Rapid adjustments are responses to forcing agents that cause a perturbation to the top of atmosphere energy budget but are uncoupled to changes in surface warming. Different mechanisms are ...responsible for these adjustments for a variety of climate drivers. These remain to be quantified in detail. It is shown that rapid adjustments reduce the effective radiative forcing (ERF) of black carbon by half of the instantaneous forcing, but for CO2 forcing, rapid adjustments increase ERF. Competing tropospheric adjustments for CO2 forcing are individually significant but sum to zero, such that the ERF equals the stratospherically adjusted radiative forcing, but this is not true for other forcing agents. Additional experiments of increase in the solar constant and increase in CH4 are used to show that a key factor of the rapid adjustment for an individual climate driver is changes in temperature in the upper troposphere and lower stratosphere.

Long‐term global warming can be estimated with knowledge of how climate forcing agents affect the Earth's top‐of‐atmosphere energy imbalance or effective radiative forcing. Changes in climate forcers, such as greenhouse gases, the Sun's intensity, or emission of aerosol particles, typically impose a direct change in the energy budget, termed an instantaneous radiative forcing. Further to this, a climate forcer may induce changes in the atmosphere, such as a change in thermal structure, clouds, or humidity. These changes themselves, termed rapid adjustments, contribute to the top‐of‐atmosphere energy budget. Together, the instantaneous radiative forcing plus rapid adjustments equals the effective radiative forcing. We show that for different climate forcing agents, the rapid adjustments behave very differently and are driven by different atmospheric mechanisms. For example, rapid adjustments add to the instantaneous forcing for a carbon dioxide increase, due to a cooling of the stratosphere, but oppose instantaneous forcing for black carbon, driven by a warming troposphere and lowering of cloud height. Understanding rapid adjustments gives a more complete picture of the climate effects of different climate forcers.

After psychological trauma, recurrent intrusive visual memories may be distressing and disruptive. Preventive interventions post trauma are lacking. Here we test a behavioural intervention after ...real-life trauma derived from cognitive neuroscience. We hypothesized that intrusive memories would be significantly reduced in number by an intervention involving a computer game with high visuospatial demands (Tetris), via disrupting consolidation of sensory elements of trauma memory. The Tetris-based intervention (trauma memory reminder cue plus c. 20 min game play) vs attention-placebo control (written activity log for same duration) were both delivered in an emergency department within 6 h of a motor vehicle accident. The randomized controlled trial compared the impact on the number of intrusive trauma memories in the subsequent week (primary outcome). Results vindicated the efficacy of the Tetris-based intervention compared with the control condition: there were fewer intrusive memories overall, and time-series analyses showed that intrusion incidence declined more quickly. There were convergent findings on a measure of clinical post-trauma intrusion symptoms at 1 week, but not on other symptom clusters or at 1 month. Results of this proof-of-concept study suggest that a larger trial, powered to detect differences at 1 month, is warranted. Participants found the intervention easy, helpful and minimally distressing. By translating emerging neuroscientific insights and experimental research into the real world, we offer a promising new low-intensity psychiatric intervention that could prevent debilitating intrusive memories following trauma.
LUX-ZEPLIN (LZ) is a next-generation dark matter direct detection experiment that will operate 4850 feet underground at the Sanford Underground Research Facility (SURF) in Lead, South Dakota, USA. ...Using a two-phase xenon detector with an active mass of 7 tonnes, LZ will search primarily for low-energy interactions with weakly interacting massive particles (WIMPs), which are hypothesized to make up the dark matter in our galactic halo. In this paper, the projected WIMP sensitivity of LZ is presented based on the latest background estimates and simulations of the detector. For a 1000 live day run using a 5.6-tonne fiducial mass, LZ is projected to exclude at 90% confidence level spin-independent WIMP-nucleon cross sections above 1.4 × 10−48 cm2 for a 40 GeV/c2 mass WIMP. Additionally, a 5 σ discovery potential is projected, reaching cross sections below the exclusion limits of recent experiments. For spin-dependent WIMP-neutron(-proton) scattering, a sensitivity of 2.3 × 10−43 cm2 (7.1 × 10−42 cm2) for a 40 GeV/c2 mass WIMP is expected. With underground installation well underway, LZ is on track for commissioning at SURF in 2020.
Independent studies have previously demonstrated that both the HIPK2 and BRAF genes are amplified and rearranged, respectively, in pilocytic astrocytomas (PAs). The purpose of this study was to ...further investigate the frequency of BRAF and HIPK2 alterations in PAs, the concordance of these events, and their relationship to clinical phenotype.
We performed extensive characterization by array-based copy number assessment (aCGH), HIPK2 copy number analysis, and BRAF rearrangement and mutation analysis in a set of 79 PAs, including 9 tumors from patients with neurofibromatosis type 1 (NF1).
We identified 1 of 3 previously identified BRAF rearrangements in 42/70 sporadic PAs. An additional 2 tumors with no rearrangement also exhibited BRAF mutation, including a novel 3-base insertion. As predicted from the genomic organization at this locus, 22/36 tumors with BRAF rearrangement also exhibited corresponding HIPK2 amplification. However, 14/36 tumors with BRAF rearrangement had no detectable HIPK2 gene amplification and 6/20 tumors demonstrated HIPK2 amplification without apparent BRAF rearrangement or mutation. Only 12/70 PAs lacked detectable BRAF or HIPK2 alterations. Importantly, none of the 9 PA tumors from NF1 patients exhibited BRAF rearrangement or mutation.
BRAF rearrangement represents the most common genetic alteration in sporadic, but not neurofibromatosis type 1-associated, pilocytic astrocytomas (PAs). These findings implicate BRAF in the pathogenesis of these common low-grade astrocytomas in children, and suggest that PAs arise either from NF1 inactivation or BRAF gain of function.
One third of deceased donor kidneys for transplantation in the UK are donated following cardiac death (DCD). Such kidneys have a high rate of delayed graft function (DGF) following transplantation. ...We conducted a multicenter, randomized controlled trial to determine whether kidney preservation using cold, pulsatile machine perfusion (MP) was superior to simple cold storage (CS) for DCD kidneys. One kidney from each DCD donor was randomly allocated to CS, the other to MP. A sequential trial design was used with the primary endpoint being DGF, defined as the necessity for dialysis within the first 7 days following transplant. The trial was stopped when data were available for 45 pairs of kidneys. There was no difference in the incidence of DGF between kidneys assigned to MP or CS (58% vs. 56%, respectively), in the context of an asystolic period of 15 min and median cold ischemic times of 13.9 h for MP and 14.3 h for CS kidneys. Renal function at 3 and 12 months was similar between groups, as was graft and patient survival. For kidneys from controlled DCD donors (with mean cold ischemic times around 14 h), MP offers no advantage over CS, which is cheaper and more straightforward.

