Two Leu residues and their ambient amino acid residues are known to exist in the cytosolic tail of chicken invariant chain (Ii), and these play an important role as motifs in mediating the sorting ...endocytic pathway. We performed 20 mutations via site-directed mutagenesis by the PCR megaprimer method to study the effect of some ambient amino residues of both Leu on the localization of chicken Ii. These mutated fragments were ligated to the vector pEGFP-C1. The recombinant plasmids were transiently transfected into COS-7 cells with Lipofectamine 2000. Furthermore, the fluorescence of located fusion proteins (green fluorescent protein-Ii) was observed with a fluorescence microscope. Our results indicated that 2 Leu-based motifs are required for chicken Ii intracellular localization, and both motifs independently mediate this function of the Ii. The other amino acid residues surrounding both Leu also influence Ii-induced endosomal vacuolation. In addition, we found that Pro¹⁹, which is near the Val¹⁷-Leu¹⁸ motif, was a key residue for chicken Ii intracellular localization. Not only is it critical for endocytic targeting to each Leu, but its unique mutation can also result in altering the function of chicken Ii.

The evolution of oxide particles in oxide dispersion strengthened (ODS) steels subjected to high temperature radiation is critical for maintaining their radiation resistance since the particles were ...damaged. Conventional ODS steels are often ultrafine-grained (with a grain size between 100 and 1000 nm), and the nanoparticles coarsen and their density decreases after ion irradiation. In this work, nanocrystalline (NC) ODS steels (with an average grain size of 50 nm) and ultrafine-grained (UFG) ODS steels (with an average grain size of 250 nm) were irradiated with 6 MeV Au ions at 500 °C up to 468 dpa. The nanoparticles in NC steels showed a slight decrease in diameter and an increase in number density, exhibiting completely reverse evolutions with respect to UFG steels under the same radiation conditions. The reverse evolution can be reasonably explained by the competition between radiation-driven ballistic dissolution and nanograin-localized long-range diffusion, leading to the partial conversion of Y4Zr3O12 to Y2Zr2O7. This work expands the strategy to stabilize the remaining nanoparticles in ODS alloys under radiation via nanocrystalization.

This study was aimed for development of vapour-grown carbon nano-fibre(VGCNF)-reinforced aluminium matrix composites by the low-pressure infiltration process. Pear rust, caused by Gymnosporangium asiaticum, is an important disease of pear (Pyrus spp.) in China in areas where cypress (Juniperus spp. and Sabina spp.), the alternate host, is present. ...Controlled environment experiments were conducted to study the effects of temperature, relative humidity (RH) and duration of free water on germination of teliospores and basidiospores of G. asiaticum. Teliospores, sampled from Juniperus chinensis, germinated at temperatures ranging from 5 to 28 , with an optimum between 16 and 20 . For teliospores to germinate, telial horns required free water: soaking in water for 30 sec triggered teliospore germination. After initial wetting, RH had little effect on germination of teliospores except at extreme temperatures (5 and 30 ) where germination needed near-saturation moisture. The minimum time for telial horns to produce basidiospores was about 3 h at optimum temperatures. Teliospores in all telial horns germinated at 12--4 , but at 8 and 28 only those from a third of telial horns germinated. Basidiospores germinated at 5--0 with the optimum around 15 ; they needed free water or saturated moisture to germinate. Germination in free water was about eight times greater than at 100% RH. Logistic models described well the germination dynamics of basidiospores. Basidiospore survival declined exponentially with storage time, and linearly with increasing temperature and decreasing RH. Summer mean (June, July, and August) surface air temperature (SSAT) in East Asia during the period from 1958 to 2001 has shown a warming. However, the relative contributions of external ...forcing (EF) and internal climate variability (ICV) to the SSAT changes over East Asia remain unclear. In this study, a new approach is applied to estimate the changes in the SSAT determined by the effects of EF and ICV over East Asia during the period from 1958 to 2001. Reanalysis data as well as simulated results from both global atmosphere–ocean coupled model outputs and a regional climate model (RCM) are used for this approach. The observed SSATs over East Asia have undergone a decreasing trend from 1958 to 1972 (−0.14°C decade
−1

< 0.01) and an increasing trend after 1972 (0.24°C decade

< 0.01). While these features are not captured by the reanalysis studied here, they are reproduced when the reanalysis output is downscaled using an RCM. The effects of the EF and the ICV on the SSAT can be separated based on the RCM downscaling simulation. The results show that the SSAT with EF displayed significant warming over most regions of East Asia, whereas the SSAT with ICV mainly exhibited cooling over East Asia. Furthermore, EF mainly influenced the decadal changes of the SSAT, whereas the ICV mainly influenced the interannual changes in the SSAT over East Asia. The interannual changes of the SSAT over East Asia that were influenced by the ICV are mainly manifested as the combined effects of the large-scale ocean–atmosphere circulations, which expressed 79% explanatory power on the SSAT changes.
