We present a new, universally applicable toy model of colour reconnections in hadronic final states. The model is based on hadronising strings and has one free parameter. We next present an ...implementation of this model in the PYTHIA event generator and provide several parameter sets (‘tunes’), constrained by fits to Tevatron minimum-bias data. Finally, we consider the sensitivity of a simplified top mass analysis to these effects, in exclusive semi-leptonic top events at the Tevatron. A first attempt at isolating the genuine non-perturbative effects gives an estimate of order δmtop ∼ ±0.5 GeV from non-perturbative uncertainties, and a further δmtop ∼ ±1 GeV from shower effects.
The lateral lemniscus encompasses processing stages for binaural hearing, suppressing spurious frequencies and frequency integration. Within the lemniscal fibres three nuclei can be identified, ...termed after their location as dorsal, intermediate and ventral nucleus of the lateral lemniscus (DNLL, INLL and VNLL). While the DNLL and VNLL have been functionally and anatomically characterized, less is known about INLL neurons. Here, we quantitatively describe the morphology, the cellular orientation and distribution of synaptic contact sites along dendrites in mature Mongolian gerbils. INLL neurons are largely non-inhibitory and morphologically heterogeneous with an overall perpendicular orientation regarding the lemniscal fibers. Dendritic ranges are heterogeneous and can extend beyond the nucleus border. INLL neurons receive VGluT1/2 containing glutamatergic and a mix of GABA- and glycinergic inputs distributed over the entire dendrite. Input counts suggest that numbers of excitatory exceed the inhibitory contact sites. Axonal projections indicate connectivity to ascending and descending auditory structures. Our data show that INLL neurons form a morphologically heterogeneous continuum and incoming auditory information is processed on thin dendrites of various length and biased to perpendicular orientation. Together with the different axonal projection patterns, this indicates that the INLL is a highly complex structure that might hold many unexplored auditory functions.
A method using thin boards (3 cm thick, 0.56 m2) comprising different paving materials typically used in urban environments (2 asphalt types and concrete) was employed to specifically investigate ...air-borne deposition dynamics of TSS, zinc, copper and lead. Boards were exposed at an urban car park near vehicular traffic to determine the rate of contaminant build-up over a 13-day dry period. Concentration profiles from simulated rainfall wash-off were used to determine contaminant yields at different antecedent dry days. Maximum contaminant yields after 13 days of exposure were 2.7 kg ha−1 for TSS, 35 g ha−1 zinc, 2.3 g ha−1 copper and 0.4 g ha−1 lead. Accumulation of all contaminants increased over the first week and levelled off thereafter, supporting theoretical assumptions that contaminant accumulation on impervious surfaces asymptotically approaches a maximum. Comparison of different surface types showed approximately four times higher zinc concentrations in runoff from asphalt surfaces and two times higher TSS concentrations in runoff from concrete, which is attributed to different physical and chemical compositions of the pavement types. Contaminant build-up and wash-off behaviours were modelled using exponential and saturation functions commonly applied in the US EPA's Stormwater Management Model (SWMM) showing good correlation between measured and modelled concentrations. Maximum build-up, half-saturation time, build-up rate constants and wash-off coefficients, necessary for stormwater contaminant modelling, were determined for the four contaminants studied. These parameters are required to model contaminant concentrations in urban runoff assisting in stormwater management decisions.

Insertional activation of cellular proto-oncogenes by replication-defective retroviral vectors can trigger clonal dominance and leukemogenesis in animal models and clinical trials. Here, we addressed ...the leukemogenic potential of vectors expressing interleukin-2 receptor common gamma-chain (IL2RG), the coding sequence required for correction of X-linked severe combined immunodeficiency. Similar to conventional gamma-retroviral vectors, self-inactivating (SIN) vectors with strong internal enhancers also triggered profound clonal imbalance, yet with a characteristic insertion preference for a window located downstream of the transcriptional start site. Controls including lentivirally transduced cells revealed that ectopic IL2RG expression was not sufficient to trigger leukemia. After serial bone marrow transplantation involving 106 C57Bl6/J mice monitored for up to 18 months, we observed leukemic progression of six distinct clones harboring gamma-retroviral long terminal repeat (LTR) or SIN vector insertions in Evi1 or Prdm16, two functionally related genes. Three leukemic clones had single vector integrations, and identical clones manifested with a remarkably similar latency and phenotype in independent recipients. We conclude that upregulation of Evi1 or Prdm16 was sufficient to initiate a leukemogenic cascade with consistent intrinsic dynamics. Our study also shows that insertional mutagenesis is required for leukemia induction by IL2RG vectors, a risk to be addressed by improved vector design.