A revised set of propositions about ecology in arid Australia is presented, based on research literature since publication of Stafford Smith and Morton (1990). Fourteen propositions distil our ...argument that most features of the Australian deserts are explicable in terms of two dominant physical and climatic elements: rainfall variability, leading to extended droughts and occasional flooding rains; and widespread nutrient poverty. Different landscapes within the arid zone show these features to varying degrees, and so it is important to think about different places separately when considering our propositions. Plant life-histories strongly reflect temporal patterns of soil moisture; because Australian deserts receive more variable rainfall than most others, there is a distinctive spectrum of life-histories. Low levels of phosphorus (together with abundant soil moisture on irregular occasions) favour plants producing a relative excess of carbohydrate (C). In turn, C-rich plant products sometimes lead to fire-prone ecosystems, assemblages dominated by consumers of sap and other C-based products, and abundant detritivores (particularly termites). Fluctuations in production due to variable rainfall provide openings for consumers with opportunistic life-histories, including inhabitants of extensive but ephemeral rivers and lakes. Most consumer species exhibit some dietary flexibility or utilise more dependable resources; these strategies give rise to greater stability in species dynamics and composition of assemblages than might first be imagined under the variable rainfall regime. Aboriginal people have had long-standing ecological influence as they accessed resources. For each proposition we suggest the extent to which it is ‘different’, ‘accentuated’ or ‘universal’ in comparison with other deserts of the world, recognising that this categorisation is in need of critical testing. Further tests of each proposition are also suggested to fill the many gaps that still exist in our knowledge of the structure and functioning of Australia’s deserts.

