After a star has been tidally disrupted by a black hole, the debris forms an elongated stream. We start by studying the evolution of this gas before its bound part returns to the original stellar ...pericenter. While the axial motion is entirely ballistic, the transverse directions of the stream are usually thinner due to the confining effects of self-gravity. This basic picture may also be influenced by additional physical effects such as clump formation, hydrogen recombination, magnetic fields and the interaction with the ambient medium. We then examine the fate of this stream when it comes back to the vicinity of the black hole to form an accretion flow. Despite recent progress, the hydrodynamics of this phase remains uncertain due to computational limitations that have so far prevented us from performing a fully self-consistent simulation. Most of the initial energy dissipation appears to be provided by a self-crossing shock that results from an intersection of the stream with itself. The debris evolution during this collision depends on relativistic apsidal precession, expansion of the stream from pericenter, and nodal precession induced by the black hole spin. Although the combined influence of these effects is not fully understood, current works suggest that this interaction is typically too weak to significantly circularize the trajectories, with its main consequence being an expansion of the shocked gas. Global simulations of disc formation performed for simplified initial conditions find that the debris experiences additional collisions that cause its orbits to become more circular until eventually settling into a thick and extended structure. These works suggest that this process completes faster for more relativistic encounters due to the stronger shocks involved. It is instead significantly delayed if weaker shocks take place, allowing the gas to retain large eccentricities during multiple orbits. Radiation produced as the matter gets heated by circularizing shocks may leave the system through photon diffusion and participate in the emerging luminosity. This current picture of accretion flow formation results from recent theoretical works synthesizing the interplay between different aspects of physics. In comparison, early analytical works correctly identified the essential processes involved in disc formation, but had difficulty developing analytic frameworks that accurately combined non-linear hydrodynamical processes with the underlying relativistic dynamics. However, important aspects still remain to be understood at the time of writing, due to numerical challenges and the complexity of this process.
RNA interference (RNAi)-related pathways target viruses and transposable element (TE) transcripts in plants, fungi, and ecdysozoans (nematodes and arthropods), giving protection against infection and ...transmission. In each case, this produces abundant TE and virus-derived 20-30nt small RNAs, which provide a characteristic signature of RNAi-mediated defence. The broad phylogenetic distribution of the Argonaute and Dicer-family genes that mediate these pathways suggests that defensive RNAi is ancient, and probably shared by most animal (metazoan) phyla. Indeed, while vertebrates had been thought an exception, it has recently been argued that mammals also possess an antiviral RNAi pathway, although its immunological relevance is currently uncertain and the viral small RNAs (viRNAs) are not easily detectable. Here we use a metagenomic approach to test for the presence of viRNAs in five species from divergent animal phyla (Porifera, Cnidaria, Echinodermata, Mollusca, and Annelida), and in a brown alga-which represents an independent origin of multicellularity from plants, fungi, and animals. We use metagenomic RNA sequencing to identify around 80 virus-like contigs in these lineages, and small RNA sequencing to identify viRNAs derived from those viruses. We identified 21U small RNAs derived from an RNA virus in the brown alga, reminiscent of plant and fungal viRNAs, despite the deep divergence between these lineages. However, contrary to our expectations, we were unable to identify canonical (i.e. Drosophila- or nematode-like) viRNAs in any of the animals, despite the widespread presence of abundant micro-RNAs, and somatic transposon-derived piwi-interacting RNAs. We did identify a distinctive group of small RNAs derived from RNA viruses in the mollusc. However, unlike ecdysozoan viRNAs, these had a piRNA-like length distribution but lacked key signatures of piRNA biogenesis. We also identified primary piRNAs derived from putatively endogenous copies of DNA viruses in the cnidarian and the echinoderm, and an endogenous RNA virus in the mollusc. The absence of canonical virus-derived small RNAs from our samples may suggest that the majority of animal phyla lack an antiviral RNAi response. Alternatively, these phyla could possess an antiviral RNAi response resembling that reported for vertebrates, with cryptic viRNAs not detectable through simple metagenomic sequencing of wild-type individuals. In either case, our findings show that the antiviral RNAi responses of arthropods and nematodes, which are highly divergent from each other and from that of plants and fungi, are also highly diverged from the most likely ancestral metazoan state.

The growing number of stellar-mass binary black hole mergers discovered by Advanced LIGO and Advanced Virgo are starting to constrain the binaries' origin and environment. However, we still lack ...sufficiently accurate modeling of binary formation channels to obtain strong constraints, or to identify subpopulations. One promising formation mechanism that could result in different black hole properties is binaries merging within the accretion disks of active galactic nuclei (AGNs). Here we show that the black holes' orbital alignment with the AGN disks preferentially selects heavier black holes. We carry out Monte Carlo simulations of orbital alignment with AGN disks, and find that AGNs harden the initial black hole mass function. Assuming an initial power-law mass distribution , we find that the power-law index changes by Δβ ∼ 1.3, resulting in a more top-heavy population of merging black holes. This change is independent of the mass of, and accretion rate onto, the supermassive black hole in the center of the AGN. Our simulations predict an AGN-assisted merger rate of ∼4 Gpc−3 yr−1. With its hardened mass spectra, the AGN channel could be responsible for 10%-50% of gravitational-wave detections.
