The current concept of radiobiology posits that damage to the DNA in the cell nucleus is the primary cause for the detrimental effects of radiation. However, emerging experimental evidence suggests ...that this theoretical framework is insufficient for describing extranuclear radiation effects, particularly the response of the mitochondria, an important site of extranuclear, coding DNA. Here, we discuss experimental observations of the effects of ionizing radiation on the mitochondria at (1) the DNA and (2) functional levels. The roles of mitochondria in (3) oxidative stress and (4) late radiation effects are discussed. In this review, we summarize the current understanding of targets for ionizing radiation outside the cell nucleus. Available experimental data suggest that an increase in the tumoricidal efficacy of radiation therapy might be achievable by targeting mitochondria. Likewise, more specific protection of mitochondria and its coding DNA should reduce damage to healthy cells exposed to ionizing radiation.
The 18 kDa translocator protein (TSPO), previously known as the peripheral benzodiazepine receptor, is expressed in the injured brain. It has become known as an imaging marker of “neuroinflammation” ...indicating active disease, and is best interpreted as a nondiagnostic biomarker and disease staging tool that refers to histopathology rather than disease etiology. The therapeutic potential of TSPO as a drug target is mostly based on the understanding that it is an outer mitochondrial membrane protein required for the translocation of cholesterol, which thus regulates the rate of steroid synthesis. This pivotal role together with the evolutionary conservation of TSPO has underpinned the belief that any loss or mutation of TSPO should be associated with significant physiological deficits or be outright incompatible with life. However, against prediction, full Tspo knockout mice are viable and across their lifespan do not show the phenotype expected if cholesterol transport and steroid synthesis were significantly impaired. Thus, the “translocation” function of TSPO remains to be better substantiated. Here, we discuss the literature before and after the introduction of the new nomenclature for TSPO and review some of the newer findings. In light of the controversy surrounding the function of TSPO, we emphasize the continued importance of identifying compounds with confirmed selectivity and suggest that TSPO expression is analyzed within specific disease contexts rather than merely equated with the reified concept of “neuroinflammation.”
Background: The debate on using concrete versus abstract materials in learning mathematics has been longstanding. For decades, research has focused on the physical characteristics of materials when ...defining them as concrete or abstract. Aims: This study extends the field by proposing a two-dimensional classification, which defines materials as concrete or abstract based on the two dimensions of representation, namely object (i.e., appearance) and language (i.e., label). Sample: A total of 120 university students participated in the study. Methods: Participants were randomly assigned to learn the concept of modular arithmetic with one of four types of learning materials: concrete object labelled with concrete language, concrete object labelled with abstract language, abstract object labelled with concrete language and abstract object labelled with abstract language. They were also divided into high and low maths anxiety groups. Results: Results showed that the students who learnt with abstract objects, regardless of the level of maths anxiety, outperformed their peers who learnt with concrete objects. However, for students with low maths anxiety only, those who learnt with materials labelled with abstract language showed better far-transfer performance compared with those who learnt with materials labelled with concrete language. Conclusions: The findings offer a new direction in the conceptualization of concrete and abstract learning materials by specifying the dimensions of representation.
The evolutionarily conserved peripheral benzodiazepine receptor (PBR), or 18-kDa translocator protein (TSPO), is thought to be essential for cholesterol transport and steroidogenesis, and thus life. ...TSPO has been proposed as a biomarker of neuroinflammation and a new drug target in neurological diseases ranging from Alzheimer's disease to anxiety. Here we show that global C57BL/6-Tspo(tm1GuWu(GuwiyangWurra))-knockout mice are viable with normal growth, lifespan, cholesterol transport, blood pregnenolone concentration, protoporphyrin IX metabolism, fertility and behaviour. However, while the activation of microglia after neuronal injury appears to be unimpaired, microglia from (GuwiyangWurra)TSPO knockouts produce significantly less ATP, suggesting reduced metabolic activity. Using the isoquinoline PK11195, the ligand originally used for the pharmacological and structural characterization of the PBR/TSPO, and the imidazopyridines CLINDE and PBR111, we demonstrate the utility of (GuwiyangWurra)TSPO knockouts to provide robust data on drug specificity and selectivity, both in vitro and in vivo, as well as the mechanism of action of putative TSPO-targeting drugs.
This study investigated the relationships of four executive functioning skills (including verbal working memory, spatial working memory, inhibitory control, and cognitive flexibility) with young ...children’s mental computation and applied mathematical problem‐solving. Two hundred and twenty‐five Chinese kindergarteners were tested with a battery of general cognitive, executive functioning and mathematics skills. Results showed that when children’s age, gender, non‐verbal intelligence, and listening comprehension skills were controlled, verbal working memory and cognitive flexibility were significant correlates of mental computation, whereas verbal working memory, spatial working memory, and cognitive flexibility were significant correlates of applied mathematical problem‐solving. Inhibitory control was not significantly associated with the two domains of mathematics under investigation. The findings highlight the differential roles of different executive functioning skills in early mathematical skills and offer practical implication for helping young children in learning complex mathematical skills.
