Metamonads are a diverse group of heterotrophic microbial eukaryotes adapted to living in hypoxic environments. All metamonads but one harbour metabolically altered ‘mitochondrion-related organelles’ ...(MROs) with reduced functions, however the degree of reduction varies. Here, we generate high-quality draft genomes, transcriptomes, and predicted proteomes for five recently discovered free-living metamonads. Phylogenomic analyses placed these organisms in a group we name the ‘BaSk’ (Barthelonids+Skoliomonads) clade, a deeply branching sister group to the Fornicata, a phylum that includes parasitic and free-living flagellates. Bioinformatic analyses of gene models shows that these organisms are predicted to have extremely reduced MRO proteomes in comparison to other free-living metamonads. Loss of the mitochondrial iron-sulfur cluster assembly system in some organisms in this group appears to be linked to the acquisition in their common ancestral lineage of a SUF-like minimal system Fe/S cluster pathway by lateral gene transfer. One of the isolates, Skoliomonas litria, appears to have lost all other known MRO pathways. No proteins were confidently assigned to the predicted MRO proteome of this organism suggesting that the organelle has been lost. The extreme mitochondrial reduction observed within this free-living anaerobic protistan clade demonstrates that mitochondrial functions may be completely lost even in free-living organisms.Mitochondria are essential cellular components that are found in animals, plants, fungi, and protists. This study reports what is believed to be the first example of complete mitochondrial loss in a free-living organism, providing insights into the evolutionary plasticity of eukaryotic cells.
Cells replicate and segregate their DNA with precision. Previous studies showed that these regulated cell-cycle processes were present in the last eukaryotic common ancestor and that their core ...molecular parts are conserved across eukaryotes. However, some metamonad parasites have secondarily lost components of the DNA processing and segregation apparatuses. To clarify the evolutionary history of these systems in these unusual eukaryotes, we generated a genome assembly for the free-living metamonad Carpediemonas membranifera and carried out a comparative genomics analysis. Here, we show that parasitic and free-living metamonads harbor an incomplete set of proteins for processing and segregating DNA. Unexpectedly, Carpediemonas species are further streamlined, lacking the origin recognition complex, Cdc6 and most structural kinetochore subunits. Carpediemonas species are thus the first known eukaryotes that appear to lack this suite of conserved complexes, suggesting that they likely rely on yet-to-be-discovered or alternative mechanisms to carry out these fundamental processes.
The fine tuning of biological electrical signaling is mediated by variations in the rates of opening and closing of gates that control ion flux through different ion channels. Human ether-a-go-go ...related gene (HERG) potassium channels have uniquely rapid inactivation kinetics which are critical to the role they play in regulating cardiac electrical activity. Here, we exploit the K+ sensitivity of HERG inactivation to determine structures of both a conductive and non-conductive selectivity filter structure of HERG. The conductive state has a canonical cylindrical shaped selectivity filter. The non-conductive state is characterized by flipping of the selectivity filter valine backbone carbonyls to point away from the central axis. The side chain of S620 on the pore helix plays a central role in this process, by coordinating distinct sets of interactions in the conductive, non-conductive, and transition states. Our model represents a distinct mechanism by which ion channels fine tune their activity and could explain the uniquely rapid inactivation kinetics of HERG.HERG channel inactivation is critical for normal heart rhythm. Authors determine structures of open and non-conducting states of HERG and identify a key role for S620 on the pore helix in coordinating transitions between open and inactivated states.
Commercially-available “stability monitors” based on in situ atmospheric radon progeny measurements remain underutilised as a tool for urban pollution studies, due in part to difficulties experienced ...in relating their standard output directly to the atmospheric mixing state in a consistent manner. The main confounding factor has been a lack of attention to the fact that the observed near-surface atmospheric radon concentration includes large synoptic and fetch-related components in addition to the local stability influence. Here, a technique recently developed for stability classification using a research-quality dual-flow-loop two-filter radon detector is adapted for use with a commercially-available radon-based stability monitor. Performance of the classification scheme is then tested in Lanzhou, China, a topographically-complex region renowned for low mean annual wind speeds (0.8 m s−1) and winter stagnation episodes. Based on an 11-month composite, a factor of seven difference is estimated between peak NOx concentrations in the city's industrial region and a rural background location under stable conditions. The radon-based scheme is evaluated against the Pasquil-Gifford “radiation” (PGR) scheme, and assigns pollutant concentrations more consistently between defined atmospheric stability states than the PGR scheme. Furthermore, the PGR scheme consistently underestimates all peak pollutant concentrations under stable conditions compared with the radon-based scheme, in some cases (e.g. CO in the industrial region) by 25%.

