored for their ameliorative effects against various metabolic diseases. Of interest is gallic acid, a trihydroxybenzoic acid that has progressively demonstrated robust anti-obesity capabilities in various experimental models. In addition to reducing excessive lipid storage in obese subjects, gallic acid has been shown to specifically target the adipose tissue to suppress lipogenesis, improve insulin signaling, and concomitantly combat raised pro-inflammatory response and oxidative stress. This review will revise mechanisms involved in the pathophysiological effects of inflammation and oxidative stress in an obese state. To better inform on its therapeutic potential and improvement of human health, available evidence reporting on the anti-obesity properties of gallic acid and its derivatives will be discussed, with emphases on its modulatory effect on molecular mechanisms involved in insulin signaling, inflammation and oxidative stress.
Parkinson’s disease (PD) is the second most prevalent neurodegenerative disorder currently affecting the ageing population. Although the aetiology of PD has yet to be fully elucidated, environmental ...factors such as exposure to the naturally occurring neurotoxin rotenone has been associated with an increased risk of developing PD. Rotenone inhibits mitochondrial respiratory chain (MRC) complex I activity as well as induces dopaminergic neuronal death. The aim of the present study was to investigate the underlying mechanisms of rotenone-induced mitochondrial dysfunction and oxidative stress in an in vitro SH-SY5Y neuronal cell model of PD and to assess the ability of pre-treatment with Coenzyme Q10 (CoQ10) to ameliorate oxidative stress in this model. Spectrophotometric determination of the mitochondrial enzyme activities and fluorescence probe studies of reactive oxygen species (ROS) production was assessed. Significant inhibition of MRC complex I and II–III activities was observed, together with a significant loss of neuronal viability, CoQ10 status, and ATP synthesis. Additionally, significant increases were observed in intracellular and mitochondrial ROS production. Remarkably, CoQ10 supplementation was found to reduce ROS formation. These results have indicated mitochondrial dysfunction and increased oxidative stress in a rotenone-induced neuronal cell model of PD that was ameliorated by CoQ10 supplementation.
Menaquinone-7 (MK7) is a member of the vitamin K family in which interest has considerably increased over the last decade, mainly due to its beneficial role in human health. MK7 can be produced by ...synthesis or fermentation, and its purity profile can differ depending on methodologies and extraction procedures. Finished formulations show a high heterogeneity of purity profiles, as well as frequent discrepancies in the nominal content, compared to the actual title. The present study compared purity profiles of different raw material and related them to their stability in normal (12 months/25 °C/60%RH) and accelerated conditions (6 months/40 °C/75% RH) in order to test their performance in the presence of different common excipients. Results showed higher purity profile results in enhanced stability, and this could explain title discrepancies found in finished products, which are present on the market worldwide.
Coastal salt marshes are valuable and critical components of tidal landscapes, currently threatened by increasing rates of sea level rise, wave-induced lateral erosion, decreasing sediment supply, ...and human pressure. Halophytic vegetation plays an important role in salt-marsh erosional and depositional patterns and marsh survival. Mapping salt-marsh halophytic vegetation species and their fractional abundance within plant associations can provide important information on marsh vulnerability and coastal management. Remote sensing has often provided valuable methods for salt-marsh vegetation mapping; however, it has seldom been used to assess the fractional abundance of halophytes. In this study, we developed and tested a novel approach to estimate fractional abundance of halophytic species and bare soil that is based on Random Forest (RF) soft classification. This approach can fully use the information contained in the frequency of decision tree “votes” to estimate fractional abundance of each species. Such a method was applied to WorldView-2 (WV-2) data acquired for the Venice lagoon (Italy), where marshes are characterized by a high diversity of vegetation species. The proposed method was successfully tested against field observations derived from ancillary field surveys. Our results show that the new approach allows one to obtain high accuracy (6.7% < root-mean-square error (RMSE) < 18.7% and 0.65 < R2 < 0.96) in estimating the sub-pixel fractional abundance of marsh-vegetation species. Comparing results obtained with the new RF soft-classification approach with those obtained using the traditional RF regression method for fractional abundance estimation, we find a superior performance of the novel RF soft-classification approach with respect to the existing RF regression methods. The distribution of the dominant species obtained from the RF soft classification was compared to the one obtained from an RF hard classification, showing that numerous mixed areas are wrongly labeled as populated by specific species by the hard classifier. As for the effectiveness of using WV-2 for salt-marsh vegetation mapping, feature importance analyses suggest that Yellow (584–632 nm), NIR 1 (near-infrared 1, 765–901 nm) and NIR 2 (near-infrared 2, 856–1043 nm) bands are critical in RF soft classification. Our results bear important consequences for mapping and monitoring vegetation-species fractional abundance within plant associations and their dynamics, which are key aspects in biogeomorphic analyses of salt-marsh landscapes.
