The island fox (Urocyon littoralis catalinae) population on Santa Catalina Island, California, USA declined precipitously in 1999 with an approximate 95% reduction on their eastern range, an area ...representing 87% of the island. During this investigation, between October 1999 and April 2000, evidence of live foxes dramatically decreased. The only carcass recovered during the decline succumbed to a co-infection of canine distemper virus (CDV) and toxoplasmosis. Sequence analysis of the viral P gene, derived by polymerase chain reaction, indicated that the virus was closely related to CDV from a mainland USA raccoon (Procyon lotor). Nine of 10 foxes trapped in 1999–2000, on the eastern portion of the island after the decline, had serologic evidence of exposure to CDV, whereas only four of 19 foxes trapped in this region in 1998 had antibodies reactive against CDV. The confirmation of CDV in one deceased fox, evidence of exposure to CDV in east-end foxes in 1999–2000 compared to 1998, and documentation of raccoon introductions to the island, implicates canine distemper as the cause of the population decline.

Viral hemagglutinin (H)-gene sequence was up to 99% similar to strains circulating contemporaneously in regional wildlife. CDV lesions in raccoons and skunk were primarily encephalitis. A few affected felids had at least one previous vaccination for CDV, while most felids at the center were vaccinated during the outbreak.

The cheetah (Acinonyx jubatus)is highly endangered because of loss of habitat in the wild and failure to thrive in captivity. Cheetahs in zoos reproduce poorly and have high prevalences of unusual ...diseases that cause morbidity and mortality. These diseases are rarely observed in free-ranging cheetahs but have been documented in cheetahs that have been captured and held in captive settings either temporarily or permanently. Because captivity may be stressful for this species and stress is suspected as contributing to poor health and reproduction, this study aimed to measure chronic stress by comparing baseline concentrations of fecal corticoid metabolites and adrenal gland morphology between captive and free-ranging cheetahs. Additionally, concentrations of estradiol and testosterone metabolites were quantified to determine whether concentrations of gonadal steroids correlated with corticoid concentration and to assure that corticosteroids in the free-ranging samples were not altered by environmental conditions. Concentrations of fecal corticoids, estradiol, and testosterone were quantified by radioimmunoassay in 20 free-ranging and 20 captive cheetahs from samples collected between 1994 and 1999. Concentrations of baseline fecal corticoids were significantly higher (p=0.005) in captive cheetahs (196.08±36.20 ng/g dry feces) than free-ranging cheetahs (71.40±14.35 ng/g dry feces). Testosterone concentrations were lower in captive male cheetahs (9.09±2.84 ng/g dry feces) than in free-ranging cheetahs (34.52±12.11 ng/g dry feces), which suggests suppression by elevated corticoids in the captive males. Evidence for similar suppression of estradiol concentrations in females was not present. Adrenal corticomedullary ratios were determined on midsagittal sections of adrenal glands from 13 free-ranging and 13 captive cheetahs obtained between 1991 and 2002. The degree of vacuolation of cortical cells in the zona fasciculata was graded for each animal. Corticomedullary ratios were larger (p=0.05) in captive cheetahs; however, there was no difference (p=0.31) in the degree of corticocyte vacuolation between the two populations. These data provide both morphologic and functional evidence suggestive of chronic stress in captive cheetahs. Further research into the role of hypercortisolemia in the pathogenesis of the reproductive abnormalities and unusual diseases of captive cheetahs is needed.
During a population decline or disease outbreak, the true risk of specific diseases to a wild population is often difficult to determine because of a lack of baseline disease information. To better ...understand the risk of disease in an endangered and scientifically important population of chimpanzees (Pan trogylodytes schweinfurthii), a health monitoring program was initiated in Gombe National Park, Tanzania. As part of this health monitoring program, comprehensive necropsies with histopathology were conducted on chimpanzees (n = 11; 5 male, 6 female), ranging in age from fetal to 44 yr, that were found dead between August 2004 and January 2010. In contrast to previous reports, respiratory disease was not noted as a cause of morbidity or mortality. Trauma was the most common cause of death in these 11 chimpanzees. All of the chimpanzees greater than 1 yr of age had intestinal and mesenteric parasitic granulomas associated with true strongyles consistent with Oesophagostomum spp. The relative numbers of granulomas increased with age and, in some cases, may have been a cause of weight loss and diarrhea. Simian immunodeficiency virus (SIV)cpz infection was documented in four deceased apes, all of whom exhibited varying amounts of lymphoid depletion including two females with marked CD4+ T cell loss consistent with end-stage SIVmac or human immunodeficiency virus infections. Myocardial megalokaryosis was common in chimpanzees greater than 1 mo of age; yet myocardial interstitial fibrosis, a common lesion in captive chimpanzees, was uncommon and only noted in two aged chimpanzees. These findings provide important information on causes of morbidity and mortality in wild chimpanzees, information that can be used to interpret findings during population declines and lead to better management of this population in the context of disease risk.
