Traumatic brain injury (TBI) is a leading cause of morbidity and mortality. The innate and adaptive immune responses play an important role in the pathogenesis of TBI. Gamma-delta (γδ) T cells have ...been shown to affect brain immunopathology in multiple different conditions, however, their role in acute and chronic TBI is largely unknown. Here, we show that γδ T cells affect the pathophysiology of TBI as early as one day and up to one year following injury in a mouse model. TCRδ
mice are characterized by reduced inflammation in acute TBI and improved neurocognitive functions in chronic TBI. We find that the Vγ1 and Vγ4 γδ T cell subsets play opposing roles in TBI. Vγ4 γδ T cells infiltrate the brain and secrete IFN-γ and IL-17 that activate microglia and induce neuroinflammation. Vγ1 γδ T cells, however, secrete TGF-β that maintains microglial homeostasis and dampens TBI upon infiltrating the brain. These findings provide new insights on the role of different γδ T cell subsets after brain injury and lay down the principles for the development of targeted γδ T-cell-based therapy for TBI.
Aims.
We construct the molecular mass function using the bivariate
K
-band-mass function (BMF) of the
Herschel
Reference Survey (HRS), which is a volume-limited sample that has already been widely ...studied at the entire electromagnetic spectrum.
Methods.
The molecular mass function was derived from the
K
-band and the gas mass cumulative distribution using a copula method, which is described in detail in our previous papers.
Results.
The H
2
mass is relatively strongly correlated with the
K
-band luminosity because of the tight relation between the stellar mass and the molecular gas mass within the sample with a scatter, which is likely due to those galaxies which have lost their molecular content because of environmental effects or because of a larger gas consumption due to past star formation processes. The derived H
2
MF samples the molecular mass range from ∼4 × 10
6
M
⊙
to ∼10
10
M
⊙
, and when compared with theoretical models, it agrees well with the theoretical predictions at the lower end of the mass values; whereas at masses larger than 10
10
M
⊙
, the HRS sample may miss galaxies with a large content of molecular hydrogen and the outcomes are not conclusive. The value of the local density of the molecular gas mass inferred from our analysis is ∼1.5 × 10

, and it is compared with the results at larger redshifts, confirming the lack of strong evolution for the molecular mass density between
z
= 0 and
z
= 4.
Conclusions.
This is the first molecular mass function that has been derived on a complete sample in the local Universe, which can be used as a reliable calibration at redshift
z
= 0 for models aiming to predict the evolution of the molecular mass density.
The physical and chemical properties of cold and dense molecular clouds are key to understanding how stars form. Using the IRAM 30 m and NRO 45 m telescopes, we carried out a Multiwavelength ...line-Imaging survey of the 70 m-dArk and bright clOuds (MIAO). At a linear resolution of 0.1-0.5 pc, this work presents a detailed study of parsec-scale CO depletion and HCO+ deuterium (D-) fractionation toward four sources (G11.38+0.81, G15.22-0.43, G14.49-0.13, and G34.74-0.12) included in our full sample. In each source with T < 20 K and nH ∼ 104-105 cm−3, we compared pairs of neighboring 70 m bright and dark clumps and found that (1) the H2 column density and dust temperature of each source show strong spatial anticorrelation; (2) the spatial distribution of CO isotopologue lines and dense gas tracers, such as 1-0 lines of H13CO+ and DCO+, are anticorrelated; (3) the abundance ratio between C18O and DCO+ shows a strong correlation with the source temperature; (4) both the C18O depletion factor and D-fraction of HCO+ show a robust decrease from younger clumps to more evolved clumps by a factor of more than 3; and (5) preliminary chemical modeling indicates that chemical ages of our sources are ∼8 × 104 yr, which is comparable to their free-fall timescales and smaller than their contraction timescales, indicating that our sources are likely dynamically and chemically young.

Filamentary substructures labeled as F1–F6 were extracted in L1251, with F1 being associated with L1251-A hosting L1251-1. The peak positions of dense gas traced by HCO
+
are misaligned relative to those of the dust clumps. Episodic outflows are common in this region. The twisted morphology of F1 and velocity distribution along L1251-A may originate from stellar feedback. SCCC in L1251-1 may have been caused by outflow activities originated from the infrared source IRS1. The signposts of ongoing SCCC and the broadened line widths of C
3
S and C
4
H in L1251-1 as well as the distribution of HC
3
N are also related to outflow activities in this region. L1251-1 (IRS1) together with the previously identified SCCC source IRS3 demonstrate that L1251-A is an excellent region to study SCCC.
