Scania has been working with statistics for a long time but has invested in becoming a data driven company more recently and uses data science in almost all business functions. The algorithms ...developed by the data scientists need to be optimized to be fully utilized and traditionally this is a manual and time consuming process. What this thesis investigates is if and how well evolutionary algorithms can be used to automate the optimization process. The evaluation was done by implementing and analyzing four variations of genetic algorithms with different levels of complexity and tuning parameters. The algorithm subject to optimization was XGBoost, a gradient boosted tree model, applied to data that had previously been modelled in a competition. The results show that evolutionary algorithms are applicable in finding good models but also emphasizes the importance of proper data preparation.

De algoritmer som utvecklas av data scientists måste optimeras för att kunna utnyttjas till fullo och detta är traditionellt sett en manuell och tidskrävade process. Detta examensarbete utreder om och hur väl evolutionära algoritmer kan användas för att automatisera optimeringsprocessen. Utvärderingen gjordes genom att implementera och analysera fyra varianter avgenetiska algoritmer med olika grader av komplexitet och trimningsparameterar. Algoritmen som var målet för optimering var XGBoost, som är en gradient boosted trädbaserad modell. Denna applicerades på data som tidigare hade modellerats i entävling. Resultatet visar att evolutionära algoritmer är applicerbara i att hitta bra modellermen påvisar även hur fundamentalt det är att arbeta med databearbetning innan modellering.
Denna studie undersöker aktierekommendationer publicerade av svenska journalister anställda på två av Sveriges största affärstidningar. Studien syftar till att undersöka huruvida dessa ...rekommendationer leder till en abnormal avkastning vid publiceringen, samt om den abnormala avkastningen sedan består eller faller tillbaka, för att kunna dra slutsatser kring relevansen av rekommendationen. För att undersöka detta har en eventstudie utförts där marknadsmodellen har använts för att beräkna den abnormala avkastningen. Resultaten visar på att aktierekommendationerna leder till en statistiskt signifikant reaktion på publiceringsdagen där det uppmättes en abnormal avkastning med ett genomsnitt på 1,66 %. Vidare så visade resultaten på att den abnormala avkastningen bestod, prisnivån höll sig kvar på den nya nivån de efterföljande 20 dagarna. Detta tyder på att det finns en relevans och ny information i aktierekommendationerna. Vid test att utesluta säljrekommendationer från urvalet erhölls liknande resultat. För att kunna dra mer generella slutsatser om den svenska marknaden skulle det behövas ett större urval av rekommendationer samt ha med fler faktorer såsom handelsvolym i framtida forskning.

Arbetet i skadeområdet styrs ...av tydliga föreskrifter och rutiner. Då allvarlig händelse inträffar relativt sällan kan ambulanspersonalen uppleva en osäkerhet att samverka på skadeområdet. Inom diskursetiken ges det goda argumentet företräde framför maktförhållanden och manipulation i syfte att nå enighet och rationell förståelse. Patientperspektivet ska genomsyra de beslut som fattas i skadeområdet vilket möjliggörs genom enighet mellan de samverkande organisationerna. Syfte: Att undersöka ambulanspersonalens erfarenhet att samverka med polis och räddningstjänst i skadeområde. Metod: Studien utfördes genom en integrativ litteraturstudie. Systematiska databassökningar genomfördes i databaserna PubMed, CINAHL och PsycINFO. Resultatet baseras på 15 vetenskapliga artiklar som bedöms vara av måttlig till hög kvalitet. Analysen genomfördes enligt Whittemore och Knafls (2005) metodbeskrivning. Resultat: Samverkan mellan organisationerna innebar utmaningar i att skapa balans mellan det egna och det gemensamma ansvaret. Ledarskapet hade i hög grad inverkan på arbetet i skadeområdet, men att iklä sig rollen som sjukvårdsledare förknippades ofta med osäkerhet hos ambulanspersonalen. Kunskap, kommunikation och en känsla av tillhörighet skapade förutsättningar för samverkan mellan organisationerna. Resultatet presenteras under fyra huvudteman: "Vi" och "dem", Härska utan att söndra, Konsten att kommunicera och Övning ger färdighet. Konklusion: Förmågan att kommunicera och att ha förståelse för varandra skapar förutsättningar för samverkan mellan ambulans, polis och räddningstjänst. Genes in the Ras pathway have somatic mutations in at least 60 % of colorectal cancers. Despite activating the same pathway, the BRAF V600E mutation and the prevalent mutations in codon 12 and 13 of ...KRAS have all been linked to different clinical outcomes, but the molecular mechanisms behind these differences largely remain to be clarified.