Comparative analysis of evolutionarily conserved neuronal circuits between phylogenetically distant mammals highlights the relevant mechanisms and specific adaptations to information processing. The ...medial nucleus of the trapezoid body (MNTB) is a conserved mammalian auditory brainstem nucleus relevant for temporal processing. While MNTB neurons have been extensively investigated, a comparative analysis of phylogenetically distant mammals and the spike generation is missing. To understand the suprathreshold precision and firing rate, we examined the membrane, voltage-gated ion channel and synaptic properties in
(bat) and in
(rodent) of either sex. Between the two species, the membrane properties of MNTB neurons were similar at rest with only minor differences, while larger dendrotoxin (DTX)-sensitive potassium currents were found in gerbils. Calyx of Held-mediated EPSCs were smaller and frequency dependence of short-term plasticity (STP) less pronounced in bats. Simulating synaptic train stimulations in dynamic clamp revealed that MNTB neurons fired with decreasing success rate near conductance threshold and at increasing stimulation frequency. Driven by STP-dependent conductance decrease, the latency of evoked action potentials increased during train stimulations. The spike generator showed a temporal adaptation at the beginning of train stimulations that can be explained by sodium current inactivation. Compared with gerbils, the spike generator of bats sustained higher frequency input-output functions and upheld the same temporal precision. Our data mechanistically support that MNTB input-output functions in bats are suited to sustain precise high-frequency rates, while for gerbils, temporal precision appears more relevant and an adaptation to high output-rates can be spared.
Neurons in the mammalian medial nucleus of the trapezoid body (MNTB) convey precise, faithful inhibition vital for binaural hearing and gap detection. The MNTB's structure and function appear evolutionarily well conserved. We compared the cellular physiology of MNTB neurons in bat and gerbil. Because of their adaptations to echolocation or low frequency hearing both species are model systems for hearing research, yet with largely overlapping hearing ranges. We find that bat neurons sustain information transfer with higher ongoing rates and precision based on synaptic and biophysical differences in comparison to gerbils. Thus, even in evolutionarily conserved circuits species-specific adaptations prevail, highlighting the importance for comparative research to differentiate general circuit functions and their specific adaptations.
In order to evaluate the influence of substrate composition on stormwater treatment and hydraulic effectiveness, mesocosm-scale (180 L, 0.17 m(2)) laboratory rain gardens were established. Saturated ...(constant head) hydraulic conductivity was determined before and after contaminant (Cu, Zn, Pb and nutrients) removal experiments on three rain garden systems with various proportions of organic topsoil. The system with only topsoil had the lowest saturated hydraulic conductivity (160-164 mm/h) and poorest metal removal efficiency (Cu ≤ 69.0% and Zn ≤ 71.4%). Systems with sand and a sand-topsoil mix demonstrated good metal removal (Cu up to 83.3%, Zn up to 94.5%, Pb up to 97.3%) with adequate hydraulic conductivity (sand: 800-805 mm/h, sand-topsoil: 290-302 mm/h). Total metal amounts in the effluent were <50% of influent amounts for all experiments, with the exception of Cu removal in the topsoil-only system, which was negligible due to high dissolved fraction. Metal removal was greater when effluent pH was elevated (up to 7.38) provided by the calcareous sand in two of the systems, whereas the topsoil-only system lacked an alkaline source. Organic topsoil, a typical component in rain garden systems, influenced pH, resulting in poorer treatment due to higher dissolved metal fractions.
