In this study, we ...evaluated their effect on the expression of SIRT1 and TGF‐β1 in peripheral blood as well as their participation in the formation of DNA–protein complexes in a pancreas‐derived cell line. It has been described that SIRT1 and TGF‐β1 participate in cell growth and regulation of production and secretion of insulin in the pancreas. Anthropometric and biochemical profiles of 127 adults were measured. Genotypes for rs3758391 and rs1800470 were determined using TaqMan assays. Expression analysis of SIRT1 and TGF‐β1 were performed using real‐time PCR. Gene expression of these genes increased 1.8 ± 0.6‐ and 1.3 ± 0.6‐fold in patients carrying the TT genotype of rs3758391 and rs1800470 when compared to carriers of the CC genotype. Then, we tested whether these single‐nucleotide polymorphisms (SNPs) (and rs932658, which is in linkage disequilibrium with rs3758391) are located in regulatory DNA‐protein binding sites by electrophoretic mobility shift assays using nuclear extract from the pancreas‐derived cell line BxPC‐3. The electrophoretic mobility shift assay showed no binding of nuclear proteins to DNA. In conclusion, the genotypes of rs3758391 and rs1800470 are associated with modifications in the expression of the genes SIRT1 and TGF‐β1, respectively, but none of the tested SNPs are located in regulatory DNA‐protein binding sites.
The organic cation transporters OCT1 and OCT2 and the multidrug and toxin extrusion transporter MATE1, encoded by the SLC22A1, SLC22A2, and SLC47A1 genes, respectively, are responsible for the ...absorption of metformin in enterocytes, hepatocytes, and kidney cells. The aim of this study was to evaluate whether genetic variations in the SLC22A1, SLC22A2, and SLC47A1 genes could be associated with an altered response to metformin in patients with type 2 diabetes mellitus. A cohort study was conducted in 308 individuals with a diagnosis of type 2 diabetes mellitus of less than 3 years and who had metformin monotherapy. Three measurements of blood glycated hemoglobin (HbA1c) were obtained at the beginning of the study and after 6 and 12 months. Five polymorphisms were analyzed in the SLC22A1 (rs622342, rs628031, rs594709), SLC22A2 (rs316019), and SLC47A1 (rs2289669) genes by real‐time polymerase chain reaction. The results showed a significant association among genotypes CC‐rs622342 (β = 1.36; P < .001), AA‐rs628031 (β = 0.98; P = .032), and GG‐rs594709 (β = 1.21; P = .016) in the SLC22A1 gene with an increase in HbA1c levels during the follow‐up period. Additionally, a significant association was found in the CGA and CAG haplotypes with an increase in HbA1c levels compared to the highest‐frequency haplotype (AGA). In conclusion, the genetic variation in the SLC22A1 gene was significantly related to the variation of the HbA1c levels, an important indicator of glycemic control in diabetic patients. This information may contribute to identifying patients with an altered response to metformin before starting their therapy.
Abstract
This study aimed to evaluate the effects of adding 0% (T1), 6% (T2), 12% (T3) and 18% (T4) of a mixture (83% dried citrus pulp (DCP) and 17% eggshell) into a sorghum-soybean meal-limestone ...based diet for laying quails, on production efficiency and quality of egg. Thirty-two quails were individually housed and randomly assigned (n = 8) to the four experimental diets, during the first 5 week of the laying cycle. Quail LW and feed intake were recorded weekly. Produced eggs were weighed (n = 457) and egg mass production recorded. Proportions of albumen, yolk and shell, egg yolk color, and shell thickness, were determined (n = 55/treatment). On week 5, total excreta of 24 laying quails (n = 6) were collected throughout 96 h, to determine metabolizable utilization of DM, energy, crude protein and ash. Statistical analyses were performed with SPSS® software. Initial and final LW, feed intake and egg production were similar (P >0.05) among treatments. Egg mass was higher (P < 0.05) in T2 than T4. Egg weight of quails fed T1 and T2 was higher than others (P < 0.05). The eggshell of quails fed T2 was thicker (248 vs 236 μm; P < 0.05) than T1. Eggshell proportion of quails fed the mixture was higher (P < 0.05) than in T1 (11.3 vs 10.5%). Yolk proportion of T1 was higher (P < 0.05). The b* parameter was lower (P < 0.05) for T1. Metabolizable utilization of dietary DM (72.1%) and energy (68.8%) of T1 was 13% higher (P < 0.05) than T4, however protein (avg 41.4%) and ash (avg 46.7%) utilization were similar (P > 0.05) among treatments. In conclusion, adding up to 12% of the mixture (83% DCP and 17% eggshell) in quail diets, resulted in similar egg production parameters, and metabolizable utilization of dietary DM, crude protein, energy and ash, however improved thickness of eggshell, compared to the control diet.
