Acute monocytic leukemia is a type of myeloid leukemia that develops in monocytes. The current clinical therapies for leukemia are unsatisfactory due to their side effects and nonspecificity toward ...target cells. Some lectins display antitumor activity and may specifically recognize cancer cells by binding to carbohydrate structures on their surface. Therefore, this study evaluated the response of the human monocytic leukemia cell lines THP‐1 to the Olneya tesota PF2 lectin. The induction of apoptosis and reactive oxygen species production in PF2‐treated cells was evaluated by flow cytometry, and the lectin‐THP‐1 cell interaction and mitochondrial membrane potential were evaluated by confocal fluorescence microscopy. PF2 genotoxicity was evaluated by DNA fragmentation analysis via gel electrophoresis. The results showed that PF2 binds to THP‐1 cells, triggers apoptosis and DNA degradation, changes the mitochondrial membrane potential, and increases reactive oxygen species levels in PF2‐treated THP‐1 cells. These results suggest the potential use of PF2 for developing alternative anticancer treatments with enhanced specificity.
A green method for synthesizing gold nanoparticles is proposed using hydroethanolic extract of Vitex mollis fruit (Vm extract) as a reducer and stabilizer. The formation of gold nanoparticles ...synthesized with Vm extract (AuVmNPs) was monitored by measuring the ultraviolet–visible spectra. The morphology and crystalline phase were determined using scanning electron microscopy, X-ray diffraction, and high-resolution transmission electron microscopy. Synthesized nanoparticles were generally spherical, and the size distribution obtained by transmission electron microscopy shows two populations with mean sizes of 12.5 and 22.5 nm. Cell viability assay using MTT and cellular apoptosis studies using annexin V on human umbilical vein endothelial cells (HUVECs) and the human mammary epithelial cell line (MCF10A) indicate that AuVmNPs have low toxicity. Cell migration tests indicate that AuVmNPs significantly inhibit HUVEC cell migration in a dose-dependent manner. The evaluation of the localization of AuVmNPs in HUVECs using confocal laser scanning microscopy indicates that nanoparticles penetrate cells and are found in the cytosol without preferential distribution and without entering the nucleus. The inhibitory effect on cellular migration and low toxicity suggest AuVmNPs as appropriate candidates in future studies of antiangiogenic activity.
Type 2 diabetes (T2D) is a complex metabolic ailment marked by a global high prevalence and significant attention in primary healthcare settings due to its elevated morbidity and mortality rates. The ...pathophysiological mechanisms underlying the onset and progression of this disease remain subjects of ongoing investigation. Recent evidence underscores the pivotal role of the intricate intercellular communication network, wherein cell-derived vesicles, commonly referred to as extracellular vesicles (EVs), emerge as dynamic regulators of diabetes-related complications. Given that the protein cargo carried by EVs is contingent upon the metabolic conditions of the originating cells, particular proteins may serve as informative indicators for the risk of activating or inhibiting signaling pathways crucial to the progression of T2D complications.
In this study, we conducted a systematic review to analyze the published evidence on the proteome of EVs from the plasma or serum of patients with T2D, both with and without complications (PROSPERO: CRD42023431464).
Nine eligible articles were systematically identified from the databases, and the proteins featured in these articles underwent Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis. We identified changes in the level of 426 proteins, with CST6, CD55, HBA1, S100A8, and S100A9 reported to have high levels, while FGL1 exhibited low levels.
These proteins are implicated in pathophysiological mechanisms such as inflammation, complement, and platelet activation, suggesting their potential as risk markers for T2D development and progression. Further studies are required to explore this topic in greater detail.
Serum ferritin is a widely-used marker of inflammation in prediabetes, diabetes and atherosclerosis. In these cases, progressive endothelial damage may involve the participation of toll-like ...receptors (TLR). The aim of this study was to determine the expression of TLR2 and TLR4 in peripheral blood mononuclear cell (PBMC)-derived CD14+ cells from subjects with prediabetes and with a high level of serum ferritin both at baseline and after in vitro cell stimulation with glycated ferritin. Blood samples were drawn from 22 subjects (13 with prediabetes and 9 with normoglycemia). Serum ferritin levels were measured by ELISA, while the expression of TLR2 and TLR4 in PBMC-derived CD14+ cells was determined by flow cytometry. IL-6 and IL-8 cytokines in PBMC-derived CD14+ supernatants were measured by ELISA. Subjects with prediabetes had a higher baseline expression of TLR4 in PBMC-derived CD14+ cells than was observed in cells from normoglycemic subjects (p<0.05). Glycated ferritin increased the expression of both TLR2 and TLR4 as well as IL-6 and IL-8 in PBMC-derived CD14+ cells from subjects with prediabetes when compared to normoglycemic subjects (p<0.05). We concluded that in prediabetes, the increased basal expression of TLR4 could be part of the low-grade inflammation, which could be increased by glycated ferritin.

