Individuals with chronic lung disease (eg, cystic fibrosis (CF)) often receive antimicrobial therapy including aminoglycosides resulting in ototoxicity. Extended high-frequency audiometry has ...increased sensitivity for ototoxicity detection, but diagnostic audiometry in a sound-booth is costly, time-consuming and requires a trained audiologist. This cross-sectional study analysed tablet-based audiometry (Shoebox MD) performed by non-audiologists in an outpatient setting, alongside home web-based audiometry (3D Tune-In) to screen for hearing loss in adults with CF.
Hearing was analysed in 126 CF adults using validated questionnaires, a web self-hearing test (0.5 to 4 kHz), tablet (0.25 to 12 kHz) and sound-booth audiometry (0.25 to 12 kHz). A threshold of ≥25 dB hearing loss at ≥1 audiometric frequency was considered abnormal. Demographics and mitochondrial DNA sequencing were used to analyse risk factors, and accuracy and usability of hearing tests determined.
Prevalence of hearing loss within any frequency band tested was 48%. Multivariate analysis showed age (OR 1.127; (95% CI: 1.07 to 1.18; p value<0.0001) per year older) and total intravenous antibiotic days over 10 years (OR 1.006; (95% CI: 1.002 to 1.010; p value=0.004) per further intravenous day) were significantly associated with increased risk of hearing loss. Tablet audiometry had good usability, was 93% sensitive, 88% specific with 94% negative predictive value to screen for hearing loss compared with web self-test audiometry and questionnaires which had poor sensitivity (17% and 13%, respectively). Intraclass correlation (ICC) of tablet versus sound-booth audiometry showed high correlation (ICC >0.9) at all frequencies ≥4 kHz.
Adults with CF have a high prevalence of drug-related hearing loss and tablet-based audiometry can be a practical, accurate screening tool within integrated ototoxicity monitoring programmes for early detection.

We review lattice results related to pion, kaon, D-meson, B-meson, and nucleon physics with the aim of making them easily accessible to the nuclear and particle physics communities. More ...specifically, we report on the determination of the light-quark masses, the form factor f+(0) arising in the semileptonic K→π transition at zero momentum transfer, as well as the decay constant ratio fK/fπ and its consequences for the CKM matrix elements Vus and Vud. Furthermore, we describe the results obtained on the lattice for some of the low-energy constants of SU(2)L×SU(2)R and SU(3)L×SU(3)R Chiral Perturbation Theory. We review the determination of the BK parameter of neutral kaon mixing as well as the additional four B parameters that arise in theories of physics beyond the Standard Model. For the heavy-quark sector, we provide results for mc and mb as well as those for D- and B-meson decay constants, form factors, and mixing parameters. These are the heavy-quark quantities most relevant for the determination of CKM matrix elements and the global CKM unitarity-triangle fit. We review the status of lattice determinations of the strong coupling constant αs. Finally, in this review we have added a new section reviewing results for nucleon matrix elements of the axial, scalar and tensor bilinears, both isovector and flavor diagonal.