A comparative study and evaluation of boundary layer height (BLH) estimation was conducted during an experimental campaign conducted at the Cape Grim Air Pollution station, Australia, from 1 June to ...13 July 2019. The temporal and spatial distributions of BLH were studied using data from a ceilometer, sodar, in situ meteorological measurements, and back-trajectory analyses. Generally, the BLH under continental sources is lower than that under marine sources. The BLH is featured with a shallow depth of 515 ± 340 m under the Melbourne/East Victoria continental source. Especially the mixed continental sources (Melbourne/East Victoria and Tasmania direction) lead to a rise in radon concentration and lower BLH. In comparison, the boundary layer reaches a higher averaged BLH value of 730 ± 305 m when marine air is prevalent. The BLH derived from ERA5 is positively biased compared to the ceilometer observations, except when the boundary layer is stable. The height at which wind profiles experience rapid changes corresponds to the BLH value. The wind flow within the boundary layer increased up to ∼200 m, where it then meandered up to ∼300 m. Furthermore, the statistic shows that BLH is positively associated with near-surface wind speed. This study firstly provides information on boundary layer structure in Cape Grim and the interaction with wind, which may aid in further evaluating their associated impacts on the climate and ecosystem.
Radon (Rn) is a radioactive, colourless, odourless, noble gas that decays rapidly. It’s most stable isotope, 222Rn, has a half-life of around 3.8 days. Atmospheric radon measurements play an ...important role in understanding our atmospheric environments. Naturally occurring radon can be used as an atmospheric tracer for airmass tracking, to assist in modelling boundary layer development, and is important for understanding background radiation levels and personal exposure to natural radiation. The daughter products from radon decay also play an important role when measuring fine particle pollution using beta-attenuation monitors (BAM). Beta radiation from the 222Rn decay chain interferes with BAM measurements of fine particles; thus, some BAMs incorporate radon measurements into their sampling systems. BAMs are ubiquitous in air quality monitoring networks globally and present a hitherto unexplored source of dense, continuous radon measurements. In this paper, we compare in situ real world 222Rn measurements from a high quality ANSTO dual flow loop, dual filter radon detector, and the radon measurements made by a commercial BAM instrument (Thermo 5014i). We find strong correlations between systems for hourly measurements (R2 = 0.91), daily means (R2 = 0.95), hour of day (R2 = 0.72–0.94), and by month (R2 = 0.83–0.94). The BAM underestimates radon by 22–39%; however, the linear response of the BAM measurements implies that they could be corrected to reflect the ANSTO standard measurements. Regardless, the radon measurements from BAMs could be used with correction to estimate local mixed layer development. Though only a 12-month study at a single location, our results suggest that radon measurements from BAMs can complement more robust measurements from standard monitors, augment radon measurements across broad regions of the world, and provide useful information for studies using radon as a tracer, particularly for boundary layer development and airmass identification.
Abstract Metamonads are a large and exclusively anaerobic group of protists. Additionally, they are one of the three clades proposed to ancestrally possess an “excavate” cell morphology, with a ...conspicuous ventral groove accompanied by a posterior flagellum with a vane. Here, we cultivate and characterize four anaerobic bacterivorous flagellates from hypersaline and alkaline soda lake environments, which represent a novel clade. Small subunit ribosomal RNA (SSU rRNA) gene phylogenies support recent phylogenomic analyses in placing them as the sister of barthelonids, a group that is itself sister to or deeply branching within Fornicata (Metamonada). The new isolates have a distinctive morphology: the hunchbacked cell body is traversed by a narrow ventral groove ending in a large opening to a conspicuous recurrent cytopharynx. The right margin of the groove is defined by a thin “lip.” The posterior flagellum bears a wide ventral‐facing vane. The narrow ventral groove and elongate cytopharynx are shared with barthelonids. We describe one isolate as Skoliomonas litria , gen. et sp. nov. Further investigation of their mitochondrial‐related organelles (MROs) and detailed ultrastructural studies would be important to understanding the adaptation to anaerobic conditions in Metamonads—especially fornicates—as well as the evolution of the “excavate” cell architecture.
