Memory Thy-1(+)CD8(+) T cells specific for the influenza A virus nucleoprotein (NP(366-374)) peptide were sorted after staining with the D(b)NP(366) tetramer, labeled with CFSE, and transferred into ...normal Thy-1.2(+) recipients. The donor D(b)NP(366)(+) T cells recovered 2 days later from the spleens of the Thy-1.2(+) hosts showed the CD62L(low)CD44(high)CD69(low) phenotype, characteristic of the population analyzed before transfer, and were present at frequencies equivalent to those detected previously in mice primed once by a single exposure to an influenza A virus. Analysis of CFSE-staining profiles established that resting tetramer(+) T cells divided slowly over the next 30 days, while the numbers in the spleen decreased about 3-fold. Intranasal infection shortly after cell transfer with a noncross-reactive influenza B virus induced some of the donor D(b)NP(366)(+) T cells to cycle, but there was no increase in the total number of transferred cells. By contrast, comparable challenge with an influenza A virus caused substantial clonal expansion, and loss of the CFSE label. Unexpectedly, the recruitment of naive Thy-1.2(+)CD8(+)D(b)NP(366)(+) host D(b)NP(366)(+) T cells following influenza A challenge was not obviously diminished by the presence of the memory Thy-1.1(+)CD8(+)D(b)NP(366)(+) donor D(b)NP(366)(+) set. Furthermore, the splenic response to an epitope (D(b)PA(224)) derived from the influenza acid polymerase (PA(224-233)) was significantly enhanced in the mice given the donor D(b)NP(366)(+) memory population. These experiments indicate that an apparent recall response may be comprised of both naive and memory CD8(+) T cells.
BackgroundOptimal feeding of very low birthweight (VLBW <1500 g)/very preterm (gestation <32 weeks) infants in resource-limited settings in sub-Saharan Africa (sSA) is critical to reducing high ...mortality and poor outcomes.ObjectiveTo review evidence on feeding of VLBW/very preterm infants relevant to sSA.MethodsWe searched the Cochrane Database of Systematic Reviews, Embase, PubMed and Cumulative Index to Nursing and Allied Health Literature (CINAHL) from inception to July 2019 to identify reviews of randomised and quasi-randomised controlled trials of feeding VLBW/very preterm infants. We focused on interventions that are readily available in sSA. Primary outcomes were weight gain during hospital stay and time to achieve full enteral feeds (120 mL/kg/day). Secondary outcomes were growth, common morbidities, mortality, duration of hospital stay and cognitive development. Quality of evidence (QOE) was assessed using the Measurement Tool to Assess Systematic Reviews (AMSTAR2).ResultsEight systematic reviews were included. Higher feed volume of day 1 (80 mL/kg) reduced late-onset sepsis and time to full enteral feeds, and higher feed volume (up to 300 mL/kg/day) improved weight gain without adverse events (QOE: low–moderate). Rapid advancement of feeds (30–40 mL/kg/day) was not associated with harm. Breast milk fortification with energy and protein increased growth and with prebiotics increased growth and reduced duration of admission (QOE: low–very low) and did not result in harm. Evidence regarding feeding tube placement and continuous versus bolus feeds was insufficient to draw conclusions. We found no reviews meeting our selection criteria regarding when to start feeds, use of preterm formula, cup-and-spoon feeding or gravity versus push feeds and none of the reviews included trials from low-income countries of sSA.ConclusionsThe evidence base informing feeding of VLBW/very preterm babies in resource-limited settings in sSA is extremely limited. Pragmatic studies are needed to generate evidence to guide management and improve outcomes for these highly vulnerable infants.PROSPERO registration numberCRD42019140204.

This 2020 International Consensus on Cardiopulmonary Resuscitation and Emergency Cardiovascular Care Science With Treatment Recommendations (CoSTR) for pediatric life support is based on the most ...extensive evidence evaluation ever performed by the Pediatric Life Support Task Force. Three types of evidence evaluation were used in this review: systematic reviews, scoping reviews, and evidence updates. Per agreement with the evidence evaluation recommendations of the International Liaison Committee on Resuscitation, only systematic reviews could result in a new or revised treatment recommendation.