Multiple stable states are established in coastal tidal wetlands (marshes, mangroves, deltas, seagrasses) by ecological, hydrological, and geomorphological feedbacks. Catastrophic shifts between ...states can be induced by gradual environmental change or by disturbance events. These feedbacks and outcomes are key to the sustainability and resilience of vegetated coastlines, especially as modulated by human activity, sea level rise, and climate change. Whereas multiple stable state theory has been invoked to model salt marsh responses to sediment supply and sea level change, there has been comparatively little empirical verification of the theory for salt marshes or other coastal wetlands. Especially lacking is long-term evidence documenting if or how stable states are established and maintained at ecosystem scales. Laboratory and field-plot studies are informative, but of necessarily limited spatial and temporal scope. For the purposes of long-term, coastal-scale monitoring, remote sensing is the best viable option. This review summarizes the above topics and highlights the emerging promise and challenges of using remote sensing-based analyses to validate coastal wetland dynamic state theories. This significant opportunity is further framed by a proposed list of scientific advances needed to more thoroughly develop the field.

) suggest that Landsat 8 OLI can be used to map and monitor soil salinity in the coastal zone of the Mekong ...River Delta. The authors use empirical correlations between the near-infrared (NIR) band, or vegetation indexes containing the NIR band, and soil salinity. We show that within the coastal portion of the Mekong Delta, extensively ponded due to widespread shrimp farming, about 90% of Landsat 8 pixels are fully or partially covered by water. We then find that, due to strong NIR radiation absorption, NIR reflectance from ponded pixels decreases linearly with increasing water percentage cover, while no significant correlation is found between reflectance and soil salinity. Through detailed new analyses, we conclude that NIR reflectance attenuation cannot be ascribed to vegetation stress caused by soil salinity, but rather to the presence of water ponds. We also show that a similar behavior exists in ponded freshwater inland areas, confirming that the NIR absorption exerted by water is independent of salinity.
Evidence on the beneficial effects of resveratrol supplementation on cardiovascular disease-related profiles in patients with type 2 diabetes (T2D) is conflicting, while its impact on renal function ...and blood pressure measurements remains to be established in these patients. The current meta-analysis included randomized controlled trials (RCTs) reporting on the impact of resveratrol supplementation on markers of renal function and blood pressure in patients with T2D on hypoglycemic medication. Electronic databases such as MEDLINE, Cochrane Library, Scopus, and EMBASE were searched for eligible studies from inception up to June 2020. The random and fixed effects model was used in the meta-analysis. A total of five RCTs met the inclusion criteria and involved 388 participants with T2D. Notably, most of the participants were on metformin therapy, or metformin in combination with other hypoglycemic drugs such as insulin and glibenclamide. Pooled estimates showed that resveratrol supplementation in patients with T2D lowered the levels of fasting glucose (SMD: -0.06 95% CI: -0.24, 0.12;
= 4%, p = 0.39) and insulin (SMD: -0.08 95% CI: -0.50, 0.34,
= 73%, p = 0.002) when compared to those on placebo. In addition, supplementation significantly lowered systolic blood pressure (SMD: -5.77 95% CI: -8.61, -2.93,
= 66%, p = 0.02) in these patients. Although resveratrol supplementation did not affect creatinine or urea levels, it reduced the total protein content (SMD: -0.19 95% CI: -0.36, -0.02;
= 91%, p = 0.001). In all, resveratrol supplementation in hypoglycemic therapy improves glucose control and lowers blood pressure; however, additional evidence is necessary to confirm its effect on renal function in patients with T2D.