•Hong Kong reported more than 1000 COVID-19 cases between January and May 2020.•The COVID-19 epidemic in Hong Kong was characterized by several waves of imported cases followed by limited local ...transmissions.•A series of public health intervention were implemented in Hong Kong in response to the local and overseas disease situations.•The multi-pronged strategy of border control, social distancing, enhanced surveillance, and contact tracing resulted in the successful prevention of a community-wide outbreak and containment of the epidemic in Hong Kong.
Hong Kong, a Special Administrative Region of China, recorded its first confirmed coronavirus disease 2019 (COVID-19) case on 23 January 2020. We reviewed the case epidemiology and the various public health measures implemented from January to May 2020.
The epidemiological and clinical characteristics of the cases recorded in different phases of the epidemic were described and compared, and the effectiveness of the public health measures implemented were reviewed using the changes in the daily number of confirmed cases and the interval from symptom onset to hospital admission.
Between January and May 2020, 1084 confirmed COVID-19 cases were reported, about 70% of which had a history of travel during the incubation period. The case fatality ratio was 0.4%. The local epidemic progressed through four phases: (1) preparedness and imported infection from mainland China, (2) local transmission, (3) imported infection from overseas countries associated with local transmission, and (4) controlled imported infection with limited local transmission, with an eventual reduction of the daily case number and minimization of the onset-to-admission interval. Various public health measures, including enhanced surveillance, border control, and social distancing, were introduced in phases in response to the prevailing local and global situations.
The overall containment strategy in Hong Kong led to a stabilization of the number of cases and the absence of a community-wide outbreak during the 4.5 m after the first case was reported. This strategy of containment might serve as an example for future planning of preparedness and response against novel infectious agents.
We can make sense of how many a number represents by mapping the symbolic number word system onto the non-symbolic, approximate number system. This study adopted an intervention design to examine ...whether counting is essential in driving the formation of such symbolic-non-symbolic mappings. We compared kindergarteners' mapping ability after reading stories (1) without cardinal labels, or (2) with cardinal labels, or (3) with cardinal labels and verbal counting. Results showed that children who had counted when reading the stories showed better mapping between number words and their approximate representations than their peers in the other two conditions - suggesting that counting plays a role in developing early symbolic-non-symbolic mappings. Such findings provides empirical support for parents and early educators to enhance young children's mappings between numbers words and approximate representations through counting activities - such as counting the items in the story books while reading to children.
This study characterized the in-pipe thermal signature of external pipewall thinning in steel pipes, a common problem that is caused by external corrosion in hostile underground environment. A model ...system was prepared to imitate the underground environment by milling several holes of various sizes and residual thicknesses into a mild steel plate. Wall thinning was investigated using active infrared thermography. The non-defective side of the steel plate was heated to 27.4 °C through the application of a thermal energy pulse while the ambient temperature was 22°C. Thermograms were captured inside the pipe at a frequency of 0.02 seconds for 5 min. The images of the thinned surface were processed in two steps. First, the peak contrast time algorithm was used to estimate the residual thickness. Second, Gaussian adaptive thresholding was used to estimate the size of the holes. The maximum observable defects had a diameter of 5 mm and a residual thickness of 3 mm. The type of defect interface (steel–sand or steel–air) had no significant effect on the estimation of residual thickness or size. This study developed a rapid approach in classifying defect's residual thickness by only utilizing two well-known parameters from infrared images – defect's peak thermal contrast and estimated area. Thus, the feasibility of non-destructive, in-pipe, quantitative IR thermographic analysis of buried metal pipelines is demonstrated.
•Peak thermal contrast and measured area are adequate to classify defect severity.•Adaptive thresholding underestimates defect area at most 25%.•Type of interfaces had no significant effect on estimating residual thickness.•Type of interfaces had no significant effect on measuring defect size.
IMU-based human joint motion acquisition system is attractive for real-time control and monitoring in the emerging wearable technology due to its portability. However, in practical applications, it ...heavily suffers from long-term drift, magnetic interference and inconsistency of rotational reference frames, which causes precision degradation. In this letter, a novel on-line IMU-based human gait estimation framework was proposed to obtain the joint rotational angles directly under the kinematic constraints between multiple body segments, whereas traditional methods need to estimate the orientation of each individual segment. This framework consists of an on-line algorithm to align IMU frames with human joints and motion estimation algorithms for hip and knee without the aid of magnetometer. Both a 2-DoF robot and human gait tests were performed to validate the proposed method as compared with the predictions from commercial IMUs, joint encoders and an optical tracking system. The outcome demonstrated its advantages of adaptive alignment, drift rejection and low computational cost, which alleviates the practical barriers faced by human motion data collection in the wearable devices.