Phytophthora ramorum (Oomycetes) is an emerging plant pathogen in forests in southwestern Oregon (Curry County). Moreover, since 2003 it has been repeatedly isolated from plants in Oregon nurseries. ...In this study, we analysed the genetic diversity of the P. ramorum population in Oregon from 2001 to 2004 by using microsatellites. A total of 323 isolates (272 from the infested forest; 51 from nurseries) were screened at 10 loci. The overall P. ramorum population in Oregon is characterized by low genetic diversity and has all the hallmarks of an introduced organism. All isolates within the A2 mating type belonged to the same clonal lineage and no recombinant genotypes were found. The forest population (24 genotypes) was dominated by a single multilocus genotype which persisted over years, indicating that eradication efforts in the forest have not completely eliminated inoculum sources. In contrast, genotypic evidence suggests that eradication was effective in nurseries. In 2003 and 2004, a total of 11 genotypes were found in the nurseries (one belonged to the European lineage of P. ramorum) but no genotype was recovered in both sampling years. Significant differentiation and low gene flow were detected between nursery and forest populations. Only two nursery genotypes were also found in the forest, and then at low frequency. Thus, the nursery infestation is not caused by the genotypes observed in Curry County, but likely resulted through introduction of novel genotypes from nurseries out-of-state. This highlights the continued importance of sanitation and quarantine in nurseries to prevent further introduction and spread of P. ramorum.
Using HISQ $N_f=2+1+1$ MILC ensembles with five different values of the lattice spacing, including four ensembles with physical quark masses, we have performed the most precise computation to date of ...the $K opiell u$ vector form factor at zero momentum transfer, $f_+^{K^0pi^-}(0)=0.9696(15)_ ext{stat}(12)_ ext{syst}$. This is the first calculation that includes the dominant finite-volume effects, as calculated in chiral perturbation theory at next-to-leading order. Our result for the form factor provides a direct determination of the Cabibbo-Kobayashi-Maskawa matrix element $|V_{us}|=0.22333(44)_{f_+(0)}(42)_ ext{exp}$, with a theory error that is, for the first time, at the same level as the experimental error. The uncertainty of the semileptonic determination is now similar to that from leptonic decays and the ratio $f_{K^+}/f_{pi^+}$, which uses $|V_{ud}|$ as input. Our value of $|V_{us}|$ is in tension at the 2--$2.6sigma$ level both with the determinations from leptonic decays and with the unitarity of the CKM matrix. In the test of CKM unitarity in the first row, the current limiting factor is the error in $|V_{ud}|$, although a recent determination of the nucleus-independent radiative corrections to superallowed nuclear $ eta$ decays could reduce the $|V_{ud}|^2$ uncertainty nearly to that of $|V_{us}|^2$. Alternative unitarity tests using only kaon decays, for which improvements in the theory and experimental inputs are likely in the next few years, reveal similar tensions. As part of our analysis, we calculated the correction to $f_+^{Kpi}(0)$ due to nonequilibrated topological charge at leading order in chiral perturbation theory, for both the full-QCD and the partially-quenched cases. We also obtain the combination of low-energy constants in the chiral effective Lagrangian $C_{12}^r+C_{34}^r-(L_5^r)^2(M_ ho)=(2.92pm0.31)cdot10^{-6}$.
Pitt‐Hopkins syndrome (PTHS) is a rare neurodevelopmental disorder caused by mutations of the transcription factor 4 (Tcf4) gene. Individuals with PTHS often suffer from severe abdominal bloating and ...constipation. In this short communication, we discuss two individuals with PTHS who died unexpectedly due to gastrointestinal complications. We aim to increase awareness among healthcare professionals who care for individuals with PTHS, to ensure adequate screening and management of gastrointestinal symptoms in this population. Moreover, we discuss how fatal gastrointestinal complications may be related to PTHS and provide an overview of the literature.
In Europe, most reported human cases of babesiosis have been attributed, without strong molecular evidence, to infection with the bovine parasite Babesia divergens. We investigated the first known ...human cases of babesiosis in Italy and Austria, which occurred in two asplenic men. The complete 18S ribosomal RNA (18S rRNA) gene was amplified from specimens of their whole blood by polymerase chain reaction (PCR). With phylogenetic analysis, we compared the DNA sequences of the PCR products with those for other Babesia spp. The DNA sequences were identical for the organism from the two patients. In phylogenetic analysis, the organism clusters with B. odocoilei, a parasite of white-tailed deer; these two organisms form a sister group with B. divergens. This evidence indicates the patients were not infected with B. divergens but with an organism with previously unreported molecular characteristics for the 18S rRNA gene.

We use lattice QCD to calculate the form factors f+(q2) and f0(q2) for the semileptonic decay Bs→Kℓν. Our calculation uses six MILC asqtad 2+1 flavor gauge-field ensembles with three lattice ...spacings. At the smallest and largest lattice spacing the light-quark sea mass is set to 1/10 the strange-quark mass. At the intermediate lattice spacing, we use four values for the light-quark sea mass ranging from 1/5 to 1/20 of the strange-quark mass. We use the asqtad improved staggered action for the light valence quarks, and the clover action with the Fermilab interpolation for the heavy valence bottom quark. We use SU(2) hard-kaon heavy-meson rooted staggered chiral perturbation theory to take the chiral-continuum limit. A functional z expansion is used to extend the form factors to the full kinematic range. We present predictions for the differential decay rate for both Bs→Kμν and Bs→Kτν. We also present results for the forward-backward asymmetry, the lepton polarization asymmetry, ratios of the scalar and vector form factors for the decays Bs→Kℓν and Bs→Dsℓν. Our results, together with future experimental measurements, can be used to determine the magnitude of the Cabibbo-Kobayashi-Maskawa matrix element |$V_{ub}$|.
We report the status of an ongoing lattice-QCD calculation of form factors for exclusive semileptonic decays of B mesons with both charged currents (

). The results are important for constraining or revealing physics beyond the Standard Model. This work uses MILC’s (2+1 + 1)-flavor ensembles with the HISQ action for the sea and light valence quarks and the clover action in the Fermilab interpretation for the

quark. Simulations are carried out at three lattice spacings down to 0.088 fm, with both physical and unphysical sea-quark masses. We present preliminary results for correlation-function fits.