Storm runoff is a major vector for transporting urban contaminants, especially metals, and continues to be a leading cause of urban waterways degradation. Stormwater treatment systems in New Zealand ...and Australia are primarily designed to remove total suspended solids and heavy metals to low levels, principally through bioinfiltration. In Christchurch, the second largest city in New Zealand, more than two thirds of the water, including stormwater, infrastructure is currently being rebuilt following the devastating 2010–2011 earthquakes. Despite increased use of bioinfiltration systems for this purpose, there is a dearth of knowledge about their treatment performance or water quality dynamics. This paper reports enhanced treatment efficacy in bioinfiltration stormwater systems by including an alkaline waste product, mussel shells, in the substrates. Experimental systems with mussel shells significantly increased the metal removal efficacy, hardness, and pH, which also have implications for reducing the potential ecotoxicological effects of stormwater. Mussel shell systems resulted in lower dissolved metal fractions in the treated effluent because metals shifted to the particulate states facilitated by hardness buffering. This resulted in greater metal removal afforded by increased filtration. Using locally available waste products can reduce the amount and transport impacts of waste going to landfills and offset costs associated with the construction of stormwater treatment systems, while concurrently improving stormwater treatment. The long-term capacity of such systems to enhance metal removal using waste mussel shells should be examined by monitoring larger pilot-scale systems in situ under different seasonal events.
Waterways can contribute to the beauty and livelihood of urban areas, but maintaining their hydro-ecosystem health is challenging because they are often recipients of contaminated water from ...stormwater runoff and other discharges. Public awareness of local waterways' health and community impacts to these waterways is usually poor due to of lack of easily available information. To improve community awareness of water quality in urban waterways in New Zealand, a web portal was developed featuring a real-time waterways monitoring system, a public forum, historical data, interactive maps, contaminant modelling scenarios, mitigation recommendations, and a prototype contamination alert system. The monitoring system featured in the web portal is unique in the use of wireless mesh network technology, direct integration with online modelling, and a clear target of public engagement. The modelling aims to show the origin of contaminants within the local catchment and to help the community prioritize mitigation efforts to improve water quality in local waterways. The contamination alert system aims to keep managers and community members better informed and to provide a more timely response opportunity to avert any unplanned or accidental contamination of the waterways. Preliminary feedback has been positive and is being supported by local and regional authorities. The system was developed in a cost-effective manner providing a community focussed solution for quantifying and mitigating key contaminants in urban catchments and is applicable and transferable to other cities with similar stormwater challenges.
Bats are a natural reservoir for many viruses and are considered to play an important role in the interspecies transmission of viruses. To analyze the susceptibility of bat airway cells to infection ...by viruses of other mammalian species, we developed an airway organoid culture model derived from airways of Carollia perspicillata. Application of specific antibodies for fluorescent staining indicated that the cell composition of organoids resembled those of bat trachea and lungs as determined by immunohistochemistry. Infection studies indicated that
bat airway organoids (AOs) from the trachea or the lung are highly susceptible to infection by two different porcine influenza A viruses. The bat AOs were also used to develop an air-liquid interface (ALI) culture system of filter-grown epithelial cells. Infection of these cells showed the same characteristics, including lower virulence and enhanced replication and release of the H1N1/2006 virus compared to infection with H3N2/2007. These observations agreed with the results obtained by infection of porcine ALI cultures with these two virus strains. Interestingly, lectin staining indicated that bat airway cells only contain a small amount of alpha 2,6-linked sialic acid, the preferred receptor determinant for mammalian influenza A viruses. In contrast, large amounts of alpha 2,3-linked sialic acid, the preferred receptor determinant for avian influenza viruses, are present in bat airway epithelial cells. Therefore, bat airway cells may be susceptible not only to mammalian but also to avian influenza viruses. Our culture models, which can be extended to other parts of the airways and to other species, provide a promising tool to analyze virus infectivity and the transmission of viruses both from bats to other species and from other species to bats.
We developed an organoid culture system derived from the airways of the bat species
. Using this cell system, we showed that the airway epithelium of these bats is highly susceptible to infection by influenza viruses of other mammalian species and thus is not a barrier for interspecies transmission. These organoids provide an almost unlimited supply of airway epithelial cells that can be used to generate well-differentiated epithelial cells and perform infection studies. The establishment of the organoid model required only three animals, and can be extended to other epithelia (nose, intestine) as well as to other species (bat and other animal species). Therefore, organoids promise to be a valuable tool for future zoonosis research on the interspecies transmission of viruses (e.g., bat → intermediate host → human).
