The goal of this study ...was to create a novel Cuproptosis-related lncRNA signature that may be utilized to predict survival and immunotherapy in HCC patients. Cuproptosis-associated lncRNAs and differentially expressed lncRNAs between HCC tumor tissue and normal tissue were discovered first. By LASSO-Cox analysis, the overlapping lncRNAs were then utilized to build a Cuproptosis-associated lncRNA signature, which might be used to predict patient prognosis and responsiveness to immune checkpoint blockade (ICB) therapy. Differences in the infiltration of immune cell subpopulations between high and low-risk score subgroups were also analyzed. Moreover, a nomogram based on the Cuproptosis-associated lncRNA signature and clinical features was developed and demonstrated to have good predictive potential. Finally, qRT-PCR was performed in HerpG2 and MHCC-97H cell lines to explore whether these lncRNAs were indeed involved in the process of Cuproptosis. In summary, we created a prognostic lncRNA profile linked to Cuproptosis to forecast response to immunotherapy, which may provide a new potential non-apoptotic therapeutic perspective for HCC patients.

The k-nearest neighbor (KNN) model is an effective statistical model applied in short-term traffic forecasting that can provide reliable data to guide travelers. This study proposes an improved KNN model to enhance forecasting accuracy based on spatiotemporal correlation and to achieve multistep forecasting. The physical distances among road segments are replaced with equivalent distances, which are defined by the static and dynamic data collected from real road networks. The traffic state of a road segment is described by a spatiotemporal state matrix instead of only a time series as in the original KNN model. The nearest neighbors are selected according to the Gaussian weighted Euclidean distance, which adjusts the influences of time and space factors on spatiotemporal state matrices. The forecasting accuracies of the improved KNN and of four other models are compared, and experimental results indicate that the improved KNN model is more appropriate for short-term traffic multistep forecasting than the other models are. This study also discusses the application of the improved KNN model in a time-varying traffic state.
The vibration and radiation noise characteristics of the gear transmission system are different under different traction conditions, and the gear modification optimization scheme based on a single ...working condition is not suitable for the operating environment under all working conditions. To modify the traction gear of a high-speed EMU, an optimized design scheme for noise reduction under multiple working conditions is proposed. A modification plan of the tooth direction in conjunction with the tooth shape was devised using a parametric model of the EMU's traction gear transmission system. The radiation noise of the gear transmission system after modification was solved using the acoustic boundary element method under different working conditions. A gear noise prediction model based on the random forest was proposed, and a gear modification parameter combination was constructed to minimize radiation noise. Then, the optimal design scheme of multi-condition modification combination parameters is obtained with the weight of the running time and acoustic contribution under different working conditions. The grey correlation degree evaluation model is established to verify that the multi-condition modification optimization design method can make the traction gear of EMU obtain satisfactory transmission performance and noise reduction effect under different working conditions.

Hepatocellular carcinoma (HCC) with cancer cells under endoplasmic reticulum (ER) stress has a poor prognosis. This study is aimed at discovering credible biomarkers for predicting the prognosis of ...HCC based on ER stress-related genes (ERSRGs). We constructed a novel four-ERSRG prognostic risk model, including PON1, AGR2, SSR2, and TMCC1, through a series of bioinformatic approaches, which can accurately predict survival outcomes in HCC patients. Higher risk scores were linked to later grade, recurrence, advanced TNM stage, later T stage, and HBV infection. In addition, 20 fresh frozen tumors and normal tissues from HCC patients were collected and used to validate the genes expressed in the signature by qRT-PCR and immunohistochemical (IHC) assays. Moreover, we found the ER stress-related signature could reflect the infiltration levels of different immune cells in the tumor microenvironment (TME) and forecast the efficacy of immune checkpoint inhibitor (ICI) treatment. Finally, we created a nomogram incorporating this ER stress-related signature. In conclusion, our constructed four-gene risk model associated with ER stress can accurately predict survival outcomes in HCC patients, and the model’s risk score is associated with the poor clinical classification.

In Neurospora crassa, deletion of cre-1 results in increased secretion of amylase and β-galactosidase.
Here we show that a strain carrying a deletion of cre-1 has increased cellulolytic activity and increased expression of cellulolytic genes during growth on crystalline cellulose (Avicel). Constitutive expression of cre-1 complements the phenotype of a N. crassa Δcre-1 strain grown on Avicel, and also results in stronger repression of cellulolytic protein secretion and enzyme activity. We determined the CRE-1 regulon by investigating the secretome and transcriptome of a Δcre-1 strain as compared to wild type when grown on Avicel versus minimal medium. Chromatin immunoprecipitation-PCR of putative target genes showed that CRE-1 binds to only some adjacent 5'-SYGGRG-3' motifs, consistent with previous findings in other fungi, and suggests that unidentified additional regulatory factors affect CRE-1 binding to promoter regions. Characterization of 30 mutants containing deletions in genes whose expression level increased in a Δcre-1 strain under cellulolytic conditions identified novel genes that affect cellulase activity and protein secretion.
Our data provide comprehensive information on the CRE-1 regulon in N. crassa and contribute to deciphering the global role of carbon catabolite repression in filamentous ascomycete fungi during plant cell wall deconstruction.