One year of radon, benzene and carbon monoxide (CO) concentrations were analysed to characterise the combined influences of variations in traffic density and meteorological conditions on urban air ...quality in Bern, Switzerland. A recently developed radon-based stability categorisation technique was adapted to account for seasonal changes in day length and reduction in the local radon flux due to snow/ice cover and high soil moisture. Diurnal pollutant cycles were shown to result from an interplay between variations in surface emissions (traffic density), the depth of the nocturnal atmospheric mixing layer (dilution) and local horizontal advection of cleaner air from outside the central urban/industrial area of this small compact inland city. Substantial seasonal differences in the timing and duration of peak pollutant concentrations in the diurnal cycle were attributable to changes in day length and the switching to/from daylight-savings time in relation to traffic patterns. In summer, average peak benzene concentrations (0.62 ppb) occurred in the morning and remained above 0.5 ppb for 2 hours, whereas in winter average peak concentrations (0.85 ppb) occurred in the evening and remained above 0.5 ppb for 9 hours. Under stable conditions in winter, average peak benzene concentrations (1.1 ppb) were 120% higher than for well-mixed conditions (0.5 ppb). By comparison, summertime peak benzene concentrations increased by 53% from well-mixed (0.45 ppb) to stable nocturnal conditions (0.7 ppb). An idealised box model incorporating a simple advection term was used to derive a nocturnal mixing length scale based on radon, and then inverted to simulate diurnal benzene and CO emission variations at the city centre. This method effectively removes the influences of local horizontal advection and stability-related vertical dilution from the emissions signal, enabling a direct comparison with hourly traffic density. AbstractObjectiveTo investigate the risks of ovarian, breast, and corpus uteri cancer in women who have had assisted reproduction.DesignLarge, population based, data linkage cohort study.Setting and ...participantsAll women who had assisted reproduction in Great Britain, 1991-2010, as recorded by the Human Fertilisation and Embryology Authority (HFEA).InterventionsHFEA fertility records for cohort members were linked to national cancer registrations.Main outcome measuresObserved first diagnosis of ovarian, breast, and corpus uteri cancer in cohort members were compared with age, sex, and period specific expectation. Standardised incidence ratios (SIRs) were calculated by use of age, sex, and period specific national incidence rates.Results255 786 women contributed 2 257 789 person years’ follow-up. No significant increased risk of corpus uteri cancer (164 cancers observed v 146.9 cancers expected; SIR 1.12, 95% confidence interval 0.95 to 1.30) was found during an average of 8.8 years’ follow-up. This study found no significantly increased risks of breast cancer overall (2578 v 2641.2; SIR 0.98, 0.94 to 1.01) or invasive breast cancer (2272 v 2371.4; SIR 0.96, 0.92 to 1.00). An increased risk of in situ breast cancer (291 v 253.5; SIR 1.15, 1.02 to 1.29; absolute excess risk (AER) 1.7 cases per 100 000 person years, 95% confidence interval 0.2 to 3.2) was detected, associated with an increasing number of treatment cycles (P=0.03). There was an increased risk of ovarian cancer (405 v 291.82; SIR 1.39, 1.26 to 1.53; AER 5.0 cases per 100 000 person years, 3.3 to 6.9), both invasive (264 v 188.1; SIR 1.40, 1.24 to 1.58; AER 3.4 cases per 100 000 person years, 2.0 to 4.9) and borderline (141 v 103.7; SIR 1.36, 1.15 to 1.60; AER 1.7 cases per 100 000 person years, 0.7 to 2.8). Increased risks of ovarian tumours were limited to women with endometriosis, low parity, or both. This study found no increased risk of any ovarian tumour in women treated because of only male factor or unexplained infertility.ConclusionsNo increased risk of corpus uteri or invasive breast cancer was detected in women who had had assisted reproduction, but increased risks of in situ breast cancer and invasive and borderline ovarian tumours were found in this study. Our results suggest that ovarian tumour risks could be due to patient characteristics, rather than assisted reproduction itself, although both surveillance bias and the effect of treatment are also possibilities. Ongoing monitoring of this population is essential.
