Background and Aims
Virus detection is essential in programs in which virus‐tested plants are required. The aim of this study was to establish in vitro biological methods for indexing grapevine ...leafroll‐associated virus‐3 (GLRaV‐3) in red‐ and white‐berried grapevines.
Methods and Results
Healthy, red‐berried grapevine Cabernet Sauvignon CaSa (H) was used as a rootstock in micrografting experiments with virus‐infected Cabernet Sauvignon CaSa (VI) and Chardonnay Ch (VI) scions. Vascular bundles connecting the rootstock and scions developed, and 100% of the micrografts survived 3 and 4 weeks after micrografting, respectively. Symptoms of GLRaV‐3 were expressed in 80 and 20% of CaSa (H) rootstocks micrografted with CaSa (VI) and with Ch (VI) scions, respectively, 12 weeks after micrografting. The rootstock CaSa (H) that had been micrografted with CaSa (VI) and Ch (VI) scions for 5 weeks was subsequently cultured on half‐strength Murashige and Skoog medium supplemented with 75 mmol/L NaCl to induce salt stress. The GLRaV‐3 symptoms were expressed in 88% of micrografts CaSa (VI)/CaSa (H) and 85% of micrografts Ch (VI)/CaSa (H) after 5 weeks of salt stress. Reddish‐purple leaf coloration and downward rolling were observed in CaSa (H) rootstocks that were micrografted with CaSa (VI) scions, while leaf downward rolling and yellowing were seen in CaSa (H) rootstocks that were micrografted with Ch (VI) scions.
Conclusions
Salt stress improved in vitro biological indexing of GLRaV‐3 in grapevines.
Significance of the Study
Establishment of in vitro micrografting and salt stress provided alternative methods for indexing of GLRaV‐3 in red‐ and white‐berried grapevines.
The initial reinforcing properties of drugs of abuse, such as cocaine, are largely attributed to their ability to activate the mesolimbic dopamine system. Resulting increases in extracellular ...dopamine in the nucleus accumbens (NAc) are traditionally thought to result from cocaine's ability to block dopamine transporters (DATs). Here we demonstrate that cocaine also interacts with the immunosurveillance receptor complex, Toll-like receptor 4 (TLR4), on microglial cells to initiate central innate immune signaling. Disruption of cocaine signaling at TLR4 suppresses cocaine-induced extracellular dopamine in the NAc, as well as cocaine conditioned place preference and cocaine self-administration. These results provide a novel understanding of the neurobiological mechanisms underlying cocaine reward/reinforcement that includes a critical role for central immune signaling, and offer a new target for medication development for cocaine abuse treatment.

During the current COVID-19 pandemic lay rescuers should consider compressions and public-access defibrillation. Lay rescuers who are willing, trained and able to do so, should consider providing rescue breaths to infants and children in addition to chest compressions. Healthcare professionals should use personal protective equipment for aerosol generating procedures during resuscitation and may consider defibrillation before donning personal protective equipment for aerosol generating procedures.
Aims
This study examined the biotransformation pathway of ginsenoside Rb1 by the fungus Esteya vermicola CNU 120806.
Methods and Results
Ginsenosides Rb1 and Rd were extracted from the root of Panax ...ginseng. Liquid fermentation and purified enzyme hydrolysis were employed to investigate the biotransformation of ginsenoside Rb1. The metabolites were identified and confirmed using NMR analysis as gypenoside XVII and gypenoside LXXV. A mole yield of 95·4% gypenoside LXXV was obtained by enzymatic conversion (pH 5·0, temperature 50°C). Ginsenoside Rd was used to verify the transformation pathway under the same reaction condition. The product Compound K (mole yield 49·6%) proved a consecutive hydrolyses occurred at the C‐3 position of ginsenoside Rb1.
Conclusions
Strain CNU 120806 showed a high degree of specific β‐glucosidase activity to convert ginsenosides Rb1 and Rd to gypenoside LXXV and Compound K, respectively. The maximal activity of the purified glucosidase for ginsenosides transformation occurred at 50°C and pH 5·0. Compared with its activity against pNPG (100%), the β‐glucosidase exhibited quite lower level of activity against other aryl‐glycosides. Enzymatic hydrolysate, gypenoside LXXV and Compound K were produced by consecutive hydrolyses of the terminal and inner glucopyranosyl moieties at the C‐3 carbon of ginsenoside Rb1 and Rd, giving the pathway: ginsenoside Rb1→ gypenoside XVII → gypenoside LXXV; ginsenoside Rd→F2→Compound K, but did not hydrolyse the 20‐C, β‐(1‐6)‐glucoside of ginsenoside Rb1 and Rd.