To characterize the similarities and differences between common activating KRAS mutations and between KRAS and BRAF mutations, we used genome editing to engineer KRAS G12C/D/V and G13D mutations in colorectal cancer cells that had their mutant BRAF V600E allele removed and subjected them to transcriptome sequencing, global proteomics and metabolomics analyses.
By intersecting differentially expressed genes, proteins and metabolites, we uncovered (i) two-fold more regulated genes and proteins when comparing KRAS to BRAF mutant cells to those lacking Ras pathway mutation, (ii) five differentially expressed proteins in KRAS mutants compared to cells lacking Ras pathway mutation (IFI16, S100A10, CD44, GLRX and AHNAK2) and 6 (CRABP2, FLNA, NXN, LCP1, S100A10 and S100A2) compared to BRAF mutant cells, (iii) 19 proteins expressed differentially in a KRAS mutation specific manner versus BRAF V600E cells, (iv) regulation of the Integrin Linked Kinase pathway by KRAS but not BRAF mutation, (v) regulation of amino acid metabolism, particularly of the tyrosine, histidine, arginine and proline pathways, the urea cycle and purine metabolism by Ras pathway mutations, (vi) increased free carnitine in KRAS and BRAF mutant RKO cells.
This comprehensive integrative -omics analysis confirms known and adds novel genes, proteins and metabolic pathways regulated by mutant KRAS and BRAF signaling in colorectal cancer. The results from the new model systems presented here can inform future development of diagnostic and therapeutic approaches targeting tumors with KRAS and BRAF mutations.
Multigene expression signatures provide a molecular subdivision of early breast cancer associated with patient outcome. A gap remains in the validation of such signatures in clinical treatment groups ...of patients within population-based cohorts of unselected primary breast cancer representing contemporary disease stages and current treatments. A cohort of 3520 resectable breast cancers with RNA sequencing data included in the population-based SCAN-B initiative (ClinicalTrials.gov ID NCT02306096) were selected from a healthcare background population of 8587 patients diagnosed within the years 2010-2015. RNA profiles were classified according to 19 reported gene signatures including both gene expression subtypes (e.g. PAM50, IC10, CIT) and risk predictors (e.g. Oncotype DX, 70-gene, ROR). Classifications were analyzed in nine adjuvant clinical assessment groups: TNBC-ACT (adjuvant chemotherapy, n = 239), TNBC-untreated (n = 82), HER2+/ER- with anti-HER2+ ACT treatment (n = 110), HER2+/ER+ with anti-HER2 + ACT + endocrine treatment (n = 239), ER+/HER2-/LN- with endocrine treatment (n = 1113), ER+/HER2-/LN- with endocrine + ACT treatment (n = 243), ER+/HER2-/LN+ with endocrine treatment (n = 423), ER+/HER2-/LN+ with endocrine + ACT treatment (n = 433), and ER+/HER2-/LN- untreated (n = 200). Gene signature classification (e.g., proportion low-, high-risk) was generally well aligned with stratification based on current immunohistochemistry-based clinical practice. Most signatures did not provide any further risk stratification in TNBC and HER2+/ER- disease. Risk classifier agreement (low-, medium/intermediate-, high-risk groups) in ER+ assessment groups was on average 50-60% with occasional pair-wise comparisons having <30% agreement. Disregarding the intermediate-risk groups, the exact agreement between low- and high-risk groups was on average ~80-95%, for risk prediction signatures across all assessment groups. Outcome analyses were restricted to assessment groups of TNBC-ACT and endocrine treated ER+/HER2-/LN- and ER+/HER2-/LN+ cases. For ER+/HER2- disease, gene signatures appear to contribute additional prognostic value even at a relatively short follow-up time. Less apparent prognostic value was observed in the other groups for the tested signatures. The current study supports the usage of gene expression signatures in specific clinical treatment groups within population-based breast cancer. It also stresses the need of further development to reach higher consensus in individual patient classifications, especially for intermediate-risk patients, and the targeting of patients where current gene signatures and prognostic variables provide little support in clinical decision-making.