This pilot study aimed to determine if increased serum ferritin (SF) is associated with cardiovascular risk factors in patients with prediabetes.
Eighteen patients with prediabetes and 36 subjects ...without prediabetes (control), non-white Hispanic, non-indigenous origin, Mexican mestizo descent were included. Participants had no inflammation, or vascular complications. SF and metabolic markers were evaluated in both groups.
SF and oxidized low-density lipoprotein (oxLDL) were increased in prediabetes subjects. Moreover, in prediabetes and control groups as a whole, natural logarithm (ln)-SF correlated with oxLDL and ln-oxLDL/LDL after adjustment for sex, ln-age, ln-fasting plasma glucose (FPG), ln-body mass index, ln-triglyceride (TG), total cholesterol (TC), and high-density lipoproteins. Finally, ln-SF was an independent contributor to ln-oxLDL/LDL ratio in control and prediabetes subjects (β = 0.2915) after the introduction of potential confounders such as FPG, TC, TG, and hypertension.
The results of this study indicate that hyperferritinemia is associated with oxLDL, considered one of the main cardiovascular risk factors, which allows us to suggest that an increase in SF could contribute to the progression of prediabetes, prior to the appearance of diabetes. Further research is required to establish a causal relationship of iron disruption metabolism in oxLDL generation under prediabetes conditions.

Cada año unos trece mil jornaleros indígenas laboran en campos agrícolas de Sonora, pero poco se sabe acerca de su incidencia de tuberculosis, y cómo sus percepciones y las del personal de salud se ...relacionan con la enfermedad. Este es un estudio trasversal, realizado de 2005 a 2010, para describir esa incidencia en dichos trabajadores, que fue superior al promedio nacional y estatal, en dicho periodo; también se exploran los factores socioculturales asociados a la ocurrencia de tuberculosis. En los registros médicos de los pacientes no se documenta el grupo étnico al que pertenecen, lo que puede subestimar la carga de la tuberculosis en estos indígenas, que trabajan en Sonora, y que es elevada. Existen brechas entre el modelo biomédico y el sistema de creencias de los pacientes, que pueden mermar las acciones de control del padecimiento.

La concurrencia de COVID-19 y padecimientos crónicos no transmisibles son capaces de deteriorar la calidad de vida relacionada con la salud (CVRS) de las personas, pero poco se ...ha investigado al respecto en México. Objetivo. Examinar la CVRS de pacientes recuperados de COVID-19, estratificados por su severidad clínica. Material y métodos. Encuesta transversal en 144 adultos mayores de 18 años, con diagnóstico confirmado de infección por SARS-CoV-2 en localidades de Sonora. La CVRS fue examinada mediante el cuestionario SF-36 y comparada de acuerdo con la gravedad de las manifestaciones clínicas. Resultados. 131 sujetos se clasificaron con COVID-19 no grave y 13 con un cuadro grave. La media global (76.4 ± 18.4) de la CVRS no fue estadísticamente diferente entre los pacientes agrupados por su severidad clínica; sin embargo, los pacientes con una comorbilidad tuvieron la menor puntuación (72.00 ± 20.74), especialmente los que padecen diabetes mellitus tipo 2 DM2 (66.33 ± 23.75). También hubo menores puntajes en la función física (58.8 ± 29.6) y el dolor corporal (69.8 ± 20.2). Conclusiones. No se observó una diferencia significativa en la CVRS de pacientes de acuerdo con la severidad de las manifestaciones clínicas de COVID-19.

Each year about 13,000 indigenous laborers migrate to work in the agricultural fields of Sonora, but little is known about their incidence of tuberculosis (tb) or how their perceptions and those of ...health personnel are related to the disease. Using tools from epidemiology and medical anthropology, we describe the epidemiological profile of tb during 2005-2010 in three agricultural fields in Sonora, and explore how social and cultural factors relate to tb occurrence. The incidence of tb in these communities is four times the national and state average. No ethnic group is documented in medical records, so tb may be underestimated in indigenous people. tb in this population exceeds the national rate, and there are gaps between the biomedical model and the belief systems of patients that reduce the effectiveness of disease control actions.

Introduction. The simultaneity of COVID-19 and chronic noncommunicable diseases can impair the health-related quality of life (HRQoL) of individuals, however limited research has been conducted in Mexico. Objective. Examine the HRQoL of patients recovered from COVID-19, ranked by clinical severity. Methods. Cross-sectional survey of 144 adults over 18 years old, with a confirmed diagnosis of infection caused by SARS-CoV-2 in localities in Sonora. HRQoL was examined using the SF-36 questionnaire and compared according to severity of clinical manifestations. Results. 131 subjects were classified with non-severe COVID-19 and 13 as a severe case. The overall mean (76.4± 18.4) HRQoL was not statistically different between patients grouped by clinical severity; however, patients with a comorbidity had the lowest score (72.00 ± 20.74), especially those with TD2 (66.33 ± 23.75). There were also lower scores for physical function (58.8± 29.6) and body pain (69.8±20.2). Conclusions. There was no significant difference in HRQoL for patients according to the severity of clinical manifestations of COVID-19.
El comportamiento epidemiológico de la fiebre manchada por Rickettsia rickettsii constituye un desafío para los sistemas de salud del continente americano. Es un padecimiento de relevancia médica por ...la letalidad que provoca si no es diagnosticado ni tratado oportunamente. Aunque cualquier persona es susceptible a la infección, algunos grupos poblacionales son más vulnerables debido a un mayor contacto con la garrapata transmisora, entre ellos los niños, quienes tienen mayor morbilidad por lo que se asocian con resultados fatales. En su origen participa una multitud de factores biológicos, ecológicos y sociales, interrelacionados complejamente, y cuyo abordaje requiere de intervenciones integradas y multidisciplinarias. La incidencia de la enfermedad puede continuar aumentando en la región, de modo que su ocurrencia actual constituye un llamado urgente para la acción regional. Acciones preventivas que disminuyan el contacto con garrapatas e incrementen la sospecha temprana de la enfermedad, son prioritarias en la agenda de salud de varias naciones de las Américas.