ABSTRACT
Hydrogen radio recombination lines (RRLs) are one of the major diagnostics of the physical properties of H ii regions. We use RRL H40α, He40α, and 3 mm continuum emission to investigate the ...properties of a large sample of resolved UC H ii regions identified in the ATOMS survey. In total, we identify 94 UC H ii regions from H40α emission. The basic parameters for these UC H ii regions, such as electron density, emission measure, electron temperature, ionic abundance ratio (n$_{\rm He^+}$/n$_{\rm H^+}$), and line width are derived. The median electron density and the median n$_{\rm He^+}$/n$_{\rm H^+}$ ratio of these UC H ii regions derived from RRLs are ∼9000 cm−3 and 0.11, respectively. Within UC H ii regions, the n$_{\rm He^+}$/n$_{\rm H^+}$ ratios derived from the intensity ratio of the He40α and H40α lines seems to be higher in the boundary region than in the centre. The H40α line width is mainly broadened by thermal motion and microturbulence. The electron temperature of these UC H ii regions has a median value of ∼6700 K, and its dependence on galactocentric distance is weak.
Ommochromes are major insect pigments involved in coloration of compound eyes, eggs, epidermis and wings. In the silkworm Bombyx mori, adult compound eyes and eggs contain a mixture of the ommochrome ...pigments such as ommin and xanthommatin. Here, we identified the gene involved in ommochrome biosynthesis by positional cloning of B. mori egg and eye color mutant pink-eyed white egg (pe). The recessive homozygote of pe has bright red eyes and white or pale pink eggs instead of a normal dark coloration due to the decrease of dark ommochrome pigments. By genetic linkage analysis, we narrowed down the pe-linked region to ~258 kb, containing 17 predicted genes. RNA sequencing analyses showed that the expression of one candidate gene, the ortholog of Drosophila haem peroxidase cardinal, coincided with egg pigmentation timing, similar to other ommochrome-related genes such as Bm-scarlet and Bm-re. In two pe strains, a common missense mutation was found within a conserved motif of B. mori cardinal homolog (Bm-cardinal). RNA interference-mediated knockdown and transcription activator-like effector nuclease (TALEN)-mediated knockout of the Bm-cardinal gene produced the same phenotype as pe in terms of egg, adult eye and larval epidermis coloration. A complementation test of the pe mutant with the TALEN-mediated Bm-cardinal-deficient strain showed that the mutant phenotype could not be rescued, indicating that Bm-cardinal is responsible for pe. Moreover, knockdown of the cardinal homolog in Tribolium castaneum also induced red compound eyes. Our results indicate that cardinal plays a major role in ommochrome synthesis of holometabolous insects.
An assay using a mouse antisialyl Lewis X (sLeX) antibody (CSLEX-1) is used clinically for screening and monitoring patients with breast cancer in Japan. However, the IgM isoform of CSLEX-1 is not ...preferred for the assay because the bulkiness of IgM generally causes poor accessibility to the antigen. To solve this problem, we developed an antisLeX mouse/human chimeric IgG antibody, CH-CSLEX-1, using transgenic silkworms. The performance of a homologous sandwich ELISA of CH-CSLEX1 was then evaluated.
To generate CH-CSLEX-1, we used a GAL4/UAS binary gene expression system in transgenic silkworms. The reactivities of CSLEX-1 and CH-CSLEX-1 were determined in a Biacore analysis. To confirm antigen specificity, 3 antigens sLeX, sLeA, and Lewis Y (LeY) were used.
CH-CSLEX-1 formed correctly as an IgG class of immunoglobulin molecule with an isoelectric point close to the predicted value. The best combination for capturing and probing in a sandwich ELISA was determined as a homologous combination of CH-CSLEX-1. The CH-CSLEX-1 assay specifically detected sLeX, but not sLeA and LeY. A correlation analysis with 107 human samples showed good concordance between the conventional CSLEX-1 assay (homologous sandwich ELISA using CSLEX-1) and the CH-CSLEX-1 assay (r = 0.98). Moreover, the CH-CSLEX-1 assay was not affected by either human antimouse IgG antibodies (HAMA IgG) or HAMA IgM.