Significance and Impact of the Study
The results showed an important practical application on the preparation of gypenoside LXXV. Additionally, this study for the first time provided a high efficient preparation method for gypenoside LXXV without further conversion, which also gives rise to a potential commercial enzyme application.
This paper is the first in a series in which we perform an extensive comparison of various galaxy-based cluster mass estimation techniques that utilize the positions, velocities and colours of ...galaxies. Our primary aim is to test the performance of these cluster mass estimation techniques on a diverse set of models that will increase in complexity. We begin by providing participating methods with data from a simple model that delivers idealized clusters, enabling us to quantify the underlying scatter intrinsic to these mass estimation techniques. The mock catalogue is based on a Halo Occupation Distribution (HOD) model that assumes spherical Navarro, Frenk and White (NFW) haloes truncated at R
200, with no substructure nor colour segregation, and with isotropic, isothermal Maxwellian velocities. We find that, above 1014M⊙, recovered cluster masses are correlated with the true underlying cluster mass with an intrinsic scatter of typically a factor of 2. Below 1014M⊙, the scatter rises as the number of member galaxies drops and rapidly approaches an order of magnitude. We find that richness-based methods deliver the lowest scatter, but it is not clear whether such accuracy may simply be the result of using an over-simplistic model to populate the galaxies in their haloes. Even when given the true cluster membership, large scatter is observed for the majority non-richness-based approaches, suggesting that mass reconstruction with a low number of dynamical tracers is inherently problematic.
We theoretically consider the superconductivity of the topological half-Heusler semimetals YPtBi and LuPtBi. We show that pairing occurs between j=3/2 fermion states, which leads to qualitative ...differences from the conventional theory of pairing between j=1/2 states. In particular, this permits Cooper pairs with quintet or septet total angular momentum, in addition to the usual singlet and triplet states. Purely on-site interactions can generate s-wave quintet time-reversal symmetry-breaking states with topologically nontrivial point or line nodes. These local s-wave quintet pairs reveal themselves as d-wave states in momentum space. Furthermore, due to the broken inversion symmetry in these materials, the s-wave singlet state can mix with a p-wave septet state, again with topologically stable line nodes. Our analysis lays the foundation for understanding the unconventional superconductivity of the half-Heuslers.
Electrocyclic reactions are characterized by the concerted formation and cleavage of both σ and π bonds through a cyclic structure. This structure is known as a pericyclic transition state for ...thermal reactions and a pericyclic minimum in the excited state for photochemical reactions. However, the structure of the pericyclic geometry has yet to be observed experimentally. We use a combination of ultrafast electron diffraction and excited state wavepacket simulations to image structural dynamics through the pericyclic minimum of a photochemical electrocyclic ring-opening reaction in the molecule α-terpinene. The structural motion into the pericyclic minimum is dominated by rehybridization of two carbon atoms, which is required for the transformation from two to three conjugated π bonds. The σ bond dissociation largely happens after internal conversion from the pericyclic minimum to the electronic ground state. These findings may be transferrable to electrocyclic reactions in general.
Ambient fine particulate matter (PM2.5) is a leading environmental risk factor for premature mortality. We use aerosol optical depth (AOD) retrieved from two satellite instruments, MISR and SeaWiFS, ...to produce a unified 15-year global time series (1998–2012) of ground-level PM2.5 concentration at a resolution of 1° x 1°. The GEOS-Chem chemical transport model (CTM) is used to relate each individual AOD retrieval to ground-level PM2.5. Four broad areas showing significant, spatially coherent, annual trends are examined in detail: the Eastern U.S. (−0.39 ± 0.10 μg m–3 yr–1), the Arabian Peninsula (0.81 ± 0.21 μg m–3 yr–1), South Asia (0.93 ± 0.22 μg m–3 yr–1) and East Asia (0.79 ± 0.27 μg m–3 yr–1). Over the period of dense in situ observation (1999–2012), the linear tendency for the Eastern U.S. (−0.37 ± 0.13 μg m–3 yr–1) agrees well with that from in situ measurements (−0.38 ± 0.06 μg m–3 yr–1). A GEOS-Chem simulation reveals that secondary inorganic aerosols largely explain the observed PM2.5 trend over the Eastern U.S., South Asia, and East Asia, while mineral dust largely explains the observed trend over the Arabian Peninsula.