We systematically measure the superconducting (SC) and mixed state properties of high-quality CsV3Sb5 sin-gle crystals with Tc ~ 3.5K.We find that the upper critical field Hc2(T) exhibits a large ...anisotropic ratio of Hc2ab/Hc2c ~ 9 at zero temperature and fitting its temperature dependence requires a minimum two-band effective model.Moreover,the ratio of the lower critical field,Hc1ab/Hc1c,is also found to be larger than 1,which indicates that the in-plane energy dispersion is strongly renormalized near Fermi energy.Both Hc1 (T) and SC diamagnetic signal are found to change little initially below Tc ~ 3.5 K and then to increase abruptly upon cooling to a char-acteristic temperature of ~2.8 K.Furthermore,we identify a two-fold anisotropy of in-plane angular-dependent magnetoresistance in the mixed state.Interestingly,we find that,below the same characteristic T ~ 2.8 K,the orientation of this two-fold anisotropy displays a peculiar twist by an angle of 60° characteristic of the Kagome geometry.Our results suggest an intriguing superconducting state emerging in the complex environment of Kagome lattice,which,at least,is partially driven by electron-electron correlation.
A vacuum pressure standard called an optical pressure standard (OPS) has been developed at the National Institute of Metrology (NIM) based on optical refractometry using nitrogen gas. The OPS ...consists of a dual Fabry-Pérot (FP) cavity made from ultra-low expansion glass, a copper chamber housing the FP cavity, a temperature control system, and an optical system. Two tunable diode lasers at a wavelength of 633 nm were frequency-locked to the measurement cavity and the reference cavity, and their frequencies were used to sense the pressure within the copper chamber. The performance of the FP cavity was investigated including the coefficient of thermal expansion and creeping effects. The temperature uniformity assessed for the copper chamber was within 1 mK. The so-called heat-island effect was elucidated and avoided for all measurements performed, and the influence of outgassing and leakage was considered. Non-linear and hysteresis behavior was observed for the deformation of the reference cavity. A direct comparison between the OPS and the NIM primary piston gauge showed no deviations greater than 10 parts per million for the pressures assessed in the range from 20 kPa to 100 kPa. The uncertainty of the OPS was evaluated to be (0.13Pa)2+(23·10−6p)20.5(k=2).

Filamentous fungi that thrive on plant biomass are the major producers of hydrolytic enzymes used to decompose lignocellulose for biofuel production. Although induction of cellulases is regulated at ...the transcriptional level, how filamentous fungi sense and signal carbon-limited conditions to coordinate cell metabolism and regulate cellulolytic enzyme production is not well characterized. By screening a transcription factor deletion set in the filamentous fungus Neurospora crassa for mutants unable to grow on cellulosic materials, we identified a role for the transcription factor, VIB1, as essential for cellulose utilization. VIB1 does not directly regulate hydrolytic enzyme gene expression or function in cellulosic inducer signaling/processing, but affects the expression level of an essential regulator of hydrolytic enzyme genes, CLR2. Transcriptional profiling of a Δvib-1 mutant suggests that it has an improper expression of genes functioning in metabolism and energy and a deregulation of carbon catabolite repression (CCR). By characterizing new genes, we demonstrate that the transcription factor, COL26, is critical for intracellular glucose sensing/metabolism and plays a role in CCR by negatively regulating cre-1 expression. Deletion of the major player in CCR, cre-1, or a deletion of col-26, did not rescue the growth of Δvib-1 on cellulose. However, the synergistic effect of the Δcre-1; Δcol-26 mutations circumvented the requirement of VIB1 for cellulase gene expression, enzyme secretion and cellulose deconstruction. Our findings support a function of VIB1 in repressing both glucose signaling and CCR under carbon-limited conditions, thus enabling a proper cellular response for plant biomass deconstruction and utilization.

A highly sensitive hollow-core photonic crystal fiber (HC-PCF) gas-sensing network based on intracavity absorption is designed and experimentally verified. The capacity of the multichannel sensing ...network is expanded by time division multiplexing and wavelength division multiplexing technology. The voltage gradient method is employed in the wavelength scanning process of Fabry-Perot (F-P) filter to enhance the detection efficiency up to six times. The proposed sensing network has 16 sensing points. Experimental results show that the minimum detection limit (MDL) of this sensing system is 25.91 ppm and 26.85 ppm at the acetylene gas absorption peaks of 1530.371 nm and 1531.588 nm, respectively. As far as we know, it is the first time to obtain an intracavity sensing network via the application of an optical switch and DWDM at the same time. The sensing network can be used for high-capacity, low-concentration dangerous gas detection. It has great potential in environmental monitoring, industrial manufacturing, safety inspection and similar occasions.
