The M2e peptide of the H5N1 A/Chicken/Kurgan/05/2005 avian influenza virus was successfully synthesized in transgenic tobacco plants. The amino-terminal segment of the M2 protein, comprised of 22, ...30, or 43 amino acids, including the M2e peptide (M122, M130, and M143 variants, respectively), was translationally fused with the N-terminus of β-glucuronidase in the pBI121 plant expression vector. The nucleotide sequence of the target fragment was synthesized by ligation from synthetic oligonucleotides; its codon composition was adapted for expression in plants. Tobacco plants were successfully transformed with the obtained vectors (pBIM122, pBIM130, and pBIM143, respectively). In the plants transformed with pBIM143, the Ml43-β-glucuronidase fusion protein was not produced, probably due to the presence of the M2 protein transmembrane domain (25–43 aa of M2) in this construct. In the pBIM122- and pBIMl30-transformed plants, the target M2e peptide was expressed as a component of the Ml22-β-glucuronidase and M130-β-glucuronidase fusion proteins, respectively, as was detected by Western blot analysis. These proteins were detected as bands of the expected size without apparent degradation. As a result, the M2e peptide of the H5N1 avian influenza virus was successfully synthesized for the first time in nuclear-transformed transgenic plants. The results obtained in this study will be used for developing a transgenic plant-based edible antiinfluenza vaccine.

Here, ...we present room-temperature coherent control of high-dimensional quantum bits, the so-called qudits, associated with vacancy-related spins in silicon carbide enriched with nuclear spin-free isotopes. In addition to the excitation of a spectrally narrow qudit mode at the pump frequency, several other modes are excited in the electron spin resonance spectra whose relative positions depend on the external magnetic field. We develop a theory of multipole spin dynamics and demonstrate selective quantum control of homogeneous spin packets with sub-MHz spectral resolution. Furthermore, we perform two-frequency Ramsey interferometry to demonstrate absolute dc magnetometry, which is immune to thermal noise and strain inhomogeneity.
A periodically driven system with spatial asymmetry can exhibit a directed motion facilitated by thermal or quantum fluctuations. This so-called ratchet effect has fascinating ramifications in ...engineering and natural sciences. Graphene is nominally a symmetric system. Driven by a periodic electric field, no directed electric current should flow. However, if the graphene has lost its spatial symmetry due to its substrate or adatoms, an electronic ratchet motion can arise. We report an experimental demonstration of such an electronic ratchet in graphene layers, proving the underlying spatial asymmetry. The orbital asymmetry of the Dirac fermions is induced by an in-plane magnetic field, whereas the periodic driving comes from terahertz radiation. The resulting magnetic quantum ratchet transforms the a.c. power into a d.c. current, extracting work from the out-of-equilibrium electrons driven by undirected periodic forces. The observation of ratchet transport in this purest possible two-dimensional system indicates that the orbital effects may appear and be substantial in other two-dimensional crystals such as boron nitride, molybdenum dichalcogenides and related heterostructures. The measurable orbital effects in the presence of an in-plane magnetic field provide strong evidence for the existence of structure inversion asymmetry in graphene.

The novel microwave wideband quasi-elliptic bandpass filter, in which the initially uncoupled strip transmission lines are used is considered in the present study. The proposed filter is based on the ...reentrant structure in which the metallic body with a floating potential is asymmetrical. Generally speaking, the internal and external dielectric fillings must be of unequal relative permittivity. As a result, the additional symmetrical transmission zeros are reached at the lower and upper stop bands of the pass band leading to the quasi-elliptic response function that improves the bandpass filter selectivity. The general TEM circuit model for the proposed filter, in terms of the multi-port series connection, is presented and then used to predict the initial electrical and geometrical quantities. A printed circuit prototype has been manufactured using a common printed circuit board implementation. The measured results are in an acceptable agreement with the simulation-derived ones, and that improves the degree of freedom in the frequency selective unit design.

