Human adipose-derived mesenchymal progenitor cells (haMPCs) are stem cells with multiple differentiation potential and immunomodulatory function. Re-Join® comprises in vitro expanded haMPCs from ...adipose tissue of patients combined with cell suspension solution. This study was undertaken to evaluate the efficacy and safety of Re-Join® in patients with symptomatic knee osteoarthritis (OA).
Patients with Kellgren-Lawrence grade 1-3 knee OA were recruited from two centers and randomized to receive intra-articular injection of Re-Join® or HA. Pain and function were assessed by using WOMAC score, VAS, and SF-36. Magnetic resonance imaging (MRI) analysis was performed to measure cartilage repair. Adverse events (AEs) were collected.
Fifty-three patients were randomized. Significant improvements in WOMAC, VAS, and SF-36 scores were observed in both groups at months 6 and 12 compared with baseline. Compared with the HA group, significantly more patients achieved 50% improvement of WOMAC and a trend of more patients achieved a 70% improvement rate in Re-Join® group after 12 months. Meanwhile, there was notably more increase in articular cartilage volume of both knees in the Re-Join® group than in the HA group after 12 months as measured by MRI. AEs were comparable between two groups. Most AEs were mild and moderate except one SAE of right knee joint infection in the HA group.
Significant improvements in joint function, pain, quality of life, and cartilage regeneration were observed in Re-Join®-treated knee OA patients with good tolerance in a period of 12 months.

Deep foundation pit settlement prediction based on machine learning is widely used for ensuring the safety of construction, but previous studies are limited to not fully considering the spatial ...correlation between monitoring points. This paper proposes a transformer-based deep learning method that considers both the spatial and temporal correlations among excavation monitoring points. The proposed method creates a dataset that collects all excavation monitoring points into a vector to consider all spatial correlations among monitoring points. The deep learning method is based on the transformer, which can handle the temporal correlations and spatial correlations. To verify the model's accuracy, it was compared with an LSTM network and an RNN-LSTM hybrid model that only considers temporal correlations without considering spatial correlations, and quantitatively compared with previous research results. Experimental results show that the proposed method can predict excavation deformations more accurately. The main conclusions are that the spatial correlation and the transformer-based method are significant factors in excavation deformation prediction, leading to more accurate prediction results.

The present study documents a pronounced interdecadal change in summer rainfall over southern China around 1992/93 and explores the plausible reasons for this change. The summer rainfall is ...persistently below normal during 1980–92 and above normal during 1993–2002. Coherent changes in atmospheric circulation are identified over East Asia and the South China Sea (SCS)–western North Pacific (WNP). The increase in rainfall is accompanied by an increase in lower-level convergence, midtropospheric ascent, and upper-level divergence over southern China. The changes in lower-level winds feature two anomalous anticyclones: one over the SCS–subtropical WNP, and the other over north China–Mongolia. The outflows from the two anomalous anticyclones converge over southern China, leading to anomalous moisture convergence, enhanced ascent, and increased rainfall. The development of the northern anticyclone is related to an increase in the Tibetan Plateau snow cover in the preceding winter–spring that leads to a contrast in temperature change between the plateau and the surrounding regions. The relatively small temperature change over the plateau, coupled with increases in temperature to the west and the east, leads to an increase in surface pressure extending northward from the plateau. The development of the southern anticyclone is related to an increase in sea surface temperature in the equatorial Indian Ocean that enhances lower-level convergence and ascent. The accompanying upper-level divergent flows from the tropical Indian Ocean to the SCS–WNP lead to the development of anomalous descent and lower-level anomalous anticyclone over the SCS–WNP.

The clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR‐associated (Cas) systems have recently received notable attention for their applications in nucleic acid detection. ...Despite many attempts, the majority of current CRISPR‐based biosensors in infectious respiratory disease diagnostic applications still require target preamplifications. This study reports a new biosensor for amplification‐free nucleic acid detection via harnessing the trans‐cleavage mechanism of Cas13a and ultrasensitive graphene field‐effect transistors (gFETs). CRISPR Cas13a‐gFET achieves the detection of SARS‐CoV‐2 and respiratory syncytial virus (RSV) genome down to 1 attomolar without target preamplifications. Additionally, we validate the detection performance using clinical SARS‐CoV‐2 samples, including those with low viral loads (Ct value >30). Overall, these findings establish our CRISPR Cas13a‐gFET among the most sensitive amplification‐free nucleic acid diagnostic platforms to date.
CRISPR Cas13a‐gFET biosensors are reported for target amplification‐free and ultrasensitive detection of SARS‐CoV‐2 and respiratory syncytial virus. The sensor harnesses the trans‐cleavage mechanism of Cas13a and the high sensitivity of graphene field‐effect transistors (gFETs). The sensor performance was validated using synthetic targets, heat‐inactivated virus samples, and clinical samples, showing a sensitivity comparable to that of RT‐qPCR.
Learning fine-grained image similarity is a challenging task. It needs to capture between-class and within-class image differences. This paper proposes a deep ranking model that employs deep learning ...techniques to learn similarity metric directly from images. It has higher learning capability than models based on hand-crafted features. A novel multiscale network structure has been developed to describe the images effectively. An efficient triplet sampling algorithm is also proposed to learn the model with distributed asynchronized stochastic gradient. Extensive experiments show that the proposed algorithm outperforms models based on hand-crafted visual features and deep classification models.
β‐Carotene displays antioxidant and anti‐inflammatory activities and prevents the development of cancer. Ulcerative colitis (UC) is a kind of inflammatory bowel disease that is accompanied by a ...certain risk of colon cancer. However, the role of β‐carotene in the modulation of gut microbiota and UC improvement is unclear. In this research, the properties of β‐carotene on anti‐inflammatory and the composition of gut microbiota were evaluated in a rat model of UC induced by dextran sulfate sodium (DSS). The results revealed that β‐carotene significantly (p < 0.05) decreased the severity of colitis in rats, as assessed using body weight (6.00 ± 1.73%), colon length (22.23 ± 0.53%), and disease activity index, and improved the structure of the colon damaged. Moreover, colonic levels of proinflammatory cytokines were significantly lower following β‐carotene supplementation. β‐Carotene intervention also lowered the expression levels of phosphorylated p65 (0.60 ± 0.02), p38 (0.57 ± 0.00), Erk (0.63 ± 0.04), and JNK (0.70 ± 0.00). The result of the relative abundance of gut microbiota showed that DSS administration significantly changed the microbial structure at the phylum and genus levels of rats. Furthermore, β‐carotene treatment significantly increased the abundance of Faecalibacterium, the levels of which negatively correlated with the levels of inflammatory cytokines. Faecalibacterium may be a potential target in the alleviation of DSS‐induced UC. β‐Carotene can alleviate DSS‐induced UC through the regulation of gut microbiota. This study provides a reference for the rational use of β‐carotene in the treatment of UC.

Structure reconstruction induced by the migration of Li, O, and transition metal (TM) ions plays a key role in the performance of Ni‐based cathodes, yet their interactions are still poorly ...understood. This work investigates systematically the structure transformation of the high‐temperature lithiation in air and oxygen rich atmosphere, charging process, and long‐term storage. Structural and electrochemical characterization of Li‐free/Li‐containing phases (Ni0.92Co0.04Mn0.04(OH)2 and LixNi0.92Co0.04Mn0.04Oy) and a series of detailed analyses provide an in‐depth understanding of the structural reconstruction induced by the interaction of Li, O, and TM ions, i.e., the shift of Li/TM ions in the lattice leads to structural reconstruction via a layered structure with oxygen vacancies.
Structural reconstruction during the synthesis, storage, and cycling of Ni‐rich cathode materials is mainly driven by oxygen vacancies in the layered structure. This result provides a theoretical guidance for the calcination and modification of cathode materials, i.e., the calcination environment should ensure sufficient oxygen, and the doped elements should be selected with strong TM‐O bonds.
The pathogenesis of thyroid dysgenesis (TD) is not well understood. Here, using a combination of single-cell RNA and spatial transcriptome sequencing, we identify a subgroup of NF-κB-activated ...thyrocytes located at the center of thyroid tissues in postnatal mice, which maintained a partially mesenchymal phenotype. These cells actively protruded out of the thyroid primordium and generated new follicles in zebrafish embryos through continuous tracing. Suppressing NF-κB signaling affected thyrocyte migration and follicle formation, leading to a TD-like phenotype in both mice and zebrafish. Interestingly, during thyroid folliculogenesis, myeloid cells played a crucial role in promoting thyrocyte migration by maintaining close contact and secreting TNF-α. We found that cebpa mutant zebrafish, in which all myeloid cells were depleted, exhibited thyrocyte migration defects. Taken together, our results suggest that myeloid-derived TNF-α-induced NF-κB activation plays a critical role in promoting the migration of vertebrate thyrocytes for follicle generation.
The aim of this study was to clarify the effect of 5-aminosalicylic acid (5-ASA) treatment on gut bacterial microbiota in patients with ulcerative colitis (UC).
A total of 57 UC patients, including ...20 untreated and 37 5-ASA-treated, were recruited into an exploration cohort. We endoscopically collected both non-inflamed and inflamed mucosal samples from all patients, and compared the gut bacterial profiles using
sequencing. Ten untreated UC patients were then treated with 5-ASA and subsequently recruited for an independent validation study to confirm the acquired data.
In untreated UC patients, compared with those in non-inflamed mucosae, Firmicutes (such as
) were decreased and Proteobacteria (e.g.,
) were increased in the inflamed mucosae. Compared with the inflamed mucosae of untreated UC patients, there was a higher abundance of Firmicutes (e.g.,
) and lower Proteobacteria (
) in the inflamed mucosae of 5-ASA treated UC patients. In the validation cohort, after administration of 5-ASA, bacterial alteration was consistent with these data. Furthermore, there was a skewed negative correlation between
and bacterial genera of Firmicutes in the inflamed mucosae. 5-ASA treatment decreased the strength of bacterial correlation and weakened the skewed negative correlation pattern.
The microbial dysbiosis (mainly characterized by an increased abundance in the
genus) and the skewed negative correlation between
and bacterial genera of Firmicutes are two characteristics of the inflamed mucosae of UC patients. 5-ASA treatment decreases
and weakens the skewed correlations, which may be related to its treatment efficiency.
