Motion sickness (MS) is an acute disorder that occurs in healthy individuals worldwide regardless of gender, age, or ethnicity. Our study used a mouse model to rule out the effects of any ...psychological factors related to MS and EA. Subjects were randomly separated into four groups, namely the control group (Con), motion sickness inducing group (MS), mentioning sickness inducing with electroacupuncture treatment group (EA) and motion sickness inducing only in TRPV1 knockout mice group (TRPV1
). The consumption of kaolin, a non-nutrient substance, was measured as a behavior observed response of an emetic reflex in a murine model. This behavior is referred to as pica behavior. Our results showed that pica behavior was observed in the MS group. Moreover, kaolin consumption in the EA group decreased to the average baseline of the control group. A similar result was observed in TRPV1 null mice. We also observed an increase of TRPV1 and related molecules in the thalamus, hypothalamic and brain stem after MS stimulation and a significant decrease in the EA and TRPV1 null groups. This is the first study to demonstrate that TRPV1 pathways are possibly associated with mechanisms of MS, and can be attended through EA or TRPV1 genetic manipulation.
Caffeoylquinic acids, coumarins and dicaffeoyl derivatives are considered to be three kinds of the most abundant bioactive components in
, an anti-inflammatory herb mainly found in Southern Asia. The ...combined anti-inflammatory effect of three typical constituents C + R + I (chlorogenic acid + rosmarinic acid + isofraxidin) from this plant has been investigated. The result implies that targeting the MAPK-NF-κB pathway would be one of the major mechanisms involved, using LPS stimulated RAW 264.7 cells as
model and LPS-induced acute lung injury in mice as
model. C + R + I can significantly suppress the levels of nitric oxide (NO), pro-inflammatory cytokines, and inhibit iNOS and COX-2 expression in LPS-treated RAW264.7 macrophage cells. Western blot analysis showed that C + R + I suppressed phosphorylation of NF-κB and MAPK, including phosphorylation of p65-NF-κB, IKB, ERK, JNK and P38. Besides, C + R + I suppressed MPO protein expression, but promoted SOD and HO-1 expression, and the related targets for C, R, and I were also predicted by molecular docking. This indicated that C + R + I could alleviate oxidative stress induced by LPS, which were further verified in the
model of mice with acute lung injury through the measurement of corresponding inflammatory mediators and the analysis of immunehistochemistry.
Advances in static random access memory (SRAM)-CIM devices are meant to increase capacity while improving energy efficiency (EF) and reducing computing latency (<inline-formula> <tex-math ...notation="LaTeX">T_{\mathrm {AC}} </tex-math></inline-formula>). This work presents a novel SRAM-CIM structure using: 1) a segmented-bitline charge-sharing (SBCS) scheme for multiply-and-accumulate (MAC) operations with low energy consumption and a consistently high signal margin across MAC values; 2) a bitline-combining (BL-CMB) scheme to reduce the number of analog-to-digital converters (ADCs) and, thereby, provide options in determining a tradeoff between EF and inference accuracy; 3) a source-injection local-multiplication cell (SILMC) connected to two types of global-bitline-switch to support the SBCS and BL-CMB schemes with consistent signal margin against process variation in transistors; and 4) prioritized-hybrid ADC to suppress area and power overhead for analog readout operations. We fabricated a 28-nm 384-kb SRAM-CIM macro using foundry-provided compact-6T cells supporting MAC operations with 16 accumulations of 8-b input and 8-b weight with near-full precision output (20 b). This macro achieved <inline-formula> <tex-math notation="LaTeX">T_{\mathrm {AC}} </tex-math></inline-formula> of 7.2 ns and EF of 22.75 TOPS/W performing 8-b-MAC operations.
Hydrogen peroxide is commonly used in the food processing industry as a chlorine-free bleaching and sterilizing agent, but excessive amounts of residual hydrogen peroxide have led to cases of food ...poisoning. Here we describe the development of a novel nonenzymatic colorimetric method for the determination of residual hydrogen peroxide in foods and agricultural products. Nitrophenylboronic acids chemoselectively react with hydrogen peroxide under alkaline conditions to produce yellow nitrophenolates. Of the three nitrophenylboronic acid isomers tested, the p-isomer displayed the highest sensitivity for hydrogen peroxide and the fastest reaction kinetics. The reaction product, p-nitrophenolate, has an absorption maximum at 405 nm and a good linear correlation between the hydrogen peroxide concentration and the A 405 values was obtained. We successfully applied this convenient and rapid method for hydrogen peroxide determination to samples of dried bean curds and disposable chopsticks, thereby demonstrating its potential in foods and agricultural industries.