The mouse/human chimeric antibody CH-CSLEX-1 allowed the establishment of a highly specific sandwich ELISA for sLeX that was not affected by HAMA.
We present the results of CS J = 2 → 1 mapping observations toward 39 massive star-forming regions selected from the previous CO line survey of cold IRAS sources with high-velocity CO flows along the ...Galactic plane. All sources are detected in CS J = 2 → 1, showing the existence of CS clumps around the IRAS sources. However, one-third of the sources are not deeply embedded in the dense clumps by comparison of the central powering IRAS sources and the morphologies of CS clumps. Physical parameters of the dense molecular clumps are presented. We have identified 12 high-mass protostellar object (HMPO) candidates by checking the association between the dense cores and the IRAS sources, the detection of water maser, and the radio properties toward the IRAS sources. We find that the HMPO sources are characterized by low FIR luminosity to virial mass ratios since they are in very early evolutionary stages when the massive protostars have not reached their full luminosities, which are typical for zero-age main-sequence stars. Large turbulent motion in the HMPO sources may be largely due to the large kinetic energy ejected by the central protostars formed in the dense clumps. However, alternative means or undetected outflows may also be responsible for the turbulence in the clumps.

all-sky survey provided a large catalog of ...Galactic cold clouds and clumps that have been the target of several follow-up surveys.
Aims.
We aim to characterize a diverse selection of dense, potentially star-forming cores, clumps, and clouds within the Milky Way in terms of their dust emission and SF activity.
Methods.
We studied 53 fields that have been observed in the JCMT SCUBA-2 continuum survey SCOPE and have been mapped with
Herschel
. We estimated dust properties by fitting
Herschel
observations with modified blackbody functions, studied the relationship between dust temperature and dust opacity spectral index
β
, and estimated column densities. We extracted clumps from the SCUBA-2 850 μm maps with the FellWalker algorithm and examined their masses and sizes. Clumps are associated with young stellar objects found in several catalogs. We estimated the gravitational stability of the clumps with virial analysis. The clumps are categorized as unbound starless, prestellar, or protostellar.
Results.
We find 529 dense clumps, typically with high column densities from (0.3–4.8) × 10
22
cm
−2
, with a mean of (1.5 ± 0.04) ×10
22
cm
−2
, low temperatures (
T
∼ 10–20 K), and estimated submillimeter
β
= 1.7 ± 0.1. We detect a slight increase in opacity spectral index toward millimeter wavelengths. Masses of the sources range from 0.04
M
⊙
to 4259
M
⊙
. Mass, linear size, and temperature are correlated with distance. Furthermore, the estimated gravitational stability is dependent on distance, and more distant clumps appear more virially bound. Finally, we present a catalog of properties of the clumps.
Conclusions.
Our sources present a large array of SF regions, from high-latitude, nearby diffuse clouds to large SF complexes near the Galactic center. Analysis of these regions will continue with the addition of molecular line data, which will allow us to study the densest regions of the clumps in more detail.
Gamma-delta (γδ) T cells are a major cell population in the intestinal mucosa and are key mediators of mucosal tolerance and microbiota composition. Little is known about the mechanisms by which ...intestinal γδ T cells interact with the gut microbiota to maintain tolerance.
We found that antibiotic treatment impaired oral tolerance and depleted intestinal γδ T cells, suggesting that the gut microbiota is necessary to maintain γδ T cells. We also found that mice deficient for γδ T cells (γδ
) had an altered microbiota composition that led to small intestine (SI) immune dysregulation and impaired tolerance. Accordingly, colonizing WT mice with γδ
microbiota resulted in SI immune dysregulation and loss of tolerance whereas colonizing γδ
mice with WT microbiota normalized mucosal immune responses and restored mucosal tolerance. Moreover, we found that SI γδ T cells shaped the gut microbiota and regulated intestinal homeostasis by secreting the fecal micro-RNA let-7f. Importantly, oral administration of let-7f to γδ
mice rescued mucosal tolerance by promoting the growth of the γδ
-microbiota-depleted microbe Ruminococcus gnavus.
Taken together, we demonstrate that γδ T cell-selected microbiota is necessary and sufficient to promote mucosal tolerance, is mediated in part by γδ T cell secretion of fecal micro-RNAs, and is mechanistically linked to restoration of mucosal immune responses. Video Abstract.
