Urbanization is a global process contributing to the loss and fragmentation of natural habitats. Many studies have focused on the biological response of terrestrial taxa and habitats to urbanization. ...However, little is known regarding the consequences of urbanization on freshwater habitats, especially small lentic systems. In this study, we examined aquatic macro‐invertebrate diversity (family and species level) and variation in community composition between 240 urban and 782 nonurban ponds distributed across the United Kingdom. Contrary to predictions, urban ponds supported similar numbers of invertebrate species and families compared to nonurban ponds. Similar gamma diversity was found between the two groups at both family and species taxonomic levels. The biological communities of urban ponds were markedly different to those of nonurban ponds, and the variability in urban pond community composition was greater than that in nonurban ponds, contrary to previous work showing homogenization of communities in urban areas. Positive spatial autocorrelation was recorded for urban and nonurban ponds at 0–50 km (distance between pond study sites) and negative spatial autocorrelation was observed at 100–150 km and was stronger in urban ponds in both cases. Ponds do not follow the same ecological patterns as terrestrial and lotic habitats (reduced taxonomic richness) in urban environments; in contrast, they support high taxonomic richness and contribute significantly to regional faunal diversity. Individual cities are complex structural mosaics which evolve over long periods of time and are managed in diverse ways. This facilitates the development of a wide range of environmental conditions and habitat niches in urban ponds which can promote greater heterogeneity between pond communities at larger scales. Ponds provide an opportunity for managers and environmental regulators to conserve and enhance freshwater biodiversity in urbanized landscapes whilst also facilitating key ecosystem services including storm water storage and water treatment.
The effect of an increase in atmospheric aerosol concentrations on the distribution and radiative properties of Earth’s clouds is the most uncertain component of the overall global radiative forcing ...from preindustrial time. General circulation models (GCMs) are the tool for predicting future climate, but the treatment of aerosols, clouds, and aerosol–cloud radiative effects carries large uncertainties that directly affect GCM predictions, such as climate sensitivity. Predictions are hampered by the large range of scales of interaction between various components that need to be captured. Observation systems (remote sensing, in situ) are increasingly being used to constrain predictions, but significant challenges exist, to some extent because of the large range of scales and the fact that the various measuring systems tend to address different scales. Fine-scale models represent clouds, aerosols, and aerosol–cloud interactions with high fidelity but do not include interactions with the larger scale and are therefore limited from a climatic point of view. We suggest strategies for improving estimates of aerosol–cloud relationships in climate models, for new remote sensing and in situ measurements, and for quantifying and reducing model uncertainty.
Despite the existence of well‐established international environmental and nature conservation policies (e.g., the Ramsar Convention and Convention on Biological Diversity) ponds are largely missing ...from national and international legislation and policy frameworks. Ponds are among the most biodiverse and ecologically important freshwater habitats, and their value lies not only in individual ponds, but more importantly, in networks of ponds (pondscapes). Ponds make an important contribution to society through the ecosystem services they provide, with effective conservation of pondscapes essential to ensuring that these services are maintained. Implementation of current pond conservation through individual site designations does not function at the landscape scale, where ponds contribute most to biodiversity. Conservation and management of pondscapes should complement current national and international nature conservation and water policy/legislation, as pondscapes can provide species protection in landscapes where large‐scale traditional conservation areas cannot be established (e.g., urban or agricultural landscapes). We propose practical steps for the effective incorporation or enhancement of ponds within five policy areas: through open water sustainable urban drainage systems in urban planning, increased incentives in agrienvironment schemes, curriculum inclusion in education, emphasis on ecological scale in mitigation measures following anthropogenic developments, and the inclusion of pondscapes in conservation policy.
The distribution of macroinvertebrates in the heads and tails of riffles were examined in an in situ field experiment under stable baseflow conditions. Paired colonisation cylinders were used to ...examine the influence of vertical hydraulic exchange (upwelling and downwelling) and horizontal interstitial flow on the patterns of sedimentation and invertebrate colonisation. Sedimentation rates were greatest in cylinders permitting vertical and horizontal flow (VHE cylinders), and were significantly lower (29%) in cylinders where only vertical flow and ingress of fine sediment were possible (VE cylinders). The results demonstrate that horizontal interstitial flows represent an important pathway for fine sediment transport. Differences in fine sediment accumulation were also observed between riffle heads and tails. Significantly higher sedimentation rates were recorded in riffle tails, with the macroinvertebrate communities characterised by larger proportions of fine sediment tolerant taxa. In contrast, riffle head communities were characterised by greater proportions of sediment sensitive taxa, and in the case of VHE cylinders, shredders and EPT taxa. The results demonstrate that spatial differences in fine sediment deposition are evident at the riffle scale as a function of vertical and horizontal subsurface flows and that these factors play a key role in the distribution of macroinvertebrate fauna.