Objectives: Physical exercise significantly impacts the biochemistry of the organism. Ubiquinone is a key component of the mitochondrial respiratory chain and ubiquinol, its reduced and active form, ...is an emerging molecule in sport nutrition. The aim of this study was to evaluate the effect of ubiquinol supplementation on biochemical and oxidative stress indexes after an intense bout of exercise.
Methods: 21 male young athletes (26 + 5 years of age) were randomized in two groups according to a double blind cross-over study, either supplemented with ubiquinol (200 mg/day) or placebo for 1 month. Blood was withdrawn before and after a single bout of intense exercise (40 min run at 85% maxHR). Physical performance, hematochemical parameters, ubiquinone/ubiquinol plasma content, intracellular reactive oxygen species (ROS) level, mitochondrial membrane depolarization, paraoxonase activity and oxidative DNA damage were analyzed.
Results: A single bout of intense exercise produced a significant increase in most hematochemical indexes, in particular CK and Mb while, on the contrary, normalized coenzyme Q
10
plasma content decreased significantly in all subjects. Ubiquinol supplementation prevented exercise-induced CoQ deprivation and decrease in paraoxonase activity. Moreover at a cellular level, in peripheral blood mononuclear cells, ubiquinol supplementation was associated with a significant decrease in cytosolic ROS while mitochondrial membrane potential and oxidative DNA damage remained unchanged.
Discussion: Data highlights a very rapid dynamic of CoQ depletion following intense exercise underlying an increased demand by the organism. Ubiquinol supplementation minimized exercise-induced depletion and enhanced plasma and cellular antioxidant levels but it was not able to improve physical performance indexes or markers of muscular damage.

There is an increasing interest in the use of nutraceuticals and plant-derived bioactive compounds from foods for their potential health benefits. For example, as a major active ingredient found from ...cruciferous vegetables like broccoli, there has been growing interest in understanding the therapeutic effects of sulforaphane against diverse metabolic complications. The past decade has seen an extensive growth in literature reporting on the potential health benefits of sulforaphane to neutralize pathological consequences of oxidative stress and inflammation, which may be essential in protecting against diabetes-related complications. In fact, preclinical evidence summarized within this review supports an active role of sulforaphane in activating nuclear factor erythroid 2-related factor 2 or effectively modulating AMP-activated protein kinase to protect against diabetic complications, including diabetic cardiomyopathy, diabetic neuropathy, diabetic nephropathy, as well as other metabolic complications involving non-alcoholic fatty liver disease and skeletal muscle insulin resistance. With clinical evidence suggesting that foods rich in sulforaphane like broccoli can improve the metabolic status and lower cardiovascular disease risk by reducing biomarkers of oxidative stress and inflammation in patients with type 2 diabetes. This information remains essential in determining the therapeutic value of sulforaphane or its potential use as a nutraceutical to manage diabetes and its related complications. Finally, this review discusses essential information on the bioavailability profile of sulforaphane, while also covering information on the pathological consequences of oxidative stress and inflammation that drive the development and progression of diabetes.

Aspalathin is a rooibos flavonoid with established blood glucose lowering properties, however, its efficacy to moderate complications associated with hepatic insulin resistance is unknown. To study ...such effects, C3A liver cells exposed to palmitate were used as a model of hepatic insulin resistance. These hepatocytes displayed impaired substrate metabolism, including reduced glucose transport and free fatty acid uptake. These defects included impaired insulin signaling, evident through reduced phosphatidylinositol-4,5-bisphosphate 3-kinase/ protein kinase B (PI3K/AKT) protein expression, and mitochondrial dysfunction, depicted by a lower mitochondrial respiration rate. Aspalathin was able to ameliorate these defects by correcting altered substrate metabolism, improving insulin signaling and mitochondrial bioenergetics. Activation of 5'-adenosine monophosphate-activated protein kinase (AMPK) may be a plausible mechanism by which aspalathin increases hepatic energy expenditure. Overall, these results encourage further studies assessing the potential use of aspalathin as a nutraceutical to improve hepatocellular energy expenditure, and reverse metabolic disease-associated complications.