ABSTRACT
Young dense massive star clusters are promising environments for the formation of intermediate mass black holes (IMBHs) through collisions. We present a set of 80 simulations carried out ...with nbody6++gpu of 10 models of compact $\sim 7 \times 10^4 \, \mathrm{M}_{\odot }$ star clusters with half-mass radii Rh ≲ 1 pc, central densities $\rho _\mathrm{core} \gtrsim 10^5 \, \mathrm{M}_\odot \, \mathrm{pc}^{-3}$, and resolved stellar populations with 10 per cent primordial binaries. Very massive stars (VMSs) up to $\sim 400 \, \mathrm{M}_\odot$ grow rapidly by binary exchange and three-body scattering with stars in hard binaries. Assuming that in VMS–stellar black hole (BH) collisions all stellar material is accreted on to the BH, IMBHs with masses up to $M_\mathrm{BH} \sim 350 \, \mathrm{M}_\odot$ can form on time-scales of ≲15 Myr, as qualitatively predicted from Monte Carlo mocca simulations. One model forms an IMBH of 140 $\mathrm{M_{\odot }}$ by three BH mergers with masses of 17:28, 25:45, and 68:70 $\mathrm{M_{\odot }}$ within ∼90 Myr. Despite the stochastic nature of the process, formation efficiencies are higher in more compact clusters. Lower accretion fractions of 0.5 also result in IMBH formation. The process might fail for values as low as 0.1. The IMBHs can merge with stellar mass BHs in intermediate mass ratio inspiral events on a 100 Myr time-scale. With 105 stars, 10 per cent binaries, stellar evolution, all relevant dynamical processes, and 300 Myr simulation time, our large suite of 80 simulations indicate another rapid IMBH formation channel in young and compact massive star clusters.
Insect galls are dramatic examples of extended phenotypes: although composed of host plant tissues, their development is largely controlled by insect genes. Adaptive explanations for gall traits ...should thus be expressed in terms of impacts on insect fitness, but the extent to which interspecific variation in gall structure is adaptive, and the possible selective pressures driving diversification in gall form remain controversial. In colonial aphids and thrips, gall structures probably diversified in response to selection for enhancement of the surface area available for feeding. In other taxa, such as gall wasps and gall midges, diversity is expressed predominantly in non-nutritive tissues, particularly those on the gall surface. All natural enemies attack the occupants of closed galls by penetrating gall tissue, and modifications that reduce enemy attack rates should thus be favoured. Recent studies of intraspecific variation in gall form strongly support a defensive role for several traits, but, to date, there is little empirical support for enemies as a cause of interspecific variation in gall form. Selection imposed by enemies nevertheless remains the most probable adaptive explanation for the evolution of diversity. We suggest that this hypothesis has yet to be tested explicitly, and discuss the requirements for an appropriate cross-species analysis.
Galls are plant tissues whose development is induced by another organism for the inducer's benefit. 30,000 arthropod species induce galls, and in most cases the inducing effectors and target plant ...systems are unknown. Cynipid gall wasps are a speciose monophyletic radiation that induce structurally complex galls on oaks and other plants. We used a model system comprising the gall wasp Biorhiza pallida and the oak Quercus robur to characterise inducer and host plant gene expression at defined stages through the development of galled and ungalled plant tissues, and tested alternative hypotheses for the origin and type of galling effectors and plant metabolic pathways involved. Oak gene expression patterns diverged markedly during development of galled and normal buds. Young galls showed elevated expression of oak genes similar to legume root nodule Nod factor-induced early nodulin (ENOD) genes and developmental parallels with oak buds. In contrast, mature galls showed substantially different patterns of gene expression to mature leaves. While most oak transcripts could be functionally annotated, many gall wasp transcripts of interest were novel. We found no evidence in the gall wasp for involvement of third-party symbionts in gall induction, for effector delivery using virus-like-particles, or for gallwasp expression of genes coding for plant hormones. Many differentially and highly expressed genes in young larvae encoded secretory peptides, which we hypothesise are effector proteins exported to plant tissues. Specifically, we propose that host arabinogalactan proteins and gall wasp chitinases interact in young galls to generate a somatic embryogenesis-like process in oak tissues surrounding the gall wasp larvae. Gall wasp larvae also expressed genes encoding multiple plant cell wall degrading enzymes (PCWDEs). These have functional orthologues in other gall inducing cynipids but not in figitid parasitoid sister groups, suggesting that they may be evolutionary innovations associated with cynipid gall induction.

Aims
Cannabidiol (CBD) is a cannabis‐derived medicinal product with potential application in a wide‐variety of contexts; however, its effective dose in different disease states remains unclear. This ...review aimed to investigate what doses have been applied in clinical populations, in order to understand the active range of CBD in a variety of medical contexts.
Methods
Publications involving administration of CBD alone were collected by searching PubMed, EMBASE and ClinicalTrials.gov.
Results
A total of 1038 articles were retrieved, of which 35 studies met inclusion criteria covering 13 medical contexts. Twenty‐three studies reported a significant improvement in primary outcomes (e.g. psychotic symptoms, anxiety, seizures), with doses ranging between <1 and 50 mg/kg/d. Plasma concentrations were not provided in any publication. CBD was reported as well tolerated and epilepsy was the most frequently studied medical condition, with all 11 studies demonstrating positive effects of CBD on reducing seizure frequency or severity (average 15 mg/kg/d within randomised controlled trials). There was no signal of positive activity of CBD in small randomised controlled trials (range n = 6–62) assessing diabetes, Crohn's disease, ocular hypertension, fatty liver disease or chronic pain. However, low doses (average 2.4 mg/kg/d) were used in these studies.
Conclusion
This review highlights that CBD has a potential wide range of activity in several pathologies. Pharmacokinetic studies as well as conclusive phase III trials to elucidate effective plasma concentrations within medical contexts are severely lacking and highly encouraged.
