This paper focuses on the US Billion-dollar Weather/Climate Disaster report by the National Oceanic and Atmospheric Administration’s National Climatic Data Center. The current methodology for the ...production of this loss dataset is described, highlighting its strengths and limitations including sources of uncertainty and bias. The Insurance Services Office/Property Claims Service, the US Federal Emergency Management Agency’s National Flood Insurance Program and the US Department of Agriculture’s crop insurance program are key sources of quantified disaster loss data, among others. The methodology uses a factor approach to convert from insured losses to total direct losses, one potential limitation. An increasing trend in annual aggregate losses is shown to be primarily attributable to a statistically significant increasing trend of about 5 % per year in the frequency of billion-dollar disasters. So the question arises of how such trend estimates are affected by uncertainties and biases in the billion-dollar disaster data. The net effect of all biases appears to be an underestimation of average loss. In particular, it is shown that the factor approach can result in a considerable underestimation of average loss of roughly 10–15 %. Because this bias is systematic, any trends in losses from tropical cyclones appear to be robust to variations in insurance participation rates. Any attribution of the marked increasing trends in crop losses is complicated by a major expansion of the federally subsidized crop insurance program, as a consequence encompassing more marginal land. Recommendations concerning how the current methodology can be improved to increase the quality of the billion-dollar disaster dataset include refining the factor approach to more realistically take into account spatial and temporal variations in insurance participation rates.
Membrane gas separation is a mature and expanding technology. However, the availability of better membrane materials would promote faster growth. In this Perspective we analyze the state of the art ...of membrane materials, including polymers and hybrid materials, as well as the current issues and barriers, and finally, we outline future research directions in membrane science. Development of new membrane materials for large scale separations will rely on a multidisciplinary approach that embraces the broad fields of chemical and materials engineering, polymer science, and materials chemistry.
During germ cell and preimplantation development, mammalian cells undergo nearly complete reprogramming of DNA methylation patterns. We profiled the methylomes of human and chimp sperm as a basis for ...comparison to methylation patterns of ESCs. Although the majority of promoters escape methylation in both ESCs and sperm, the corresponding hypomethylated regions show substantial structural differences. Repeat elements are heavily methylated in both germ and somatic cells; however, retrotransposons from several subfamilies evade methylation more effectively during male germ cell development, whereas other subfamilies show the opposite trend. Comparing methylomes of human and chimp sperm revealed a subset of differentially methylated promoters and strikingly divergent methylation in retrotransposon subfamilies, with an evolutionary impact that is apparent in the underlying genomic sequence. Thus, the features that determine DNA methylation patterns differ between male germ cells and somatic cells, and elements of these features have diverged between humans and chimpanzees.

Summary In this report we review the health effects of three short-lived greenhouse pollutants—black carbon, ozone, and sulphates. We undertook new meta-analyses of existing time-series studies and ...an analysis of a cohort of 352 000 people in 66 US cities during 18 years of follow-up. This cohort study provides estimates of mortality effects from long-term exposure to elemental carbon, an indicator of black carbon mass, and evidence that ozone exerts an independent risk of mortality. Associations among these pollutants make drawing conclusions about their individual health effects difficult at present, but sulphate seems to have the most robust effects in multiple-pollutant models. Generally, the toxicology of the pure compounds and their epidemiology diverge because atmospheric black carbon, ozone, and sulphate are associated and could interact with related toxic species. Although sulphate is a cooling agent, black carbon and ozone could together exert nearly half as much global warming as carbon dioxide. The complexity of these health and climate effects needs to be recognised in mitigation policies.

