Immunocytochemistry: an indispensable technique in routine cytology
Immunocytology is today accepted as an indispensable adjunct to cytomorphology. It has led to a dramatic ...increase in diagnostic accuracy and also allowed the identification of markers both for prognosis and targeted therapies. Most commercially available antibodies will perform in a reproducible and reliable way provided that the cytological specimen has been prepared and fixed properly. In this review various aspects of immunocytochemistry such as preparation of cytological specimens, fixation and choice of antibodies will be discussed. The specificity of the most commonly used antibodies is summarized and staining panels for various tumours are suggested. In addition, the use of markers for targeted therapy and theranostics is discussed, as well as a brief section on the identification of infectious agents.
Calcium pyrophosphate dihydrate deposition (CPDD) is the accepted name for a disease that mainly occurs in elderly patients. This disease affects many joints in particular the knee joint. CPDD is ...extremely rare in the temporomandibular joint (TMJ) with only few cases reported in the English literature. Herein, we present a case of an 89 years old woman with a radiological diagnosis of chondrosarcoma of TMJ. Fine‐needle aspiration cytology however showed crystals, multinucleated giant cells and macrophages which allowed a correct diagnosis of CPDD.
This essay solders together a historical case study of advertising on the China mainland in the early twentieth century with a series of generalizable appraisals and determinations to establish how ...the catachresis
is evental. The event of
emerged historically in part because commercial advertising alleges that it did, and in part because at roughly the same time a subjective claim—“I am a woman”—began to be voiced. Although registered orthographically,
女性 is not simply a rhetorical category or a natural fact; it is not the effect of abyssal sex difference or an array of performatively accreted social subjects. It is a historical novelty. This essay universalizes a case rooted in Chinese- and Japanese-language archives and historical specificities, particularly the international advertising ephemera that presents a multiple (Badiou’s language for a set or “context”) or site where a historical event could have erupted and, in this case, did erupt. Ephemera are useful in historical analysis, as Walter Benjamin suggested a century ago, because historical events lie immanent in detritus as potential dialectical images, which the determined historian can extricate. Reading Badiou’s philosophy creatively, the essay makes the case that
or
is an emergent, historical, modern, and universal truth; direct critique of Badiou also drives home the point that philosophy (or “theory”) is vulnerable to history and that historically, factually, actually, and in truth, women and feminism are foundations of modernity no matter how modernity is qualified.
The fruit canning industry processes large quantities of the clingstone varieties of peach (Prunus persica L. Batch). The occurrence of split-pit formation--the opening of the pit and sometimes ...splitting of the fruit--causes deterioration of canned fruit quality. The frequency of split-pit formation is influenced by genetic and environmental factors. To increase understanding of the molecular mechanisms underlying split-pit formation in peach, we cloned and characterized the PPERFUL and PPERSHP genes that are homologues to the genes FRUITFULL and SHATTERPROOF, respectively, which are involved in fruit splitting (pod shattering) in Arabidopsis thaliana. The deduced amino acid sequences of the two genes had high homology with members of the MADS-box family of transcription factors, and particularly with other members of the FUL-like family of A-type MADS-box proteins and PLENA-like family of C-type MADS-box proteins, respectively. PPERFUL and PPERSHP were expressed throughout fruit development from full anthesis until fruit harvest. Differences in the mRNA abundance of each gene were compared in a split-pit sensitive and a split-pit resistant variety. Results suggested that temporal regulation of PPERFUL and PPERSHP expression may have an effect on the split-pit process.
Cotton (
Gossypium hirsutum
) wilt caused by
Fusarium oxysporum
f. sp. v
asinfectum
(
Fov
) is considered as a major threat for commercial cotton production worldwide. Relative expression ratios of ...two key pathogenesis-related (
PR
) genes (
PR-3
and
PR-10
) and a detoxification gene (
GST18
) were compared between a fully susceptible (“LACTA”) and a partially field-resistant (“EMERALD”) cultivar after challenging with an Australian
Fov
isolate, as well as after pre-treatments with chemical inducers of defense such as BION® (a chemical analog of salicylic acid) and methyl-jasmonate (MeJA) prior to
Fov
inoculation. It was demonstrated that in both hypocotyls and roots of “EMERALD”, all
PR
genes were over-expressed after inoculation with
Fov
but not in the fully susceptible cultivar.
