Single-layer MoS2 is an attractive semiconducting analogue of graphene that combines high mechanical flexibility with a large direct bandgap of 1.8 eV. On the other hand, bulk MoS2 is an indirect ...bandgap semiconductor similar to silicon, with a gap of 1.2 eV, and therefore deterministic preparation of single MoS2 layers is a crucial step toward exploiting the large direct bandgap of monolayer MoS2 in electronic, optoelectronic, and photovoltaic applications. Although mechanical and chemical exfoliation methods can be used to obtain high quality MoS2 single layers, the lack of control in the thickness, shape, size, and position of the flakes limits their usefulness. Here we present a technique for controllably thinning multilayered MoS2 down to a single-layer two-dimensional crystal using a laser. We generate single layers in arbitrary shapes and patterns with feature sizes down to 200 nm and show that the resulting two-dimensional crystals have optical and electronic properties comparable to that of pristine exfoliated MoS2 single layers.
A full appreciation of the role played by gas metallicity (Z), star formation rate (SFR) and stellar mass (M
*) is fundamental to understanding how galaxies form and evolve. The connections between ...these three parameters at different redshifts significantly affect galaxy evolution, and thus provide important constraints for galaxy evolution models. Using data from the Sloan Digital Sky Survey-Data Release 7 (SDSS-DR7) and the Galaxy and Mass Assembly (GAMA) surveys, we study the relationships and dependences between SFR, Z and M
*, as well as the Fundamental Plane for star-forming galaxies. We combine both surveys using volume-limited samples up to a redshift of z 0.36. The GAMA and SDSS surveys complement each other when analysing the relationships between SFR, M
* and Z. We present evidence for SFR and metallicity evolution to z ∼ 0.2. We study the dependences between SFR, M
*, Z and specific SFR (SSFR) on the M
*-Z, M
*-SFR, M
*-SSFR, Z-SFR and Z-SSFR relations, finding strong correlations between all. Based on those dependences, we propose a simple model that allows us to explain the different behaviour observed between low- and high-mass galaxies. Finally, our analysis allows us to confirm the existence of a Fundamental Plane, for which M
* = f(Z, SFR) in star-forming galaxies.
Articular cartilage lesions are a particular challenge for regenerative medicine strategies as cartilage function stems from a complex depth-dependent organization. Tissue engineering scaffolds that ...vary in morphology and function offer a template for zone-specific cartilage extracellular matrix (ECM) production and mechanical properties. We fabricated multi-zone cartilage scaffolds by the electrostatic deposition of polymer microfibres onto particulate-templated scaffolds produced with 0.03 or 1.0mm3 porogens. The scaffolds allowed ample space for chondrocyte ECM production within the bulk while also mimicking the structural organization and functional interface of cartilage’s superficial zone. Addition of aligned fibre membranes enhanced the mechanical and surface properties of particulate-templated scaffolds. Zonal analysis of scaffolds demonstrated region-specific variations in chondrocyte number, sulfated GAG-rich ECM, and chondrocytic gene expression. Specifically, smaller porogens (0.03mm3) yielded significantly higher sGAG accumulation and aggrecan gene expression. Our results demonstrate that bilayered scaffolds mimic some key structural characteristics of native cartilage, support in vitro cartilage formation, and have superior features to homogeneous particulate-templated scaffolds. We propose that these scaffolds offer promise for regenerative medicine strategies to repair articular cartilage lesions.
Background Oral immunotherapy (OIT) is an effective experimental food allergy treatment that is limited by treatment withdrawal and the frequent reversibility of desensitization if interrupted. Newly ...diagnosed preschool children may have clinical and immunological characteristics more amenable to treatment. Objective We sought to test the safety, effectiveness, and feasibility of early OIT (E-OIT) in the treatment of peanut allergy. Methods We enrolled 40 children aged 9 to 36 months with suspected or known peanut allergy. Qualifying subjects reacted to peanut during an entry food challenge and were block-randomized 1:1 to receive E-OIT at goal maintenance doses of 300 or 3000 mg/d in a double-blinded fashion. The primary end point, sustained unresponsiveness at 4 weeks after stopping early intervention oral immunotherapy (4-SU), was assessed by double-blinded, placebo-controlled food challenge either upon achieving 4 prespecified criteria, or after 3 maintenance years. Peanut-specific immune responses were serially analyzed. Outcomes were compared with 154 matched standard-care controls. Results Of 40 consented subjects, 3 (7.5%) did not qualify. Overall, 29 of 37 (78%) in the intent-to-treat analysis achieved 4-SU (300-mg arm, 17 of 20 85%; 3000 mg, 12 of 17 71%, P = .43) over a median of 29 months. Per-protocol, the overall proportion achieving 4-SU was 29 of 32 (91%). Peanut-specific IgE levels significantly declined in E-OIT-treated children, who were 19 times more likely to successfully consume dietary peanut than matched standard-care controls, in whom peanut-specific IgE levels significantly increased (relative risk, 19.42; 95% CI, 8.7-43.7; P < .001). Allergic side effects during E-OIT were common but all were mild to moderate. Conclusions At both doses tested, E-OIT had an acceptable safety profile and was highly successful in rapidly suppressing allergic immune responses and achieving safe dietary reintroduction.

