Pancreatic carcinoma (PC) is a lethal cancer. Gut microbiota is associated with some risk factors of PC, e.g. obesity and types II diabetes. However, the specific gut microbial profile in clinical PC ...in China has never been reported. This prospective study collected 85 PC and 57 matched healthy controls (HC) to analyze microbial characteristics by MiSeq sequencing. The results showed that gut microbial diversity was decreased in PC with an unique microbial profile, which partly attributed to its decrease of alpha diversity. Microbial alterations in PC featured by the increase of certain pathogens and lipopolysaccharides-producing bacteria, and the decrease of probiotics and butyrate-producing bacteria. Microbial community in obstruction cases was separated from the un-obstructed cases.

Importantly, based on 40 genera associated with PC, microbial markers achieves a high classification power with AUC of 0.842. In conclusion, gut microbial profile was unique in PC, providing a microbial marker for non-invasive PC diagnosis.
The product of the Nijmegen breakage syndrome gene (NBS1) plays crucial roles in DNA damage response through its association with many proteins, including MRE11 and RAD50. However, it remains to be ...determined exactly how NBS1 accumulates at or near DNA double-strand breaks. Here we report that MDC1 directly binds to NBS1 and targets NBS1 to the sites of DNA damage. The MDC1-NBS1 interaction occurs through a specific region (residues 200-420) of MDC1, which contains multiple consensus casein kinase 2 (CK2) phosphorylation sites. In addition, this interaction requires both the forkhead-associated (FHA) and tandem BRCA1 C-terminal (BRCT) domains of NBS1. Disruption of the MDC1-NBS1 interaction results in failure of NBS1 accumulation at DNA double-strand breaks and impairment of intra-S checkpoint activation. These studies provide important mechanistic insights as to how MDC1 regulates NBS1 and the intra-S-phase checkpoint in response to DNA damage.
Abstract
In order to explore the influence of weak interlayer on blasting characteristics in natural rock mass, by using the particle flow code (PFC
2D
), a single hole blasting numerical model of ...hard rock with soft interlayer is established. The blasting experiments at different positions and thicknesses of weak interlayer are carried out. Then an in-depth analysis from the perspectives of crack effect, stress field and energy field is made. Results showed that: (i) When the explosion is initiated outside the weak interlayer, if the interlayer is located within about twice the radius of the crushing area, the closer the interlayer is to the blast hole, the higher the damage degree of the rock mass around the blast hole. And the number of cracks will increase by about 1–2% when the distance between the weak interlayer and the blast hole decreases by 0.5 m. (ii) When detonating outside the weak interlayer, if the interlayer is within about 4 times radius of the crushing area, the hard rock on both sides of the weak interlayer will in a high stress state. Under the same case, the peak kinetic energy and peak friction energy will increase by about 23 and 13%, respectively, and the peak strain energy will increase by about 218 kJ for every 0.1 m increase in the thickness of the weak interlayer.
A dynamic stall will cause dramatic changes in the aerodynamic performance of the blade, resulting in a sharp increase in the blade vibration load. The bionic leading-edge airfoil with different ...waviness ratios, inspired by the humpback whales flipper, is adopted to solve this problem. In this study, based on the NACA0015 airfoil, the three-dimensional unsteady numerical simulation and sliding mesh technique are used to reveal the flow control mechanism on the dynamic stall of the bionic wavy leading edge. The effects of the waviness ratio on the dynamic stall characteristics of the airfoil are also investigated. The results show that the peak drag coefficient is dramatically reduced when a sinusoidal leading edge is applied to the airfoil. Although the peak lift coefficient is also reduced, the reduction is much smaller. When the waviness ratio R is 0.8, the peak drag coefficient of the airfoil is reduced by 17.14% and the peak lift coefficient of the airfoil is reduced by 9.20%. The dynamic hysteresis effect is improved gradually with an increasing waviness ratio. For the bionic airfoil with R = 1.0, the area of the hysteresis loop is the smallest.
Neurogenesis in the adult brain comprises the entire set of events of neuronal development. It begins with the division of precursor cells to form a mature, integrated, and functioning neuronal ...network. Adult neurogenesis is believed to play an important role in animals’ cognitive abilities, including learning and memory. In the present study, significant neuronal differentiation-promoting activity of 80% (v/v) ethanol extract of P. cocos (EEPC) was found in Neuro-2a cells and mouse cortical neural stem/progenitor cells (NSPCs). Subsequently, a total of 97 compounds in EEPC were identified by UHPLC-Q-Exactive-MS/MS. Among them, four major compounds—Adenosine; Choline; Ethyl palmitoleate; and L-(-)-arabinitol—were further studied for their neuronal differentiation-promoting activity. Of which, choline has the most significant neuronal differentiation-promoting activity, indicating that choline, as the main bioactive compound in P. cocos, may have a positive effect on learning and memory functions. Compared with similar research literature, this is the first time that the neuronal differentiation-promoting effects of P. cocos extract have been studied.
Plant polyphenols are rich sources of natural anti-oxidants and prebiotics. After ingestion, most polyphenols are absorbed in the intestine and interact with the gut microbiota and modulated ...metabolites produced by bacterial fermentation, such as short-chain fatty acids (SCFAs). Dietary polyphenols immunomodulatory role by regulating intestinal microorganisms, inhibiting the etiology and pathogenesis of various diseases including colon cancer, colorectal cancer, inflammatory bowel disease (IBD) and colitis. Foodomics is a novel high-throughput analysis approach widely applied in food and nutrition studies, incorporating genomics, transcriptomics, proteomics, metabolomics, and integrating multi-omics technologies. In this review, we present an overview of foodomics technologies for identifying active polyphenol components from natural foods, as well as a summary of the gastrointestinal protective effects of polyphenols based on foodomics approaches. Furthermore, we critically assess the limitations in applying foodomics technologies to investigate the protective effect of polyphenols on the gastrointestinal (GI) system. Finally, we outline future directions of foodomics techniques to investigate GI protective effects of polyphenols. Foodomics based on the combination of several analytical platforms and data processing for genomics, transcriptomics, proteomics and metabolomics studies, provides abundant data and a more comprehensive understanding of the interactions between polyphenols and the GI tract at the molecular level. This contribution provides a basis for further exploring the protective mechanisms of polyphenols on the GI system.