In this paper, a novel synthetization approach is proposed for filter-integrated wideband impedance transformers (ITs). The original topology consists of N cascaded coupled line sections (CLSs) with ...2N characteristic impedance parameters. By analyzing these characteristic impedances, a Chebyshev response can be derived to consume N + 2 design conditions. To optimize the left N − 2 variable parameters, CLSs were newly substituted by transmission lines (TLs) to consume the remaining variable parameters and simplify the circuit topology. Therefore, there are totally 2N − N − 2 substituting possibilities. To verify the proposed approach, 25 cases are listed under the condition of N = 5, and 7 selected cases are compared and discussed in detail. Finally, a 75–50 Ω IT with 100% fractional bandwidth and 20 dB bandpass return loss (RL) is designed and fabricated. The measured results meet the circuit simulation and the EM simulation accurately.
Cancer-associated fibroblasts (CAFs) play a crucial role in cancer progression and even initiation. However, the origins of CAFs in various cancer types remain controversial, and one of the important ...hypothesized origins is through epithelial-mesenchymal transition (EMT) from cancer cells. In this study, we investigated whether the HEp-2 laryngeal cancer cells are able to generate CAFs via EMT during tumor formation, which is now still unknown. The laryngeal xenografted tumor model was established by inoculating the HEp-2 laryngeal cancer cell line in nude mice. Primary cultured CAFs from the tumor nodules and matched normal fibroblasts (NFs) from the adjacent connective tissues were subcultured, purified, and verified by immunofluorescence. Migration, invasion, and proliferation potentials were compared between the CAFs and NFs. A co-culture of CAFs with HEp-2 cells and a co-injection of CAFs with HEp-2 cells in nude mice were performed to examine the cancer-promoting potential of CAFs to further verify their identity. Karyotypic analyses of the CAFs, NFs, and HEp-2 cells were conducted. A co-culture of NFs with HEp-2 cells was also performed to examine the expression of activated markers of CAFs. A pathological examination confirmed that the laryngeal xenografted tumor model was successfully established, containing abundant CAFs. Immunocytochemical staining verified the purities and identities of the CAFs and NFs. Although the CAFs manifested higher migration, invasion, proliferation, and cancer-promoting capacities compared with the NFs, an analysis of chromosomes revealed that both the CAFs and NFs showed typical normal mouse karyotypes. In addition, the NFs co-cultured with HEp-2 cells did not show induced expressions of activated markers of CAFs. Our findings reveal that the CAFs in the HEp-2 established laryngeal xenografted tumor are not of laryngeal cancer origin but of mouse origin, indicating that the HEp-2 laryngeal cancer cells cannot generate their own CAFs via EMT in this model.

Recent studies have found compared with insulin glargine (IGlar), insulin degludec/aspart (IDeg/Asp) may provide adequate glycemic control and prevent hypoglycemia events in type 2 diabetes mellitus ...(T2DM). Consequently, we performed a meta-analysis to appraise and compare the efficiency and safety of IDeg/Asp and IGlar in the treatment of T2DM. We sought the databases including PubMed, Embase, Scopus, Cochrane library to confirm related articles which inspected the effect of IDeg/Asp versus IGlar for the treatment of T2DM until May 2021. Finally, six randomized controlled trials (RCTs) of 1,346 patients were included. The results showed that IDeg/Asp significantly decreased the mean hemoglobin A1c (HbA1c) level but was prone to serious adverse events, and IGlar increased the nocturnal confirmed hypoglycemia events. Besides, there were no significant changes in other indicators, including mean fasting plasma glucose (FPG) level, nine-point self-measured plasma glucose (SMPG) level, and adverse events. What’s more, we found that there was no significant difference in the occurrence of hypoglycemia overall, but our subgroup analysis of confirmed hypoglycemia revealed the population in this subgroup (duration of diabetes ≤11 years) might has its particularity effecting the hypoglycemia outcome. Concerning efficiency, IDeg/Asp may have advantages in controlling the mean HbA1c level. Regarding safety, IGlar might increase the risk of nocturnal confirmed hypoglycemia. Further evidence is needed to compare better the efficiency and safety of IDeg/Asp versus IGlar therapy.
