Most patients with solitary bone plasmacytomas (SBP) progress to multiple myeloma (MM) after definitive radiation therapy as their primary treatment. Whether the presence of high-risk (HR) ...cytogenetic abnormalities by fluorescence in situ hybridization (FISH) in the clonal plasma cells, obtained either directly from the diagnostic SBP tissue or the corresponding bone marrow examination at the time of diagnosis, is associated with a shorter time to progression (TTP) to MM is unknown. This study evaluated all patients diagnosed with SBP at the Mayo Clinic from January 2012 to July 2022. The presence of del(17p), t(14;16), t(4;14), or +1q (gain or amplification) by FISH in clonal plasma cells was defined as HR. A total of 114 patients were included in this cohort, and baseline FISH was available for 55 patients (48%), of which 22 were classified as HR (40%). The median TTP to MM for patients with SBP and HR FISH was 8 months (95% confidence interval CI, 6.3-26) compared with 42 months (95% CI, 25-not reached NR) in patients with SBP without HR FISH (P < .001). In a multivariate analysis, only HR FISH was a significant predictor for shorter TTP to MM, independent of minimal marrow involvement and an abnormal serum free light chain ratio at diagnosis. Deletion (17p) and gain 1q abnormalities were the most common FISH abnormalities responsible for the short TTP to MM. Thus, assessing for HR FISH abnormalities in clonal plasma cells derived from either the diagnostic SBP tissue or the staging bone marrow examination of patients with newly diagnosed SBP is feasible and prognostic for a shorter TTP to MM.
Fused deposition modelling (FDM) is a technique of additive manufacturing used to fabricate a 3D (three-dimensional) model with layer-by-layer deposition of required materials with less material ...wastage. FDM is used to make any objects with a meager cost, but also there are some negative points related to less strength, less accuracy, and less surface finish. In this study, acrylonitrile butadiene styrene (ABS) is printed using an FDM printer to investigate the effects of various changing parameters like nozzle temperature (°C), infill pattern, and printing speed (mm/s) on surface roughness and thickness measurement. Experiments are designed using the Taguchi L9 orthogonal array method and ANOVA method. For obtaining an increase in surface roughness, the most influencing factor is printing speed with 83.41% contribution, and the effect of nozzle temperature is 9.04%. Lesser printing speed enhances the surface finish and, in the case of thickness and outer dimension of all the printed samples, results are almost constant. Regression analysis is performed to formulate the single-objective equations, and a genetic algorithm (GA) is applied to optimize the values of process parameters.
Sensors are used in various areas and have numerous applications including military surveillance, weather monitoring, maritime communication etc. Most of the applications need the multimedia sensors ...with directional sensing range of the sensors to perform advance task. Additionally, there are some constraints on the targets namely Q-constraints, such that each target must be covered by different number of sensors. The crucial issue in this case is deployment of minimum number of multimedia sensors to satisfy the Q-constraints. The optimization of the placement of the sensors can result in reduced number of sensors needed for the coverage of the specified targets. In this paper, the placement of the sensors is optimized using the Brain Storm Optimization Algorithm (BSO) which is inspired by the human brainstorming process. BSO algorithm efficiently designed for the considered problem to identify the optimal position of the sensors. The performance of the proposed algorithm is verified by simulation results, which performs better than the random deployment of the sensors.
Pediatric tuberculous meningitis (TBM) commonly causes death or disability. In adults, high-dose rifampicin may reduce mortality. The role of fluoroquinolones remains unclear. There have been no ...antimicrobial treatment trials for pediatric TBM.
TBM-KIDS was a phase 2 open-label randomized trial among children with TBM in India and Malawi. Participants received isoniazid and pyrazinamide plus: (i) high-dose rifampicin (30 mg/kg) and ethambutol (R30HZE, arm 1); (ii) high-dose rifampicin and levofloxacin (R30HZL, arm 2); or (iii) standard-dose rifampicin and ethambutol (R15HZE, arm 3) for 8 weeks, followed by 10 months of standard treatment. Functional and neurocognitive outcomes were measured longitudinally using Modified Rankin Scale (MRS) and Mullen Scales of Early Learning (MSEL).
Of 2487 children prescreened, 79 were screened and 37 enrolled. Median age was 72 months; 49%, 43%, and 8% had stage I, II, and III disease, respectively. Grade 3 or higher adverse events occurred in 58%, 55%, and 36% of children in arms 1, 2, and 3, with 1 death (arm 1) and 6 early treatment discontinuations (4 in arm 1, 1 each in arms 2 and 3). By week 8, all children recovered to MRS score of 0 or 1. Average MSEL scores were significantly better in arm 1 than arm 3 in fine motor, receptive language, and expressive language domains (P < .01).