To evaluate the effect of oxytocin as a cervical dilator, a study was carried out on nulliparous goats inseminated transcervically at the beginning of the breeding season. One hundred sixteen ...nulliparous goats with a mean live weight of 33.4 ± 0.68 kg and an age of 13.7 ± 0.37 months were used. The goats were exposed to active bucks of proven fertility for a period of 14 d in order to induce oestrus. One week later, the Ovsynch protocol was applied, which consisted of the application of 20 mg of gonadorelin (Day Zero), 0.075 mg of cloprostenol (Day 7) and of a second dose of 20 mg of gonadorelin applied on Day 9. Artificial insemination (AI) was performed 16 hr later. Three treatments were evaluated: T1 = 50 IU saline, T2 = 25 IU oxytocin; T3 = 50 IU of oxytocin, intravenously applied 10–15 min before AI. The time required to inseminate each treated goat from groups T2 and T3 was 49.56 and 56.25 s, respectively, versus 85.78 s needed for the goats from group T1 (p < .0001). In the T1 group of goats, the insemination catheter was inserted 2.1 cm into the cervical canal and in goats from groups T2 and T3 it reached 3.41 and 3.77 cm into the cervical canal, respectively (p = .02). Pregnancy rates and prolificacy (kids/doe) were higher (p = .02) for groups T2 (82.93%; 1.16) and T3 (76.92%; 1.21) respectively than for control goats (61.11%; 0.69). In conclusion, the intravenous administration of oxytocin led to greater dilation and depth of cervical penetration, obtaining higher pregnancy rates and prolificacy.
Abstract
The objective of this study was to evaluate the effect of supplementing 10% of sorghum grain (SG), soybean meal (SBM) or dried citrus pulp (DCP) upon dry matter digestibility (DMD), organic ...matter digestibility (OMD), and metabolizable energy content (ME) of diets based on dried leaves of two shrub species (Acacia farnesiana and Acacia rigidula) and one grass (Cenchrus ciliaris) browsed by small ruminants. Twelve experimental diets were elaborated using three forage sources: C. ciliaris, A. farnesiana and A. rigidula and evaluated either alone (0% supplementation) or supplemented with 10% of SG (9.0% CP, 22.3% NDF), SBM (42.8% CP, 16.3% NDF) or DCP (4.9% CP, 19.1% NDF). In vitro DMD (Daisy incubator, ANKOM) at 48 h, and in vitro gas production (GP, in calibrated 100 mL glass syringes) at 24 h, were determined by incubating samples in an inoculum prepared by using rumen liquor of two rumen fistulated sheep and artificial saliva in a 1:4 ratio. OMD and ME content were calculated from the gas production, crude protein, ash, and ether extract content. Data were evaluated according to a 3 x 4 factorial experiment. There were no significant (P > 0.05) forage source x supplement interaction effects. Leaves of A. farnesiana had higher (P < 0.05) DMD (64%), GP (26 mL), ME (1910 kcal ME/kg DM), and OMD (53%) than other forages (mean 47% DMD, 17 mL GP, 1232 kcal ME/kg DM, and 39% DMO). Supplements increased (P < 0.05) DMD (average +8%), but only DCP increased (P < 0.05) the GP (+23%) of evaluated forages. Leaves of A. farnesiana had higher DMD (P < 0.05; 64%). There were no effects (P >0.05) of the 10% added supplements in ME content or DMO. In conclusion, leaves of A. farnesiana and DCP are valuable sources of nutrients and energy for small ruminants in northeastern Mexico.

The fast development of today’s healthcare and the need to extract new medical knowledge from exponentially-growing volumes of standardized Electronic Health Records data, as required by studies in Precision Medicine, brings up a challenge that may probably only be addressed using NoSQL DBMSs, due to the non-optimal performance of traditional relational DBMSs on standardized data; and these database systems operated by semantic archetype-based query languages, because of the expected generalized extension of standardized EHR systems. An AQL into MongoDB interpreter has been developed to its first version. It translates system-independent AQL queries posed on ISO/EN 13606 standardized EHR extracts into the NoSQL MongoDB query language. The new interpreter has the advantages of both the archetype-based system-independent AQL queries and the dual-model-based standardized EHR extracts stored on document-centric NoSQL DBMSs, such as MongoDB. AQL queries are independent of applications, programming languages and system environments due to the use of the dual model, but EHR extracts featuring this model are best persisted on document-based NoSQL databases. Consequently, the interpreter allows us to query standardized EHR extracts semantically, and also affording optimal performance.
The purpose of this study was to assess the prevalence, associated factors, and impact on mortality of primary respiratory disease in a large systemic lupus erythematosus (SLE) retrospective cohort.