A novel conceptual model linking anoxia, phosphorus (P), nitrogen (N), iron (Fe) and sulphate to the formation of noxious filamentous and colonial cyanobacteria blooms is presented that ...reconciles seemingly contradictory ideas about the roles of P, N and Fe in bloom formation.
The model has several critical concepts: (i) P regulates biomass and productivity in fresh waters until excessive loading renders a system N‐limited or light‐limited, but it is the availability of ferrous ions (Fe2+) that regulates the ability of cyanobacteria to compete with its eukaryotic competitors; (ii) Fe2+ diffusing from anoxic sediments is a major Fe source for cyanobacteria, which acquire it by migrating downwards into Fe2+‐rich anoxic waters from oxygenated waters; and (iii) subsequent cyanobacterial siderophore production provides a supply of Fe3+ for reduction at cyanobacteria cell membranes that leads to very low Fe3+ concentrations in the mixing zone.
When light and temperature are physiologically suitable for cyanobacteria growth, bloom onset is regulated by the onset of internal Fe2+ loading which in turn is controlled by anoxia, reducible Fe content of surface sediments and sulphate reduction rate.
This conceptual model provides the basis for improving the success of approaches to eutrophication management because of its far‐reaching explanatory power over the wide range of conditions where noxious cyanobacteria blooms have been observed.
Carbonaceous aerosol is an important component that influences the environment, climate, and human health. Organic and elemental carbon (OC and EC) are the two main constituents of carbonaceous ...aerosols that have opposite, i.e., cooling versus warming, effects on the Earth's radiation balance. Knowledge on the variability of OC/EC splits measured by different thermal/optical protocols is useful for understanding the uncertainty in the climate models. This study shows good correlations within OC or EC (r2 > 0.83, P < 0.001) across the IMPROVE, IMPROVE_A, and EUSAAR_2 protocols for both ambient aerosol samples and biomass burning samples. However, EC concentrations differ by more than two folds, and OC/EC ratios differ up to a factor of 2.7. The discrepancies were attributed to the selection between the reflectance and transmittance corrections and the different peak inert-atmosphere temperature. The IMPROVE and IMPROVE_A protocols also quantified different char and soot concentrations, two subtypes of EC with distinct chemical and optical properties. Char, but not soot, was found to correlate with the humic-like substances (HULIS) content in the samples, suggesting that both char and HULIS originate mainly from biomass burning. A one-year (2012–2013) ambient aerosol monitoring in Xi'an, China, shows that OC, EC, and char displayed winter highs and summer lows, while soot had no seasonal trend. The char/soot ratios showed a “single peak” in winter, while OC/EC ratios exhibited “dual peak” feature due to the influence of secondary organic aerosol formation. In addition to commonly measured OC and EC, we recommend both char and soot from a common reference method to be considered in the chemical transport and climate models.

Primary progressive aphasia (PPA) has been proposed to comprise 3 discrete clinical subtypes: semantic, agrammatic/nonfluent, and logopenic. Recent consensus recommendations suggest a diagnostic ...framework based primarily on clinical and neuropsychological findings to classify these variants. Our objective was to evaluate the extent to which patients with PPA would conform to the proposed tripartite system and whether the clustering pattern of elements of the linguistic profile suggests discrete clinical syndromes.
A total of 46 patients with PPA were prospectively recruited to the Cambridge Longitudinal Study of PPA. Sufficient data were collected to assess all consensus-proposed diagnostic domains. By comparing patients' performances against those of 30 age- and education-matched healthy volunteers, z scores were calculated, and values of 1.5 SDs outside control participants' means were considered abnormal. Raw test scores were used to undertake a principal factor analysis to identify the clustering pattern of individual measures.
Of the patients, 28.3%, 26.1%, and 4.3% fitted semantic, nonfluent/agrammatic, and logopenic categories respectively, and 41.3% did not fulfill the diagnostic recommendations for any of the 3 proposed variants. There was no significant between-group difference in age, education, or disease duration. Furthermore, the outcome of the factor analysis was in keeping with discrete semantic and nonfluent/agrammatic syndromes but did not support a logopenic variant.
Taken together, the results of this prospective data-driven study suggest that although a substantial proportion of patients with PPA have neither the semantic nor the nonfluent variants, they do not necessarily conform to a discrete logopenic variant.
Gonadal steroids are potent regulators of adult neurogenesis. We previously reported that androgens, such as testosterone (T) and dihydrotestosterone (DHT), but not estradiol, increased the survival ...of new neurons in the dentate gyrus of the male rat. These results suggest androgens regulate hippocampal neurogenesis via the androgen receptor (AR). To test this supposition, we examined the role of ARs in hippocampal neurogenesis using 2 different approaches. In experiment 1, we examined neurogenesis in male rats insensitive to androgens due to a naturally occurring mutation in the gene encoding the AR (termed testicular feminization mutation) compared with wild-type males. In experiment 2, we injected the AR antagonist, flutamide, into castrated male rats and compared neurogenesis levels in the dentate gyrus of DHT and oil-treated controls. In experiment 1, chronic T increased hippocampal neurogenesis in wild-type males but not in androgen-insensitive testicular feminization mutation males. In experiment 2, DHT increased hippocampal neurogenesis via cell survival, an effect that was blocked by concurrent treatment with flutamide. DHT, however, did not affect cell proliferation. Interestingly, cells expressing doublecortin, a marker of immature neurons, did not colabel with ARs in the dentate gyrus, but ARs were robustly expressed in other regions of the hippocampus. Together these studies provide complementary evidence that androgens regulate adult neurogenesis in the hippocampus via the AR but at a site other than the dentate gyrus. Understanding where in the brain androgens act to increase the survival of new neurons in the adult brain may have implications for neurodegenerative disorders.
