Metaplastic thymoma (MT) is a very uncommon thymoma type, with biphasic differentiation as one of its histological characteristics. This histological pattern, however, can also be mistaken for type A ...thymoma and the A component in type AB thymoma.
Postoperative specimens were collected from five MT and four type A thymomas with a retrospective analysis involving immunohistochemistry, fluorescence in situ hybridization (FISH) and next-generation sequencing (NGS).
The histological morphology of the MT overlapped with that of the type A thymoma. With immunostains, the former's spindle cell components expressed vimentin and EMA, but not CD20. In MT, 3/5 cases had the nuclear expression of YAP1. The spindle cell component of the type A thymoma was found to express CD20. In all five cases of MT, FISH detection revealed YAP1-MAML2 fusion, which was not found in any type A thymoma cases. NGS sequencing confirmed YAP1-MAML2 rearrangement in all five cases of MT, and mutations in POLE and HRAS were also found in two cases, respectively. GTF2I c.74146970 T > A mutations were found in all cases of type A thymoma, and HRAS and NRAS mutations were found in two cases, but no YAP1-MAML2 rearrangement was evident.
For the diagnosis and differential diagnosis of challenging cases, the YAP1-MAML2 rearrangement and GTF2I mutation were both significant molecular events specific to MT and type A thymoma, respectively.
A heterohexameric complex composed of minichromosome maintenance protein 2-7 (MCM2-7), which acts as a key replicative enzyme in eukaryotes, is crucial for initiating DNA synthesis only once per cell ...cycle. The MCM complex remains inactive through the G1 phase, until the S phase, when it is activated to initiate replication. During the transition from the G1 to S phase, the MCM undergoes multisite phosphorylation, an important change that promotes subsequent assembly of other replisome members. Phosphorylation is crucial for the regulation of MCM activity and function. MCMs can be phosphorylated by multiple kinases and these phosphorylation events are involved not only in DNA replication but also cell cycle progression and checkpoint response. Dysfunctional phosphorylation of MCMs appears to correlate with the occurrence and development of cancers. In this review, we summarize the currently available data regarding the regulatory mechanisms and functional consequences of MCM phosphorylation and seek the probability that protein kinase inhibitor can be used therapeutically to target MCM phosphorylation in cancer.
This paper presents a dual-band high-power digital polar Doherty power amplifier (PA). A current-mode parallel-combining transformer (PCT) power combiner is introduced for Doherty operation, which ...realizes simultaneous dual-band coverage, high output power, and backoff efficiency enhancement with an ultra-compact single-transformer footprint. The PA is implemented in 55-nm CMOS and occupies 1.11 mm 2 die area. It achieves 28.9-dBm peak <inline-formula> <tex-math notation="LaTeX">P_{\mathrm {out}} </tex-math></inline-formula> with 36.8% PAE in low band (LB) and 27.0-dBm peak <inline-formula> <tex-math notation="LaTeX">P_{\mathrm {out}} </tex-math></inline-formula> with 25.4% PAE in high band (HB). For 12-subcarrier 180-kHz bandwidth cellular narrowband Internet-of-Things (NB-IoT) signals, the peak <inline-formula> <tex-math notation="LaTeX">P_{\mathrm {avg}} </tex-math></inline-formula> of 24.4 dBm with average PAE of 29.5% at 850 MHz and the peak <inline-formula> <tex-math notation="LaTeX">P_{\mathrm {avg}} </tex-math></inline-formula> of 23.0 dBm with average PAE of 17.9% at 1.7 GHz are obtained with −21.6-dB error vector magnitude (EVM). Moreover, the PA achieves 20.8-dBm <inline-formula> <tex-math notation="LaTeX">P_{\mathrm {avg}} </tex-math></inline-formula>, 22.7% average PAE, and −30.5-dB EVM for 20-MHz 256-QAM WLAN signal.