Brain development has largely been studied through unimodal analysis of neuroimaging data, providing independent results for structural and functional data. However, structure clearly impacts ...function and vice versa, pointing to the need for performing multimodal data collection and analysis to improve our understanding of brain development, and to further inform models of typical and atypical brain development across the lifespan. Ultimately, such models should also incorporate genetic and epigenetic mechanisms underlying brain structure and function, although currently this area is poorly specified. To this end, we are reporting here a multi-site, multi-modal dataset that captures cognitive function, brain structure and function, and genetic and epigenetic measures to better quantify the factors that influence brain development in children originally aged 9–14 years. Data collection for the Developmental Chronnecto-Genomics (Dev-CoG) study (http://devcog.mrn.org/) includes cognitive, emotional, and social performance scales, structural and functional MRI, diffusion MRI, magnetoencephalography (MEG), and saliva collection for DNA analysis of single nucleotide polymorphisms (SNPs) and DNA methylation patterns. Across two sites (The Mind Research Network and the University of Nebraska Medical Center), data from over 200 participants were collected and these children were re-tested annually for at least 3 years. The data collection protocol, sample demographics, and data quality measures for the dataset are presented here. The sample will be made freely available through the collaborative informatics and neuroimaging suite (COINS) database at the conclusion of the study.
Physical activity has a protective effect against cardiovascular disease (CVD) in high-income countries, where physical activity is mainly recreational, but it is not known if this is also observed ...in lower-income countries, where physical activity is mainly non-recreational. We examined whether different amounts and types of physical activity are associated with lower mortality and CVD in countries at different economic levels.
In this prospective cohort study, we recruited participants from 17 countries (Canada, Sweden, United Arab Emirates, Argentina, Brazil, Chile, Poland, Turkey, Malaysia, South Africa, China, Colombia, Iran, Bangladesh, India, Pakistan, and Zimbabwe). Within each country, urban and rural areas in and around selected cities and towns were identified to reflect the geographical diversity. Within these communities, we invited individuals aged between 35 and 70 years who intended to live at their current address for at least another 4 years. Total physical activity was assessed using the International Physical Activity Questionnaire (IPQA). Participants with pre-existing CVD were excluded from the analyses. Mortality and CVD were recorded during a mean of 6·9 years of follow-up. Primary clinical outcomes during follow-up were mortality plus major CVD (CVD mortality, incident myocardial infarction, stroke, or heart failure), either as a composite or separately. The effects of physical activity on mortality and CVD were adjusted for sociodemographic factors and other risk factors taking into account household, community, and country clustering.
Between Jan 1, 2003, and Dec 31, 2010, 168 916 participants were enrolled, of whom 141 945 completed the IPAQ. Analyses were limited to the 130 843 participants without pre-existing CVD. Compared with low physical activity (<600 metabolic equivalents MET × minutes per week or <150 minutes per week of moderate intensity physical activity), moderate (600–3000 MET × minutes or 150–750 minutes per week) and high physical activity (>3000 MET × minutes or >750 minutes per week) were associated with graded reduction in mortality (hazard ratio 0·80, 95% CI 0·74–0·87 and 0·65, 0·60–0·71; p<0·0001 for trend), and major CVD (0·86, 0·78–0·93; p<0·001 for trend). Higher physical activity was associated with lower risk of CVD and mortality in high-income, middle-income, and low-income countries. The adjusted population attributable fraction for not meeting the physical activity guidelines was 8·0% for mortality and 4·6% for major CVD, and for not meeting high physical activity was 13·0% for mortality and 9·5% for major CVD. Both recreational and non-recreational physical activity were associated with benefits.
Higher recreational and non-recreational physical activity was associated with a lower risk of mortality and CVD events in individuals from low-income, middle-income, and high-income countries. Increasing physical activity is a simple, widely applicable, low cost global strategy that could reduce deaths and CVD in middle age.