Fov
inoculation did not significantly affect
GST18
expression in both cultivars. Exogenous application of each defense elicitor, prior to
Fov
inoculation, resulted in up-regulation of the three genes in root tissues of the fully susceptible cultivar. BION® application did not influence
PR-3
expression in hypocotyls of both cultivars, whereas MeJA application resulted in induction of
PR-3
in both cultivars. Furthermore, in hypocotyls of “LACTA”, over-expression of
PR-10
was recorded after treatment with each chemical inducer. This pathogen exhibited different ability in eliciting oxidative burst in roots of the two cotton cultivars used in our analysis.
Most participating countries have now adopted a triple assessment approach, i.e. clinical, imaging and pathology, to breast diagnosis, with FNAC as the first-line pathological investigation in both ...screening and symptomatic populations, with the exception of microcalcifications. Pathologists specialized in cytopathology are best qualified to collect and interpret FNAC samples, but this is not always possible or practical. Radiologists involved in breast imaging should ensure that they have the necessary skills to carry out FNAC under all forms of image guidance. Best results are achieved by a combination of both techniques, as shown in the image-guided FNAC in the presence of the cytopathologist. The majority of European countries use similar reporting systems for breast FNAC (C1-C5), in keeping with European Guidelines for Quality Assurance in Breast Cancer Screening and Diagnosis, although some still prefer descriptive reporting only. When triple assessment is concordant, final treatment may proceed on the basis of FNAC, without a tissue biopsy. ER and PR assessment can be done safely on FNAC material. However, not all institutions may have expertise in doing this. HER-2 protein expression on direct cytological preparations is insufficiently reliable for clinical use, although its use for FISH is possible, if expertise is available. The majority of participants practise a degree of one-stop diagnosis with a cytopathologist present in the out-patient clinic. Formal recognition of the importance of the time spent outside the laboratory, both for cytopathologist and cytotechnologist, is necessary in order to ensure appropriate resourcing. The use of core biopsy (CB) has increased, although not always for evidence-based reasons. CB and FNAC are not mutually exclusive. FNAC should be used in diagnosis of benign, symptomatic lesions and CB in microcalcifications, suspicious FNAC findings and malignancies where radiology cannot guarantee stromal invasion.
Background: The incidence of non-Hodgkin lymphoma and prevalence of obesity are increasing globally. A suggested positive association between obesity and risk of non-Hodgkin lymphoma has prompted us ...to investigate the relationship between body mass index (BMI) and risk of malignant lymphoma subtypes in a population-based case–control study. Methods: Telephone interviews were conducted with 3055 case patients with non-Hodgkin lymphoma and 618 case patients with Hodgkin lymphoma diagnosed between October 1, 1999, and August 30, 2002, and 3187 population-based control subjects. The interviews assessed current height, normal adult weight, and other possible risk factors. Multivariable odds ratios (ORs) and corresponding 95% confidence intervals (CIs) for risk of lymphoma were estimated by unconditional logistic regression. All statistical tests were two-sided. Results: BMI was not associated with risk of overall non-Hodgkin lymphoma or of Hodgkin lymphoma (for example, comparing the highly obese group BMI ≥35.0 kg/m2 with the normal-weight group BMI = 18.5–24.9 kg/m2, OR for risk of non-Hodgkin lymphoma = 0.9, 95% CI = 0.6 to 1.3; Ptrend across all categories of BMI = .27). BMI was also not associated with risk of any non-Hodgkin lymphoma subtype evaluated, although there was some evidence of a positive association with risk of diffuse large B-cell lymphoma (for example, comparing the highly obese group with the normal-weight group, OR for diffuse large B-cell lymphoma = 1.5, 95% CI = 0.9 to 2.4; Ptrend =.05). Conclusions: Excess weight does not appear to be associated with an increased risk of malignant lymphoma in general, or with a risk of most major lymphoma subtypes. Hence, the growing incidence of obesity is unlikely to be an important contributor to the increasing incidence of non-Hodgkin lymphoma worldwide.