Актуальность. В структуре современного фармацевтического рынка неуклонно растет доля препаратов на основе лекарственного растительного сырья. Широкий спектр их действия объясняется ...многокомпонентностью состава биологически активных веществ, одновременным присутствием поликомпонентных соединений различной природы. Мягкое терапевтическое действие, малая токсичность, редкое возникновение побочных явлений, экономическая доступность – это немногие из достоинств лечебно-профилактических средств природного происхождения. По мнению исследователей, немаловажным звеном является выбранный метод извлеченияуказанных биологически активных веществ. В частности это касается разработки технологии и вида лекарственной формы. Традиционные технологии, позволяющие получать многокомпонентные смеси неполярных соединений сложного состава, в большинстве случаев связаны с использование достаточно «тяжелых» органических растворителей. Цель исследования: проведение микробиологических исследований сверхкритического СО2экстракта ромашки аптечной для разработки рекомендаций его возможного использования в составе комбинированных мягких лекарственных форм для лечения раневого процесса. Материалы и методы. Материалами исследования стал нативный СО2 екстракт ромашки.Определение антимикробной активности СО2 екстракта ромашкипроводили микробиологическим методом диффузии в агар на твердой питательной среде (в соответствии с требованиями Государственной Фармакопеи Украины), который основан на способности антибактериального вещества угнетать рост микроорганизмов. Антимикробную активность экстракта изучали на питательной среде – соево-казеиновом агаре на разных тест-штаммах микроорганизмов.Статистическую обработку полученных результатов проводили припомощи пакета Statistica 6.0 (StatSoftInc., USA). Результатысчитались статистически достоверными при P< 0,05. Результаты. В статье представлены результаты антимикробной активности СО2экстракта Ромашки аптечной методом диффузии в агар.Из полученных данных следует, что тест-штаммы Bacillus subtilis и Staphylococсusaureus наиболее чуствительны к действию указанного фитоэкстракта и имеют зоны задержки роста от 10 до 15 мм. По отношению к тест-штаммам Pseudomonasaeruginosae, Candidaalbicans и Saccharomycescerevisiae зон задержки роста не выявленно.Таким образом, на основании проведенных экспериментальных исследованийin vitroустановлено, что СО2фитоэкстракт ромашки проявляет антимикробную активность в отношении тест-штаммовBacillus subtilis АЕС 6633 и StaphylococсusaureusATCC 6538. Выводы. Проведенные авторами комплексные исследования (литературный скрининг, экспериментальная работа) стали логичным продолжением внимания научной общественности многих стран мира к чистым технологиям. Поэтому современные исследователи при создании новых лекарственных фитопрепаратов отдают предпочтение новым высокоэффективным и экологически безопасным технологиям. Представленные результаты наглядно доказывают, что сверхкритический СО2экстракт ромашки, проявляет определенную антимикробную активность (до 15 мм) по отношению к Bacillus subtilis и Staphylococсusaureus. Это подтверждает возможность использовать указанный фитоэкстракт в качестве антимикробного компонента в составе комбинированных мягких лекарственных форм для наружного применения,вчастности для лечения раневого процесса.
The purpose of the work was to study the histological structure of the greater omentum in the people of second mature and elderly age in normal condition and in peritonitis. Materials and methods. ...The materials of the study were 40 greater omenta of deceased people (20 males and 20 females) of the second mature and elderly age without pathology in the abdominal cavity and 10 greater omenta of deceased people from peritonitis taken in accordance with the cooperation agreement. Pieces of greater omenta were fixed with a 10% solution of neutral formalin. Paraffin blocks were made according to the traditional method, followed by making sections that were stained with hematoxylin and eosin. Histological specimens were studied using a light microscope "MICROmed" XS-3320 in transmitted light using an increase of ×40, ×100, ×400 with the photographing of micropreparations on the camera Ningero Sheng Heng Optics and Electronics Co. Results and discussion. The findings of the study showed that the histological structure of the tissue of the greater omentum in women and men of these ages has common structural features. Histological sections stained with hematoxylin and eosin revealed that normally the outside of the greater omentum is covered with a solid layer of mesotheliocytes, located on the basal plate, deeper there were “milk spots”, white adipocytes, which formed individual fat particles, separated by connective tissue fibers. “Milk spots” do not have capsules, in contrast to lymph nodes, and contain lymphocytes and macrophages that emerge from their fenestrated postcapillary venules and migrate into the abdominal cavity through “stigmas”. Normally, active and inactive “milk spots” are present in the greater omentum in adults. “Milk spots” had an oval, irregular and spherical shape and different sizes. In the thickness of the tissue of the greater omentum there were lymph nodes, which, in contrast to the "milk spots" on the outside were covered with a connective tissue capsule. In the thickness of the nodule, trabeculae extend from the capsule. Marginal and medullary sinuses were determined. Lymphocytes were the main cellular component of lymph nodes. Conclusion. In peritonitis, disruption of the continuous mesothelial cover of the greater omentum, resulted from disintegration of mesothelial cells; some areas of their adhesion and desquamation were observed. Areas with inflammatory infiltration, plethora of arterial and venous microvessels and an increase in active “milk spots” with a high content of lymphocytes were found in the fat particles of the greater omentum