An increasing number of observations have shown that gaseous debris discs are not an exception. However, until now, we only knew of cases around A stars. Here we present the first detection of 12CO ...(2-1) disc emission around an F star, HD 181327, obtained with the Atacama Large Millimeter/submillimeter Array (ALMA) observations at 1.3 mm. The continuum and CO emission are resolved into an axisymmetric disc with ring-like morphology. Using a Markov chain Monte Carlo method coupled with radiative transfer calculations, we study the dust and CO mass distribution. We find the dust is distributed in a ring with a radius of 86.0 +/- 0.4 au and a radial width of 23.2 +/- 1.0 au. At this frequency, the ring radius is smaller than in the optical, revealing grain size segregation expected due to radiation pressure. We also report on the detection of low-level continuum emission beyond the main ring out to approximately 200 au. We model the CO emission in the non-local thermodynamic equilibrium regime and we find that the CO is co-located with the dust, with a total CO gas mass ranging between 1.2 x 10(exp -6) solar mass and 2.9 x 10(exp -6) solar mass, depending on the gas kinetic temperature and collisional partners densities. The CO densities and location suggest a secondary origin, i.e. released from icy planetesimals in the ring. We derive a CO+CO2 cometary composition that is consistent with Solar system comets. Due to the low gas densities, it is unlikely that the gas is shaping the dust distribution.
Feeding highly fermentable diets to ruminants is one strategy to increase energy intake. The increase in short-chain fatty acid (SCFA) production and reduced ruminal pH associated with highly ...fermentable diets imposes a challenge to the metabolism and the regulation of intracellular pH homeostasis of ruminal epithelia. The ruminal epithelia respond to these challenges in a coordinated manner. Whereas the enlargement of absorptive surface area is well documented, emerging evidence at the mRNA and transporter and enzyme activity levels indicate that changes in epithelial cell function may be the initial response. It is not surprising that gene expression analysis has identified pathways involved in fatty acid metabolism, ion transport, and intracellular homeostasis to be the pathways dominantly affected during adaptation and after adaptation to a highly fermentable diet. These findings are important because the intraepithelial metabolism of SCFA, particularly butyrate, helps to maintain the concentration gradient between the cytosol and lumen, thereby facilitating absorption. Butyrate metabolism also controls the intracellular availability of butyrate, which is widely regarded as a signaling molecule. Current data indicate that for butyrate metabolism, 3-hydroxy-3-methylglutaryl-CoA synthase and acetyl-CoA acetyltransferase are potential regulatory points with transient up- and downregulation during diet adaptation. In addition to nutrient transport and utilization, genes involved in the maintenance of cellular tight junction integrity and induction of inflammation have been identified as differentially expressed genes during adaptation to highly fermentable diets. This may have important implications on ruminal epithelial barrier function and the inflammatory response often associated with subacute ruminal acidosis. The objective of this review is to summarize ruminal epithelial adaptation to highly fermentable diets focusing on the changes at the enzyme and transporter activity levels, as well as the underlying molecular changes at the mRNA and protein expression levels.
Articular cartilage is comprised of zones that vary in architecture, extracellular matrix composition, and mechanical properties. Here, we designed and engineered a porous zonal microstructured ...scaffold from a single biocompatible polymer (poly ϵ-caprolactone) using multiple fabrication strategies: electrospinning, spherical porogen leaching, directional freezing, and melt electrowriting. With this approach we mimicked the zonal structure of articular cartilage and produced a stiffness gradient through the scaffold which aligns with the mechanics of the native tissue. Chondrocyte-seeded scaffolds accumulated extracellular matrix including glycosaminoglycans and collagen II over four weeks in vitro. This prompted us to further study the repair efficacy in a skeletally mature porcine model. Two osteochondral lesions were produced in the trochlear groove of 12 animals and repaired using four treatment conditions: (1) microstructured scaffold, (2) chondrocyte seeded microstructured scaffold, (3) MaioRegen™, and (4) empty defect. After 6 months the defect sites were harvested and analyzed using histology, micro computed tomography, and Raman microspectroscopy mapping. Overall, the scaffolds were retained in the defect space, repair quality was repeatable, and there was clear evidence of osteointegration. The repair quality of the microstructured scaffolds was not superior to the control based on histological scoring; however, the lower score was biased by the lack of histological staining due to the limited degradation of the implant at 6 months. Longer follow up studies (e.g., 1 yr) will be required to fully evaluate the efficacy of the microstructured scaffold. In conclusion, we found consistent scaffold retention, osteointegration, and prolonged degradation of the microstructured scaffold, which we propose may have beneficial effects for the long-term repair of osteochondral defects.