In a pediatric TBM trial, functional outcomes were excellent overall. The trend toward higher frequency of adverse events but better neurocognitive outcomes in children receiving high-dose rifampicin requires confirmation in a larger trial.

Variability analysis revealed that the oil content had high GCV with high broad sense heritability. High PCV was observed among the other yield related traits which indicates that these traitsare highly influenced by the environmental factors.
The objective of the present study was to investigate the effect of ofloxacin (10 mg/kg) on the pharmacokinetics of meloxicam (0.5 mg/kg) in goats, when administered concurrently by intravenous ...route. Meloxicam concentration in plasma was measured using HPLC assay. The pharmacokinetics of meloxicam were best described by the two-compartment open model. Following concurrent administration of meloxicam and ofloxacin, the mean plasma level of meloxicam was only found to be significantly higher (P < 0.05) at 15 min compared to its alone administration. Statistical analysis of data revealed that there were no statistically significant differences in PK parameters between the two treatments, except K . Therefore, the results of the present study suggest that concurrent administration of ofloxacin and meloxicam in goats does not require any adjustment in dosage regimens. Based on PK determinants, meloxicam may be administered by i.v. route at 0.86 and 0.65 mg/kg, as loading and maintenance doses respectively, and is to be repeated at 8 h intervals. Malnutrition has emerged as one of the major health problems worldwide. Traditional yellow maize has low provitamin-A (proA) content and its genetic base in proA biofortification breeding program of ...subtropics is extremely narrow. To diversify the proA rich germplasm, 10 elite low proA inbreds were crossed with a proA rich donor (HP702-22) having mutant crtRB1 gene. The F2 populations derived from these crosses were genotyped using InDel marker specific to crtRB1. Severe marker segregation distortion was observed. Seventeen crtRB1 inbreds developed through marker-assisted pedigree breeding and seven inbreds generated using marker-assisted backcross breeding were characterized using 77 SSRs. Wide variation in gene diversity (0.08 to 0.79) and dissimilarity coefficient (0.28 to 0.84) was observed. The inbreds were grouped into three major clusters depicting the existing genetic diversity. The crtRB1-based inbreds possessed high β-carotene (BC: 8.72μg/g), β-cryptoxanthin (BCX: 4.58μg/g) and proA (11.01μg/g), while it was 2.35μg/g, 1.24μg/g and 2.97μg/g in checks, respectively. Based on their genetic relationships, 15 newly developed crtRB1-based inbreds were crossed with five testers (having crtRB1 gene) using line × tester mating design. 75 experimental hybrids with crtRB1 gene were evaluated over three locations. These experimental hybrids possessed higher BC (8.02μg/g), BCX (4.69μg/g), proA (10.37μg/g) compared to traditional hybrids used as check (BC: 2.36 μg/g, BCX: 1.53μg/g, proA: 3.13μg/g). Environment and genotypes × environment interaction had minor effects on proA content. Both additive and dominance gene action were significant for proA. The mean proportion of proA to total carotenoids (TC) was 44% among crtRB1-based hybrids, while 11% in traditional hybrids. BC was found to be positively correlated with BCX (r = 0.68) and proA (r = 0.98). However, no correlation was observed between proA and grain yield. Several hybrids with >10.0 t/ha grain yield with proA content >10.0 μg/g were identified. This is the first comprehensive study on development of diverse proA rich maize hybrids through marker-assisted pedigree breeding approach. The findings provides sustainable and cost-effective solution to alleviate vitamin-A deficiency.

Earlier we have reported mercury-induced alterations in functional dynamics of buck spermatozoa through free radicals-mediated oxidative stress and spontaneous acrosome reaction. Based on our earlier ...findings, we aimed to investigate the effect of mercury exposure on motility, kinematic patterns, DNA damage, apoptosis and ultra-structural alterations in goat spermatozoa following in vitro exposure to different concentrations (0.031-1.25 µg/ml) of mercuric chloride for 15 min and 3 h. Following exposure of sperm cells to 0.031 µg/ml of mercuric chloride for 3 h, livability and motility of sperms was significantly reduced along with altered kinematic patterns, significant increase in per cent necrotic sperm cells and number of cells showing DNA damage; and this effect was dose- and time-dependent. Contrary to up-regulation of Bax gene after 3 h in control group, there was significant increase in expression of Bcl-2 in mercury-treated groups. Transmission electron microscopy studies revealed rifts and nicks in plasma and acrosomal membrane, mitochondrial sheath, and collapsed mitochondria with loss of helical organization of mitochondria in the middle piece of spermatozoa. Our findings evidently suggest that mercury induces necrosis instead of apoptosis and targets the membrane, acrosome, mid piece of sperms; and the damage to mitochondria seems to be responsible for alterations in functional and kinematic attributes of spermatozoa.