AIM:To investigate the effects of tectorigenin on human hepatocellular carcinoma(HCC)HepG2 cells.METHODS:Tectorigenin, one of the main components of rhizome of Iris tectorum, was prepared by simple ...methods,such as extraction, filtration, concentration, precipitation and recrystallization.HepG2 cells were incubated with tectorigenin at different concentrations, and their viability was assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay.Apoptosis was detected by morphological observation of nuclear change, agarose gel electrophoresis of DNA ladder, and flow cytometry with Hoechst 33342,Annexin V-EGFP and propidium iodide staining.Generation of reactive oxygen species was quantified using DCFH-DA.Intracellular Ca2+was monitored by Fura 2-AM.Mitochondrial membrane potential was monitored using Rhodamine 123.Release of cytochrome c from mitochondria to cytosol was detected by Western blotting.Activities of caspase-3,-8 and-9 were investigated by Caspase Activity Assay Kit.RESULTS:The viability of HepG2 cells treated by tectorigenin decreased in a concentration-and timedependent manner. The concentration that reduced the number of viable HepG2 cells by 50%(IC50)after 12,24 and 48 h of incubation was 35.72 mg/L,21.19 mg/L and 11.06 mg/L,respectively.However,treatment with tectorigenin at 20 mg/L resulted in a very slight cytotoxicity to L02 cells after incubation for 12,24 or 48 h.Tectorigenin at a concentration of 20 mg/L greatly inhibited the viability of HepG2 cells and induced the condensation of chromatin and fragmentation of nuclei.Tectorigenin induced apoptosis of HepG2 cells in a time-and dose-dependent manner.Compared with the viability rate, induction of apoptosis was the main mechanism of the anti-proliferation effect of tectorigenin in HepG2 cells.Furthermore, tectorigenininduced apoptosis of HepG2 cells was associated with the generation of reactive oxygen species, increased intracellularCa2+i,loss of mitochondrial membrane potential, translocation of cytochrome c,and activation of caspase-9 and-3.CONCLUSION:Tectorigenin induces apoptosis of HepG2 cells mainly via mitochondrial-mediated pathway, and produces a slight cytotoxicity to L02 cells.
Peanut (
Arachis hypogaea
L.) skins are primarily an industrial by-product with only small quantities utilized in animal feed or dropped directly. Due to peanut skins being a rich source of phenolic ...compounds with antioxidant activity and low cost, they have a great potential utility as food preservative. Herein, the chemical composition and antioxidant activity of extracts obtained from peanut skins prepared with 95% ethanol by ultrasound extraction also needed to be established. The crude peanut skin extracts of extraction yield, total phenols and flavonoids, DPPH scavenging activity, ferrous-ion chelating activity, reducing power, Trolox equivalent antioxidant capacity (TEAC) and accelerated oxidation of button mushroom (
Agaricus bisporus
) and sacha inchi (
Plukenetia volubilis
L.) oil were evaluated. To understand the actual antioxidant effects of peanut skin extracts, the button mushroom and sacha inchi oil were added in various doses of peanut skin extracts under air and pure oxygen ambient with the isothermal temperature of 30 and 60.5 °C by conducting isothermal calorimeter (TAM Air) analyses, respectively. Overall, the peanut skin extracts displayed beneficial antioxidant properties in button mushrooms and sacha inchi oil, which could be used as a natural antioxidant of agricultural waste reduction and added value to circular economy and sustainable development in food industries for SDG12 by responsible consumption and production.
To investigate the association between radiotherapy (RT) and thoracic vertebral fractures in esophageal squamous cell carcinoma (ESCC) and explore the risk factors of thoracic vertebral fracture in ...ESCC who underwent RT. This retrospective cohort study including 602 consecutive ESCC patients examined the association between RT and thoracic vertebral fractures using multivariable Cox proportional hazard models and relevant risk factors of thoracic vertebral fractures based on clinical and RT parameters in patients with ESCC. Followed for a median follow-up of 24 months, 54 patients had thoracic vertebral fractures. The multivariable analysis revealed RT as an independent risk factor after adjusting for clinical risk factors. Univariable analyses associated a 5-Gy increase in vertebral dose to single vertebrae and a 1-time increase in RT fraction with higher risk of vertebral fracture. Adding RT factors (vertebral dose and fraction) and mean vertebral hounsfield unit to the Cox models containing conventional clinical risk factors significantly improved the χ
2
value for predicting vertebral fractures (all
P
< .001). This study revealed RT, as well as increased vertebral dose and RT fractions, as a significant, consistent, and strong vertebral fracture predictor in ESCC. Combined vertebral dose, RT fractions, and vertebral hounsfield unit provided optimal risk stratification for ESCC patients.