Purpose
Oestrogen receptor-positive (ER+) and human epidermal receptor 2-negative (HER2–) breast cancers are classified as Luminal A or B based on gene expression, but immunohistochemical markers are ...used for surrogate subtyping. The aims of this study were to examine the agreement between molecular subtyping (MS) and surrogate subtyping and to identify subgroups consisting mainly of Luminal A or B tumours.
Methods
The cohort consisted of 2063 patients diagnosed between 2013–2017, with primary ER+/HER2– breast cancer, analysed by RNA sequencing. Surrogate subtyping was performed according to three algorithms (St. Gallen 2013, Maisonneuve and our proposed Grade-based classification). Agreement (%) and kappa statistics (κ) were used as concordance measures and ROC analysis for luminal distinction. Ki67, progesterone receptor (PR) and histological grade (HG) were further investigated as surrogate markers.
Results
The agreement rates between the MS and St. Gallen 2013, Maisonneuve and Grade-based classifications were 62% (κ = 0.30), 66% (κ = 0.35) and 70% (κ = 0.41), respectively. PR did not contribute to distinguishing Luminal A from B tumours (auROC = 0.56). By classifying HG1-2 tumours as Luminal A-like and HG3 as Luminal B-like, agreement with MS was 80% (κ = 0.46). Moreover, by combining HG and Ki67 status, a large subgroup of patients (51% of the cohort) having > 90% Luminal A tumours could be identified.
Conclusions
Agreement between MS and surrogate classifications was generally poor. However, a post hoc analysis showed that a combination of HG and Ki67 could identify patients very likely to have Luminal A tumours according to MS.
Digital games have become a central part of contemporary culture and society. At the same time digital games provide numerous challenges for collections, preservation efforts, documentation, and ...exhibitions. This article investigates the challenges and opportunities implicit in LAM convergence and collaboration with actors outside of the LAM-sector itself. These actors are stakeholders of various kinds within game culture: game makers and industry, players, and rogue archives. More specifically, we turn to the collaboration in two Nordic museums in their work with digital games: The Finnish Museum of Games and the National Swedish Museum of Science and Technology. We draw on their actual efforts at collaboration between LAM-institutions and outside stakeholders and analyze them through the lens of political participation and agonistic pluralism. These concepts come from an interpretation of the participatory agenda in cultural policy that aims to resolve inconsistencies in the participatory agenda specifically around neoliberal logics of participation. The paper asks: How can the preservation of digital games be supported through participation of stakeholders inside and outside the LAM sector, and what policy changes would such collaborations require? This paper concludes that political participation and agonistic pluralism are useful concepts for the modeling and understanding of game preservation and provide a possible solution for the paradoxes of the participatory agenda in Nordic cultural policy. Our comparison of the work in two museums shows that approaches that empower participants can lead to successful and surprising exhibitions not possible without the sharing of curatorial power. Policy regulating LAM-institutions should change in order to accommodate players, makers, and rogue archives as participants in game preservation efforts. For the future the participatory agenda in cultural policy should be interpreted through the lens of political participation and agonistic pluralism as calling for truly empowering participants in order to elevate participation in game preservation from lucky accidents to a political participation policy.