Systematic reviews performed for this 2020 CoSTR for pediatric life support included the topics of sequencing of airway-breaths-compressions versus compressions-airway-breaths in the delivery of pediatric basic life support, the initial timing and dose intervals for epinephrine administration during resuscitation, and the targets for oxygen and carbon dioxide levels in pediatric patients after return of spontaneous circulation. The most controversial topics included the initial timing and dose intervals of epinephrine administration (new treatment recommendations were made) and the administration of fluid for infants and children with septic shock (this latter topic was evaluated by evidence update). All evidence reviews identified the paucity of pediatric data and the need for more research involving resuscitation of infants and children.
The international field campaign called the Convective and Orographically-induced Precipitation Study (COPS) took place from June to August 2007 in southwestern Germany/eastern France. The ...overarching goal of COPS is to advance the quality of forecasts of orographically-induced convective precipitation by four-dimensional observations and modeling of its life cycle.
COPS was endorsed as one of the Research and Development Projects of the World Weather Research Program (WWRP), and combines the efforts of institutions and scientists from eight countries. A strong collaboration between instrument principal investigators and experts on mesoscale modeling has been established within COPS. In order to study the relative importance of large-scale and small-scale forcing leading to convection initiation in low mountains, COPS is coordinated with a one-year General Observations Period in central Europe, the WWRP Forecast Demonstration Project MAP D-PHASE, and the first summertime European THORPEX Regional Campaign. Furthermore, the Atmospheric Radiation Measurement program Mobile Facility operated in the central COPS observing region for nine months in 2007.
The article describes the scientific preparation of this project and the design of the observation systems. COPS will rest on three pillars: A unique synergy of observing systems, the next-generation high-resolution mesoscale models with improved model physics, and advanced data assimilation and ensemble prediction systems. These tools will be used to separate and to quantify errors in quantitative precipitation forecasting as well as to study the predictability of convective precipitation.

The star HD 193322 is a remarkable multiple system of massive stars that lies at the heart of the cluster Collinder 419. Here we report on new spectroscopic observations and radial velocities of the ...narrow-lined component Ab1 which we use to determine its orbital motion around a close companion Ab2 (P = 312 days) and around a distant third star Aa (P = 35 years). We have also obtained long baseline interferometry of the target in the K' band with the CHARA Array which we use in two ways. First, we combine published speckle interferometric measurements with CHARA separated fringe packet measurements to improve the visual orbit for the wide Aa,Ab binary. Second, we use measurements of the fringe packet from Aa to calibrate the visibility of the fringes of the Ab1,Ab2 binary, and we analyze these fringe visibilities to determine the visual orbit of the close system. The two most massive stars, Aa and Ab1, have masses of approximately 21 and 23 M , respectively, and their spectral line broadening indicates that they represent extremes of fast and slow projected rotational velocity, respectively.
Pre-exercise carbohydrate (CHO) ingestion is an effective strategy for reducing the occurrence of hypoglycemia during or after exercise in individuals with type 1 diabetes (T1DM). The metabolic ...effects of ingestion of different CHOs for glycemic or performance gains have been under-researched. This study compared metabolic responses and fuel use during sub-maximal and high-intensity performance running following pre-exercise ingestion of waxy barley starch (WBS) or dextrose (DEX) in T1DM.
Seven participants attended the laboratory on two separate occasions following preliminary testing. On each visit participants consumed either 0.6 g/kg body mass of DEX or WBS 2 hours before a 26-minute discontinuous incremental treadmill protocol (4-minute running: 1.5-min rest) finishing at 80±4% V̇O2peak followed by a 10-min performance run on a non-motorized treadmill. Capillary blood samples were taken at rest, during and following exercise and analyzed for glucose (BG) and acid-base variables. Data (mean ± SEM) were analyzed using repeated measures ANOVA (P<0.05).