The primary end points were the safety and tolerability of hESC-derived RPE cell administration. We also investigated evidence of the survival of transplanted cells and measured retinal structure and function using microperimetry and spectral-domain OCT.
Focal areas of subretinal hyperpigmentation developed in all participants in a dose-dependent manner in the recipient retina and persisted after withdrawal of systemic immunosuppression. We found no evidence of uncontrolled proliferation or inflammatory responses. Borderline improvements in best-corrected VA in 4 participants either were unsustained or were matched by a similar improvement in the untreated contralateral eye. Microperimetry demonstrated no evidence of benefit at 12 months in the 12 participants. In one instance at the highest dose, localized retinal thinning and reduced sensitivity in the area of hyperpigmentation suggested the potential for harm. Participant-reported quality of life using the 25-item National Eye Institute Visual Function Questionnaire indicated no significant change.
Subretinal hyperpigmentation is consistent with the survival of viable transplanted hESC-derived RPE cells, but may reflect released pigment in their absence. The findings demonstrate the value of detailed analysis of spatial correlation of retinal structure and function in determining with appropriate sensitivity the impact of cell transplantation and suggest that intervention in early stage of disease should be approached with caution. Given the slow rate of progressive degeneration at this advanced stage of disease, any protection against further deterioration may be evident only after a more extended period of observation.
Bio-optical and physical measurements were collected in the Mississippi Sound (Northern Gulf of Mexico) during the spring of 2018 as part of the Integrated Coastal Bio-Optical Dynamics project. The ...goal was to examine the impact of atmospheric and tidal fronts on fine-scale physical and bio-optical property distributions in a shallow, dynamic, coastal environment. During a 25-day experiment, eight moorings were deployed in the vicinity of a frontal zone. For a one-week period in the middle of the mooring deployment, focused ship sampling was conducted with aircraft and unmanned aerial vehicle overflights, acquiring hyperspectral optical and thermal data. The personnel in the aircraft located visible color fronts indicating the convergence of two water masses and directed the ship to the front. Dye releases were performed on opposite sides of a front, and coincident aircraft and unmanned aerial vehicle overflights were collected to facilitate visualization of advection/mixing/dispersion processes. Radiometric calibration of the optical hyperspectral sensor was performed. Empirical Line Calibration was also performed to atmospherically correct the aircraft imagery using in situ remote sensing reflectance measurements as calibration sources. Bio-optical properties were subsequently derived from the atmospherically corrected aircraft and unmanned aerial vehicle imagery using the Naval Research Laboratory Automated Processing System.
Observations of sea surface and land–near-surface merged temperature anomalies are used to monitor climate variations and to evaluate climate simulations; therefore, it is important to make analyses ...of these data as accurate as possible. Analysis uncertainty occurs because of data errors and incomplete sampling over the historical period. This manuscript documents recent improvements in NOAA’s merged global surface temperature anomaly analysis, monthly, in spatial 5° grid boxes. These improvements allow better analysis of temperatures throughout the record, with the greatest improvements in the late nineteenth century and since 1985. Improvements in the late nineteenth century are due to improved tuning of the analysis methods. Beginning in 1985, improvements are due to the inclusion of bias-adjusted satellite data. The old analysis (version 2) was documented in 2005, and this improved analysis is called version 3.

DNA methylation has been implicated as an epigenetic component of mechanisms that stabilize cell-fate decisions. Here, we have characterized the methylomes of human female hematopoietic ...stem/progenitor cells (HSPCs) and mature cells from the myeloid and lymphoid lineages. Hypomethylated regions (HMRs) associated with lineage-specific genes were often methylated in the opposing lineage. In HSPCs, these sites tended to show intermediate, complex patterns that resolve to uniformity upon differentiation, by increased or decreased methylation. Promoter HMRs shared across diverse cell types typically display a constitutive core that expands and contracts in a lineage-specific manner to fine-tune the expression of associated genes. Many newly identified intergenic HMRs, both constitutive and lineage specific, were enriched for factor binding sites with an implied role in genome organization and regulation of gene expression, respectively. Overall, our studies represent an important reference data set and provide insights into directional changes in DNA methylation as cells adopt terminal fates.

During development, mammalian germ cells reprogram their epigenomes via a genome-wide erasure and de novo rewriting of DNA methylation marks. We know little of how methylation patterns are ...specifically determined. The piRNA pathway is thought to target the bulk of retrotransposon methylation. Here we show that most retrotransposon sequences are modified by default de novo methylation. However, potentially active retrotransposon copies evade this initial wave, likely mimicking features of protein-coding genes. These elements remain transcriptionally active and become targets of piRNA-mediated methylation. Thus, we posit that these two waves play essential roles in resetting germ cell epigenomes at each generation.