We have developed a simple and convenient method for the synthesis of 3-aryl- and 3-hetaryl-1,2,4-oxadiazin-5-ones bearing an easily functionalizable (methoxycarbonyl)methyl group at position 6 via ...the reaction of aryl or hetaryl amidoximes with maleates or fumarates. The conditions for this reaction were optimized. Different products can be synthesized selectively in good yields depending on the base used and the ratio of reactants: substituted (1,2,4-oxadiazin-6-yl)acetic acids, corresponding methyl esters, or hybrid 3-(aryl)-6-((3-(aryl)-1,2,4-oxadiazol-5-yl)methyl)-4H-1,2,4-oxadiazin-5(6H)-ones. The reaction is tolerant to substituents’ electronic and steric effects in amidoximes. As a result, a series of 2-(5-oxo-3-(p-tolyl)-5,6-dihydro-4H-1,2,4-oxadiazin-6-yl)acetic acids, their methyl esters, and 1,2,4-oxadiazoles based on them were prepared and characterized by HRMS, 1H, and 13C NMR spectroscopy. The structures of three of them were elucidated with X-ray diffraction.
MicroRNAs (miRNAs) are promising biomarkers in cancer research. Quantitative PCR (qPCR), also known as real-time PCR, is the most frequently used technique for measuring miRNA expression levels. The ...use of this technique, however, requires that expression data be normalized against reference genes. The problem is that a universal internal control for quantitative analysis of miRNA expression by qPCR has yet to be known. The aim of this work was to find the miRNAs with stable expression in the thyroid gland, brain and bone marrow according to NanoString nCounter miRNA quantification data. As a results, the most stably expressed miRNAs were as follows: miR-361-3p, -151a-3p and -29b-3p in the thyroid gland; miR-15a-5p, -194-5p and -532-5p in the brain; miR-140-5p, -148b-3p and -362-5p in bone marrow; and miR-423-5p, -28-5p and -532-5p, no matter what tissue type. These miRNAs represent promising reference genes for miRNA quantification by qPCR.

Ранній неонатальний сепсис здебільшого має бактеріальну природу (у 84% випадків), у 12% – грибкову, а в 4% – вірусну та паразитарну. За даними Європейського центру контролю захворювань та ...профілактики (CDC), виникнення сепсису в новонароджених тісно пов’язане з масою тіла при народженні, гестаційним віком, а також із кольором шкіри. Встановлено, що найтяжчий стан спостерігається в дітей з масою тіла 400–1500 г і гестаційним віком менше 30 тижнів. Паразитарний сепсис у новонароджених належить до тяжких форм дитячого сепсису з високим рівнем летальності. Клінічний випадок. Наведено досвід успішного лікування 2 дітей з тяжким паразитарним сепсисом в умовах пологового будинку, коли пацієнти були нетранспортабельними. На початку лікування детально вивчено анамнез кожної вагітності. Обом пацієнтам, починаючи з першої доби, проведено комплексне інструментальне дослідження (ультразвукове і рентгенологічне), а також лабораторні дослідження крові (полімеразна ланцюгова реакція, імуноферментний аналіз, коагулограма, С-реактивний білок, прокальцитонін, загальні біохімічні тести). Першу дитину, передчасно народжену на 30-му тижні вагітності з вагою 1070 г, лікували від внутрішньоутробного хламідіозного сепсису. Другу, народжену в строк, яка страждала на внутрішньоутробний токсоплазменний сепсис і резус-конфлікт, почали лікувати внутрішньоутробно – на 30-му тижні вагітності. Обох дітей лікувала багатопрофільна команда, діти одужали. Висновки. Позитивний результат лікування демонструє ефективність патогенетичного підходу до лікування тяжких форм сепсису в новонароджених. Комплексна спеціалізована протизапальна терапія і стимуляція захисних механізмів дає змогу вилікувати новонароджених і завчасно народжених дітей, навіть із паразитарним сепсисом. Дослідження проведено відповідно до принципів Гельсінської декларації. На проведення досліджень отримано згоду батьків дітей. Автори заявляють про відсутність конфлікту інтересів.