BG reached similar peak values one hour after CHO ingestion and immediate pre-sub-maximal exercise BG were comparable. Resting CHO oxidation was elevated and lipid oxidation lower under WBS (P<0.05). There were no metabolic or cardio-respiratory differences during the sub-maximal exercise (P>0.05). In the final quartile of the performance run, a greater distance was completed under WBS (WBS 323±21 vs. DEX 301±20 m, P=0.02).
Consumption of WBS demonstrated similar hyperglycemic responses to dextrose ingestion but a greater rate of CHO use at rest. Interestingly, T1DM individuals displayed an improved performance at the latter stages of a high-intensity run test.
The Wiskott-Aldrich syndrome (WAS) is an X-linked disorder characterized by immune dysfunction, thrombocytopenia, and eczema. We used a murine model created by knockout of the WAS protein gene (WASP) ...to evaluate the potential of gene therapy for WAS. Lethally irradiated, male WASP— animals that received transplants of mixtures of wild type (WT) and WASP— bone marrow cells demonstrated enrichment of WT cells in the lymphoid and myeloid lineages with a progressive increase in the proportion of WT T-lymphoid and B-lymphoid cells. WASP— mice had a defective secondary T-cell response to influenza virus which was normalized in animals that received transplants of 35% or more WT cells. The WASP gene was inserted into WASP— bone marrow cells with a bicistronic oncoretroviral vector also encoding green fluorescent protein (GFP), followed by transplantation into irradiated male WASP— recipients. There was a selective advantage for gene-corrected cells in multiple lineages. Animals with higher proportions of GFP+ T cells showed normalization of their lymphocyte counts. Gene-corrected, blood T cells exhibited full and partial correction, respectively, of their defective proliferative and cytokine secretory responses to in vitro T-cell–receptor stimulation. The defective secondary T-cell response to influenza virus was also improved in gene-corrected animals.
We investigated the hypothesis that different prefrontal brain systems (i.e., dorsal vs. ventral) and sex contribute differentially to cognitive deficit in schizophrenia. Performance was assessed ...among clinically stable, chronic schizophrenic outpatients and matched normal control subjects on olfactory identification on the University of Pennsylvania Smell Identification Test (UPSIT) and on executive functions using the Wisconsin Card Sorting Test (WCST). Patients were impaired on both tests compared to controls, and male schizophrenics were impaired on the WCST compared to female schizophrenics. The pattern of results suggests that gender differences on the UPSIT are mildly accentuated in schizophrenia. The data support our previous study indicating that UPSIT performance is largely independent of the executive or attentional deficits typically associated with schizophrenia, with the exception of verbal ability. Further research with larger samples is required to test the hypothesis that there is a severely impaired subgroup of male patients with diffuse prefrontal dysfunctions.
Toward the end of the Moore's-law era, increases in system complexity will rely more heavily on packaging technology. Systems will increasingly comprise multiple chips that must be linked by ...high-speed data channels carrying a substantial fraction of on-chip bandwidth. To take advantage of inexpensive organic packages and conventional printed circuit (PC) boards, data links that are both energy and pin efficient are needed. A link between neighboring packages is by far the more challenging application due to increased cross-talk, signal attenuation, and reflections from impedance discontinuities. The combination of signal integrity challenges and production margining requires increased amplitude, equalization, ESD protection, and PVT-tolerant circuit design techniques.
OBJECTIVE:To assess the prevalence of undiagnosed cardiovascular disease (CVD) in a cohort of male steelworkers in South Wales, UK.
METHODS:Male steel industry workers (n = 221) with no prior ...diagnosis of CVD or diabetes accepted a CVD risk assessment within the work environment. Demographic, anthropometric, family, and medical histories were all recorded and capillary blood samples obtained. The 10-year CVD risk was predicted using the QRISK2–2012 algorithm.
RESULTS:Up to 81.5% of workers were either overweight or obese. More than 20% of workers were found to have diastolic hypertension, high total cholesterol, and/or a total cholesterol/high-density lipoprotein ratio of six or more. Over one quarter of workers assessed had an increased 10-year CVD risk.
CONCLUSIONS:Despite a physically demanding occupation, risk assessment in the workplace uncovered significant occult factors in CVD risk in a sample of male heavy industry workers.