Gastric cancer is a leading cause of mortality due to neoplastic disease. Although early detection of gastric cancers can decrease the mortality rate, it remains a diagnostic challenge because of the ...lack of effective biomarkers. In this study, fifteen gastric cancer patients and ten healthy subjects were recruited to assess novel serum biomarkers for gastric cancer using antibody microarray technology. ELISA was utilized to validate the antibody array results. As a result, compared to the controls, eleven cytokines were found to be significantly increased in gastric cancer, including interferon gamma receptor 1 (IFNGR1), neurogenic locus notch homolog protein 3 (Notch‐3), tumor necrosis factor receptor superfamily member 19L (TNFRSF19L), growth hormone receptor (GHR), signaling lymphocytic activation molecule family 8 (SLAMF8), folate receptor beta (FR‐beta), integrin alpha 5, galectin‐8, erythropoietin‐producing hepatocellular A1 (EphA1), epiregulin, and fibroblast growth factor 12 (FGF‐12) with P < 0.05. ELISA validation supported the results of the antibody array. More importantly, most of these eleven cytokines, including IFNGR1, TNFRSF19L, GHR, SLAMF8, FR‐beta, and integrin alpha 5 were discovered to be elevated in gastric cancer serum samples for the first time in this study, suggesting that these proteins may serve as novel biomarkers for the early diagnosis and prognosis determination of gastric cancer.
Eleven cytokines were found significantly increased in gastric cancer including IFN‐gamma R1, Notch‐3, RELT, GHR, BLAME, FOLR2, Integrin alpha 5, Galectin‐8, EphA1, Epiregulin and FGF‐12, which suggests these eleven cytokines may play important roles in the pathogenesis of gastric cancer.
HBeAg seroconversion is an important intermediate outcome in HBeAg-positive chronic hepatitis B (CHB) patients. This study aimed to compare the effect of nucleos(t)ide analogs (NAs) on HBeAg ...seroconversion in treating CHB with lamivudine, adefovir, telbivudine, entecavir, and tenofovir.
Network meta-analysis of NA treatment-induced HBeAg seroconversion after 1-2 years of treatment was performed. In addition, NA treatment-induced HBeAg seroconversion after 3-5 years of treatment was systematically evaluated.
A total of 31 articles were included in this study. Nine and five studies respectively reporting on 1- and 2-year treatment were included in our network meta-analysis. In addition, 6, 5, and 5 studies, respectively reporting on 3-, 4-, and 5-year treatment were included in our systematic evaluation. Telbivudine showed a significantly higher HBeAg seroconversion rate after a 1 year treatment period compared to the other NAs (odds ratio (OR) = 3.99, 95% CI 0.68-23.6). This was followed by tenofovir (OR = 3.36, 95% CI 0.70-16.75). Telbivudine also showed a higher seroconversion rate compared to the other NAs after a 2 year treatment period, (OR = 1.38, 95% CI 0.92-2.22). This was followed by entecavir (OR = 1.14, 95% CI 0.72-1.72). No significant difference was observed between spontaneous induction and long-term telbivudine treatment-induced HBeAg seroconversion. However, entecavir and tenofovir treatment-induced HBeAg seroconversions were significantly lower than spontaneous seroconversion.
Long-term treatment with potent anti-HBV drugs, especially tenofovir and entecavir, may reduce HBeAg seroconversion compared with spontaneous HBeAg seroconversion rate. Telbivudine treatment, whether short term or long term, is associated with higher HBeAg seroconversion compared with the other NAs. However, the high rates of drug resistance likely limit the application of telbivudine.

Accurate and timely crop type mapping and rotation monitoring play a critical role in crop yield estimation, soil management, and food supplies. To date, to our knowledge, accurate mapping of crop ...types remains challenging due to the intra-class variability of crops and labyrinthine natural conditions. The challenge is further complicated for smallholder farming systems in mountainous areas where field sizes are small and crop types are very diverse. This bottleneck issue makes it difficult and sometimes impossible to use remote sensing in monitoring crop rotation, a desired and required farm management policy in parts of China. This study integrated Sentinel-1 and Sentinel-2 images for crop type mapping and rotation monitoring in Inner Mongolia, China, with an extensive field-based survey dataset. We accomplished this work on the Google Earth Engine (GEE) platform. The results indicated that most crop types were mapped fairly accurately with an F1-score around 0.9 and a clear separation of crop types from one another. Sentinel-1 polarization achieved a better performance in wheat and rapeseed classification among different feature combinations, and Sentinel-2 spectral bands exhibited superiority in soybean and corn identification. Using the accurate crop type classification results, we identified crop fields, changed or unchanged, from 2017 to 2018. These findings suggest that the combination of Sentinel-1 and Sentinel-2 proved effective in crop type mapping and crop rotation monitoring of smallholder farms in labyrinthine mountain areas, allowing practical monitoring of crop rotations.