...All adult patients in the RELESSER-TRANS (Registry of Systemic Lupus Erythematosus Patients of the Spanish Society of Rheumatology SER, cross-sectional phase) registry were retrospectively investigated for the presence of primary pleuropulmonary manifestations.
In total 3215 patients were included. At least one pleuropulmonary manifestation was present in 31% of patients. The most common manifestation was pleural disease (21%), followed by lupus pneumonitis (3.6%), pulmonary thromboembolism (2.9%), primary pulmonary hypertension (2.4%), diffuse interstitial lung disease (2%), alveolar hemorrhage (0.8%), and shrinking lung syndrome (0.8%). In the multivariable analysis, the variables associated with the development of pleuropulmonary manifestation were older age at disease onset (odds ratio (OR) 1.03, 95% confidence interval (CI) 1.02-1.04), higher SLEDAI (Systemic Lupus Erythematosus Disease Activity Index) scores (OR 1.03, 95% CI 1.00-1.07), the presence of Raynaud's phenomenon (OR 1.41, 95% CI 1.09-1.84), secondary antiphospholipid syndrome (OR 2.20, 95% CI 1.63-2.97), and the previous or concomitant occurrence of severe lupus nephritis, (OR 1.48, 95% CI 1.12-1.95) neuropsychiatric manifestations (OR 1.49, 95% CI 1.11-2.02), non-ischemic cardiac disease (OR 2.91, 95% CI 1.90-4.15), vasculitis (OR 1.81, 95% CI 1.25-2.62), hematological manifestations (OR 1.31, 95% CI 1.00-1.71), and gastrointestinal manifestations, excluding hepatitis (OR 2.05, 95% CI 1.14-3.66). Anti-RNP positivity had a clear tendency to significance (OR 1.32, 95% CI 1.00-1.75; P = 0.054). The development of pleuropulmonary manifestations independently contributes to a diminished survival (hazard ratio of 3.13). However, not all complications will influence the prognosis in the same way. Whereas the occurrence of pleural disease or pulmonary thromboembolism has a minimal impact on the survival of these patients, the remaining manifestations have a major impact on mortality.
Except for pleural disease, the remaining respiratory manifestations are very uncommon in SLE (<4%). Pleuropulmonary manifestations independently contributed to a decreased survival in these patients.
The clinical manifestation of median nerve entrapment at the carpal tunnel level is known as carpal tunnel syndrome (CTS). Electroneurography (ENG) is considered the gold standard in CTS evaluation. ...We conducted a retrospective study and analyzed some clinical and demographic variables, relating them to the degree of neuropathy using ENG, to better understand the role of ENG in this very common disease. We studied 816 patients referred to our service for neurographic evaluation. Their symptoms were classified as compatible with CTS (cCTS) (
= 646) and atypical for CTS (aCTS) (
= 170). A blind ENG was performed on 797 patients. Patient characteristics were coded as variables and analyzed to study whether they could predict neuropathy severity (sensory and motor involvement or grade ≥ 3 in our classification). We found a correlation between typical symptomatology, age over 50 years, male gender, positivity of Phalen's maneuver and Tinel's sign, and a neuropathy grade ≥ 3. We also found a correlation with CTS in the contralateral hand if the other hand showed neuropathy, despite the lack of symptoms in this hand. We propose a practical algorithm for ENG referral based on clinical symptoms, demographic factors, and neurophysiological variables.
An experiment was conducted to evaluate the effect of a polyherbal mixture based on Withania somnifera, Ocimum tenuiflorum, Tinospora cordifolia and Emblica officinalis, on lamb's growth and blood ...metabolites. Thirty-two lambs (initial body weight BW 25 ± 4.1 kg) were randomly assigned to the treatments which consisted of the polyherbal ImmuPlus at dietary concentrations of 0, 0.1, 0.2 and 0.3% of dry matter (DM) for 60 days. There was a quadratic response to polyherbal in average daily gain (p < 0.01) and feed conversion (p < 0.05). The optimal concentration estimated by regression for maximum ADG was 0.14 ± 0.08% DM of ImmuPlus. Ruminal DM digestibility was not affected but pH was reduced linearly (p < 0.05). Ruminal N-NH3 (p < 0.01) and total VFA concentration showed a quadratic increment (p < 0.05) whereas butyrate proportion was increased linearly (p < 0.05). There was no effect in most blood metabolites related to energy metabolism, but uric acid was linearly reduced (p < 0.01) and bilirubin showed a quadratic decrement (p < 0.05). Mean corpuscular haemoglobin (p < 0.01) increased linearly (p < 0.10), lymphocytes and segmented neutrophils showed a quadratic response (p < 0.01) and platelets showed a linear reduction (linear p < 0.05). The inclusion of ImmuPlus improved the daily gain in finishing lambs and feed efficiency at 0.1% DM without altering blood chemistry and blood biometry.