Abstract Extracellular vesicles (EVs) are natural nanoparticles that mediate intercellular transfer of RNA and proteins and are of great medical interest; serving as novel biomarkers and potential ...therapeutic agents. However, there is little consensus on the most appropriate method to isolate high-yield and high-purity EVs from various biological fluids. Here, we describe a systematic comparison between two protocols for EV purification: ultrafiltration with subsequent liquid chromatography (UF-LC) and differential ultracentrifugation (UC). A significantly higher EV yield resulted from UF-LC as compared to UC, without affecting vesicle protein composition. Importantly, we provide novel evidence that, in contrast to UC-purified EVs, the biophysical properties of UF-LC-purified EVs are preserved, leading to a different in vivo biodistribution, with less accumulation in lungs. Finally, we show that UF-LC is scalable and adaptable for EV isolation from complex media types such as stem cell media, which is of huge significance for future clinical applications involving EVs. Deposition of fine sediment that fills interstitial spaces in streambed substrates is widely acknowledged to have significant negative effects on macroinvertebrate communities, but the temporal ...consistency of clogging effects is less well known. In this study the effects of experimentally enhanced fine sediment content on aquatic invertebrates were examined over 126days in two lowland UK streams. Taxonomic approaches indicated significant differences in macroinvertebrate community structure associated with sediment treatment (clean or sedimented substrates), although the effects were variable on some occasions. The degree of separation between clean and sedimented communities was strong within seven of the nine sampling periods with significant differences in community composition being evident. EPT taxa and taxon characterised as sensitive to fine sediment demonstrated strong responses to enhanced fine sediment loading. Faunal traits also detected the effects of enhanced fine sediment loading but the results were not as consistent or marked. More widely, the study highlights the temporal dynamics of sedimentation effects upon macroinvertebrate communities and the need to consider faunal life histories when examining the effects of fine sediment loading pressures on lotic ecosystems.

A greater understanding and effective management of biological invasions is a priority for biodiversity conservation globally. Many freshwater ecosystems are experiencing the colonization and spread ...of multiple co-occurrent alien species. Here the implications of both the relative abundance and richness of alien invaders on aquatic macroinvertebrate taxonomic and functional richness, ecosystem quality, and functional redundancy are assessed using long-term data from rivers in England. Based on the most common aquatic invaders, results indicated that their richness, rather than abundance, was the most important factor negatively affecting aquatic macroinvertebrate biodiversity. However, the response of functional redundancy was negatively affected by invader abundance at the river basin scale. The response of communities varied as the number of invading taxa increased, with the most marked reductions following the colonization of the first few invaders. Results indicate that different facets of multiple biological invasions influence distinct aspects of aquatic biodiversity. Preventing the establishment of new invaders and limiting invader taxa richness within a community should therefore be a conservation priority. These findings will assist river scientists in understanding mechanisms driving changes in biodiversity and facilitate the testing of ecological theories while also ensuring environmental managers and regulators can prioritize conservation / management opportunities.
Advances in metacommunity theory have made a significant contribution to understanding the drivers of variation in biological communities. However, there has been limited empirical research exploring ...the expression of metacommunity theory for two fundamental components of beta diversity: nestedness and species turnover. In this paper, we examine the influence of local environmental and a range of spatial variables (hydrological connectivity, proximity and overall spatial structure) on total beta diversity and the nestedness and turnover components of beta diversity for the entire macroinvertebrate community and active and passively dispersing taxa within pond habitats. High beta diversity almost entirely reflects patterns of species turnover (replacement) rather than nestedness (differences in species richness) in our dataset. Local environmental variables were the main drivers of total beta diversity, nestedness and turnover when the entire community was considered and for both active and passively dispersing taxa. The influence of spatial processes on passively dispersing taxa, total beta diversity and nestedness was significantly greater than for actively dispersing taxa. Our results suggest that species sorting (local environmental variables) operating through niche processes was the primary mechanism driving total beta diversity, nestedness and turnover for the entire community and active and passively dispersing taxa. In contrast, spatial factors (hydrological connectivity, proximity and spatial eigenvectors) only exerted a secondary influence on the nestedness and turnover components of beta diversity.
The hyporheic zone of river ecosystems provides a habitat for a diverse macroinvertebrate community that makes a vital contribution to ecosystem functioning and biodiversity. However, effective ...methods for sampling this community have proved difficult to establish, due to the inaccessibility of subsurface sediments. The aim of this study was to compare the two most common semi-quantitative macroinvertebrate pump-sampling techniques: Bou-Rouch and vacuum-pump sampling. We used both techniques to collect replicate samples in three contrasting temperate-zone streams, in each of two biogeographical regions (Atlantic region, central England, UK; Continental region, southeast France). Results were typically consistent across streams in both regions: Bou-Rouch samples provided significantly higher estimates of taxa richness, macroinvertebrate abundance, and the abundance of all UK and eight of 10 French common taxa. Seven and nine taxa which were rare in Bou-Rouch samples were absent from vacuum-pump samples in the UK and France, respectively; no taxon was repeatedly sampled exclusively by the vacuum pump. Rarefaction curves (rescaled to the number of incidences) and non-parametric richness estimators indicated no significant difference in richness between techniques, highlighting the capture of more individuals as crucial to Bou-Rouch sampling performance. Compared to assemblages in replicate vacuum-pump samples, multivariate analyses indicated greater distinction among Bou-Rouch assemblages from different streams, as well as significantly greater consistency in assemblage composition among replicate Bou-Rouch samples collected in one stream. We recommend Bou-Rouch sampling for most study types, including rapid biomonitoring surveys and studies requiring acquisition of comprehensive taxon lists that include rare taxa. Despite collecting fewer macroinvertebrates, vacuum-pump sampling remains an important option for inexpensive and rapid sample collection.