Increased glycolysis in tumor cells is associated with increased risk of tumor progression and mortality. Therefore, disruption of glycolysis, one of the main sources of cellular energy supply, can ...serve as a target for suppressing tumor growth and progression. Of note, hexokinase-2 (HK2) plays vital roles in glucose metabolism. Moreover, the expression of HK2 alters the metabolic phenotype and supports the continuous growth of tumor cells, making it an attractive target for cancer therapy. Quercetin (QUE), a bioactive flavonoid, has a profound anti-tumor effect on hepatocellular carcinoma (HCC), but the precise underlying mechanism of this effect is unclear. In the present study, we reported that QUE inhibited the proliferation of HCC cells that relied on aerobic glycolysis. We further found that QUE could decrease the protein levels of HK2 and suppress the AKT/mTOR pathway in HCC cells. In addition, QUE significantly restrained the growth of HCC xenografts and decreased HK-2 expression in vivo. Taken together, we have revealed that QUE suppresses the progression of HCC by inhibiting HK2-dependentglycolysis, which may have a promising potential to be an effective treatments for HCC, especially for those patients with high HK2 expression.
ABSTRACTThe cellular automata (CA) model is the predominant method for predicting land use and land cover (LULC) changes. The accuracy of this model critically depends on well-defined transition ...rules, which encapsulate the local dynamics of complex systems and facilitate the manifestation of organized global patterns. While current studies largely concentrate on land use transition matrices as core elements of these rules, exclusive reliance on these matrices is insufficient for capturing the full spectrum of land use change potential. Addressing this gap, our research introduces the adaptive transition probability matrix with quality seeds (ATPMS) model, which incorporates both the Markov model and the genetic algorithm (GA) into the traditional CA framework. Furthermore, an artificial neural network (ANN) is utilized to determine land suitability. Implemented in Beijing, Wuhan, and the Pearl River Delta (PRD), our results indicate that the ATPMS-ANN-CA model surpasses the standard Markov-ANN-CA model in various validation metrics, displaying improvements in overall accuracy (OA) by 0.03% to 0.74% and figure of merit (FoM) by 3.67% to 63.14%. Additionally, the ATPMS-ANN-CA model excels in providing detailed landscape analysis.
Tigecycline is one of the few therapeutic options for treating infections caused by some multi-drug resistant pathogens, such as Klebsiella pneumoniae. However, tigecycline-resistant K. pneumoniae ...has been discovered recently in China. From 2009 to 2013, nine tigecycline-resistant K. pneumoniae isolates were identified in our hospital. Six of nine strains were identified before using tigecycline. To investigate the efflux-mediated resistance mechanisms of K. pneumoniae, the expression of efflux pump genes (acrA, acrB, tolC, oqxA and oqxB) and pump regulators (acrR, marA, soxS, rarA, rob and ramA) were examined by real-time RT-PCR. Molecular typing of the tigecycline resistant strains was performed. ST11 was the predominant clone of K. pneumoniae strains, while ST1414 and ST1415 were novel STs. Efflux pump inhibitor (EPI)-carbonyl cyanide chlorophenylhydrazone (CCCP) was able to reverse the resistance patterns of 5 resistant K. pneumoniae strains. In comparison with strain A111, a tigecycline-susceptible strain (negative control), we found that the expression levels of efflux pump genes and pump regulators were higher in a majority of resistant strains. Higher expression levels of regulators rarA (2.41-fold, 9.55-fold, 28.44-fold and 18.31-fold, respectively) and pump gene oqxB (3.87-fold, 31.96-fold, 50.61-fold and 29.45-fold, respectively) were observed in four tigecycline resistant strains (A363, A361, A368, A373, respectively). Increased expression of acrB was associated with ramA and marA expression. To our knowledge, studies on tigecycline resistance mechanism in K. pneumoniae are limited especially in China. In our study, we found that both efflux pump AcrAB-TolC and OqxAB contributed to tigecycline resistance in K. pneumoniae isolates.