The reproductive tracts of three captive male aardvark ( Orycteropus afer) were evaluated to characterize the gross and histological anatomy, with correlations to ultrasonographic and computed ...tomographic imaging. Observations were made from a reproductive tract examined at necropsy, with subsequent evaluation of tissues histologically. Two living specimens were evaluated via ultrasonography with a 10-MHz linear transducer. One living animal was also evaluated via computed tomography. Prominent external scent glands were present at the base of the prepuce. Testicles were present internally at the level of the inguinal canal and capable of sliding into a subcutaneous position. Accessory sex glands consisted of seminal vesicles, prostate gland, and bulbourethral glands, with histological characteristics similar to other species. Ultrasonography was an effective tool for evaluation of internal and external reproductive structures, while the usefulness of computed tomography was limited in the evaluation of pelvic organs due to artifact from nearby bony structures. While a larger study population is desirable, this report provides an important comparative anatomical reference and will help improve the clinical management and care of this species.
To investigate cases of acute oxalate nephrosis without evidence of ethylene glycol exposure, archived data and tissues from cheetahs (Acinonyx jubatus) from North America (n = 297), southern Africa ...(n = 257), and France (n = 40) were evaluated. Renal and gastrointestinal tract lesions were characterized in a subset of animals with (n = 100) and without (n = 165) oxalate crystals at death. Crystals were confirmed as calcium oxalate by Raman spectroscopy in 45 of 47 cheetahs tested. Crystals were present in cheetahs from 3.7 months to 15.9 years old. Cheetahs younger than 1.5 years were less likely to have oxalates than older cheetahs (P = .034), but young cheetahs with oxalates had more oxalate crystals than older cheetahs (P < .001). Cheetahs with oxalate crystals were more likely to have renal amyloidosis, interstitial nephritis, or colitis and less likely to have glomerular loop thickening or gastritis than those without oxalates. Crystal number was positively associated with renal tubular necrosis (P ≤ .001), regeneration (P = .015), and casts (P ≤ .001) but inversely associated with glomerulosclerosis, renal amyloidosis, and interstitial nephritis. Crystal number was unrelated to the presence or absence of colitis and was lower in southern African than American and European animals (P = .01). This study found no evidence that coexisting chronic renal disease (amyloidosis, interstitial nephritis, or glomerulosclerosis), veno-occlusive disease, gastritis, or enterocolitis contributed significantly to oxalate nephrosis. Oxalate-related renal disease should be considered as a potential cause of acute renal failure, especially in young captive cheetahs. The role of location, diet, stress, and genetic predisposition in the pathogenesis of oxalate nephrosis in cheetahs warrants further study.
Summary Tuberculosis is an important health concern for Asian elephant ( Elephas maximus ) populations worldwide, however, mechanisms underlying susceptibility to Mycobacterium tuberculosis are ...unknown. Proliferative responses assessed via brominated uridine incorporation and cytokine expression measured by real-time RT-PCR were evaluated in peripheral blood mononuclear cell (PBMC) cultures from 8 tuberculosis negative and 8 positive Asian elephants. Cultures were stimulated with Mycobacterium bovis purified protein derivative (PPD-B), M. tuberculosis culture filtrate protein (CFP)-10, and Mycobacterium avium PPD (PPD-A). Following stimulation with PPD-B, proliferation was higher ( α = 0.005) in positive samples; no significant differences were detected following CFP-10 or PPD-A stimulation. Tumor necrosis factor (TNF)-α, interleukin (IL)-12, and interferon (IFN)-γ expression was greater in samples from positive elephants following stimulation with PPD-B ( α = 0.025) and CFP-10 ( α = 0.025 TNF-α and IL-12; α = 0.005 IFN-γ). Stimulation with PPD-A also produced enhanced IL-12 expression in positive samples ( α = 0.025). Findings suggested that differences in immune cell function exist between tuberculosis positive and negative elephants. Proliferative responses and expression of TNF-α, IL-12, and IFN-γ in response to stimulation with PPD-B and CFP-10 differ between tuberculosis positive and negative elephants, suggesting these parameters may be important to tuberculosis immunopathogenesis in this species.
Four fishers (Martes pennanti) from an insular population in the southern Sierra Nevada Mountains, California, USA died as a consequence of an infection with canine distemper virus (CDV) in 2009. ...Three fishers were found in close temporal and spatial relationship; the fourth fisher died 4 mo later at a 70 km distance from the initial group. Gross lesions were restricted to hyperkeratosis of periocular skin and ulcera-tion of footpads. All animals had necrotizing bronchitis and bronchiolitis with syncytia and intracytoplasmic inclusion bodies. Inclusion bodies were abundant in the epithelia of urinary bladder and epididymis but were infrequent in the renal pelvis and the female genital epithelia. No histopathologic or immu-nohistochemical evidence for virus spread to the central nervous system was found. One fisher had encephalitis caused by Sarcocystis neurona and another had severe head trauma as a consequence of predation. The H gene nucleo-tide sequence of the virus isolates from the first three fishers was identical and was 99.6% identical to the isolate from the fourth fisher. Phylogenetically, the isolates clustered with other North American isolates separate from classical European wildlife lineage strains. These data suggest that the European wildlife lineage might consist of two separate subgroups that are genetically distinct and endemic in different geographic regions. The source of infection as well as pertinent transmission routes remained unclear. This is the first report of CDV in fishers and underscores the significance of CDV as a pathogen of management concern.