The objective of this cross-sectional study was to evaluate the accuracy of fecal consistency scoring as a measure of fecal dry matter (DM) in dairy calves. This study was conducted at a commercial ...grain-fed veal facility in southwestern Ontario. A total of 160 calves arrived at the facility in 2 groups of 80 calves each. Calves were fed milk replacer twice daily at 0700 and 1700 h and had ad libitum access from arrival onward to water through nipple drinkers and starter through a shared trough. Fecal consistency scores were evaluated once daily in the first 28 d after arrival before milk feeding. The fecal consistency scoring was conducted using a 4-level scoring scale: 0 = normal (firm but not hard); 1 = soft (does not hold form, piles but spreads slightly); 2 = runny (spreads readily); and 3 = watery (liquid consistency, splatters). Fecal samples were collected from all calves via rectal palpation on d 1, 7, 14, and 21 at 0900 h for determination of fecal DM. Mixed repeated measures linear regression models were built to assess the accuracy of fecal consistency scoring in predicting fecal DM. Over 4 selected time points (d 1, 7, 14, and 21) the 160 calves were observed, 382 (61.6%) had a fecal consistency score of 0, 121 (19.5%) had a score of 1, 85 (13.7%) had a score of 2, and 32 (5.2%) had a score of 3. A fecal score of 0 had a fecal DM of 25.1 ± 8.4%, whereas a fecal score of 1 had a DM of 21.8 ± 8.2%. With respect to calves that had a fecal score of 2 or 3, their fecal DM was 16.0 ± 11.1% and 10.7 ± 6.9%, respectively. In evaluating the pairwise comparisons generated in the repeated measures model that controlled for day of sampling, a fecal score of 0 had a 3.2%, 8.1%, and 12.0% higher fecal DM, respectively, when compared with those that had a fecal score of 1, 2, and 3. In addition, calves with a fecal score of 1 had a 5.0% and 8.8% higher fecal DM than calves with a fecal score of 2 and 3, respectively. Finally, calves with a fecal score of 2 had a 3.8% higher fecal DM than those with a fecal score of 3. This study confirms that using observational fecal consistency scoring can accurately predict diarrhea or a decline in fecal DM.
Aims. We investigate the evolution of X-ray selected tidal disruption events. Methods. New events are found in near real-time data from XMM-Newton slews, and are monitored by multi-wavelength ...facilities. Results. In August 2016, X-ray emission was detected from the galaxy XMMSL2 J144605.0+685735 (also known as 2MASX 14460522+6857311), that was 20 times higher than an upper limit from 25 years earlier. The X-ray flux was flat for ∼100 days and then fell by a factor of 100 over the following 500 days. The UV flux was stable for the first 400 days before fading by a magnitude, while the optical (U,B,V) bands were roughly constant for 850 days. Optically, the galaxy appears to be quiescent, at a distance of 127 ± 4 Mpc (z = 0.029 ± 0.001) with a spectrum consisting of a young stellar population of 1–5 Gyr in age, an older population, and a total stellar mass of ∼6 × 109 M⊙. The bolometric luminosity peaked at Lbol ∼ 1043 ergs s−1 with an X-ray spectrum that may be modelled by a power law of Γ ∼ 2.6 or Comptonisation of a low-temperature thermal component by thermal electrons. We consider a tidal disruption event to be the most likely cause of the flare. Radio emission was absent in this event down to < 10 μJy, which limits the total energy of a hypothetical off-axis jet to E < 5 × 1050 ergs. The independent behaviour of the optical, UV, and X-ray light curves challenges models where the UV emission is produced by reprocessing of thermal nuclear emission or by stream-stream collisions. We suggest that the observed UV emission may have been produced from a truncated accretion disc and the X-rays from Compton upscattering of these disc photons.
